Regulation of intrinsic and synaptic properties of neonatal rat trigeminal motoneurons by metabotropic glutamate receptors.

We studied how metabotropic glutamate receptor (mGluR) activation modifies the synaptic and intrinsic membrane properties of neonatal rat trigeminal motoneurons using the broad-spectrum mGluR agonist (1S,3R)-1-amino-1,3-cyclopentane-dicarboxylic acid [(1S,3R)-ACPD], group I/II antagonist (+/-)-alpha-methyl-4-carboxy-phenylglycine (MCPG), and group III agonist L-2-amino-4-phosphonobutanoate (L-AP4). (1S,3R)-ACPD depressed excitatory transmission to trigeminal motoneurons presynaptically and postsynaptically via presynaptic inhibition and by reducing the currents carried by ionotropic glutamate

Metabotropic glutamate receptor agonist ACPD inhibits some, but not all, muscarinic-sensitive K+ conductances in basolateral amygdaloid neurons.

Muscarinic agonists produce membrane depolarization and losses of spike frequency accommodation and the slow afterhyperpolarization (AHP) when applied to neurons of the basolateral amygdala (BLA). Underlying these changes are the muscarinic-induced inhibitions of several K+ conductances, including the voltage-activated M-current (IM), a slowly decaying Ca(2+)-activated current (IAHP), a voltage-insensitive leak current (ILeak), and the hyperpolarization-activated inward rectifier current (IIR). Similar depolarizations and losses of the slow AHP have been observed in other neuronal cell types f

Inhibition of adenosine 3',5'-cyclic monophosphate phosphodiesterase by CD-349, a novel 1,4-dihydropyridine derivative: effect of EGTA on the inhibitory activity.

CD-349 (2-nitratopropyl 3-nitratopropyl 2,6-dimethyl-4-(3-nitrophenyl)-1,4-dihydropyridine-3,5-dicarboxylate) inhibited the activity of cyclic AMP phosphodiesterase (PDE) from the porcine coronary artery more effectively than that from the myocardium. Other dihydropyridines, nicardipine and nifedipine, were not tissue selective in inhibiting the cyclic AMP PDE from both sources. CD-349 inhibited cyclic AMP PDE noncompetitively with apparent inhibition constant (K1) values of 6.6 and 4.6 microM for high and low affinity constant (Km) enzymes, respectively. Basal activity of coronary arterial cy

Effectiveness and Safety of Bempedoic Acid in Taiwanese Patients With Hypercholesterolemia: A Pragmatic Phase 4 Study (CLEAR Taiwan Study).

BACKGROUND: Elevated low-density lipoprotein cholesterol (LDL-C) is a risk factor for cardiovascular disease. Despite available lipid-lowering therapies, lipid control remains suboptimal. Bempedoic acid offers a nonstatin oral treatment for hypercholesterolemia. However, real-world data in Asia are limited. The study aimed to investigate the effectiveness and safety of bempedoic acid in Taiwan. METHODS: This prospective, pragmatic phase 4 study enrolled 180 patients with inadequately controlled hypercholesterolemia to receive bempedoic acid for 12 weeks in addition to background lipid-lowerin

Inhibition of ATP citrate lyase does not affect muscle mitochondrial activity in apoE-/- mice.

INTRODUCTION: Lipid-lowering therapy is a cornerstone in preventing coronary disease. Bempedoic acid is recommended in patients who are unable to take statin therapy to achieve the low-density lipoprotein cholesterol goal. Based on the hypothesis that statins impact skeletal mitochondrial activity, this study aimed to evaluate the effect of bempedoic acid on this aspect in apoE-/- mice fed a high-fat high-cholesterol (HFHC) diet and compare it to the results obtained using atorvastatin. METHODS: Female apoE-/- mice. The following techniques were used: En face analysis of aorta, mitostress ana

Triple Versus Dual Lipid-Lowering Therapy in Acute Coronary Syndrome: The ES-BempedACS Randomized Clinical Trial.

BACKGROUND: Current guidelines recommend a stepwise strategy to achieve low-density lipoprotein cholesterol (LDL-C) goals after acute coronary syndrome (ACS). Earlier intensive strategies based on a combination of lipid-lowering therapies (LLTs) could be useful from the onset of ACS. However, the role of bempedoic acid in ACS, particularly when combined with high-intensity statins and ezetimibe, remains uncertain. The aim of ES-BempedACS (Efficacy and Security of Bempedoic Acid in Acute Coronary Syndrome) was to compare the efficacy and safety of triple LLT (high-dose, high-intensity statin+ez

New and Emerging LDL Cholesterol-Lowering Drugs.

The role of low-density lipoprotein cholesterol (LDL-C) in the pathophysiology of atherosclerosis is well recognized, and the use of LDL-C lowering medications has led to a significant reduction of cardiovascular risk in both primary and secondary prevention. Statins are the standard of care and their use is supported by extensive evidence demonstrating their effectiveness in lowering LDL-C and in reducing the risk for cardiovascular disease. However, many individuals at risk for cardiovascular disease fail to achieve LDL-C goals. In addition, several patients are intolerant to statins due to

Dimethyloxalylglycine improves functional recovery through inhibiting cell apoptosis and enhancing blood-spinal cord barrier repair after spinal cord injury.

PURPOSE: The secondary damage of spinal cord injury (SCI) starts from the collapse of the blood spinal cord barrier (BSCB) to chronic and devastating neurological deficits. Thereby, the retention of the integrity and permeability of BSCB is well-recognized as one of the major therapies to promote functional recovery after SCI. Previous studies have demonstrated that activation of hypoxia inducible factor-1α (HIF-1α) provides anti-apoptosis and neuroprotection in SCI. Endogenous HIF-1α, rapidly degraded by prolylhydroxylase, is insufficient for promoting functional recovery. Dimethyloxalylglyci

LDL-C target attainment and treatment costs in high-risk and very-high-risk patients with or without bempedoic acid: A Spanish cohort simulation.

Achieving low-density lipoprotein cholesterol (LDL-C) targets remains challenging for Spanish patients at high (HR) or very-high (VHR) cardiovascular risk, despite treatment with statins and/or ezetimibe (EZE). Escalation to PCSK9 inhibitors (PCSK9i) or inclisiran (INC) is limited due to budgetary concerns. This study evaluates the impact of incorporating bempedoic acid (BA) into lipid-lowering treatment algorithms on LDL-C target attainment and treatment costs. A Monte Carlo simulation used real-world data from 34,967 Spanish adults with HR (29%) or VHR (71%) and uncontrolled LDL-C despite ≥4

Aerosol-Administered Adelmidrol Attenuates Lung Inflammation in a Murine Model of Acute Lung Injury.

Acute lung injury (ALI) is a common and devastating clinical disorder with a high mortality rate and no specific therapy. The pathophysiology of ALI is characterized by increased alveolar/capillary permeability, lung inflammation, oxidative stress and structural damage to lung tissues, which can progress to acute respiratory distress syndrome (ARDS). Adelmidrol (ADM), an analogue of palmitoylethanolamide (PEA), is known for its anti-inflammatory and antioxidant functions, which are mainly due to down-modulating mast cells (MCs) and promoting endogenous antioxidant defense. The aim of this stud

Plasmodesmata Localizing Proteins Regulate Transport and Signaling during Systemic Acquired Immunity in Plants.

Systemic acquired resistance (SAR) in plants is mediated by the signaling molecules azelaic acid (AzA), glycerol-3-phosphate (G3P), and salicylic acid (SA). Here, we show that AzA and G3P transport occurs via the symplastic route, which is regulated by channels known as plasmodesmata (PD). In contrast, SA moves via the extracytosolic apoplast compartment. We found that PD localizing proteins (PDLP) 1 and 5 were required for SAR even though PD permeability in pdlp1 and 5 mutants was comparable to or higher than wild-type plants, respectively. Furthermore, PDLP function was required in the recip

Optimal management of papulopustular rosacea: rationale for combination therapy.

The pathophysiology of papulopustular rosacea (PPR) is primarily characterized by inflammation associated with several factors such as abnormal innate immune response, neurovascular dysregulation, stratum corneum barrier dysfunction, and depletion of antioxidant reserve, with no definitive evidence supporting an underlying microbial etiology. Several molecular inflammatory pathways have now been identified that enable the development of therapeutic agents that target the signs and symptoms of disease by modifying specific pathophysiological mechanisms. Available evidence demonstrates that topi
