Comparison of syndromic versus laboratory-confirmed diagnosis of Neisseria gonorrhoeae and Treponema palladium, infections at the selected health centers in Addis Ababa, Ethiopia.

BACKGROUND: Sexually transmitted infections (STIs) are major infectious diseases worldwide. Around one million people get STIs every day and among them a high burden of the diseases seen in Sub-Saharan African countries. In most developing countries, including Ethiopia, STIs are diagnosed only using syndromic methods, although there seems to be no consensus between syndromic and laboratory-based research. OBJECTIVE: To compare the effectiveness of a syndromic versus laboratory-based approach in the diagnosis of sexually transmitted infections, especially Neisseria gonorrhoeae (NG) and Trepone

Multiplex RT-PCR provides improved diagnosis of skin and nail dermatophyte infections compared to microscopy and culture: a laboratory study and review of the literature.

Dermatophytes are the most common cause of superficial mycosis, estimated to affect 20% to 25% of the general population. We assessed the performance of a novel real-time polymerase chain reaction (RT-PCR) multiplex assay for diagnosis of dermatophytosis. To evaluate sensitivity and specificity, 10 known bacteria and 10 known fungi commonly found on skin, as well as 105 samples with culture confirmed dermatophytosis were tested using Dermatophyte and Fungi assay (AusDiagnostics, Sydney, Australia), a novel multiplex assay for diagnosis of dermatophytosis in skin and nail. This was followed by

Feeding behaviour of Glossina pallidipes and g. morsitans centralis on Boran cattle infected with trypanosoma congolense or T. vivax under laboratory conditions.

In field studies, tsetse flies (Diptera: Glossinidae) feed more successfully on cattle infected with Trypanosoma congolense Broden (Kinetoplastida: Trypanosomatidae) than on cattle infected with T. vivax Ziemann or uninfected cattle. Here we describe the first laboratory investigation of this phenomenon. In the first experiment, caged Glossina pallidipes Austen were fed for 1 and 5 min on a Boran steer infected with T. congolense clone IL 1180 and on an uninfected steer. Feeding success was recorded in this way five times over several weeks. The same protocol was subsequently used in three add

Prolongation and enhancement of Nippostrongylus infection in the laboratory rat by a heterologous antiserum to rat peritoneal cells--a possible role for pharmacologically-active cells in immunity.

A rabbit anti-rat peritoneal cell (APCS) serum significantly suppresses (P<0.001) the immune response to Nippostrongylus brasiliensis infection in the rat and the infection can be prolonged by such an antiserum. The antiserum appeared to contain antibodies directed against the surface antigens of pharmacologically-active cells such as the mast cell. Prolongation of infection following treatment with APCS was accompanied by a reduction in the number of eosinophils, basophils and neutrophils in the peripheral circulation. The number of circulating peripheral lymphocytes was not significantly aff

A Bayesian latent class estimation of the diagnostic accuracy of clinical examination and laboratory assays to identify bovine ephemeral fever virus infection in South African cattle.

Bovine ephemeral fever (BEF) is an economically important vector-borne viral disease of cattle and water buffalo in Africa, Australia and parts of Asia. The control of BEF is centred around vaccination, and therefore accurate, early identification of disease outbreaks are key to minimize its economic and welfare impact. In Africa, control programs are hampered by limited diagnostic capabilities and poor infrastructure for rapid transportation of diagnostic specimens. The primary objective of this study was to estimate the sensitivity (Se) and specificity (Sp) of four tests, namely clinical exa

Surprising uncovering of gram-negative diplococci from blood cultures: First identification of Pseudoroseomonas aerofrigidensis as probable bloodstream infection - clinical and laboratory approach towards an unknown pathogen.

Identification of pathogens has been accelerated by MALDI-TOF, including very rare bacteria, which can challenge routine diagnostics by uncommon features. Moreover, rare or only recently discovered microorganisms often lack experience or data on antibiotic sensitivity. A 67-year-old patient presented with a feverish clinical picture. Initial Gram-staining of bacteria in positive blood cultures showed gram-negative diplococci suggesting highly pathogenic bacteria. In further identification a 2017 newly discovered species, Pseudoroseomonas aerofrigidensis, was identified by MALDI. We present the

Comparison of 4 laboratory tests for the detection of bovine rabies viral infection in Paraguay: fluorescent antibody test, rapid detection test, histologic lesions, and RT-PCR.

Rabies virus (RABV; Lyssavirus rabies) is a neurotropic virus that can be transmitted to mammals by the hematophagous bat Desmodus rotundus. An accurate, accessible method for the detection of RABV in cattle is necessary in Paraguay; thus, we evaluated the detection of RABV using 4 techniques: fluorescent antibody test (FAT), immunochromatography rapid detection test (RDT; Anigen Rapid Rabies Ag test kit; Bionote), a reverse-transcription PCR (RT-PCR) assay, and histologic lesions in different portions of the CNS of 49 Paraguayan cattle to determine the most sensitive and specific technique. At-Home Specimen Self-Collection and Self-Testing for Sexually Transmitted Infection Screening Demand Accelerated by the COVID-19 Pandemic: a Review of Laboratory Implementation Issues.

The coronavirus disease 2019 (COVID-19) pandemic reduced the sexually transmitted infection (STI) testing volume due to social-distancing and stay-at-home orders, among other reasons. These events highlighted previously known benefits of at-home STI self-testing or specimen self-collection and accelerated testing demand via telemedicine. We review testing outside traditional clinical settings. We focus on three curable bacterial STIs among the top 10 U.S. nationally notifiable conditions with screening recommendations: syphilis, gonorrhea (Neisseria gonorrhoeae, also known as the gonococcus [G

Real-time PCR for detection of Strongyloides stercoralis in human stool samples from Côte d'Ivoire: diagnostic accuracy, inter-laboratory comparison and patterns of hookworm co-infection.

Human infections with the helminth species Strongyloides stercoralis encompass a wide clinical spectrum, ranging from asymptomatic carriage to life-threatening disease. The diagnosis of S. stercoralis is cumbersome and the sensitivity of conventional stool microscopy is low. New molecular tools have been developed to increase sensitivity. We compared the diagnostic accuracy of real-time PCR with microscopy for the detection of S. stercoralis and hookworm in human stool samples, and investigated the inter-laboratory agreement of S. stercoralis-specific real-time PCR in two European laboratories
