Comparative insecticidal properties of two nucleopolyhedrovirus vectors encoding a similar toxin gene chimer.

Laboratory, greenhouse and field studies were conducted to characterize the insecticidal properties of genetically altered forms of Autograp … l to the macrolide, spinosad, and only slightly less than that of selected pyrethroid and carbamate insecticides. Overall, results from these studies indicate that, because of host range differences between the two wild-type viruses, HzNPV is the better vectoring agent (versus AcNPV) for designing recombinant clones as insecticides targeted at the multi-specie …

Amplification of a serine esterase gene is involved in insecticide resistance in Sri Lankan Culex tritaeniorhynchus.

Culex tritaeniorhynchus, the major vector of Japanese encephalitis in Sri Lanka, is resistant to organophosphorus insecticides, with a 10-fold resistance ratio at the LC50 for chlorpyrifos, and a high heterogenelty factor in the insect field population. The major mechanism of resistance in this species, as in the mosquito C. quinquefasciatus, is elevation of esterase activity. Basic biochemical, immunological and molecular analysis suggests that the C. tritaeniorhynchus CtrEstbeta1 gene is orthologous to the C. quinquefasciatus amplified Estbetas. The Estbeta2(1) antiserum cross-reacts strongl…

The Mossie-Buster: a hose-driven insecticide delivery tool for the control of container-breeding mosquitoes.

A novel insecticide delivery tool, the Mossie-Buster, was recently developed to control larval populations from urban breeding sites in Townsville, Australia. This functional and user-friendly control device directly targets the main breeding sites of Aedes aegypti (L.) with a focused delivery of insecticides. The Mossie-Buster comprises a commercially available device and attachments that mix an insecticide solution into the flow of water emitted from a hose. Attached to the device is a trigger for controlled insecticide release. Preliminary laboratory and field trials demonstrated the tool t…

Do insecticide-treated curtains reduce all-cause child mortality in Burkina Faso?

To evaluate whether insecticide-treated netting (ITN) reduces child mortality in different epidemiological settings, 4 large, randomized, controlled trials were conducted in Africa. Here we report the findings from the trial in Burkina Faso, in an area of hyperendemic and markedly seasonal malaria transmission. The trial involved 158 villages, with a total population of some 90,000, grouped into 16 geographical clusters. Ascertainment of mortality among children aged 6-59 months began in early 1993. In June/July 1994, 8 of the clusters, randomly selected, received permethrin-treated curtains.

Mode of action of an insecticidal peptide toxin from the venom of a weaving spider (Diguetia canities).

A recently discovered spider toxin (DTX9.2) induced a rapid paralysis when injected into insects. In neurophysiological experiments, DTX9.2 elevated spike discharges in sensory nerves and at the neuromuscular junction, and also caused a depolarization of the membrane potential in cockroach giant axons. All these effects were reversed by treatment with the specific sodium channel blocker, tetrodotoxin. These findings suggest that DTX9.2 acts upon the voltage-dependent sodium channels of insect nerve membrane.

Insecticide barrier spraying for the control of sand fly vectors of cutaneous leishmaniasis in rural Guatemala.

An initial evaluation of insecticide barrier spraying directed against sand fly vectors of cutaneous leishmaniasis was done in a nonclimax forested area with heavy undergrowth in Peten, Guatemala. A 100 m-wide swath of vegetation was sprayed once with a 1:3 mixture of cyfluthrin insecticide and a palm oil carrier using back-pack sprayers to simulate a central cantonment area in one site while another site remained as an untreated control. Prior to spraying and throughout 87 days post-treatment, sand fly populations were monitored at both sites with light traps set at ground and canopy levels a

The protective effect of bed nets impregnated with pyrethroid insecticide and vaccination against Japanese encephalitis.

A population-based case-control study to evaluate the protective effect of bed nets impregnated with pyrethroid insecticide and of vaccination against Japanese encephalitis was carried out in Gusi county, Henan province, China from June to September 1991; 50 cases and 100 matched controls were studied. Bed nets impregnated with pyrethroid insecticide greatly decreased the risk of infection among children under 10 years old, and vaccine efficacy was 78% (95% CI 16%-94%). We suggest that impregnated bed nets could be used as a quick response during outbreaks of Japanese encephalitis.

Specificity domain localization of Bacillus thuringiensis insecticidal toxins is highly dependent on the bioassay system.

The Bacillus thuringiensis crylA(a) and crylA(c) gene specificity regions were probed by creating and testing hybrid toxins both in vivo and in vitro against cultured insect cells or dissociated midgut epithelial cells. Toxin threshold dose determinations revealed that CrylA(c) is highly active against cultured Choristoneura fumiferana cells (CF-1) whereas CrylA(a) is nontoxic. In live insect bioassays, a reversed order of toxicity was observed. Hybrid analysis revealed that the CrylA(c) toxicity-determining region is located between codons 258 and 510. Two smaller subsections of this region (

Isolation and sequencing of insecticidal peptides from the primitive hunting spider, Plectreurys tristis (Simon).

About 50 peptide toxins were purified from venom of the primitive hunting spider, Plectreurys tristis. Bioassay by injection into larval Heliothis virescens (tobacco budworm) allowed selection of nine toxins for analysis of amino acid sequences. Total sequences were determined for six of the more insecticidal peptides (46-49 amino acids) and three contained free carboxyl-terminal amino acids. These toxins (plectoxins) are paralytic and/or lethal when injected into insect pests such as the lepidopterous larvae H. virescens, Spodoptera exigua (beet armyworm), and Manduca sexta (tobacco hornworm)

Identification of insecticidal peptides from venom of the trap-door spider, Aptostichus schlingeri (Ctenizidae).

Nine insecticidal peptides were isolated from the venom of the trap-door spider, Aptostichus schlingeri. Seven of these toxins cause flaccid paralysis of insect larvae within 10 min of injection and all were lethal within 24 hr. The complete amino acid sequences (32-76 residues) of six peptides are presented. The identified peptides contain three or four disulfide bonds and the larger peptides (74-76 residues) are quite similar in sequence with carboxyl termini as free acids, not amidated.

Curtatoxins. Neurotoxic insecticidal polypeptides isolated from the funnel-web spider Hololena curta.

Three polypeptide neurotoxins (curtatoxins) were isolated from the venom of the spider Hololena curta by reverse-phase high performance liquid chromatography, gel permeation, and ion-exchange chromatography. The purified toxins induced an immediate paralysis in the cricket Acheta domestica that resulted in desiccation and death of the insect within 24-48 h (LD50 congruent to 4-20 micrograms/g); this toxic effect is consistent with irreversible presynaptic neuromuscular blockade. Curtatoxins are a class of sequence-related, cysteine-rich, carboxyl-terminal amidated polypeptides of 36 to 38 amin

Control of horn flies on cattle with insecticide pellets fired from a CO2-operated pistol.

A method is needed to control pyrethroid-resistant horn flies (Haematobia irritans) which would involve the intermittent application of insecticides to cattle without their confinement for treatment. Our objective was to test a CO2 gas-operated pistol, designed to shoot marking pellets, to determine the number of pellet applications required to obtain season-long control. Also, since horn fly resistance to pyrethroid insecticides is widespread, we tested ten other insecticides as concentrates in pellets. Six pellet pistol applications of 38% permethrin provided 84% control of horn flies for 15…

A sampling method to determine insecticide residues on surfaces and its application to food-handling establishments.

Known amounts of acephate, chlorpyrifos, and diazinon were applied to Formica, unfinished plywood, stainless steel, and vinyl tile. Cotton-ball and dental wick materials were dipped in 2-propanol and "swiped" over the treated surface area two time. More acephate was found on the second swipe compared to the first from vinyl tile, similar amounts on both swipes from plywood, and less on the second swipe from formica and stainless steel. The ratio of chlorpyrifos on Swipe 1 compared to Swipe 2 found with cotton-ball on both formica and stainless steel surfaces was equivalent (6:1), but a conside

Insights into the toxicity, enzyme activity, and molecular docking of chemical and bio-insecticides against the fall armyworm, Spodoptera frugiperda.

The fall armyworm, Spodoptera frugiperda J.E. Smith (Lepidoptera: Noctuidae), is a major global pest that threatens cereal crops. Insecticides remain one of the key strategies for its management. This study assessed the toxicity, developmental effects, detoxification enzyme activity, and molecular docking interactions of 2 chemical insecticides profenophos (organophosphates group) and cypermethrin (pyrethroids group) and 2 bio-insecticides (spinosad and Bacillus thuringiensis) on S. frugiperda. Bioassay results revealed that spinosad exhibited the highest toxicity, followed by cypermethrin, pr…

Field efficacy of new-generation insecticides and household residue mitigation techniques assessed by LC-MS/MS for safe tomato consumption.

The present study was conducted during 2023–24 and 2024–25 to evaluate the field efficacy of new-generation insecticides against major insect pests of tomato (Solanum lycopersicum L.) and to assess household-level residue mitigation techniques for safe tomato consumption. Field trials tested chlorantraniliprole, emamectin benzoate, spinosad, and indoxacarb at three dosages. Chlorantraniliprole 18.5% SC at the recommended 1× dose reduced Helicoverpa armigera populations by 59.82% and 51.60% across the two seasons, and aphid populations by 32.20% and 39.09%, achieving tomato yields of 17.40 t ha…

Differential chemical hazards of common insecticides to Eupelmidae parasitoids: An ecological risk assessment for integrated pest management in forest ecosystems.

Eupelmidae parasitoids (Anastatus and Mesocomys spp.) are crucial for biological pest control, yet their ecological function may be compromised by insecticide exposure within integrated pest management programs. We evaluated the lethal and sublethal hazards of five widely used insecticides (beta-cypermethrin, avermectin, imidacloprid, matrine, and chlorbenzuron) on four parasitoid species. Beta-cypermethrin exhibited high toxicity to M. albitarsis and M. trabalae, causing 100 % mortality within 72 h, while A. japonicus and A. meilingensis showed significantly greater tolerance. Hazard quotient…

Psychiatric Risk Implications From Behavioral and Neural Effects of Adolescent Exposure to Environmental Insecticides: A Systematic Review of Rodent Studies.

Adolescence is a sensitive neurodevelopmental period marked by remodeling of brain circuits that support cognitive development and emotion and behavior regulation. These maturation processes heighten psychiatric vulnerability to environmental exposures, including to toxicants such as insecticides. Epidemiological studies show widespread adolescent insecticide exposure and increasingly link this to psychiatric outcomes, but underlying neural mechanisms remain poorly understood. Preclinical studies can clarify these associations and identify insecticide-induced mechanisms that may disrupt neurod…

High resistance of Anopheles gambiae s.L. populations to public health insecticides in Ghana: contribution of domestic and agricultural pesticide use.

BACKGROUND: Malaria elimination efforts in sub-Saharan Africa are increasingly threatened by the spread of insecticide resistance in Anopheles gambiae sensu lacto (s.l.). The widespread use of insecticides in vector control and pesticides in agriculture has intensified resistance development in malaria vectors. This study examined the drivers of high insecticide resistance in Anopheles mosquitoes in Ghana's Coastal and Sahel Savannah ecological zones. METHODS: Anopheles larvae were collected from eight sites across the Coastal Savannah (Tuba, Opeibea, Nima, and Teshie) and Sahel Savannah (Kpa…

A self-assembled multicomponent RNA nano-biopesticide for increasing the susceptibility of destructive bean flower thrips to insecticides via dsNrf2.

High resistance of bean flower thrips (BFT, Megalurothrips usitatus) has led to the unscientific application of insecticides to cause famous "toxic cowpea" incidents in China. Nuclear factor erythroid 2-related factor 2 (Nrf2) plays an important role in inducing antioxidant responses and drug detoxification. Therefore, the detoxification genes may be suppressed to control insecticide resistance via Nrf2. Herein, we demonstrated that the expression of most detoxification genes and enzyme activity were remarkably suppressed via nrf2 RNAi. Subsequently, a novel hydrophilic-lipophilic diblock poly…
