A new butenolide derivative (1) featuring octyl substitution at γ-position, together with four known analogues (2-5) were isolated from marine-derived Streptomyces koyangensis SCSIO 5802. The structure of 1 was elucidated by HR-MS and NMR spectroscopic data analyses. The absolute configuration of the stereo centre in lactone ring of 1 was determined by comparison of CD spectrum with those of known compounds. Compound 1 exhibited mild antiviral activity against herpes simplex virus with EC50 value of 25.4 µM.

An enantioselective synthesis of 8-epi-xanthatin (9) has been accomplished starting from the bicyclic lactone 3, which has been used for the synthesis of other xanthanolides, sundiversifolide (4) and diversifolide (5), through a synthetic route without the use of a selenium species. Additionally we have evaluated antimicrobial activities of five natural xanthanolides and their derivatives. Although the synthetic xanthanolides did not show any activity against methicillin-resistant Staphylococcus aureus (MRSA), some of the synthetic intermediates did exhibit moderate antimicrobial activities.

Two new cardenolides (1 and 2) along with 12 known compounds were isolated from the dichloromethane extract of the leaves of Calotropis gigantea. The structural elucidation was accomplished by spectroscopic methods. Some of the isolates were evaluated for cytotoxic activity against KB, BC, and NCI-H187 cancer cell lines, and all cardenolides tested were found to possess strong inhibitory effects. The presence of a deoxysugar at C-3, a formyl group at C-10, and an alpha,beta-unsaturated gamma-lactone was crucial for cytotoxic activity.

The first total synthesis of the anti-inflammatory Ganoderma meroterpenoid dimers dispirocochlearoids A-C, along with five of their diastereomers─potential unreported natural products─has been accomplished using a convergent strategy that leveraged common intermediates derived from dayaolingzhiol M. The synthesis features several key steps: a hetero-Diels-Alder reaction, a hydrolysis/double-bond migration cascade to assemble the D/E-bicyclic core, and a condensation/intramolecular aldol/lactonization cascade that constructs the final B/C-bicyclic system of dispirocochlearoids A-C.

Thelepamide, an unique ketide-amino acid isolated from a marine annelid worm Thelepus crispus, has a unique oxazolidinone ring derived from cysteine, glycine and valine. Rareness in nature as well as promising bioactive possibility make the oxazolidinone ring an attractive synthetic target. The hydroxy oxazolidinone fragment of thelepamide was prepared by acid-catalysed N,O-acetal formation between a ketoamide and formaldehyde. The first total synthesis of the potent antibiotic berkeleylactone A is described in 10 steps with an overall yield of 9.5%. A key step of our concise route is a late-stage, highly diastereoselective, sulfa-Michael addition. The 16-membered macrocyclic lactone was formed via ring closing metathesis and subsequent chemoselective reduction. The absolute stereochemical configuration was confirmed by single-crystal X-ray analysis. Synthetic berkeleylactone A was tested against several methicillin-resistant Staphylococcus aureus strains, and its potent antibacterial activity was verified.
