Building Programs to Eradicate Toxoplasmosis Part III: Epidemiology and Risk Factors.

PURPOSE OF REVIEW: Review comprehensive data on rates of toxoplasmosis in Panama and Colombia. RECENT FINDINGS: Samples and data sets from Panama and Colombia, that facilitated estimates regarding seroprevalence of antibodies to Toxoplasma and risk factors, were reviewed. SUMMARY: Screening maps, seroprevalence maps, and risk factor mathematical models were devised based on these data. Studies in Ciudad de Panamá estimated seroprevalence at between 22 and 44%. Consistent relationships were found between higher prevalence rates and factors such as poverty and proximity to water sources. Prena

Pathogenesis of Staphylococcus aureus necrotizing pneumonia: the role of PVL and an influenza coinfection.

Only recently necrotizing pneumonia was defined as a specific disease entity that is caused by a Panton-Valentine leukocidin (PVL)-producing Staphylococcus aureus strain and is frequently preceded by an influenza infection. Necrotizing pneumonia is characterized by a sudden onset and rapid worsening of symptoms, leukopenia, airway hemorrhages, severe respiratory failure and a high mortality rate. Despite clear epidemiological data, the function of PVL in necrotizing pneumonia has been controversially discussed due to conflicting results from different disease models. Furthermore, there are man

A village-based multidisciplinary study on factors affecting the intensity of cystic echinococcosis in an endemic region of the Tibetan plateau, China.

We investigated and quantified the factors which may affect the prevalence of cystic echinococcosis caused by Echinococcus granulosus in Rangtang County using a multidisciplinary approach. From a previously performed field survey, epidemiological data were linked with environmental data. Altitude and land surface temperature were extracted from remote-sensing images. Cumulative logistic regression models were used to identify and quantify the potential risk factors. The multiple regression models confirmed that yaks (χ 2 = 4·0447, P = 0·0443), dogs (χ 2 = 8·3455, P = 0·0039) and altitude (χ2 =

Sterile insect technique with accidental releases of sterile females. Impact on mosquito-borne diseases control when viruses are circulating.

The sterile insect technique (SIT) is a technique to control some vectors of diseases by releasing sterile males. However, during these releases, sterilized females can be (accidentally) released and since only females are vectors of diseases, it is important to study their impact when arthropod viruses are circulating. To that aim, we develop and study an entomological-epidemiological model, considering either permanent or periodic releases. Qualitative analyses of the continuous and periodic models are conducted. We highlight a critical sterile males release rate, ΛMcrit, above which the con

Determination of safe levels of persistent organic pollutants in toxicology and epidemiology.

We reviewed published manuscripts from toxicology and epidemiology reporting harmful health effects and doses of persistent organic pollutants (POPs), published between 2000 and 2021. We found 42 in vitro, 32 in vivo, and 74 epidemiological studies and abstracted the dose associated with harm in a common Molar unit. We hypothesized that the dose associated with harm would vary between animal and human studies. To test this hypothesis, for each of several POPs, we assessed the significance of variation in the dose associated with a harmful effect [categorized as non-thyroid endocrine (NTE), dev

EURL ECVAM Literature Review Series on Advanced Non-Animal Models for Respiratory Diseases, Breast Cancer and Neurodegenerative Disorders.

In vivo models are used in biomedical research to reproduce human disease and develop new drugs. However, they do not mimic the disease as it occurs in humans, and their use has failed to identify novel therapies effective for many highly prevalent non-communicable diseases, such as Alzheimer's disease. Indeed, the clinical failure rate in drug development remains very high, with an overall likelihood of approval from Phase I of about 9.6%. On the other hand, human-based models, advanced imaging techniques and human epidemiological studies may increase our understanding of disease aetiology an

Multi-model forecasts of the ongoing Ebola epidemic in the Democratic Republic of Congo, March-October 2019.

The 2018-2020 Ebola outbreak in the Democratic Republic of the Congo is the first to occur in an armed conflict zone. The resulting impact on population movement, treatment centres and surveillance has created an unprecedented challenge for real-time epidemic forecasting. Most standard mathematical models cannot capture the observed incidence trajectory when it deviates from a traditional epidemic logistic curve. We fit seven dynamic models of increasing complexity to the incidence data published in the World Health Organization Situation Reports, after adjusting for reporting delays. These mo

Hourly land-use regression models based on low-cost PM monitor data.

Land-use regression (LUR) models provide location and time specific estimates of exposure to air pollution and thereby improve the sensitivity of health effects models. However, they require pollutant concentrations at multiple locations along with land-use variables. Often, monitoring is performed over short durations using mobile monitoring with research-grade instruments. Low-cost PM monitors provide an alternative approach that increases the spatial and temporal resolution of the air quality data. LUR models were developed to predict hourly PM concentrations across a metropolitan area usin

Development of an Equity-Centered Sociotechnical Architecture for Generative AI Integration in Public Health Promotion: Conceptual Framework.

This article is a viewpoint: it presents the authors' perspective, informed by a critical synthesis of the current literature at the intersection of generative AI (GenAI) technologies, public health communication, and digital ethics, rather than original empirical data or analyses. The emergence of GenAI, including large language models (LLMs), represents a profound paradigm shift in digital health communication. By moving beyond traditional information retrieval to dynamic, human-like knowledge generation, GenAI offers unprecedented opportunities for public health promotion. However, the ungu

Urinary organophosphate metabolites, DNA methylation aging, and heart disease mortality in middle-aged and older adults: an exploratory cohort study with in vitro evidence of biological plausibility.

BACKGROUND: Chronic low-dose exposure to organophosphorus pesticides (OPPs) is widespread in the general population and has been linked to heart disease mortality, yet the biological pathways connecting such exposure to fatal outcomes remain poorly defined. We examined whether urinary OPP metabolites are associated with heart disease mortality and whether DNA methylation biomarkers might contribute to this association. METHODS: We studied 502 adults aged ≥50 years from the National Health and Nutrition Examination Survey (NHANES) 1999-2002 cycles with urinary OPP metabolite measurements, DNA

Understanding mosquito host-choice behaviour: a new and low-cost method of identifying the sex of human hosts from mosquito blood meals.

BACKGROUND: Mosquito-borne diseases are a global health problem, causing hundreds of thousands of deaths per year. Pathogens are transmitted by mosquitoes feeding on the blood of an infected host and then feeding on a new host. Monitoring mosquito host-choice behaviour can help in many aspects of vector-borne disease control. Currently, it is possible to determine the host species and an individual human host from the blood meal of a mosquito by using genotyping to match the blood profile of local inhabitants. Epidemiological models generally assume that mosquito biting behaviour is random; ho…

Abdominal obesity and the risk of esophageal and gastric cardia carcinomas.

BACKGROUND: Esophageal adenocarcinoma is rapidly increasing in incidence. Body mass index (BMI) is a risk factor, but its distribution does not reflect the demographic distribution of the cancer (which is highest among White men). Abdominal obesity patterns may explain this discordance, but no studies exist to date. METHODS: Nested case-control study within 206,974 members of the Kaiser Permanente multiphasic health checkup cohort; subjects received detailed questionnaires, a standardized examination including BMI and anthropometric measurements, and follow-up of esophageal and cardia cancers

Assessing dengue transmission risk and a vector control intervention using entomological and immunological indices in Thailand: study protocol for a cluster-randomized controlled trial.

BACKGROUND: Dengue fever is the most common and widespread mosquito-borne arboviral disease in the world. There is a compelling need for cost-effective approaches and practical tools that can reliably measure real-time dengue transmission dynamics that enable more accurate and useful predictions of incidence and outbreaks. Sensitive surveillance tools do not exist today, and only a small handful of new control strategies are available. Vector control remains at the forefront for combating dengue transmission. However, the effectiveness of many current vector control interventions is fraught wi

Dengue Virus-Specific Antibodies Enhance Brazilian Zika Virus Infection.

Anti-Flavivirus antibodies are highly cross-reactive and may facilitate Zika virus (ZIKV) infection through the antibody-dependent enhancement (ADE) mechanism. We demonstrate that dengue-specific antibodies enhance the infection of a primary Brazilian ZIKV isolate in a FcγRII-expressing K562 cell line. In addition, we demonstrate that serum samples from dengue-immune pregnant women enhanced ZIKV infection. These findings highlight the need for epidemiological studies and animal models to further confirm the role of ADE in the development of congenital and neurological complications associated

Developmental origins of adult diseases.

There is considerable evidence for the fact that early life environment in human beings are associated with future development of various metabolic diseases. Fetal programming and perinatal events appear to exert effects on later life that are independent of environmental risk factors in adults. Our understanding of the underlying mechanisms are limited and remains unclear. However several animal models and epidemiological studies have shown this association, and it is assumed secondary to the penalties of developmental plasticity. In this review, we amalgamate facts from several disciplines t

[Nonpathogenic listeriae: L. innocua sp. n. (Seeliger et Schoofs, 1977) (author's transl)].

In connexion with the separation of Listeria innocua (Seeliger and Schoofs, 1977) from Listeria monocytogenes (Murray et at., Pirie, 1940), resulting epidemiological and epizootological problems are discussed. L. innocua is apathogenic when conventional experimental models for testing Listeria monocytogenes are used. Under special conditions, i.e. use of suckling mice, strain Welshimer of L. innocua produced localized encephalitis (Patoĉka et al., 1979). The peculiarities of L. innocua justify further studies at the immunological, infectious-pathological and genetical level. A description of t

An integrated modelling methodology for estimating global incidence and prevalence of hereditary spastic paraplegia subtypes SPG4, SPG7, SPG11, and SPG15.

BACKGROUND: Hereditary spastic paraplegias (HSPs) are progressively debilitating neurodegenerative disorders that follow heterogenous patterns of Mendelian inheritance. Available epidemiological evidence provides limited incidence and prevalence data, especially at the genetic subtype level, preventing a realistic estimation of the true social burden of the disease. The objectives of this study were to (1) review the literature on epidemiology of HSPs; and (2) develop an epidemiological model of the prevalence of HSP, focusing on four common HSP genetic subtypes at the country and region-level
