Internal Heat Acupuncture Treatment Inhibits Ferroptosis in the Spinal Dorsal Horn in Neurotoxin-Induced Parkinson's Disease Rats.

Parkinson’s disease (PD) is the second neurodegenerative disorder with limited therapeutic options. Ferroptosis is involved in the pathogenesis of PD. This study aimed to investigate the neuroprotective effects of internal heat acupuncture (IHA) in a PD model, focusing on its impact on ferroptosis. A neurotoxin-induced PD rat model was established using 6-hydroxydopamine (6-OHDA). These rats received IHA therapy. Behavioral tests were conducted to assess motor function and neuropathic pain. Serum inflammatory cytokines were evaluated by enzyme-linked immunosorbent assay. Ferroptosis markers an

Morphologic changes in striatonigral neurons in a rat model of levodopa-induced dyskinesia.

We aimed to elucidate morphological changes in striatonigral projection neurons in a rat model of levodopa-induced dyskinesia (LID). Male Wistar rats underwent unilateral 6-hydroxydopamine lesioning to establish a hemiparkinsonian model. At 8 weeks postoperatively, the rats were allocated to either the levodopa-treated group or the saline-treated control group. Behavioral abnormalities were quantified using the Abnormal Involuntary Movement (AIM) scores. We identified direct pathway neurons projecting to substantia nigra pars reticulata (SNr) by retrograde tracer injection. Dendritic spine mor

Measurement of Glutamate Suppression in a 6-OHDA-Induced Dopamine Deficiency Rat Model Following Acute Single-Dose L-DOPA Using GluCEST/MRS.

Background/Objectives: The Glutamate Chemical Exchange Saturation Transfer (GluCEST) technique is an advanced imaging modality that enables non-invasive glutamate quantification using MRI. Methods: This study evaluated glutamate dynamics in Parkinson's disease (PD) using a unilateral PD rat model, in which Wistar rats received 6-hydroxydopamine (6-OHDA) injections into the medial forebrain bundle, selectively eliminating dopaminergic neurons in the substantia nigra-striatum pathway. Results: The PD rat model exhibited a significant GluCEST increase (MTR Values: 3.0 ppm) compared to the sham-op

MRI-guided focused ultrasound for treating Parkinson's disease with human mesenchymal stem cells.

Parkinson's disease (PD) is a progressive disorder that affects the nervous system and causes regions of the brain to deteriorate. In this study, we investigated the effects of MR-guided focused ultrasound (MRgFUS) for the delivery of human mesenchymal stem cells (MSCs) on the 6-hydroxydopamine (6-HODA)-induced PD rat model. MRgFUS-induced blood-brain barrier (BBB) permeability modulation was conducted using an acoustic controller with the targets at the striatum (ST) and SN. Human MSCs were injected immediately before sonication. Here, we show that we can deliver human MSCs into Parkinsonian

Chemogenetic Regulation in Reprogrammed Stem Cell-derived Precursor Cells in Treating Neurodegenerative Diseases.

The integration of designer receptors exclusively activated by designer drugs (DREADD) with stem cell-based therapies presents an advanced strategy for precise neuronal modulation. Here, we utilized CRISPR-engineered human reprogrammed stem cells expressing excitatory (hM3Dq) or inhibitory (hM4Di) DREADD receptors to evaluate the functional integration and modulation of transplanted dopaminergic precursors in a murine model of Parkinson's disease (PD). Key steps included generating non-fusion DREADD-expressing stem cell lines, differentiating them into midbrain dopaminergic precursors, and tra

Nicotine Attenuates Pathogenesis of Parkinson's Disease via α7-nAChR-Mediated Lipid Metabolic Reprogramming and Anti-inflammatory Signaling.

Nicotine-an abundant tobacco alkaloid-has been epidemiologically linked to a lower incidence of Parkinson's disease (PD). In a 6‑hydroxydopamine (6‑OHDA) rat model, we found that chronic nicotine (1.5 mg kg-1 day-1) was associated with better motor performance and a smaller drop in striatal brain‑derived neurotrophic factor (BDNF) (p < 0.05). Untargeted lipidomics indicated partial normalization of 6‑OHDA‑induced changes in sphingolipids, phospholipids, and neutral lipids, alongside reduced lipid‑peroxidation markers. In SH‑SY5Y cells, nicotine attenuated 6‑OHDA‑related cytotoxicity, mitochond

Electroacupuncture suppresses motor impairments via microbiota-metabolized LPS/NLRP3 signaling in 6-OHDA induced Parkinson's disease rats.

Emerging evidence indicates that electroacupuncture (EA) exerts significant therapeutic effects on Parkinson's disease (PD)-related symptoms, with the immune mechanisms of the gut-brain axis playing a pivotal role in PD pathophysiology. This study aimed to explore whether EA mitigated PD-related symptoms and conferred neuroprotection to dopaminergic neurons in a 6-hydroxydopamine (6-OHDA) rat model by modulating the microbiota-metabolized lipopolysaccharide (LPS)/NLRP3 pathway. EA treatment ameliorated motor and anxiety symptoms in 6-OHDA rats and elevated the levels of TH. Metabolomic analysi

The GLP1R Agonist Semaglutide Inhibits Reactive Astrocytes and Enhances the Efficacy of Neural Stem Cell Transplantation Therapy in Parkinson's Disease Mice.

Cell transplantation offers a promising approach for treating Parkinson's disease (PD), but the limited survival of transplanted cells remains a major challenge. Reactive astrocytes, abundant in PD brains, may exacerbate this issue. GLP1R agonists, like semaglutide, are shown to inhibit reactive astrocytes in PD models. This study explores whether semaglutide could enhance the survival of transplanted neural stem cells (NSCs) in PD treatment. Six-hydroxydopamine-induced PD mouse models are used, with midbrain-derived NSCs transplanted into the lesioned striatum. Semaglutide is administered eve

Desmodesmus Extract as a Mitochondrion-Targeted Neuroprotective Agent in Parkinson's Disease: An In Vitro Study.

Parkinson's disease (PD) is the second most common neurodegenerative disease, and its prevalence is expected to double in the next 30 years. Currently, no effective treatment exists for Parkinson's disease. Thus, the research has focused on discovering new natural compounds with strong neuroprotective potential. This study aimed to investigate the effects of the methanol extract of Desmodesmus arthrodesmiformis EM13 (DaMe) on the mitochondrial damage pathway in an in vitro model of PD. The isolate of Desmodesmus arthrodesmiformis EM13 was first grown under appropriate culture conditions, and t

Environmental noise-induced changes to the IC-SNc circuit promotes motor deficits and neuronal vulnerability in a mouse model of Parkinson's Disease.

Emerging clinical evidence suggests a link between environmental noise and the severity of Parkinson's disease (PD). However, the effects of high-decibel noise exposure on PD and its underlying mechanisms remain unclear. In this study, we demonstrate that acute noise exposure induces reversible motor deficits in subacute low-dose 6-hydroxydopamine (6-OHDA) mice, a model of presymptomatic early-stage PD, while chronic noise exposure results in irreversible motor deficits and significant loss of substantia nigra compacta (SNc) dopaminergic (DA) neurons. Additionally, noise exposure activates the

Alpha-Synuclein Gene Alterations Modulate Tyrosine Hydroxylase in Human iPSC-Derived Neurons in a Parkinson's Disease Animal Model.

Parkinson's disease (PD) caused by SNCA gene triplication (3XSNCA) leads to early onset, rapid progression, and often dementia. Understanding the impact of 3XSNCA and its absence is crucial. This study investigates the differentiation of human induced pluripotent stem cell (hiPSC)-derived floor-plate progenitors into dopaminergic neurons. Three different genotypes were evaluated in this study: patient-derived hiPSCs with 3XSNCA, a gene-edited isogenic line with a frame-shift mutation on all SNCA alleles (SNCA 4KO), and a normal wild-type control. Our aim was to assess how the substantia nigra

Impact of 6-OHDA injection and microtrauma in the rat substantia nigra on local brain amyloid beta protein concentrations in the affected area.

Amyloid beta peptides (Aβ) are key indicators of Alzheimer's disease and are also linked to cognitive decline in Parkinson's disease (PD) and other neurodegenerative disorders. This study explored the accumulation of Aβ in a standard 6-Hydroxydopamine (6-OHDA) model of PD. We unilaterally injected 6-OHDA into the substantia nigra of Wistar rats to induce dopaminergic cell degeneration and death, a characteristic of PD. The goal was to detect Aβ protein in tissues and blood vessels showing inflammation or degeneration from the 6-OHDA injection. Our results showed that 6-OHDA injection produced

Adaptive wheel exercise for mouse models of Parkinson's Disease.

BACKGROUND: Physical exercise has been extensively studied for its therapeutic properties in neurological disease, particularly Parkinson's Disease (PD). However, the established techniques for exercise in mice are not well suited to motor-deficient disease-model animals, rely on spontaneous activity or force exercise with aversive stimuli, and do not facilitate active measurement of neurophysiology with tethered assays. Motorized wheel exercise may overcome these limitations, but has not been shown to reliably induce running in mice. NEW METHOD: We developed an apparatus and technique for in

Pioglitazone enhances brain mitochondrial biogenesis and phase II detoxification capacity in neonatal rats with 6-OHDA-induced unilateral striatal lesions.

The psychostimulant methylphenidate (MPH) is the first-line pharmacological treatment for attention-deficit/hyperactivity disorder (ADHD), but has numerous adverse side effects. The PPARγ receptor agonist pioglitazone (PIO) is known to improve mitochondrial bioenergetics and antioxidant capacity, both of which may be deficient in ADHD, suggesting utility as an adjunct therapy. Here, we assessed the effects of PIO on ADHD-like symptoms, mitochondrial biogenesis and antioxidant pathways in multiple brain regions of neonate rats with unilateral striatal lesions induced by 6-hydroxydopamine (6-OHD

Skeletal muscle sympathetic denervation disrupts the neuromuscular junction postterminal organization: A single-cell quantitative approach.

The sympathetic nervous system (SNS) regulates skeletal muscle motor innervation and stabilizes the NMJ in health, disease and aging. Previous studies using both chemical (6-hydroxydopamine, 6-OHDA) and microsurgically-induced sympathetic denervation examined the NMJ organization and transmission in the mouse; however, a detailed quantification of the postterminal on larger hindlimb muscles involved in gait mechanics and posture is lacking. The purpose of this study was to determine whether targets of the sympathetic neuron (SN) exhibiting different intrinsic composition such as the fast-twitc
