Adolescent binge drinking is associated with long-lasting adverse consequences. Preclinical studies have shown that the adolescent neuroimmune system may be functionally immature, as adolescent rats show a blunted cytokine response to a variety of challenges, including ethanol. The neuroimmune gene expression alterations seen after acute ethanol mirror those produced by the glucocorticoid receptor (GR) agonist dexamethasone (DEX), providing evidence that rapid, ethanol-induced changes in neuroimmune gene expression are mediated by corticosterone (CORT) release and subsequent GR activation. The

Computational theories of memory posit that the dentate gyrus and CA3 (CA3DG) hippocampal subfields reduce mnemonic interference via a process called pattern separation. While the CA3DG is viewed as a domain-general pattern separator, the parahippocampal and perirhinal cortices may play a role in content-specific (e.g., spatial or object-related) interference reduction. Recent work suggests that frontal and parietal control areas may allocate resources during mnemonic discrimination, but the evidence is still limited. Moreover, mnemonic discrimination tasks designed for humans almost exclusive

SIGNIFICANCE: The findings of correlations between the hippocampus and memory, but not processing speed, and fornix with both memory and processing speed in TLE suggest a specialized role of the hippocampus in memory and a broader role for the fornix in cognitive function, beyond its association with memory.

Medically intractable temporal lobe epilepsy is associated with potential neuronal and glial damage in key brain structures, including the hippocampus and amygdala. This study aimed to examine apoptosis, caspase expression, and glial activation markers in these regions, and to evaluate their associations with clinical variables. Hippocampal and amygdala tissues from 17 patients with medically intractable epilepsy and autopsy-derived non-epileptic controls were examined for cellular apoptosis (TUNEL), caspase-3, -9, and -12, as well as GFAP (astrocytic marker) and IBA1 (microglial marker) expre…

Children who undergo multiple exposures to anesthesia for surgeries and procedures are at increased risk of developing neurocognitive impairments. Growing evidence suggests that anesthesia-induced neurotoxicity and acute damage to neural cells underlie these impairments. However, the mechanisms underlying the long-term adverse effects of early anesthetic exposure remain largely unknown. Here, we show that repeated early-life exposures to sevoflurane, a commonly used anesthetic in clinical practice, lead to long-term impairments in hippocampal vascular architecture, characterized by reduced vas

Salicylate reliably induces tinnitus, yet its systemic effects on the central auditory and limbic systems remain incompletely characterized. Through integrated transcriptomic and metabolomic profiling of the rat cochlear nucleus and hippocampus, we observed pronounced region-specific remodeling following chronic tinnitus-inducing salicylate treatment. All differential and enrichment analyses were filtered using a nominal p-value cutoff (p < 0.05) without multiple-testing correction; thus, the findings should be interpreted as exploratory. We identified 150 differentially expressed genes (DEGs)…

Stress can be beneficial for adapting to dangerous situations in the short term but can be damaging in the long term, especially in the hippocampus. The noradrenergic and orexinergic systems play important roles in stress response. This study investigated the effect of noradrenergic activation on changes induced by chronic stress in the hippocampus and the involvement of orexinergic modulation in this process. This study examined five groups of 40 male Wistar rats (4 + 4 animals/ group): control, chronic stress, acute stress, control with noradrenergic activation, and chronic stress with norad…

Functional hyper-connectivity in hippocampus has been recently observed in patients with an early stage of type 2 diabetes, however, the structural connectivity between hippocampus and other regions of the brain and its effects on neuropsychological performance are still unknown. Participants (82 patients with type 2 diabetes and without peripheral microvascular complications; 75 healthy controls) underwent detailed cognitive assessment and diffusion MRI. Probabilistic tractography based on a multifiber model was performed to investigate the fiber connections between hippocampus and other regi…

Previous studies have demonstrated neuronal and microglial Fyn, a Src family kinase (SFK), and how its interactions with tau contribute to epileptogenesis. Saracatinib, a Fyn/SFK inhibitor, modifies disease progression in rat kainate (KA) epilepsy models. In this study, we investigated neuronal-specific fyn knockdown effects on Fyn-tau signaling, neurodegeneration, and gliosis using a calcium/calmodulin-dependent protein kinase II (CaMKII)-promoter-driven adeno-associated viral vector (AAV9)-mediated fyn-shRNA injection in the rat hippocampus. Eight days following AAV administration, rats rece…

OBJECTIVE: To evaluate the ability of magnetoencephalography (MEG) to detect the activity of a deep brain structure, particularly the hippocampus, using simultaneous stereotactic electroencephalography (SEEG) and MEG recordings.
METHODS: We included six patients with intractable temporal lobe epilepsy who underwent simultaneous SEEG and MEG. We recorded SEEG spikes and examined the MEG waveforms. We defined S-MEG spikes as MEG spikes synchronized with SEEG spikes in the deep brain structures. We examined the proportion of S-MEG spikes relative to SEEG spikes, the amplitude of S-MEG spikes, an…

Pain relievers such as tramadol are among the most commonly prescribed around the world. However, Long-term tramadol use has been shown to cause neurotoxicity, neuroinflammation, and cognitive impairment, which has limited its clinical use. Elderberry (EB), a plant extract widely recognized for its medicinal properties, is used in this study to investigate the effects of a plant-based diet on tramadol's adverse effects in the hippocampus region. The effects of EB oral diet on the devastating effects of tramadol on the hippocampus structure and function of 36 male albino rats were evaluated usi…

BACKGROUND: Multiple exposures to sevoflurane in developmental neonatal mice lead to neurocognitive deficits. The present study investigates the role and mechanisms of Piezo1 in sevoflurane-induced developmental neurotoxicity.
METHODS: Neonatal mice received sevoflurane on postnatal days (P) 6, 7, and 8. Piezo1 function was assessed using inhibitor GsMTx4 and agonist Yoda1. Cognitive function was evaluated via behavioral tests. Neuroinflammation was assessed by ELISA, Western blot, and immunofluorescence. Synaptic integrity was analyzed by Golgi staining.
RESULTS: Sevoflurane induced cogniti

Despite observational studies suggesting a link between chronic musculoskeletal pain (CMP) and increased risk of cognitive decline and dementia, the causal nature of this relationship remains uncertain due to potential confounding factors and reverse causality. We employed two-sample Mendelian Randomization (TSMR), bidirectional MR, mediation MR, drug-target MR, and colocalization analysis, along with gene set enrichment and protein-protein interaction (PPI) analyses. TSMR assessed the causal associations between CMP and the risk of dementia and its subtypes, including Alzheimer's disease (AD)

Neurotransmitter release consists of rapid synchronous release followed by longer-lasting asynchronous release (AR). Although the presynaptic proteins that trigger synchronous release are well understood, the mechanisms for AR remain unclear. AR is sustained by low concentrations of intracellular Ca2+ and Sr2+, suggesting the involvement of sensors with high affinities for both ions. Synaptotagmin 7 (SYT7) partly mediates AR, but substantial AR persists in the absence of SYT7. The closely related SYT3 binds Ca2+ and Sr2+ with high affinity, making it a promising candidate to mediate AR. Here,
