HDC/histamine Signaling Axis Drives Macrophage Reprogramming to Promote Angiogenesis in Hindlimb-Ischemic Mice.

Histamine is catalyzed by histidine decarboxylase (HDC), which plays important roles in many physiological and pathological processes, but its role in angiogenesis has not been thoroughly clarified. Here we report that HDC is highly expressed in Ly6C+ macrophages, rather than in endothelial cells using Hdc-GFP transgenic mice with hindlimb ischemia (HLI) mouse model. Given the whole-process promoting effect of macrophages on angiogenesis, a cluster of HDC+CXCR2+ macrophages have been identified by single-cell sequencing technology in ischemic tissue. The inactivation of HDC leads to a lack of

Newly-engineered angiopoietin-1 as a cell-priming agent for CVD.

Human peripheral blood stem cells (PBSC) that have been most frequently used for repair or regeneration in ischemic cardiovascular disease (CVD) showed limitations in their efficacy. We previously reported that angiopoietin-1 (Ang1) is the cell-priming agent to enhance the vasculogenic potential of PBSC. The limitation was the difficulty to produce Ang1 protein with high efficiency. In this study, we engineered Ang1 structure and made FVA3-Ang1 by adding VASP and COMP sequence for stable tetramer formation as well as FLAG sequence for purification in large scale production and a signal peptide

A novel subcutaneous islet transplantation method using a bioabsorbable medical device to facilitate the creation of a highly vascularized transplantation site.

Subcutaneous transplantation is garnering attention as a potential transplantation site for pluripotent stem cell-derived islet cells to address the shortage of pancreatic islet transplant donors. However, subcutaneous transplantation of cells presents challenges related to angiogenesis, which is necessary for successful islet bioproduction. This study aimed to investigate a novel method for enhancing vascularization at the transplant site and thereby promote islet engraftment using a clinically available bioabsorbable medical device. A nonabsorbable device (agarose) or a bioabsorbable device

S-nitrosoglutathione releasing nano-micron combination hydrogel enhances cutaneous wound healing via promoting angiogenesis and collagen deposition.

Nitric oxide (NO) is essential for wound healing, promoting angiogenesis and collagen deposition. This study investigates a novel dual-matrix nanocomposite hydrogel incorporating S-nitrosoglutathione (GSNO), a physiological NO donor, to enhance cutaneous wound healing. GSNO was encapsulated in ammonio methacrylate copolymer nanoparticles and embedded in an alginate-based matrix, achieving controlled NO release. GSNO-loaded nanoparticles were prepared using solvent displacement and solvent evaporation methods, resulting in spherical, well-distributed and positively charged particles. These nano

Silver Nanoparticles at Low Concentrations Embedded in ECM Promote Endothelial Monolayer Formation and Cell Migration.

Several scientific studies have reported the opposing effects of silver nanoparticles (AgNPs) on angiogenesis, ranging from proangiogenic to anti-angiogenic. The widespread use of AgNPs in biomedical applications and the variability of their effects depending on concentration and exposure conditions highlight the need for further research into their impact on vascularization and endothelial cell behavior. This study aimed to investigate the potential influence of AgNPs on human umbilical vein endothelial cells (HUVECs) using a model incorporating a thin layer of an extracellular matrix (ECM).

Semaphorin 3E-Plexin-D1 Pathway Downstream of the Luteinizing Hormone Surge Regulates Ovulation, Granulosa Cell Luteinization, and Ovarian Angiogenesis in Mice.

Ovulation is induced by the luteinizing hormone (LH) surge and accompanied by granulosa cell luteinization and ovarian angiogenesis. Semaphorin 3E (Sema3E)-Plexin-D1 pathway regulates angiogenesis in other tissues, but its role in the ovary is unknown. Evidence indicates that Sema3E-Plexin-D1 pathway plays an important role in the mouse ovary. The expression of Sema3E and its receptor, Plexin-D1, is dynamically regulated in the mouse ovary downstream of the LH surge. This regulation requires the modulation of chromatin accessibility by CCAAT/enhancer-binding proteins α and β. Intraovarian inje

Stromal cell-derived factor-encapsulated nanoparticles target ischemic myocardium and attenuate myocardial injury via proangiogenic effects.

Lipid bilayer nanoparticles (NPs) with and without stromal cell-derived factor (SDF) were created to target and treat ischemia/reperfusion (I/R)-injured myocardium. Male Wistar rats were subjected to myocardial I/R insult and, at reperfusion, randomized to receive systemic injections of 5 mL/kg PBS, 6 μg/kg of NPs, SDF, or SDF-NPs. Four days after treatment, SDF-NPs circulated and accumulated selectively in the ischemic myocardium, with an SDF concentration roughly three times that of the other three treatments. SDF-NP-treated rats had more endothelial progenitor cells (EPCs) in the blood and

Function of orthologous fibroblast growth factor 11 protein in angiogenesis and immunomodulatory after spinal cord injury.

Intracellular fibroblast growth factors (iFGFs) are emerging as promising therapeutic agents in tissue engineering and regenerative medicine. Although FGF11, a typical iFGF, plays crucial roles in cellular hypoxia adaptation, its roles in angiogenesis and immunomodulation post-spinal cord injury (SCI) remain poorly characterized. This study investigated four FGF11 orthologs: ZFgf11a/ZFgf11b from zebrafish (Danio rerio) and MFGF11a/MFGF11b from mouse (Mus musculus). In vitro, intracellularly delivered ZFgf11a and MFGF11b exhibited stronger pro-angiogenic effects compared to ZFgf11b or MFGF11a.

A novel vascularized urethra-on-a-chip model.

The male urethra transports urine and semen. Any disease of the male urethra, hindering normal voiding or ejaculation, has a major impact on quality of life. Urethral stricture disease is common and molecular research into urethral strictures is hampered by the lack of reliable models of the human urethra. The aim of this project is to develop an in vitro model system of the human urethra. We hypothesized that by using the organ-on-a-chip technology we would be able to recapitulate physiology, functionality and the biomechanical cues of the native urethra and its surrounding vascular bed. Our

GLP-1 Receptor Agonists as Emerging Modulators of Inflammation and Angiogenesis in Chronic Cutaneous Wound Healing.

Diabetic foot ulcers (DFUs) represent a clinically burdensome and pathophysiologically distinct complication of diabetes mellitus, marked by persistent inflammation, dysregulated immune signaling, impaired angiogenesis, and delayed re-epithelialization. Despite adherence to standardized wound care protocols, DFUs remain recalcitrant to healing, contributing to high rates of recurrence, infection, and lower extremity amputation. Although established in glycemic control, glucagon-like peptide-1 receptor agonists (GLP-1 RAs) exert pleiotropic immunomodulatory and regenerative effects relevant to

Angiogenic Ability of Extracellular Vesicles Derived from Angio-miRNA-Modified Mesenchymal Stromal Cells.

BACKGROUND: Regenerative therapy using extracellular vesicles (EVs) is a promising approach for the supportive treatment of chronic limb-threatening ischaemia. Herein, we examined the angiogenic potential of EVs derived from genetically modified mesenchymal stromal cells (MSCs), focusing on the angio-micro RNAs (miRNAs) in EVs. METHODS: Bone marrow-derived MSCs (BM-MSCs) were transfected with lentiviral vectors containing specific angio-miRNAs (miRNA-126, -135b, or -210), and miRNA overexpression was confirmed using quantitative polymerase chain reaction (qPCR). EVs were isolated from the BM-

Exploring the Changes in IL-6 and Related Cytokines in Angiogenesis after Tibial Transverse Transplantation in Diabetic Foot Ulcers.

OBJECTIVE: The transverse tibial transfer technique is employed primarily to treat diabetic foot ulcers (DFUs), aiming to enhance leg circulation and promote new blood vessel growth. This technique is also beneficial for various conditions associated with poor blood flow in the lower extremities. However, there is no clear molecular mechanism to explain the relationship between the transverse tibial transfer technique and angiogenesis in patients with diabetic foot. This study aims to preliminarily explore the change of IL-6 and related cytokines in promoting angiogenesis during transverse tib

Adipose-Derived Mesenchymal Stem Cells Protect Endothelial Cells from Hypoxic Injury by Suppressing Terminal UPR In Vivo and In Vitro.

Adipose-derived stem cells (ASCs) have been used as a therapeutic intervention for peripheral artery disease (PAD) in clinical trials. To further explore the therapeutic mechanism of these mesenchymal multipotent stromal/stem cells in PAD, this study was designed to test the effect of xenogeneic ASCs extracted from human adipose tissue on hypoxic endothelial cells (ECs) and terminal unfolded protein response (UPR) in vitro and in an atherosclerosis-prone apolipoprotein E-deficient mice (ApoE-/- mice) hindlimb ischemia model in vivo. ASCs were added to Cobalt (II) chloride-treated ECs; then, me

Spatiotemporal Regulation of Injectable Heterogeneous Silk Gel Scaffolds for Accelerating Guided Vertebral Repair.

Osteoporotic vertebral fracture is jeopardizing the health of the aged population around the world, while the hypoxia microenvironment and oxidative damage of bone defect make it difficult to perform effective tissue regeneration. The balance of oxidative stress and the coupling of vessel and bone ingrowth are critical for bone regeneration. In this study, an injectable heterogeneous silk gel scaffold which can spatiotemporally and sustainedly release bone mesenchymal stem cell-derived small extracellular vesicles, HIF-1α pathway activator, and inhibitor is developed for bone repair and verteb

The angiogenic neuropeptide catestatin exerts beneficial effects on human coronary vascular cells and cardiomyocytes.

INTRODUCTION: Myocardial infarction (MI) induces irreversible tissue damage, eventually leading to heart failure. Exogenous induction of angiogenesis positively influences ventricular remodeling after MI. Recently, we could show that therapeutic angiogenesis by the neuropeptide catestatin (CST) restores perfusion in the mouse hind limb ischemia model by the induction of angio-, arterio- and vasculogenesis. Thus, we assumed that CST might exert beneficial effects on cardiac cells. METHODS/RESULTS: To test the effect of CST on cardiac angiogenesis in-vitro matrigel assays with human coronary ar

Selective ROCK Inhibitor Enhances Blood Flow Recovery after Hindlimb Ischemia.

The impairment in microvascular network formation could delay the restoration of blood flow after acute limb ischemia. A high-content screen of a GSK-published kinase inhibitor library identified a set of ROCK inhibitor hits enhancing endothelial network formation. Subsequent kinase activity profiling against a panel of 224 protein kinases showed that two indazole-based ROCK inhibitor hits exhibited high selectivity for ROCK1 and ROCK2 isoforms compared to other ROCK inhibitors. One of the chemical entities, GSK429286, was selected for follow-up studies. We found that GSK429286 was ten times m

Deficiency of lncRNA SNHG12 impairs ischemic limb neovascularization by altering an endothelial cell cycle pathway.

SNHG12, a long noncoding RNA (lncRNA) dysregulated in atherosclerosis, is known to be a key regulator of vascular senescence in endothelial cells (ECs). However, its role in angiogenesis and peripheral artery disease has not been elucidated. Hind-limb ischemia studies using femoral artery ligation (FAL) in mice showed that SNHG12 expression falls readily in the acute phase of the response to limb ischemia in gastrocnemius muscle and recovers to normal when blood flow recovery is restored to ischemic muscle, indicating that it likely plays a role in the angiogenic response to ischemia.
