A note on variance estimation of the Aalen-Johansen estimator of the cumulative incidence function in competing risks, with a view towards left-truncated data.

The Aalen-Johansen estimator is the standard nonparametric estimator of the cumulative incidence function in competing risks. Estimating its variance in small samples has attracted some interest recently, together with a critique of the usual martingale-based estimators. We show that the preferred estimator equals a Greenwood-type estimator that has been derived as a recursion formula using counting processes and martingales in a more general multistate framework. We also extend previous simulation studies on estimating the variance of the Aalen-Johansen estimator in small samples to left-trun

A better lemon squeezer? Maximum-likelihood regression with beta-distributed dependent variables.

Uncorrectable skew and heteroscedasticity are among the "lemons" of psychological data, yet many important variables naturally exhibit these properties. For scales with a lower and upper bound, a suitable candidate for models is the beta distribution, which is very flexible and models skew quite well. The authors present maximum-likelihood regression models assuming that the dependent variable is conditionally beta distributed rather than Gaussian. The approach models both means (location) and variances (dispersion) with their own distinct sets of predictors (continuous and/or categorical), th

Sex-specific shape models to disentangle sex effects and age-associated anatomical variation in biventricular cardiac anatomy.

AIMS: Age- and sex-related differences in cardiac anatomy are well established using conventional cardiovascular magnetic resonance biomarkers, but their representation in 3D ventricular shape remains unclear. In pooled statistical shape models, leading modes may be dominated by between-sex anatomical variation, potentially obscuring within-sex age-associated variation. This study evaluated whether sex-specific statistical shape models provide a more interpretable representation of age-associated ventricular variation than a sex-agnostic model. METHODS: Biventricular meshes were reconstructed

Infectious disease forecasting to support public health: use of readily available methods to predict malaria and diarrhoeal diseases in Mozambique.

BACKGROUND: Mozambique faces a high burden of infectious diseases but currently has limited capacity for forecasting disease incidence. Recent improvements in disease surveillance through the National Monitoring and Evaluation System now provide weekly reports of disease incidence across the country's districts. This study focuses on using these records, specifically for malaria and diarrhoeal diseases, which together account for approximately 40% of deaths among children under five, to develop statistical forecasts and evaluate their accuracy. METHODS: We utilised a Python library for time s

Machine learning-based prediction of N2 lymph node metastasis in non-small cell lung cancer.

BACKGROUND: Lung cancer is a leading cause of cancer-related mortality worldwide. Accurate staging of mediastinal lymph nodes is a crucial step in determining appropriate treatment approaches. Current noninvasive diagnostic methods do not provide sufficient accuracy to confidently decide on surgery without histological confirmation. Our study aimed to develop a artificial intelligence model for the precise prediction of N2 lymph node metastasis. METHODS: We retrospectively analyzed 1489 patients who underwent standard cervical mediastinoscopy at our department, including 472 patients diagnose

Combining process-based and correlative models improves predictions of climate change effects on Schistosoma mansoni transmission in eastern Africa.

Currently, two broad types of approach for predicting the impact of climate change on vector-borne diseases can be distinguished: i) empirical-statistical (correlative) approaches that use statistical models of relationships between vector and/or pathogen presence and environmental factors; and ii) process-based (mechanistic) approaches that seek to simulate detailed biological or epidemiological processes that explicitly describe system behavior. Both have advantages and disadvantages, but it is generally acknowledged that both approaches have value in assessing the response of species in gen

Monte Carlo optimization based QSAR modeling of angiotensin II receptor antagonists.

The pathogenesis of essential hypertension, congestive heart failure, and reno-vascular hypertension is related to angiotensin II. This study presents QSAR modeling for a set of compounds acting as angiotensin II receptor antagonists based on the Monte Carlo optimization with molecular graph-based and SMILES notation based descriptors. Conformation independent QSAR models were developed for three random splits. Various statistical approaches were used to assess the statistical quality of the developed models, and the obtained results were very good. This study used a novel statistical metric k

Structure-activity relationships of the antimalarial agent artemisinin. 6. The development of predictive in vitro potency models using CoMFA and HQSAR methodologies.

Artemisinin (1) is a unique sesquiterpene peroxide occurring as a constituent of Artemisia annua L. Because of the effectiveness of Artemisinin in the treatment of drug-resistant Plasmodium falciparum and its rapid clearance of cerebral malaria, development of clinically useful semisynthetic drugs for severe and complicated malaria (artemether, artesunate) was prompt. However, recent reports of fatal neurotoxicity in animals with dihydroartemisinin derivatives such as artemether have spawned a renewed effort to develop nontoxic analogues of artemisinin. In our effort to develop more potent, le

Confidence Intervals for the MADET Index and Its Corresponding Classification Thresholds Using Generalized Pivotal Quantities.

In medical contexts, especially in the diagnosis process of patients into different ordered stages of a disease (e.g., not diseased, early disease, and advanced disease) based on a continuous biomarker, the concept of the receiver operating characteristic (ROC) curve has been extended to that of the ROC surface and, consequently, the corresponding diagnostic performance measures as well. Specifically, the maximum absolute determinant (MADET) has been recently proposed, extending the classic concept of the Youden index. However, no method has been proposed for constructing confidence intervals

Variations of Power Priors in Time-to-Event Analysis With Piecewise Constant Hazards Through Baseline and Outcome Similarity Driven Bounded Weighting.

Patient recruitment in clinical trials, particularly in rare diseases or pediatric populations, remains a significant challenge. Leveraging external data to supplement trial information offers a promising solution, reducing placebo allocation, and enhancing recruitment feasibility while increasing the number of patients receiving active treatment, thus improving safety evaluations. However, the success of this method depends on the alignment of external data with target data from a population characteristics perspective and scientific translation perspective. Generative estimation of conditional survival function.

We propose a deep generative approach to nonparametric estimation of conditional survival and hazard functions with censored data. The key idea of the proposed method is to first learn a conditional generator for the joint conditional distribution of the observed time and censoring indicator given covariates, and then construct the Kaplan-Meier and Nelson-Aalen estimators based on this conditional generator for conditional hazard and survival functions. Our method combines ideas from the recently developed deep generative learning and classical nonparametric estimation in survival analysis. We

Prediction intervals for random-effects meta-analysis based on confidence distributions and Edgington's method.

Statistical inference about the average effect in random-effects meta-analysis has been considered insufficient in the presence of substantial between-study heterogeneity. Predictive distributions are well suited for quantifying heterogeneity since they are interpretable on the effect scale and provide clinically relevant information about future events. We construct predictive distributions accounting for uncertainty through confidence distributions from Edgington's p-value combination method and the generalized heterogeneity statistic. Simulation results suggest that 95% prediction intervals

Density and treatment effect estimation under covariate-adaptive randomization with heavy-tailed outcomes.

Randomized experiments are the gold standard for investigating causal relationships, with comparisons of potential outcomes under different treatment groups used to estimate treatment effects. However, outcomes with heavy-tailed distributions pose significant challenges to traditional causal inference approaches. While recent studies have explored these issues under simple randomization, their application in more complex randomization designs, such as stratified randomization or covariate-adaptive randomization, has not been adequately addressed. To fill the gap, we first investigate the perfo

Integration of aggregate data in causally interpretable meta-analysis by inverse weighting.

Obtaining causally interpretable meta-analysis results is challenging when there are differences in the distribution of effect modifiers between eligible trials. To overcome this, recent work on transportability methods has considered standardizing results of individual studies over the case-mix of a target population, prior to pooling them as in a classical random-effect meta-analysis. One practical challenge, however, is that case-mix standardization often requires individual participant data (IPD) on outcome, treatments and case-mix characteristics to be fully accessible in every eligible s

The impact of coarsening an exposure on partial identifiability in instrumental variable settings.

In instrumental variable (IV) settings, such as imperfect randomized trials and observational studies with Mendelian randomization, one may encounter a continuous exposure, the causal effect of which is not of true interest. Instead, scientific interest may lie in a coarsened version of this exposure. Although there is a lengthy literature on the impact of coarsening of an exposure with several works focusing specifically on IV settings, all methods proposed in this literature require parametric assumptions. Instead, just as in the standard IV setting, one can consider partial identification v

When does adjusting covariate under randomization help? A comparative study on current practices.

PURPOSE: We aim to thoroughly compare past and current methods that leverage baseline covariate information to estimate the average treatment effect (ATE) using data from of randomized clinical trials (RCTs). We especially focus on their performance, efficiency gain, and power. METHODS: We compared 6 different methods using extensive Monte-Carlo simulation studies: the unadjusted estimator, i.e., analysis of variance (ANOVA), the analysis of covariance (ANCOVA), the analysis of heterogeneous covariance (ANHECOVA), the inverse probability weighting (IPW), the augmented inverse probability weig

An overview of Bayesian reasoning in the analysis of delay-discounting data.

Statistical inference (including interval estimation and model selection) is increasingly used in the analysis of behavioral data. As with many other fields, statistical approaches for these analyses traditionally use classical (i.e., frequentist) methods. Interpreting classical intervals and p-values correctly can be burdensome and counterintuitive. By contrast, Bayesian methods treat data, parameters, and hypotheses as random quantities and use rules of conditional probability to produce direct probabilistic statements about models and parameters given observed study data. In this work, we r

A nonparametric maximum likelihood approach for survival data with observed cured subjects, left truncation and right-censoring.

We consider observational studies in pregnancy where the outcome of interest is spontaneous abortion (SAB). This at first sight is a binary 'yes' or 'no' variable, albeit there is left truncation as well as right-censoring in the data. Women who do not experience SAB by gestational week 20 are 'cured' from SAB by definition, that is, they are no longer at risk. Our data is different from the common cure data in the literature, where the cured subjects are always right-censored and not actually observed to be cured. We consider a commonly used cure rate model, with the likelihood function tailo
