Sweet preference, sugar addiction and the familial history of alcohol dependence: shared neural pathways and genes.

Contemporary research has shown that a high number of alcohol-dependent and other drug-dependent individuals have a sweet preference, specifically for foods with a high sucrose concentration. Moreover, both human and animal studies have demonstrated that in some brains the consumption of sugar-rich foods or drinks primes the release of euphoric endorphins and dopamine within the nucleus accumbens, in a manner similar to some drugs of abuse. The neurobiological pathways of drug and "sugar addiction" involve similar neural receptors, neurotransmitters, and hedonic regions in the brain. Craving, …

Opioid receptors: from binding sites to visible molecules in vivo.

Opioid drugs such as heroin interact directly with opioid receptors whilst other addictive drugs, including marijuana, alcohol and nicotine indirectly activate endogenous opioid systems to contribute to their rewarding properties. The opioid system therefore plays a key role in addiction neurobiology and continues to be a primary focus for NIDA-supported research. Opioid receptors and their peptide ligands, the endorphins and enkephalins, form an extensive heterogeneous network throughout the central and peripheral nervous system. In addition to reward, opioid drugs regulate many functions suc…

Effects of low-dose naloxone on subjective alertness and pupil diameter in normal and narcoleptic subjects.

Recent evidence suggests that endogenous opiates may be involved in the pathophysiology of narcolepsy. To test this theory, the effect of 0.8 mg naloxone hydrochloride on pupil size and subjective alertness was measured in normal and narcoleptic subjects. Naloxone resulted in significant pupillary constriction in the normal but not in the narcoleptic subjects. The extent of contraction of the pupil light reflex was reduced significantly in the narcoleptic but not in the normal subjects. There was no effect on subjective ratings of alertness on the Stanford Sleepiness Scale or the visual analog

[Inhibitory effects of indomethacin on prolactin liberation induced with peptides with opium-like activity].

Indomethacin inhibits prolactin liberating effects by MET-enkefalin-NH2, a synthetic analogue of MET-enkefalin, both in intact and in ovariectomized, estradiol benzoate treated rats. The introduction of PGE1 increases the intensity of this effect. It is therefore possible to suppose that the PGs are involved as intermediaries of the prolactin relasing effect induced by MET-ENH-NH2.

Influence of nalmefene on energy balance and glucose regulation in Zucker rats.

It has been hypothesized that opioid peptides play a role in the development of obesity. The opiate antagonist naltrexone decreases glucose-stimulated insulin release, while the sensitivity of diabetic rats to naloxone-induced satiety is increased. These findings suggest a possible interaction between glucose concentrations and opiate antagonists in the control of food intake. In three experiments the energy balance and glucose regulatory responses of Zucker obese and lean rats to chronic administration of nalmefene, an opiate antagonist, were measured. Nalmefene, when injected subcutaneously

[Comparison of fentanyl and tramadol in pain therapy with an on-demand analgesia computer in the early postoperative phase].

17 patients undergoing cholecystectomy in non-opiate general anaesthesia received tramadol (n = 7) or fentanyl (n = 10) for immediate postoperative pain relief using the on-demand analgesia computer (ODAC). Heart rate, blood pressure, and respiratory rate were monitored at half-hourly intervals during the 6-h trial period. Arterial blood was withdrawn at hourly intervals for blood gas analyses and beta-endorphin plasma level assays. Fentanyl and tramadol serum levels were determined prior to each on-demand bolus injection during the first 2 h of the study. At the end of the trial period, the q

Characterization of peptides derived from pro-opiomelanocortin in the biological fluids of a patient with Nelson's syndrome.

Molecular forms of immunoreactive adrenocorticotropin (ACTH), beta-lipotropin (beta-LPH) (beta-endorphin (beta-END), human NH-2-terminal (hNT) of pro-opiomelanocortin (POMC), and gamma-3-melanotropin (gamma-3-MSCH) were studied in plasma, CSF and urine of a patient with Nelson's syndrome by molecular sieving and concanavalin A (Con A)-sepharose chromatography. In the culture tumor medium of the tumor cells, and in the plasma and CSF, these compounds were found mainly in forms corresponding in molecular weight to the authentic peptides, with the exception of gamma-3-MSH. Stimulation of the pitu

Centrally administered PGE2 inhibits gastric secretion in the rat by releasing vasopressin.

The pituitary mechanism of the central inhibitory effect of PGE2 on gastric secretion was investigated in rats. The intravenous (i.v.) injection of neurointermediate lobe extracts but not of anterior lobe extracts inhibited gastric secretion in pylorus-ligated rats. The i.c.v. administration of 3 micrograms of PGE2 increased the plasma level of vasopressin but had no effect on plasma beta-endorphin/beta-lipotropin. The i.v. administration of 10 micrograms d(CH2)5Tyr(Me) AVP, a vasopressin antagonist, prevented the inhibition of gastric secretion induced by i.c.v. administration of 3 micrograms

Activation of the proopiomelanocortin gene with ketoconazole as a treatment for Parkinson's disease: a new hypothesis.

Parkinson's disease is a neurodegenerative process characterized by progressive degeneration of the substantia nigra and concurrent loss of neuromelanin in these structures. The present hypothesis suggests that progression of Parkinson's disease may be reduced by enhancing the regional levels of neuroprotective factors derived from proopiomelanocortin (POMC). One practical means to accomplish this goal may be administration of ketoconazole or other inhibitors of corticosteroid synthesis at doses sufficient to stimulate hypophyseal secretion of POMC. The POMC constituents adrenocorticotropic ho

Cholecystokinin and endogenous opioid peptides: interactive influence on pain, cognition, and emotion.

It is well documented that stressful life experiences contribute to the etiology of human mood disorders. Cholecystokinin (CCK) is a neuropeptide found in high concentrations throughout the central nervous system, where it is involved in numerous physiological functions. A role for CCK in the induction and persistence of anxiety and major depression appears to be conspicuous. While increased CCK has been associated with motivational loss, anxiety and panic attacks, an increase in mesocorticolimbic opioid availability has been associated with coping and mood elevation. The close neuroanatomical

Endogenous opioids, the enteric nervous system and gut motility.

Opium alkaloids have been used for centuries as potent antidiarrheals and analgesics, their constipating action in the latter instance taken as an unwanted effect. It was only during the last decade that the physiological role of opioid peptides present in both neurons and endocrine cells of the gastrointestinal (GI) tract has been defined. The recognition of distinct opioid receptor types which may be differentially involved in the control of motility, acid and electrolyte secretion in the GI tract presently focuses the attention of researchers in this field on the identification of receptor-

Opioid neuronal denervation in Gilles de la Tourette syndrome.

Increased striatal dopaminergic functions with heightened postsynaptic receptor sensitivity has been proposed to underlie the major clinical symptoms of Tourette's syndrome (TS). The beneficial response of the majority of TS patients to haloperidol supports the hyperdopaminergic pathophysiological concept of TS. However, in 5 recently encountered TS patients, haloperidol failed to ameliorate self-injurious behavior (SIB) while the opiate antagonist, naloxone, attenuated SIB, implicating deranged endorphinergic mechanisms in the pathophysiology of this disorder. Brain damage is commonly associa

The differential effects of naloxone hydrochloride on the acquisition and maintenance of schedule-induced polydipsia.

Rats injected with the opiate antagonist, naloxone hydrochloride (10 mg/kg), 15 min prior to sessions in which they were given free food on a fixed time 75-sec schedule, displayed retarded acquisition of schedule-induced polydipsia relative to vehicle-injected subjects. Rats injected with naloxone after schedule-induced polydipsia had been acquired were unaffected, i.e., they continued to drink at control levels. Given that schedule-induced polydipsia has been considered non-opioid in nature, because of previous reports of its insensitivity to naloxone, the present report of differential effec

The effect of trichloroethylene on male sexual behavior: possible opioid role.

Trichloroethylene (TCE) is a chlorinated hydrocarbon solvent which is widely used as an industrial degreasing agent. Workers exposed to TCE often exhibit symptoms similar to those symptoms produced by narcotics. The present studies evaluated the effects of TCE exposure on measures of male sexual behavior in rats. The data indicated that TCE (1000 mg/kg, PO) 4 hours before testing produced an increased ejaculation latency effect on male copulatory behavior. Naltrexone (2.0 mg/kg, IP) given 15 minutes before testing blocked this TCE-induced effect. Animals given chronic TCE administration showed
