Chromosomal aberrations and SCE in lymphocytes of children revaccinated against smallpox.

Chromosomal changes were analysed in the peripheral lymphocytes of 14 twelve-year-old children before and 3 months and 10 months after smallpox revaccination (group A), and in peripheral lymphocytes of 8 children of the same age before and 1.5 and 8 months after revaccination (group B). A significantly increased number of aberrant cells was found after 1.5 and 3 months. In the blood samples collected 8 and 10 months after revaccination there was a decrease in the number of aberrant cells which, however, did not reach the control level. Sister chromatid exchanges (SCE) were counted in 5 childre

[Local reaction and neutralization test in smallpox vaccination after previous inoculation with MVA. Stickl's graduated vaccination (author's transl)].

Of 200 first inoculations and 8 revaccinations, the sera were examined for content of neutralizing vaccine antibodies 3 weeks after the MVA graduated inoculation. In spite of imponderables which affect the failure rate in the "clinical trial", a positive result was obtained on first vaccination in 85%. In vaccinated children with a negative neutralization test a distinct booster effect was obtained with a "main vaccination" after about six months. The same effect was seen with revaccinations. For the time being inadequately controlled cerebral convulsive diseases, severe endogenous eczema, neu

Leptospiral titres in pigs after vaccination.

Two groups of 8 pigs were vaccinated and given a booster vaccination 6 weeks later each with a commercial dual L. pomona and L. tarassovi killed vaccine. Serum from bloods collected before and up to 30 weeks after vaccination had agglutinating antibodies only after the 0ooster vaccination and then only with 1 vaccine. Titres persisted less than 8 weeks when tested against L. pomona but up to 16 weeks when tested against L. tarassovi at the 1:300 dilution and up to 20 weeks at the 1:100 dilution.

A replicating LCMV-based vaccine for the treatment of solid tumors.

Harnessing the immune system to eradicate tumors requires identification and targeting of tumor antigens, including tumor-specific neoantigens and tumor-associated self-antigens. Tumor-associated antigens are subject to existing immune tolerance, which must be overcome by immunotherapies. Despite many novel immunotherapies reaching clinical trials, inducing self-antigen-specific immune responses remains challenging. Here, we systematically investigate viral-vector-based cancer vaccines encoding a tumor-associated self-antigen (TRP2) for the treatment of established melanomas in preclinical mou

Increased neutralization and IgG epitope identification after MVA-MERS-S booster vaccination against Middle East respiratory syndrome.

Vaccine development is essential for pandemic preparedness. We previously conducted a Phase 1 clinical trial of the vector vaccine candidate MVA-MERS-S against the Middle East respiratory syndrome coronavirus (MERS-CoV), expressing its full spike glycoprotein (MERS-CoV-S), as a homologous two-dose regimen (Days 0 and 28). Here, we evaluate the safety (primary objective) and immunogenicity (secondary and exploratory objectives: magnitude and characterization of vaccine-induced humoral responses) of a third vaccination with MVA-MERS-S in a subgroup of trial participants one year after primary im

CD40 stimulation promotes human secondary immunoglobulin responses in HuPBL-SCID chimeras.

Antibodies to CD40 have been demonstrated to promote B-cell growth and differentiation in vitro. In order to determine if CD40 stimulation could promote antigen-specific human immunoglobulin (Ig) production in vivo, we examined the effects of anti-human CD40 MoAb in an in vivo system where human peripheral blood lymphocytes (huPBL) were engrafted into mice with severe combined immune deficiency (SCID). The huPBL-SCID mice were then given various doses of diphtheria-tetanus toxoid (DT) vaccine and were examined for the presence of human DT-specific antibodies by ELISA. Surprisingly, treatment w

Identification of Schistosoma mansoni antigens recognized by T cells of C57BL/6 mice immunized with gamma-irradiated cercariae.

Immunization of C57Bl/6 mice with gamma-irradiated Schistosoma mansoni cercariae induces highly significant protection against subsequent challenge with unattenuated living cercariae. The antigens that evoke T cell-mediated immunity in this model are vaccine antigen candidates since T lymphocytes mediate antibody-independent immune responses and control the production of most antibodies. To identify these antigens, spleen cells of mice immunized twice with gamma-irradiated cercariae were tested for proliferative responsiveness and production of interleukins (Il) and interferon-gamma (IFN-gamma

Determinants of BCG vaccine coverage among 0-2 months aged Somali children: Promoting equality in vaccination.

Somalia reports low overall immunization rates; the 2020 Somalia Demographic and Health Survey (SDHS) indicated that overall BCG vaccination coverage for children under five years of age was only 37.0%. However, coverage specifically for newborns and early infants remains underexplored. This study assessed the determinants of BCG vaccination among infants aged 0-2 months. Secondary data from the 2020 SDHS were analyzed for 457 mother - infant pairs. Descriptive statistics and multivariable logistic regression were used to identify factors associated with BCG uptake. BCG coverage was 79.7% (95%

Shielding the immunocompromised: COVID-19 prevention strategies for patients with primary and secondary immunodeficiencies.

The COVID-19 pandemic has significantly impacted immunocompromised patients, particularly those with inborn errors of immunity (IEI), transplant recipients, hematologic malignancies, and those undergoing treatment with immunosuppressive biologics and medications. These patients face an elevated risk of experiencing severe or even fatal consequences following SARS-CoV-2 infections. Vaccination is the primary defense against COVID-19; however, immune responses following immunization are often suboptimal in these patients, with variable specific humoral response rates. Despite the expedited regul

Microparticles entrapping pneumococcal protein SP0845 show improved immunogenicity and temperature stability.

Protein based vaccines are the most safe and affordable strategy to combat pneumococcal disease circumventing the limitations of conventional polysaccharide-based vaccines like serotype dependence, high cost and inability to be administered to immunocompromised. SP0845 is a highly conserved vaccine candidate shown to provide protection against heterologous strains of Streptococcus pneumoniae, the primal cause of pneumonia. However, the associated poor immunogenicity warrants the need for adjuvants and multiple doses to mount desired responses. The present study relates to improve the immunogen

Development of a subunit vaccine containing recombinant chicken anemia virus VP1 and pigeon IFN-γ.

Chicken anemia virus (CAV) is a severe threat to the chicken industry and causes heavy economic losses worldwide. In this study, we evaluated the immune response and protective efficacy provided by a subunit vaccine containing recombinant VP1 (rVP1) and pigeon interferon-γ (rPiIFN-γ). Results indicated that rPiIFN-γ enhanced humoral immunity elicited by rVP1 as early as 10 day after primary immunization and reach the high titer after secondary immunization. When compared to chickens immunized with rVP1, inactivated vaccine, chickens immunized with rVP1+rPiIFN-γ showed faster and higher levels

Sustained antigen-specific CD8+ T cell immunity post-mRNA booster requires notch pathway activation.

Messenger RNA (mRNA) vaccines effectively induce protective immunity, but antigen-specific CD8+ T cell responses exhibit limited persistence. In this study, we aimed to assess CD8+ T cell responses following a third dose of the Pfizer BNT162b2 COVID-19 vaccine and identify factors contributing to their longevity. Using HLA tetramers, we analyzed antigen-specific CD8+ T cells in 141 vaccinated individuals (86.5% female) and identified 2 groups: those with strong responses (strong group) and those with weak responses (weak group) 6 mo after the third mRNA vaccination. Transcriptomic analysis rev

Impact of acellular immunization against pertussis; comparative experience of four countries in North, Central and South America.

INTRODUCTION: Pertussis remains a public health concern despite widespread vaccination, due to waning immunity and asymptomatic transmission. The shift to acellular pertussis (aP) vaccines, aimed at reducing side effects, has changed disease dynamics, requiring ongoing evaluation. AREAS COVERED: We examined immunization strategies and epidemiological trends in Chile, Costa Rica, Mexico, and Panama following aP vaccine introduction. Compared vaccine coverage (VC), maternal immunization (MI), and booster strategies (BS), and explored their role in disease control, including post-COVID-19 resurg

Immunogenicity and duration of protection after yellow fever vaccine in people living with human immunodeficiency virus: a systematic review.

BACKGROUND: We lack the rationale on which to base the development of a yellow fever (YF) vaccination schedule for people living with human immunodeficiency virus (PLWHIV). OBJECTIVES: To report on the current evidence regarding the seroconversion rate and the duration of humoral protection after YF vaccine, as well as the impact of revaccination in PLWHIV. DATA SOURCES: MEDLINE, Google Scholar, LILACS and Cochrane CENTRAL were searched. METHODS: We selected studies on PLWHIV of all ages (including perinatally HIV-infected patients) and all settings (YF endemic and non-endemic zones). Inter

In Search of Factors Negatively Affecting Vaccine Immunity to Pertussis in Preschool Children Before the Administration of the First Booster.

INTRODUCTION: The top priority for active immunoprophylaxis of pertussis is the immunisation of infants as they can sometimes develop severe multiple-organ complications. OBJECTIVES: The aim of the work is the identification of factors negatively affecting vaccine immunity to pertussis in preschool children prior to the administration of the first booster. PATIENTS AND METHODS: The research was conducted on 352 children from 4.5 to 5.9 years of age who were hospitalised in the University Children's Hospital in Lublin (Poland) from 1 January 2012 to 31 December 2015. The children taking part

Need for Optimisation of Immunisation Strategies Targeting Invasive Meningococcal Disease in the Netherlands.

Invasive meningococcal disease (IMD) is a severe bacterial infectious disease with high mortality and morbidity rates worldwide. In recent years, industrialised countries have implemented vaccines targeting IMD in their National Immunisation Programmes (NIPs). In 2002, the Netherlands successfully implemented a single dose of meningococcal serogroup C conjugate vaccine at the age of 14 months and performed a single catch-up for children ≤18 years of age. Since then the disease disappeared in vaccinated individuals. Furthermore, herd protection was induced, leading to a significant IMD reductio

Irregular tick-borne encephalitis vaccination schedules: the effect of a single catch-up vaccination with FSME-IMMUN. A prospective non-interventional study.

BACKGROUND: Intervals longer than recommended are frequently encountered between doses of tick borne encephalitis virus (TBE) vaccines in both residents of and travelers to endemic regions. In clinical practice the management of individuals with lapsed TBE vaccination schedules varies widely and has in common that the underlying immunological evidence is scarce. STUDY PURPOSE AND METHODS: The aim of this study was to generate data reliable enough to derive practical recommendations on how to continue vaccination with FSME-IMMUN in subjects with an irregular TBE vaccination history.
