Changes in histological construction and decrease in 3H-QNB binding in the rat brain after prenatal X-irradiation.

To elucidate the mechanisms involved in deleterious neuronal and behavioral changes after prenatal ionizing irradiation, in vitro muscarinic acetylcholine (mACh) receptor binding and histological construction were investigated in 9-week old rat brains after 1.5 Gy X-ray exposure on embryonic day 15 (E15). A gross anatomical examination with a magnetic-resonance imaging system showed an irregular tissue construction in the hippocampus and cortex of the irradiated rat brain. Histological sections stained with hematoxylin and eosin also indicated that the structures of the hippocampus and cortex

N-WASP deficiency reveals distinct pathways for cell surface projections and microbial actin-based motility.

The Wiskott-Aldrich syndrome protein (WASP) family of molecules integrates upstream signalling events with changes in the actin cytoskeleton. N-WASP has been implicated both in the formation of cell-surface projections (filopodia) required for cell movement and in the actin-based motility of intracellular pathogens. To examine N-WASP function we have used homologous recombination to inactivate the gene encoding murine N-WASP. Whereas N-WASP-deficient embryos survive beyond gastrulation and initiate organogenesis, they have marked developmental delay and die before embryonic day 12. N-WASP is n

The embryonic development of sensory organs and the skull in the trisomy 16 mouse, an animal model for Down's syndrome.

The trisomy 16 mouse is a widely accepted animal model for the study of the embryonic development of human trisomy 21. While the development of the brain and heart has been thoroughly studied, there are hardly any data on the development of sensory organs like the eye, nose and ear. By studying scanning electron microscopic pictures and semithin sections from the tenth to the 15th day of development, we found delayed development of the nose, and, in particular, of the vomer. Sensory structures of the otic vesicle also showed a marked developmental delay. Pigmentation of the outer layer of the

Adult, fetal, and polycystic kidney expression of polycystin, the polycystic kidney disease-1 gene product.

The polycystic kidney disease-1 gene, which is mutated in the majority of patients with autosomal dominant polycystic kidney disease, has been identified. The protein encoded by this gene, polycystin, has no homology with any gene known thus far. To gain more insight into the function of polycystin, we raised antibodies against synthetic peptides and a fusion protein corresponding to the sequence of two different fragments of polycystin. Two of the antibodies were capable of immunoprecipitating an in vitro transcription and translation product corresponding to a fragment of polycystin. In the

Teratomas of the head and neck.

Teratomas of the head and neck are uncommon. They are composed of tissues from all three germ layers with varying degrees of differentiation. They arise from pluripotent stem cells and ectopic embryonic nongerm cells. Head and neck teratomas most commonly emerge during the neonatal period and are associated with airway obstruction and high rates of mortality in untreated patients. The tumors are usually benign, but when found during adulthood, they have a high incidence of malignancy. The most common site of occurrence is the cervical region and the nasopharynx. We presented a rare case of ter

[Lysosomal structures in the intestinal epithelium of mammals during their pre- and postnatal development. A micromorphometric-functional synopsis].

The first lysosomes appear in the stratified embryonic intestinal epithelium during its transition into the simple columnar form. This occurs concurrently with the initial villogenesis. Lysosomes situated basally in the epithelium are presumably the precursor of the first giant lysosomes in the lower small intestine of rodents. Immediately after establishment of the simple configuration a special form of secondary lysosomes can be observed, i.e. glycogenosomes, in the ephemerally existing huge glycogen containing areas. During subsequent fetal intestinal development one observes two events in

Tumor necrosis factor alpha is involved in mouse growth and lymphoid tissue development.

Tumor necrosis factor alpha (TNF-alpha), a major mediator of inflammation, also possesses a wide pleiotropism of actions, suggesting its involvement in physiological conditions. TNF-alpha mRNA is present in mouse embryonic tissues and also in fetal thymus and spleen. Repeated injections of a monospecific polyclonal rabbit anti-mouse TNF-alpha antibody in mice, starting either during pregnancy or at birth, led to a severe but transient growth retardation, already present at birth, reaching a 35% decrease in body weight at 3 wk, with complete recovery at 8 wk. The insulin growth factor I (IGF-I)

Vertebral hypersegmentation in a case of the VATER association.

Associations are statistical clusterings of malformations not known to be polytopic field defects, sequences, or syndromes. The VATER association is a nonrandom association of malformations including vertebral, anal, cardiovascular, tracheoesophageal, genitourinary, and limb defects. The caudal "dysplasia" sequence of lumbosacral vertebral defects, genitourinary abnormalities, and imperforate anus overlaps the VATER association. The cloacal membrane agenesis sequence is a pattern of malformations resulting in the absence of anal, genital, and urinary orifices with associated malformations in s

Teratogenic antibodies are directed against a coated-pit glycoprotein.

It has been well established that heterologous antibodies against certain tissue components may cause congenital abnormalities when injected into pregnant rats during the critical period of organogenesis. A glycoprotein antigen (gp340) of rat renal proximal tubules was isolated (C.C.K. Leung: (J. Exp. Med., 156:372-384, 1982); antibodies against gp340 were teratogenic. Indirect colloidal gold immunocytochemical method was utilized to study the ultrastructural localization of gp340. For comparative studies, both preembedding and postembedding immunostaining procedures were used. The results ind

Immunohistochemical localization of a beta-galactoside-binding lectin in rat central nervous system. I. Light- and electron-microscopical studies on developing cerebral cortex and corpus callosum.

From a lectin isolated from human brain (HBL-14), that specifically binds beta-galactosides, a polyclonal antiserum was raised that also recognizes a similar rat brain lectin (RBL-16). These antibodies allowed the immunocytochemical localization of the lectin during rat brain development by optical and electron microscopy. The presence of RBL-16, first detected at embryonic day 15, was specially increased from postnatal day 1 to 10. Its level decreased thereafter but it could still be detected in adult rat brain. The lectin, predominantly neuronal until postnatal day 13, was also present in as

Influence of an early grafted adenohypophysis on the thyroid of the chick embryo.

A hypophyseal pars distalis from an 11-day-old chick embryonic donor was implanted in the extraembryonic coelom of a 3-day-old recipient in order to evaluate the reactions of the thyroid of the host embryo and thus the functional capabilities of the grafted pituitary related to the thyroid. The graft did not modify the general growth of the embryo, but strongly influenced thyroidal development. The volume, colloid content, and radioiodide uptake of the thyroid gland were significantly increased, from Day 10 of incubation, compared with controls, as was the development of cytological structures

Histochemical enzyme-staining patterns of onchocerca volvulus microfilariae and their occurrence in different onchocerciasis areas.

Histochemical differentiation of Onchocerca volvulus microfilariae from 164 patients in West African rain-forest (Liberia), Sudan-savanna (Upper Volta), Guatemala and the Yemen has been carried out using a staining method for the demonstration of acid phosphatase. Intrauterine microfilariae showed considerable changes in their enzyme activity during embryonic development which are probably associated with the maturation of the parasite before migration to the tissues. Five distinct types of staining patterns could be distinguished among microfilariae from the skin according to the localization

Observations on the marginal ruffles of an established fibroblast-like cell line.

LW13K2 cells, a clone of a spontaneously in vitro transformed derivative of embryonic Lewis rat fibroblastic cells, were studied by phase contrast cine-light microscopy, scanning electron microscopy (SEM) and transmission electron microscopy (TEM). The ruffles found at the advancing edge of cells grown on glass substrates in vitro form and recede in a period of less than one min if they do not make an attachment of the substrate. If they fail to make an attachment they may form pinocytotic channels near the leading edge as described by Price (1972) and/or collapse, generally backwards, towards

Porcine Parvovirus.

Porcine parvovirus (PPV) is considered the main cause of reproductive disorders in pigs, which are summarized under the acronym SMEDI (stillbirth, mummification, embryonic death, and infertility). In this review the biology of the virus and its structure, pathogenic potential and strain variation, as well as the disease induced by the virus, are described. Known aspects of pathogenesis, diagnosis and prevention, particularly by vaccination, are summarized. Furthermore, in recent years 'new' parvoviruses (PPV2 to 7) have been described in pigs. They have been detected in pigs from various parts

A protein expressed in the growth cones of embryonic vertebrate neurons defines a new class of intermediate filament protein.

We have isolated and characterized cDNAs that encode a protein expressed in the axons and growth cones of a subset of Xenopus embryonic neurons. The protein is also expressed in a subset of cells of the brain, including cells in even-numbered rhombomeres, the eye, and the heart. The sequence of the cDNA suggests that the protein belongs to a new class of neural-specific intermediate filaments. Both the RNA and the protein are expressed in the neurula and persist during embryogenesis in the brain, cranial nerves, and spinal cord. Because of the predicted structure of the protein, we have named

Fibromuscular annular pancreas. A variant of pancreatic malformation?

All reported cases of annular pancreas consist of histologically normal pancreatic tissue. The most widely accepted theory of the pathogenesis of the pancreatic annulus agrees with, and is partially based on this fact. The pancreatic annulus encircling the stenotic second part of the duodenum in our patient was composed of fibromuscular tissue with lymph glands and a few pancreatic acini scattered among the smooth muscle fibers. This unique histologic variant of annular pancreas possible arose from a double fault in the ventrial anlage of the embryonal pancreas: faulty rotation (giving rise to

The Metabolic Role of Mitochondria in the Perinatal Cardiac Development and Cardiovascular Diseases.

The heart, as the central organ of the circulatory system, undergoes distinct metabolic transitions from embryonic development through postnatal maturation. A key aspect of this transition is mitochondrial maturation, which is influenced by changes in oxygen levels and metabolic substrates. These changes regulate cardiac structure and function by affecting signaling pathways and transcription factors. In cardiovascular diseases, mitochondria play a crucial role in altering metabolic patterns, contributing to pathological remodeling and worsening disease outcomes. This article explores mitochon

[Clinical guide to the management of patients with Beckwith-Wiedemann syndrome].

Beckwith-Wiedemann syndrome (BWS) is characterized by congenital overgrowth, macroglossia and omphalocele or umbilical hernia. Children with BWS may also have all or some of the following features: asymmetry (hemihypertrophy) of the limbs, torso or face, hypoglycemia, organomegaly, ear pits or creases, and embryonal tumors. The frequency of BWS is approximately 1:14,000 births. We present a guide for the management of children with BWS aimed at helping pediatricians and general practitioners or specialists in the clinical follow-up of these patients. This guide has been structured according to

Teratological research using in vitro systems. III. Embryonic organs in culture.

A search of literature published through the spring of 1986 yielded approximately 95 citations for the following embryonic organs in culture: kidney, pancreas, skin, palate, craniofacial tissue, tooth, lens, bones, digits, and liver. However, only the in vitro organ culture of the palate and tooth are reviewed in this paper. The other organ culture systems were not reported as teratogenic screens. Although some organs may have the potential for such use, many are currently used for evaluation of the pathogenesis associated with congenital abnormalities, cancer, or transplacental carcinogenesis

A histologic and embryologic analysis of so-called globulomaxillary cysts.

In an attempt to clarify the confusing concept of the so-called globulomaxillary cyst, we have reviewed all cases of this lesion occurring in the files of the CMDNJ-NJDS Biopsy Service between 1967--1977. We have discussed our cases in terms of the criteria used inconsistently in the literature of the globulomaxillary cyst: its location, the vitality of adjacent teeth, its histologic structure and its supposed origin from non-odontogenic embryonic epithelial remnants. We concluded that none of the criteria established the globulomaxillary cyst as a distinct entity and suggest that use of the t
