Diffuse large B-cell lymphoma (DLBCL) is the most common subtype of B-cell non-Hodgkin lymphoma (NHL) and is characterized by significant biological and clinical heterogeneity. The programmed death 1 (PD-1)/ programmed death-ligand 1 (PD-L1) immune checkpoint pathway plays a crucial role in tumor immune evasion; however, its diagnostic and prognostic relevance in DLBCL remains unclear. We retrospectively analyzed 66 cases of DLBCL diagnosed between 2017 and 2024 at a single institution. Immunohistochemistry was performed on formalin-fixed, paraffin-embedded tissues using antibodies against PD-

Diffuse large B-cell lymphoma (DLBCL) is a common aggressive non-Hodgkin lymphoma, frequently seen in immunocompromised individuals. This report details the case of a 40-year-old HIV-positive female admitted for abdominal pain, vomiting, and imaging suggestive of disseminated tuberculosis, who was ultimately diagnosed with DLBCL of the ileum complicated by intussusception. Initial investigations revealed anemia and thrombocytosis. Contrast-enhanced CT showed multiple necrotic granulomatous lesions and jejunoileal intussusception, mimicking tuberculosis. Surgical exploration and resection of th

Diffuse large B-cell lymphoma (DLBCL), the most prevalent non-Hodgkin lymphoma (NHL), is characterized by rapid growth and an unfavorable prognosis. Artesunate (ART), a derivative of Artemisinin, is a widely recognized antimalarial drug that displays notable antitumor properties across diverse cancers. Our previous studies have demonstrated ART's capacity to inhibit DLBCL progression via the induction of ferroptosis. However, its direct target molecules mediating this effect remained elusive. In this study, using small molecule (SM) pull-down combined with mass spectrometry analysis (LC-MS/MS)

Diffuse large B-cell lymphoma (DLBCL) presents a substantial clinical challenge due to its aggressive nature and resistance to conventional therapies, thereby developing novel treatment strategies is critical. In this work, we take advantage of the insensitivity of heat shock protein (HSP) expression in senescent tumor cells to combine mild photothermal therapy (MPTT) for efficient DLBCL treatment. Insensitivity to HSPs implies that cells are less capable of activating their usual stress protection mechanisms during MPTT, which reduces their tolerance to heat damage. Consequently, the treatmen

Diffuse large B-cell lymphoma (DLBCL) is one of the most common and lethal B-cell malignancies worldwide, with high relapse rate after standard treatment, and the relapsed cases are often difficult to treat. Ubiquitination has a pivotal role in cellular protein homeostasis and tumorigenic deubiquitinases (DUBs) stabilize oncoproteins during carcinogenesis. In this study, we have identified USP13 as one of the abnormally-overexpressed and stage-related DUBs in DLBCL critical to disease pathogenesis. Employing LC-MS/MS, Ran GTPase was characterized as potential substrate of USP13, with the inter

INTRODUCTION: Antibody-drug conjugates (ADCs) are a rapidly evolving class of anti-cancer drugs with a significant impact on management of hematological malignancies including diffuse large B-cell lymphoma (DLBCL). ADCs combine a cytotoxic drug (a.k.a. payload) attached through a linker to a monoclonal antibody specific to a particular cancer antigen. Payloads include microtubule disruptors or DNA damaging chemicals. After attaching to the antigen, the ADCs are internalized, and the payload is dissociated from ADC by lysozymes and delivered to the intended site for exerting cytotoxic effects.

PHD finger protein 19 (PHF19) is an epigenetic regulator known to be associated with the prognosis of patients in various tumours, while its impact on the prognosis of diffuse large B-cell lymphoma (DLBCL) remains unexplored. This study aimed to investigate the expression and prognostic significance of PHF19 in DLBCL. PHF19 expression was assessed in 192 DLBCL cases using immunohistochemistry. Correlations between PHF19 expression and clinicopathological parameters were analysed. Survival analysis was performed in patients receiving an anthracycline-based regimen with rituximab. Patients with relapsed/refractory diffuse large B-cell lymphoma (DLBCL) transformed from indolent B-cell lymphomas, including Richter transformation, have a poor prognosis. PD-1/PD-L1 antibodies produce modest objective and complete response rates in B-cell non-Hodgkin lymphoma as monotherapy but may synergize with immunogenic chemotherapies such as gemcitabine and oxaliplatin (GemOx). Thus, we evaluated the safety and efficacy of atezolizumab plus rituximab and GemOx (R-GemOx+Atezo) in R/R transformed DLBCL, including Richter transformation. We conducted a phase I trial including patients wit

BACKGROUND: Diffuse large B-cell lymphoma (DLBCL) is a highly heterogeneous disease with variable clinical and molecular features. Studies have highlighted the significant role of γδ T cells in the survival of leukemia patients. However, the heterogeneity of γδ T cells and their impact on clinical correlation in the peripheral blood of patients with DLBCL remain unclear.
METHOD: Single-cell RNA sequencing (scRNA-seq) was employed on 9 blood samples, sourced from 6 patients with diffuse large B-cell lymphoma (DLBCL) and 3 healthy individuals (HIs), to delineate clinically pertinent γδ T cell s

INTRODUCTION: Pheochromocytoma is a catecholamine-producing tumor arising from the adrenal medulla. When it coexists with a tumor of different origin within the same adrenal gland, it is classified as a collision tumor involving a pheochromocytoma.
CASE PRESENTATION: The left adrenal tumor was identified in an 84-year-old Japanese woman and initially considered pheochromocytoma; however, rapid growth, lymphadenopathy, reduced 123I-MIBG uptake, and intense FDG (Fluorodeoxyglucose) accumulation raised suspicion of another tumor component. The patient received cyclophosphamide, vincristine and d

Diffuse large B-cell lymphoma (DLBCL) frequently involves the abdomen and retroperitoneum, where it may present as bulky perirenal soft tissue. Spontaneous retroperitoneal hematoma is an increasingly recognized cause of flank pain and anemia, and can produce similar mass-like collections on computed tomography. Distinguishing these entities is crucial, as misclassification can delay potentially curative systemic therapy or expose patients to non-beneficial invasive procedures aimed at hemorrhage control. We describe a 58-year-old man presenting with progressive left flank and lower quadrant pa

Bone marrow necrosis (BMN) is a very rare pathological finding that may arise in the context of numerous underlying conditions. It has been linked to hematologic malignancies, solid tumors, sickle cell disease, various infections, and certain medications. Advances in hematopathology have contributed significantly to the improved identification and diagnostic accuracy of BMN. We report a case of a 75-year-old woman who was evaluated for anemia and fever of unknown origin. Histopathological examination of a bone marrow biopsy revealed widespread necrosis accompanied by infiltration with diffuse

INTRODUCTION: Diffuse large B-cell lymphoma is the most common type of non-Hodgkin lymphoma in Saudi Arabia. Osmotic demyelination syndrome (ODS) is divided into central pontine myelinolysis (CPM) and extrapontine myelinolysis. The occurrence of osmotic demyelination syndrome is classically described in the setting of rapid correction of hyponatremia. Clinical Manifestations are variable, ranging from asymptomatic cases to quadriparesis and locked-in syndrome in the severe form. METHOD: We reported a total of six cases with diffuse large B-cell lymphoma (DLBCL) that developed central pontine m

Diffuse large B-cell lymphoma (DLBCL) is a heterogeneous and prevalent subtype of aggressive non-Hodgkin lymphoma that poses diagnostic and prognostic challenges, particularly in predicting drug responsiveness. In this study, we used digital pathology and deep learning to predict responses to immunochemotherapy in patients with DLBCL. We retrospectively collected 251 slide images from 216 DLBCL patients treated with rituximab, cyclophosphamide, doxorubicin, vincristine, and prednisone (R-CHOP), with their immunochemotherapy response labels. The digital pathology images were processed using con

The most common subtype of lymphoma globally, diffuse large B cell lymphoma (DLBCL), is a leading cause of cancer death in people with HIV. The restructuring of the T cell compartment because of HIV infection and antiretroviral therapy (ART) may have implications for modern treatment selection, but current understanding of these dynamic interactions is limited. Here, we investigated the T cell response to DLBCL by sequencing the T cell receptor (TCR) repertoire in a cohort of HIV-negative (HIV-), HIV+/ART-experienced, and HIV+/ART-naive patients with DLBCL. HIV+/ART-naive tumor TCR repertoires

Glofitamab, an anti-CD20 antibody, is approved as a third-line treatment for relapsed or refractory (r/r) diffuse large-cell B lymphoma (DLBCL), achieving a complete response in nearly 40% of patients. This humanized IgG1 bispecific monoclonal antibody binds to CD20 on malignant B lymphocytes and to CD3 on cytotoxic T cells. This dual binding forms an immunological synapse, activating T lymphocytes and leading to the lysis of tumor cells. Salvage radiotherapy is also effective for r/r DLBCL, but its combination with systemic treatments like glofitamab may increase radiation-induced toxicity. W
