Abamectin has been used for decades as an insecticide and acaricide in arthropod pest management. However, there is no direct evidence from CRISPR/Cas9 studies confirming the involvement of GSTs in insect resistance to abamectin. The oriental fruit moth, Grapholita molesta, is a destructive pest of fruit trees worldwide. The role of GSTs in the oriental fruit moth remains unclear. In this study, an abamectin-resistant strain (AB-R) was derived from a susceptible laboratory strain (AB-S) of G. molesta. Synergist bioassays showed that the GST inhibitor diethyl maleate (DEM) significantly increas

BACKGROUND: Aedes aegypti (L.) is the main vector of dengue, yellow fever, Zika, and chikungunya viruses in many parts of the world, impacting millions of people worldwide each year. Insecticide-based interventions have been effective in controlling Aedes mosquito populations for several years, but in recent times, resistance to these compounds has developed, posing a global threat to the control of this mosquito.
METHODS: Ovitraps were used to collect A. aegypti eggs in the cities of Tartagal and San Ramón de la Nueva Orán (Salta), Puerto Iguazú (Misiones), and Clorinda (Formosa). World Heal

BACKGROUND: Aedes aegypti is a widespread mosquito in tropical and subtropical regions that causes significant mortality and morbidity in humans by transmitting diseases, such as dengue fever and Zika virus disease. Synthetic insecticides, such as pyrethroids, have been used to control Ae. aegypti, but these insecticides can also affect nontarget organisms and contaminate soil and water. This study aimed to investigate the mosquitocidal activity of Pseudomonas mosselii isolated from pond sludge against larvae of Ae. aegypti.
RESULTS: Based on the initial results, similar time-course profiles

Anopheles stephensi is a malaria vector that has been recently introduced into East Africa, where it threatens to increase malaria disease burden. The use of insecticides, especially pyrethroids, is still one of the primary malaria vector control strategies worldwide. The knockdown resistance (kdr) mutation in the IIS6 transmembrane segment of the voltage-gated sodium channel (vgsc) is one of the main molecular mechanisms of pyrethroid resistance in Anopheles. Extensive pyrethroid resistance in An. stephensi has been previously reported in Ethiopia. Thus, it is important to determine whether o

BACKGROUND: Increased intensity of pyrethroid resistance is threatening the effectiveness of insecticide-based interventions to control malaria in Africa. Assessing the extent of this aggravation and its impact on the efficacy of these tools is vital to ensure the continued control of major vectors. Here we took advantage of 2009 and 2014 data from Malawi to establish the extent of the resistance escalation in 2021 and assessed its impact on various bed nets performance.
METHODS: Indoor blood-fed and wild female Anopheles (An) mosquitoes were collected with an electric aspirator in Chikwawa.

It is known that a single mutation exerts moderate resistance to pesticide, while double mutations (DM) cause severe resistance problem through synergistic effect, and even result in failure application of pesticides. However, little is known about how double mutations would synergistically cause much high resistance level. In this work, computational studies were performed on the interaction of fipronil with N. lugens RDL-GABA receptors, to see how single and double mutations changed receptor structure properties and then conferred distinct resistance levels. The A2'S mutation displayed relat

Spinosad is a macrocyclic lactone insecticide that acts primarily at the nicotinic acetylcholine receptors (nAChRs) of target insects. Here we describe evidence that high levels of resistance to spinosad in the diamondback moth (Plutella xylostella) are associated with a three amino acid (3-aa) deletion in the fourth transmembrane domain (TM4) of the nAChR α6 subunit (Pxα6). Following laboratory selection with spinosad, the SZ-SpinR strain of P. xylostella exhibited 940-fold resistance to spinosad. In addition, the selected insect population had 1060-fold cross-resistance to spinetoram but, in

INTRODUCTION: The mosquito Aedes aegypti is the principal vector of dengue and yellow fever flaviviruses. Temephos is an organophosphate insecticide used globally to suppress Ae. aegypti larval populations but resistance has evolved in many locations.
METHODOLOGY/PRINCIPAL FINDINGS: Quantitative Trait Loci (QTL) controlling temephos survival in Ae. aegypti larvae were mapped in a pair of F3 advanced intercross lines arising from temephos resistant parents from Solidaridad, México and temephos susceptible parents from Iquitos, Peru. Two sets of 200 F3 larvae were exposed to a discriminating do

The larvae and adults of Aedes aegypti were tested for the potential to develop resistance to the synthetic pyrethroid, deltamethrin, alone or a combination of deltamethrin with the synergist, piperonyl butoxide (PBO). Although continuous larval selection for 40 generations resulted in 703-fold resistance, the resistance ratio in the adults was only 1.3. Similarly, adult selections with deltamethrin showed a resistance ratio of less than four after 40 generations, indicating differential response to deltamethrin selection in the two developmental stages of the insect. When the susceptible larv

Lice were collected from 71 flocks detected as infested at market inspection in 1990 and 1991, from 16 flocks where resistance was suspected and from 31 flocks from Kangaroo Island. Susceptibility to cypermethrin was measured by a treated surface technique and survival of lice at 5 ppm or greater was taken as an indication of resistance. The prevalence of resistance was 34% in louse populations from the market inspection sample, 50% in flocks in which resistance was suspected and 68% in flocks from Kangaroo Island. Most resistance factors were in the range 1 to 20 although one louse population

BACKGROUND: Genetically engineered crops producing insecticidal Bacillus thuringiensis proteins are useful for managing some key insect pests, including Helicoverpa zea (corn earworm or bollworm). In the United States, widespread practical resistance to crystalline (Cry) proteins and early warning of resistance to the vegetative insecticidal protein Vip3Aa have been reported for H. zea. To test the hypothesis that resistance to Cry1Ac or Vip3Aa causes fitness costs affecting flight and related traits in H. zea, we compared lab-selected Cry1Ac-resistant and Vip3Aa-resistant strains with their s

BACKGROUND: Insecticides in various formulations are indispensable for agricultural pest control and the prevention of vector-borne diseases. Piperonyl butoxide (PBO) is a synergist widely used to enhance the effectiveness of insecticides, notably pyrethroids, by inhibiting the detoxifying cytochrome P450 enzymes, thus reducing the capacity of insects to metabolize and resist insecticides. Recent studies have revealed an unexpectedly weak enhancement of pyrethroid efficacy by PBO, which may have serious implications for insect control. The underlying mechanism of this effect remains unclear.

Hygienic insecticides are applied directly to the living environment and are closely related to human life. Dimefluthrin (DIM) is one of the most widely used hygienic insecticides globally. However, with increasing mosquito resistance, both the concentration and duration of DIM usage have risen, prompting public concerns regarding its neurotoxic risks, especially for immunocompromised children. Therefore, this study evaluated DIM's neurotoxic risk from multiple perspectives using zebrafish larvae as a model, which was valuable for the future risk control of sanitary insecticides. The findings

Both exogenous and endogenous compounds can induce the expression of cytochrome P450 genes. The insect cytochrome P450 genes related to insecticide resistance are likely to be expressed as the "first line of defense" when challenged with insecticides. In this study, four cytochrome P450 genes, SinvCYP6B1, SinvCYP6A1, SinvCYP4C1, and SinvCYP4G15, were firstly isolated from workers of the red imported fire ant (Solenopsis invicta) through rapid amplification of cDNA ends (RACE) and sequenced. The fipronil induction profiles of the four cytochrome P450 genes and the two previously isolated CYP4AB…

The Aedes aegypti mosquito serves as a vector for several diseases, including dengue, Zika, chikungunya, and yellow fever. This species is well adapted to urban environments and poses a significant threat to public health. Some studies suggest that the gut bacteria of insect vectors may play a crucial role in developing resistance to insecticides. This study assessed the resistance of Ae. aegypti from Florencia, Caquetá, to temephos and deltamethrin and analyzed the diversity of gut bacteria in resistant larvae and adult females. Larvae exhibited resistance to temephos at a lethal concentratio

A series of novel quinazolinone derivatives targeting Aphis fabae were designed and synthesized. Their structures were confirmed through comprehensive analyses of 1H and 13C NMR and MS data. Biological assays showed that compounds 9-4, 9-7, 9-8, and 10-7 exhibit superior insecticidal activity against A. fabae compared to commercial compounds pyrifluquinazon, imidacloprid, and pymetrozine. Field trials further validated 10-7's long-lasting efficacy and enhanced control of imidacloprid-resistant aphid populations. Acute toxicity assays indicated that compound 10-7 had a low mammalian toxicity. A

Pesticides play a crucial role in agricultural pest management and crop yield preservation, yet their extensive application has led to rapid resistance evolution among pest populations. To address this challenge, insecticide resistance management (IRM) strategies aim to prevent or delay resistance evolution. Among the proposed IRM strategies, mixtures are promising but controversial. A key concern with mixtures is the potential for the accelerated evolution of multiple resistance, where selection builds linkage disequilibrium (LD) that leads to a disproportionate number of individuals carrying…
