CUBIC pathology: three-dimensional imaging for pathological diagnosis.

The examination of hematoxylin and eosin (H&E)-stained tissues on glass slides by conventional light microscopy is the foundation for histopathological diagnosis. However, this conventional method has some limitations in x-y axes due to its relatively narrow range of observation area and in z-axis due to its two-dimensionality. In this study, we applied a CUBIC pipeline, which is the most powerful tissue-clearing and three-dimensional (3D)-imaging technique, to clinical pathology. CUBIC was applicable to 3D imaging of both normal and abnormal patient-derived, human lung and lymph node tissues.…

Growth patterns: Pathology vs. Normal variation.

Growth of children and adolescents follows a universal pattern with three distinct growth spurts. Common medical practice refers to normal child growth as a health and biological advantage, and considers "pathology in growth" as a deviation from the universal pattern. Growth can differ in amplitude and tempo with considerable variation both within and between populations over time, and depending on geographic region. Local growth charts are necessary to adapt the universal pattern of growth to characteristics of a specific historic or modern population. Local charts can visualize the distinct …

Pathology of adrenal tumours: recent advances.

Changes in the nomenclature and classification of adrenal gland diseases are the result of advances in understanding the pathogenesis, germline susceptibility and the clonal-neoplastic nature of diseases of the adrenal gland. Although numerous classification systems have been proposed, the Weiss system remains the standard for distinguishing benign from malignant adult adrenal cortical tumours, but the Helsinki system and the reticulin algorithm are proving to be increasingly useful in difficult cases. Subtypes of adrenal cortical neoplasms, such as myxoid and oncocytic, as well as those occur

Molecular pathology of adenoid cystic carcinoma.

Adenoid cystic carcinoma (ACC) is a slow-growing but locally aggressive salivary gland tumor. ACC is composed of ductal/tubular epithelial cells and basal/myoepithelial cells, which form cribriform, tubular, and solid growth patterns in variable combinations and dominance. ACC from different anatomic sites have similar morphological, molecular, and genetic changes. The key molecular alteration in ACC is chromosomal fusion/rearrangement/translocation involving MYB or MYBL1, usually with NFIB as a fusion partner. In this review, we summarize the pathology and molecular alterations in ACC and the…

Pathology of ruminal acidosis in cattle.

Rumen acidosis (RA) is one of the most prevalent alimentary diseases of ruminants and is caused by excessive fermentation of feedstuff in the reticulorumen. This retrospective study aimed to characterize the microscopic lesions of acute and subacute cases of RA in cattle older than 6 months. Fifty-two RA cases with rumen fluid pH ≤5.5 at the time of necropsy were selected. Cases were classified as acute or subacute RA based on their clinical histories; 7 animals were included as controls. In rumen histologic sections, the number, length, and width of ruminal papillae and the thickness of the m

Incidence and Pathology of Aortic Regurgitation.

Aortic regurgitation (AR) is the third most common valvular heart disease, with its prevalence and severity increasing with age. AR can arise from degenerative, congenital, inflammatory and infectious aetiologies, manifesting as primary AR with intrinsic leaflet disease, secondary AR due to aortopathy or annular dilatation, or a combination. Furthermore, AR can be acute, as observed in endocarditis and aortic dissection, or chronic, as seen in calcific aortic valve disease, connective tissue disorders, or bicuspid valve phenotypes. This review discusses the aetiopathology of these various AR m…

Surgical Pathology of Primary Intestinal Myopathy.

CONTEXT.—: Pathologic evaluation of intestinal biopsies or resection specimens is often part of the diagnostic workup for patients with pseudo-obstruction or other forms of severe intestinal dysmotility. Some of these patients have one of several types of primary intestinal myopathy, but the pathologic features that identify and/or distinguish these conditions have been incompletely defined and need to be readdressed in the context of newly recognized genetic etiologies. OBJECTIVE.—: To convey a practical approach to surgical pathology diagnosis of primary intestinal myopathy based on a compr…

The comparative pathology workbench: An update.

The Comparative Pathology Workbench (CPW) is a web-browser-based visual analytics platform providing shared access to an interactive "spreadsheet" style presentation of image data and associated analysis data. The software was developed to enable pathologists and other clinical and research users to compare histopathological images of diseased and/or normal tissues between different samples of the same or different patients/species. The CPW provides a grid layout of cells in rows and columns so that images that correspond to matching data can be organized in the form of an image-enabled "sprea…

Xanthogranulomatous oophoritis: malignant appearance, benign pathology.

Xanthogranulomatous oophoritis (XO) is a rare inflammatory condition of the female genital tract that closely mimics malignancy due to its non-specific clinical presentation and mass-like appearance on imaging. It is histologically characterised by lipid-laden macrophages, chronic inflammatory cells and necrosis. We report the case of a woman in the late 60s referred to the gynaecologic oncology clinic for evaluation of a newly identified complex ovarian mass on CT. She underwent surgical intervention with histopathological examination confirming the diagnosis of XO with extension into the ute

Endothelium glutamate receptors in brain pathology.

The endothelium in brain microcirculation functions not only as a barrier but also as a signal transduction component within a system that regulates multiple vascular processes, including muscle tone, permeability, and structural integrity. The control of local blood flow is vital to ensure adequate oxygen and nutrient supply, efficient removal of catabolic waste, and the maintenance of proper brain cell function. The role of endothelial glutamate receptors in brain pathology is an emerging area of research, particularly important for understanding how these receptors contribute to neurologica…

Molecular Pathology of Mature Lymphoid Malignancies.

Lymphoid malignancies are a broad and heterogeneous group of neoplasms. In the past decade, the genetic landscape of these tumors has been explored and cataloged in fine detail offering a glimpse into the mechanisms of lymphomagenesis and new opportunities to translate these findings into patient management. A myriad of studies have demonstrated both distinctive and overlapping molecular and chromosomal abnormalities that have influenced the diagnosis and classification of lymphoma, disease prognosis, and treatment selection.

Adrenal Ganglioneuroma With Radiology-Pathology Correlation.

This report presents the case of a 62-year-old male with an incidentally discovered adrenal mass, ultimately diagnosed as a ganglioneuroma after laparoscopic adrenalectomy. Imaging findings, of a 9 cm lobulated mass with heterogeneous enhancement on MRI, were unspecific and malignancy could not be excluded. Histological examination of the specimen revealed a well-demarcated tumor composed of Schwann cells and ganglion cells, confirming the diagnosis. Postoperative follow-up showed no recurrence. This case highlights the critical role of histopathological evaluation for definitive diagnosis, as

Pathology of Gestational Trophoblastic Disease (GTD).

Gestational trophoblastic disease (GTD), comprising hydatidiform moles (HM) and gestational trophoblastic tumors (GTT), is extremely rare. HM originate from villous trophoblast and are considered preneoplastic. GTT originate from the intermediate, largely extravillous trophoblast and includes choriocarcinoma, placental site trophoblastic tumor, epitheloid trophoblastic tumor, and mixed trophoblastic tumor. The abnormal (non-molar) villous lesions, non-malignant tumour-like conditions, and non-gestational tumors add to the diagnostic dilemma. The correct diagnosis and classification of these ra

Betrayal Trauma and Histrionic Personality Pathology.

Histrionic personality disorder is among the most common forms of personality pathology, and recent research has focused on its associations with trauma. This research has focused primarily on the types of traumatic experiences people endure, highlighting the role of sexual trauma in particular. However, recent research on the association between traumatic events and histrionic personality characteristics has not taken into account the role of traumatic experiences characterized by interpersonal betrayal, which research suggests is associated with personality pathology in general. In this stud…
