In-utero exposure to maternal HIV infection alters T-cell immune responses to vaccination in HIV-uninfected infants.

OBJECTIVE: In sub-Saharan Africa, HIV-exposed uninfected (HEU) infants have higher morbidity and mortality than HIV-unexposed infants. To evaluate whether immune dysfunction contributes to this vulnerability of HEU infants, we conducted a longitudinal, observational cohort study to assess T-cell immune responses to infant vaccines (Mycobacterium bovis BCG and acellular pertussis) and staphylococcal enterotoxin B (SEB). In total, 46 HEU and 46 HIV-unexposed infants were recruited from Khayelitsha, Cape Town. METHODS: Vaccine-specific T-cell proliferation (Ki67 expression) and intracellular exp

Maternal occupational exposure to organic solvents during early pregnancy and risks of neural tube defects and orofacial clefts.

OBJECTIVES: Though toxicological experiments demonstrate the teratogenicity of organic solvents in animal models, epidemiologic studies have reported inconsistent results. Using data from the population-based National Birth Defects Prevention Study, the authors examined the relation between maternal occupational exposure to aromatic solvents, chlorinated solvents and Stoddard solvent during early pregnancy and neural tube defects (NTDs) and orofacial clefts (OFCs). METHODS: Cases of NTDs (anencephaly, spina bifida and encephalocoele) and OFCs (cleft lip ± cleft palate and cleft palate alone)

Maternal allergy acts synergistically with cigarette smoke exposure during pregnancy to induce hepatic fibrosis in adult male offspring.

Maternal environmental exposures during pregnancy are known to affect disease onset in adult offspring. For example, maternal asthma exacerbations during pregnancy can worsen adult asthma in the offspring. Cigarette smoking during pregnancy is associated with future onset of cardiovascular disease, obesity and diabetes. However, little is known about the effect of maternal environmental exposures on offspring susceptibility to liver disease. This pilot study examined the long-term effect of maternal allergen challenge and/or cigarette smoking during pregnancy on hepatic inflammation and fibros

Adverse birth outcomes and maternal exposure to trichloroethylene and tetrachloroethylene through soil vapor intrusion in New York State.

BACKGROUND: Industrial spills of volatile organic compounds (VOCs) in Endicott, New York (USA), have led to contamination of groundwater, so … We used Poisson regression to analyze births and malformations to estimate the association between maternal exposure to VOCs adjusting for sex, mother's age, race, education, parity, and prenatal care. Two exposure areas were identified based on environmental sampling data: one area was primarily contaminated with TCE, and the other with PCE. RESULTS: In the TCE- …

In utero and lactational exposure to blueberry via maternal diet promotes mammary epithelial differentiation in prepubescent female rats.

Early developmental events influence the fine tuning of later susceptibility to adult diseases. Diet is a determinant of breast cancer risk, and our previous studies showed that diet-mediated changes in transcriptional programs promote early mammary gland differentiation. Although consumption of fruits is considered to elicit multiple health benefits, little is known on whether associated bioactive components modify the early differentiation program in developing mammary glands. Here, we evaluated the hypothesis that early exposure (in utero and lactational) to blueberry through maternal diet

Plasmodium falciparum exposure in utero, maternal age and parity influence the innate activation of foetal antigen presenting cells.

BACKGROUND: Malaria in pregnancy is associated with immunological abnormalities in the newborns, such as hampered T-helper 1 responses and increased T-regulatory responses, while the effect of maternal Plasmodium falciparum infection on foetal innate immunity is still controversial. MATERIALS AND METHODS: The immunophenotype and cytokine release by dendritic cells (DC) and monocytes were evaluated in cord blood from 59 Beninese women with or without malaria infection by using flow cytometry. RESULTS: Accumulation of malaria pigment in placenta was associated with a partial maturation of cord

Maternal adrenocorticotropin, cortisol, and thyroid hormone responses to all three-trimester equivalent repeated binge alcohol exposure: ovine model.

Alcohol-mediated alterations in hypothalamic-pituitary-adrenal (HPA) and hypothalamic-pituitary-thyroid axis function are two proposed mechanisms by which alcohol causes neurodevelopmental injury to the fetus. We previously reported that third-trimester equivalent only alcohol exposure in sheep results in increases in the maternal and fetal adrenocorticotropin and cortisol levels, and decreases in the fetal thyroid hormones T(3) and T(4) and maternal T(3) levels. In this study, we wished to characterize the maternal HPA and hypothalamic-pituitary-thyroid hormone responses to repeated binge alc

Maternal adrenalectomy abrogates the effect of fetal alcohol exposure on the interleukin-1beta-induced febrile response: gender differences.

Fetal alcohol exposure (FAE) has been shown to blunt the febrile component of the primary host-defense response to infection induced experimentally by systemic administration of interleukin (IL)-1beta. Given that maternal adrenalectomy (ADX) can prevent various postnatal effects of FAE, the present experiments were designed to determine whether maternal ADX would prevent the blunted IL-1beta-induced febrile response of fetal alcohol-exposed offspring and whether the effects of maternal ADX would be gender related. Timed-pregnant rats underwent ADX or were sham-operated on gestation day (GD) 7,

Long-term learning deficits and changes in unlearned behaviors following in utero exposure to multiple daily doses of cocaine during different exposure periods and maternal plasma cocaine concentrations.

Although the possible behavioral neurotoxic effects of in utero exposure to cocaine have been the subject of numerous experiments, only a limited number of different types of animal models of cocaine exposure, critical periods, or long-term effects of such exposures have been investigated. In the present experiment, the effects of multiple daily SC exposures to cocaine (20 mg/kg/dose x 5 doses per day) were investigated when administered to gravid Sprague-Dawley CD rats on embryonic days E7-12 or E13-18 compared to weight-matched, vehicle injected, pair-fed controls. Effects of exposure were a

Maternal cyclobenzaprine exposure and risk of birth defects in the National Birth Defects Prevention Study (1997-2011) and Birth Defects Study to Evaluate Pregnancy exposureS (2014-2018).

PURPOSE: Cyclobenzaprine is a muscle relaxant indicated for acute pain. Little is known about cyclobenzaprine's safety during pregnancy. We explored the association between maternal cyclobenzaprine exposure and risk of birth defects among offspring. METHODS: We combined data from two large, multi-site, population-based case-control studies in the United States. Cases were identified from birth defects registries across 10 states; controls were liveborn infants without birth defects randomly selected from the same catchment areas. Participants reported cyclobenzaprine use during the month befo

Serum trace and heavy metal exposure and IVF/ICSI outcomes: modifying effects of maternal diet in a prospective cohort.

BACKGROUND: Environmental exposure to trace and heavy metals may influence female fertility, yet their impact on assisted reproductive outcomes remains poorly defined. Maternal diet can potentially modulate metal-related toxicity by influencing metabolic pathways and oxidative stress. OBJECTIVES: To evaluate the associations between ten serum metal concentrations and IVF/ICSI outcomes, and to investigate whether maternal dietary patterns modify these relationships. METHODS: This prospective cohort study included 396 women undergoing their first IVF/ICSI cycle. Serum levels of calcium (Ca), m

Insights from over a million births on maternal exposure to multiple pollutants and the risk of isolated ventricular septal defects.

Numerous research works have established connections between maternal exposure to specific air pollutants and ventricular septal defects (VSDs), there are still gaps in understanding the complexities of combined exposures in real-world settings. This study seeks to investigate the correlation between simultaneous exposure to five air pollutants throughout pregnancy and the likelihood of isolated VSDs occurrence. It aims to address the gap between research focusing on individual pollutants and the complexities of real-world exposure scenarios. Utilizing data from China's National Population-Bas…

Maternal mycoestrogen exposure in relation to timing of birth and placental corticotropin releasing hormone concentrations in a U.S. pregnancy cohort.

Zearalenone (ZEN) is a mycotoxin that widely contaminates human food supplies. In animal models, exposure to ZEN has extensive impacts on reproduction and pregnancy including contributing to early parturition. Here, we translate that research to humans by examining gestational exposure to ZEN in relation to the timing of birth and placental corticotropin releasing hormone (pCRH), a hormone that regulates the timing of parturition. Utilizing data from a pregnancy cohort (UPSIDE; Rochester, NY; n = 299), we investigated exposure to ZEN and its metabolites in relation to: 1) gestational age at bi

Predicting Maternal and Fetal Exposures of Elexacaftor-Tezacaftor-Ivacaftor during Pregnancy through Physiologically Based Pharmacokinetic Models.

The use of elexacaftor/tezacaftor/ivacaftor (ETI) has been associated with increased fertility in women with cystic fibrosis (CF) and is increasingly used during pregnancy to support both maternal and fetal health. However, little is known about the pharmacokinetics (PK) of ETI during pregnancy, which is crucial for optimizing its efficacy and safety. This study aimed to predict the PK of ETI during pregnancy and to determine the maternal dose required to achieve therapeutic concentrations in both the maternal and fetus. The pregnancy physiological-based pharmacokinetic (PBPK) model within the

Maternal, paternal, and dual-parental alcohol exposures result in both overlapping and distinct impacts on behavior in adolescent offspring.

Emerging research reveals that alcohol use by fathers before conception can affect the growth and development of their offspring. Here, we used a C57BL/6J mouse model to study the effects of alcohol exposure on the behavior of the first-generation (F1) offspring, comparing the impacts of alcohol exposure by mothers, fathers, and both parents. Our goal was to determine how alcohol exposure by each parent or both parents influences the behavior of the offspring. We found that adolescent male offspring of alcohol-exposed fathers showed reduced anxiety-like behaviors as they spent more time in the

Global burden of maternal hypertensive disorders (1990-2045): trends, regional disparities, and causal links to occupational exposures.

BACKGROUND: Maternal Hypertensive Disorders (MHD), encompassing gestational hypertension, chronic hypertension, preeclampsia, and eclampsia, which was a significant contributor to maternal morbidity and mortality, particularly in regions with lower socioeconomic status. METHODS: Using data from the 2021 Global Burden of Disease (GBD) study, we analyzed the burden of MHD globally. We used the slope index and concentration index to measure cross-country inequality in MHD burden, and employed a Bayesian age-period-cohort (BAPC) model to project the burden from 2022 to 2045. Additionally, we cond
