Carminic acid (CA) is a high-value natural anthraquinone pigment used in foods, cosmetics, textiles, and pharmaceuticals, but its current industrial supply depends largely on extraction from the scale insect Dactylopius coccus, creating constraints in yield, cost, sustainability, and allergen control. This review summarizes recent progress from pathway elucidation to microbial production. We first outline the structure, occurrence, applications, and biosynthetic logic of CA, emphasizing the convergence of type III polyketide assembly with insect-associated tailoring reactions, especially C-gly

Mosquitoes are the main vector of multiple diseases worldwide and transmit viral (malaria, chikungunya, encephalitis, yellow fever, as well as dengue fever), as well as bacterial diseases (tularemia). To manage the outbreak of mosquito populations, various management programs include the application of chemicals, followed by biological and genetic control. Here we aimed to focus on the role of bacterial pathogenesis and molecular tactics for the management of mosquitoes and their vectorial capacity. Bacterial pathogenesis and molecular manipulations have a substantial impact on the biology of

The Glossinavirus (Glossina pallidipes salivary gland hypertrophy virus (GpSGHV)) is a rod-shaped enveloped insect virus containing a 190,032 bp-long, circular dsDNA genome. The virus is pathogenic for the tsetse fly Glossina pallidipes and has been associated with the collapse of selected mass-reared colonies. Maintenance of productive fly colonies is critical to tsetse and trypanosomiasis eradication in sub-Saharan Africa using the Sterile Insect Technique. Proteomics, an approach to define the expressed protein complement of a genome, was used to further our understanding of the protein com

BACKGROUND: The fall armyworm, Spodoptera frugiperda, is a destructive pest threatening global food security. The sterile insect technique (SIT) is a promising eco-friendly control strategy, which relies on exploring genes regulating male fertility. Small heat shock proteins (sHSPs) act as molecular chaperones and participate in insect reproduction, yet their roles in lepidopteran dimorphic spermatogenesis remain poorly understood.
RESULTS: Here, we functionally characterized a testis-enriched sHSP gene, SfHsp20.2, in S. frugiperda. Using CRISPR/Cas9-mediated knockout, we found that loss of S

Growing global concern about the environmental risks posed by chemical insecticides has positioned RNA interference (RNAi)-based biopesticides at the forefront of research in green agricultural pest management, due to their precise target specificity and environmental safety. With ongoing technological advances and expanding use, the emergence of insect resistance to this technology has become a major constraint on their sustainable application. This paper provides a systematic review of delivery strategies and current applications of RNA-based biopesticides in pest control, and examines the m

Obesity is a major global health issue linked to metabolic disorders, chronic inflammation, and systemic complications, with high-fat diets (HFDs) playing a key role by disrupting intestinal balance and promoting oxidative stress. This study investigates Protaetia brevitarsis-derived protein hydrolysate (PBPH), an insect-derived bioactive peptide extract, as a potential intervention to counteract HFD-induced metabolic disturbances. Female ICR mice were divided into three groups: control diet, HFD, and HFD + PBPH, with PBPH (obtained by alcalase hydrolysis and ultrafiltration) administered dail

The Osiris gene family is believed to play important roles in insect biology. Previous studies mainly focused on the roles of Osiris in Drorophila, how Osiris operates during the development of other species remains largely unknown. Here, we investigated the role of LmOsi17 in development of the hemimetabolous insect Locusta migratoria. LmOsi17 was highly expressed in the intestinal tract of nymphs. Knockdown of LmOsi17 by RNA interference (RNAi) in nymphs resulted in growth defects. The dsLmOsi17-injected nymphs did not increase in body weight or size and eventually died. Immunohistochemical

Beetle Tenebrio molitor L. (Coleoptera, Tenebrionidae) is a well-known pest of grain and flour in food stores and grocery shops. Recently, commercial cultivation of the insect was started for human food and animal feed. Behaviour control of this insect using natural repellents is promising both for grain protection and commercial cultivation. We analysed if natural products of plant origin, namely essential oils (EOs), could be used for this purpose. Behavioural tests were performed using EOs of six plants: thymus (Thymus vulgaris), eucalyptus (Eucalyptus globulus), spearmint (Mentha spicata),

Phenolic compounds are the secondary metabolites (SMs) present in plants carrying different bioactivities. In the present study, we explored the influence of a phenolic compound namely phloroglucinol on oviposition behaviour and different biochemical entities of an insect pest Zeugodacus cucurbitae (Coquillett) (Diptera: Tephritidae) using artificial diet. Phloroglucinol (IUPAC name: benzene-1,3,5-triol) affected the activity of antioxidant and detoxifying enzymes viz. superoxide dismutases (SOD), catalase (CAT), ascorbate peroxidases (APOX). dehydroascorbate reductase (DHAR), peroxidases (POX

Despite their medical and economic relevance, it remains largely unknown how suboptimal temperatures affect adult insect reproduction. Here, we report an in-depth analysis of how chronic adult exposure to suboptimal temperatures affects oogenesis using the model insect Drosophila melanogaster. In adult females maintained at 18°C (cold) or 29°C (warm), relative to females at the 25°C control temperature, egg production was reduced through distinct cellular mechanisms. Chronic 18°C exposure improved germline stem cell maintenance, survival of early germline cysts and oocyte quality, but reduced

FK506 binding proteins (FKBPs) participate in regulation of diverse biological processes. However, the role of these proteins in insect-pathogenic fungi is far from well understood. To investigate the functions of FKBPs in Beauveria bassiana, a widely used entomopathogenic fungus for control of insect pests, we identify three putative FKBP genes, Bbfkbp12, Bbfkbp15, and Bbfkbp50, in the fungus. Gene-disruption experiments show that loss of Bbfkbp12 results in a significant increase of resistance of B. bassiana against the immunosuppressive compounds FK506 and rapamycin, while loss of Bbfkbp50

The rapid growth of global population is intensifying the demand for higher protein production. To satisfy the protein demand edible insects are emerging as a promising alternative protein source. Insect proteins exhibit high nutritional quality, favorable amino acid profiles, and significant functional versatility. However, their incorporation in the food requires a lot of processing to break it in to powder or other forms such as protein concentrate and isolate. The processing often applies thermal and non-thermal treatments which bring many structural modifications and alters the secondary,

The use of whole and visible insects is poorly accepted in Western countries, and this remains a significant challenge for product development. However, using insect-based protein-rich ingredients, like protein concentrate, can improve levels of consumer approval. The residual lipid content in insect protein concentrates can influence their techno-functional properties. Our study therefore aimed to evaluate the impact of the residual lipid content on the protein structure and foaming properties of a mealworm protein concentrate. Our results showed that the protein content increased from 78.01

Due to some similarity of innate immunity between insects and mammals, the study of the molecular mechanism of innate immunity in insects has become a focus of research. However, the exact molecular and cellular basis of immune system in insect remains poorly understood. Characterization of the transcriptomic response to Cd of spider is an effective approach to understanding the innate immunity mechanisms. In this study, we carried out transcriptome sequencing and gene expression analyses to develop molecular resources for Pardosa pseudoannulata venom glands with and without Cd treatments. A t

Jasmonates, together with other plant hormones, are important orchestrators of the plant immune system. The different hormone-controlled signaling pathways cross-communicate in an antagonistic or a synergistic manner, providing the plant with a powerful capacity to finely regulate its immune response. Jasmonic acid (JA) signaling is required for plant resistance to harmful organisms, such as necrotrophic pathogens and herbivorous insects. Furthermore, JA signaling is essential in interactions of plants with beneficial microbes that induce systemic resistance to pathogens and insects. The role

The scorpion depressant toxins are a group of evolutionarily conserved polypeptides targeting sodium channels, which show preferential ability to induce flaccid paralysis in insects, making them attractive candidates for the construction of transgenic plants or viral vectors to control pests. In this study, two new depressant toxin-like peptides (BmKITc and BmKITc2) differing only at position 52 (Lys for Thr) were produced in Escherichia coli. Circular dichroism analysis indicated that these two recombinant peptides display a typical structural feature similar to native scorpion toxins. They b

Locomotion of stick insects climbing over gaps of more than twice their step length has proved to be a useful paradigm to investigate how locomotor behaviour is adapted to external conditions. In this study, swing amplitudes and extreme positions of single steps from gap-crossing sequences have been analysed and compared to corresponding parameters of undisturbed walking. We show that adaptations of the basic mechanisms concern movements of single legs as well as the coordination between the legs. Slowing down of stance velocity, searching movements of legs in protraction and the generation of

We report the complete mitochondrial DNA genome of the soybean stem fly (SSF) Melanagromyza sojae from Brazil Santa Catarina state based on Illumina MiSeq sequence data. The estimated mitogenome is 15 475 base pairs (bp) (KT597923), with 13 protein-coding genes (PCGs), two ribosomal RNA (rRNA) genes, and 22 tRNAs, and an estimated 579 bp AT-rich control region. Similar to other insects, the SSF mitogenome is A-T bias with 40.9% A, 36.7% T, 13.6% C, and 8.8% G. Molecular characterization of SSF mitogenome will facilitate the development of effective molecular markers, and robust and rapid ident

Several naturally occurring amino acid derivatives display significant activities against weeds, fungi and insects: some of them have been even commercialized and are applied as crop protection agents. The 53 most important amino acid natural products with such efficacy are presented in this review together with their natural source, mode of action and biological activity. The diversity of the manifold bacterial, fungal and plantal sources of these compounds is impressive as well as their completely different structural scaffolds, ranging from cyclopeptides via unique non-proteinogenic amino a

BACKGROUND: The emergence of insecticide resistance in insects has led researchers to develop new control tools so that historic gains made in reducing disease transmission are not lost. Attractive targeted sugar baits (ATSBs) are a vector control tool being widely trialled to target insects that feed on plant sugars and blood. We designed a field-friendly, economical and more environmentally responsible sugar feeder for maintaining mosquito colonies and screening potential ATSB candidates.
METHODS: We simultaneously tested, in both male and female Anopheles gambiae mosquitoes, the effect of
