The actions and responses of hypothalamic appetite regulatory factors change markedly during the neonatal to pre-pubertal period in order to maintain appropriate metabolic and nutritional conditions. In this study, we examined the developmental changes in the hypothalamic mRNA levels of brain-derived neurotrophic factor (BDNF), which is a potent anorectic factor and the changes in the sensitivity of the hypothalamic expression of this factor to fasting during the neonatal to pre-pubertal period. Under fed conditions, hypothalamic BDNF mRNA expression decreased during development in both male a

BACKGROUND AND OBJECTIVES: Fexinidazole is a 5-nitroimidazole recently included in a clinical efficacy trial as an oral drug for the treatment of human African trypanosomiasis (HAT). Preclinical studies showed it acts as a pharmacologically active pro-drug with two key active metabolites: sulfoxide and sulfone (the most active metabolite). The present studies aimed to determine the best dose regimen for the treatment of stage 2 sleeping sickness patients, which could eventually also treat stage 1 patients.
METHODS: Fexinidazole was assessed in 154 healthy adult male subjects of sub-Saharan Af

Law enforcement officers experience substantial occupational stressors that increase vulnerability to food addiction and cardiovascular disease (CVD), which may be compounded by adverse local environments. This study examined associations among county-level environmental factors, food addiction, and cardiometabolic profiles among North Carolina law enforcement officers. Participants included 330 officers (mean age = 37.98 years; mean BMI = 30.53 kg/m2) who completed the Yale Food Addiction Scale 2.0 and underwent assessments of anthropometrics, blood pressure, blood lipids, and glucose. County

OBJECTIVES: Intrauterine exposure to gestational diabetes mellitus (GDM) increases the risk of obesity in the offspring, but little is known about the underlying neural mechanisms. The hippocampus is crucial for food intake regulation and is vulnerable to the effects of obesity. The purpose of the study was to investigate whether GDM exposure affects hippocampal functional connectivity during exposure to food cues using functional magnetic resonance imaging.
METHODS: Participants were 90 children age 7-11 years (53 females) who underwent an fMRI-based visual food cue task in the fasted state.

Suction feeding in fish has long fascinated experimental biologists because of its complex motions, intricate anatomy and vast distribution across thousands of species and nearly every aquatic habitat. Suction feeding poses three main mechanical challenges for fish. First, how do shortening muscles create three-dimensional (3D) expansion of the mouth cavity to suck in water? Second, how do muscles produce the substantial power required for fast and forceful expansion to accelerate food and water into the mouth? Third, how is water moved through the mouth so that food can be deposited in the oe

PURPOSE: Tegoprazan is a potassium-competitive acid blocker (P-CAB) that is designed to treat acid-related diseases through a fundamentally different mechanism than that of proton pump inhibitors (PPIs). Because PPIs inhibit only activated parietal cell H+/K+ adenosine triphosphatase, stimulation of parietal cells by a meal is necessary for optimal results. In contrast, P-CABs can inactivate proton pumps without acid activation and bind to both activated and inactivated adenosine triphosphatase. This study evaluates the effect of food consumption on the pharmacokinetic and pharmacodynamic prop

OBJECTIVES: Taking advantage of its food-dependent bioavailability, the present study investigated the effect of a reduced dose taken with real-life meals on the pharmacokinetics (PK) of nilotinib in chronic myeloid leukaemia (CML) patients.
METHODS: Nilotinib was taken fasted (300 mg BID, days 1-4) or with real-life meals (200 mg BID, days 5-11). Rich sampling (days 1, 3, 8, 11) allowed for non-compartmental PK analysis. Nilotinib exposure (AUC0-12 h -Cmin -Cmax ) and its intra- and interpatient variability were compared between the two regimens. Adverse events were recorded by means of a p

Emerging resistance to antimalarial agents raises the need for new drugs. ACT-451840 is a new compound with potent activity against sensitive and resistant Plasmodium falciparum strains. This was a first-in-humans single-ascending-dose study to investigate the safety, tolerability, and pharmacokinetics of ACT-451840 across doses of 10, 50, 200, and 500 mg in healthy male subjects. In the 200- and 500-mg dose groups, the effect of food was investigated, and antimalarial activity was assessed using an ex vivo bioassay with P. falciparum. No (serious) adverse events leading to discontinuation wer

Semipurified human satietin (SP hSAT) significantly (p less than 0.05) reduced food intake for each of five consecutive days when injected intraperitoneally (2 mg/kg/day). Tolerance developed rapidly during the second and third days of injection but food intake of those animals receiving the highest doses of Sp hSAT (1 and 2 mg/kg/day) remained significantly below (p less than 0.05) that of the saline-injected animals. Food intake of rats receiving doses of 0.5 and 0.25 mg/kg/day Sp hSAT was below that of the saline-treated group for the first two days of injection (p less than 0.05) but not t

An anorectic substance in human, mammalian (rat, guinea pig, rabbit, cat, dog, horse, cattle) and poultry (goose) serum, named satietin, was discovered recently. The substance was extracted and purified by gelchromatographic techniques. With an improved technique, a highly purified glycopeptide fraction, containing 87% carbohydrates (fucose, mannose, glucose and galactose) and 12.5% amino acids, was prepared from human serum and its anorectic effect was measured in fasting rats. A biological assay for satietin, expressing activity in units, was developed. The unit is equivalent to the anorecti

Ghrelin is a regulator of appetite and growth hormone secretion, rising during fasting and decreasing after food intake. Children born small for gestational age (SGA) who undergo postnatal catch-up growth are at increased risk of obesity and metabolic disturbances, which may be related to impaired ghrelin regulation. This study assessed fasting and post-glucose ghrelin responses during an oral glucose tolerance test (OGTT) in prepubertal SGA children in relation to obesity and metabolic syndrome components. Ninety-eight children aged 5-9 years were included. Anthropometry, blood pressure, fast

BACKGROUND: Similar to procedures requiring general anesthesia, current guidelines recommend fasting for 6 hours for solids and for 2 hours for liquids prior to coronary angiography, but without data supporting such recommendation. The CORO-NF study aimed at assessing whether a shorter fasting period prior to elective coronary angiography associates with improved patient satisfaction without more complications compared with the standard fasting approach.
METHODS: We conducted a single-center, randomized, prospective, pragmatic study in 2 sequential phases: a "conventional protocol phase," con

The goal of this narrative review is to summarize the effects of prolonged fasting on various metabolic health measures, including body weight, blood pressure, plasma lipids, and glycemic control. Prolonged fasting is characterized by consciously eating little to no food or caloric beverages for several days to weeks. Results reveal that prolonged fasting for 5-20 days produces potent increases in circulating ketones, and mild to moderate weight loss of 2-10%. Approximately two-thirds of the weight lost is lean mass, and one-third is fat mass. The excessive lean mass loss suggests that prolong

The raccoon dog (Nyctereutes procyonoides) is a canid species with autumnal fattening and prolonged wintertime fasting. Nonpathological body weight cycling and the ability to tolerate food deficiency make this species a unique subject for studying physiological mechanisms in energy metabolism. AMP-activated protein kinase (AMPK) is a cellular energy sensor regulating energy homeostasis. During acute fasting, AMPK promotes fatty acid oxidation and enhances glucose uptake. We evaluated the effects of prolonged fasting on muscle energy metabolism in farm-bred raccoon dogs. Total and phosphorylate
