Phylogenetic insights into Leishmania species circulating among humans, vectors, and reservoir hosts in Shiraz, Southern Iran: implications for leishmaniasis control.

Leishmaniasis remains a significant public health challenge in southern Iran, particularly in the Fars province, where Shiraz is a major focus of the disease. This study aimed to elucidate the phylogenetic relationships of Leishmania species isolated from humans, sand flies, and reservoir hosts in Shiraz and its suburbs. Cutaneous slit biopsies were collected from 350 suspected human cases of cutaneous leishmaniasis (CL), and samples were also obtained from rodents (Tatera indica) and sand flies (Phlebotomus papatasi). Parasites were cultured, and DNA was extracted for PCR amplification of the…

[Investigation and analysis of an outbreak of cutaneous leishmaniasis in Ksar Ouled Dabbab, Tataouine (Tunisia), 2012-2013].

UNLABELLED: Zoonotic cutaneous leishmaniasis (ZCL), due to Leishmania major, and chronic CL, due to L. tropica, are endemic in the governora … s (71.3%) indicated ZCL. This hypothesis is supported by the high proportions of L. major (81%) and Phlebotomus papatasi found. CONCLUSION: The epidemiologic and clinical profiles of the cases and the predominance of L. major and P. papatasi confirm that the recent outbreak in KOD was related to ZCL. It probably followed recent environmental changes and tran …

Toscana virus NSs protein promotes degradation of double-stranded RNA-dependent protein kinase.

Toscana virus (TOSV), which is transmitted by Phlebotomus spp. sandflies, is a major etiologic agent of aseptic meningitis and encephalitis in the Mediterranean. Like other members of the genus Phlebovirus of the family Bunyaviridae, TOSV encodes a nonstructural protein (NSs) in its small RNA segment. Although the NSs of Rift Valley fever virus (RVFV) has been identified as an important virulence factor, which suppresses host general transcription, inhibits transcription from the beta interferon promoter, and promotes the proteasomal degradation of double-stranded RNA-dependent protein kinase …

Sandflies (Diptera: Psychodidae) associated with epidemic cutaneous leishmaniasis in Sanliurfa, Turkey.

As part of a project to study the possible impact of environmental change on health in southeastern Turkey, we evaluated sandfly species diversity, abundance, and habitat associations in an urban area where cutaneous leishmaniasis was undergoing epidemic re-emergence. Houses and caves in and around the city of Sanliurfa, Turkey, were sampled using mechanical aspirators, sticky papers, and CDC light traps. Of 1,649 sandflies captured, including 6 Phlebotomus and 1 Sergentomyia species, nearly all were P. papatasi (Scopoli) (967) or P. sergenti Parrot (674). Sandflies were active during June-Sep…

Cutaneous Leishmaniasis in High-Altitude: A Case Report From Nepal.

Leishmaniasis is a parasitic infection caused by Leishmania and spread by the bite of a sandfly, most commonly Phlebotomus. Among the three subtypes of Leishmaniasis, cutaneous leishmaniasis is the most common form. Visceral Leishmaniasis is common in Nepal whereas few cases of cutaneous leishmaniasis have been reported in migrant Nepalese population. The most common lesion seen in cutaneous leishmaniasis is papulo-nodular although other morphological lesions can also be seen. A 30 years female currently residing in Humla presented with a history of multiple non-tender nodules with central nec…

Evaluation of a metofluthrin fan vaporizer device against phlebotomine sand flies (Diptera: Psychodidae) in a cutaneous leishmaniasis focus in the Judean Desert, Israel.

The OFF! Clip-On fan vaporizer device releasing metofluthrin was evaluated against phletobomine sand flies in the Judean Desert, Israel, in October, 2009. A total of 76,400 sand flies was collected, with male flies representing 98.3%Phlebotomus sergenti and 1.7%P. papatasi. Females comprised 43.0% of the total catch and included 6.7% blood-fed females. Similar proportions of flies were collected in both suction and sticky traps. In trials with unbaited suction traps, similar numbers of sand flies were collected in traps with a metofluthrin device, blank device, or no device (i.e., suction only…

A century of leishmaniasis in Alpes-Maritimes, France.

A century of publications on leishmaniasis in Alpes-Maritimes, in southern France, is here reviewed. Autochtonous human and canine leishmaniasis were first recognised in this département, which lies by the Mediterranean Sea and near the Italian border, in 1918 and 1925, respectively. The parasite responsible for the leishmaniasis, Leishmania infantum, is transmitted by Phlebotomus perniciosus and P. ariasi. The human leishmaniasis is zoonotic, with domestic dogs acting as the main 'reservoir' hosts. In prospective surveys over the last two decades, a mean of 12% of the domestic dogs checked in…

Assessing leishmaniasis risk in irrigated lands in Central Tunisia using visible-spectrum remote sensing data.

The expansion of irrigation in central Tunisia has reshaped landscapes, creating ecological conditions favourable to Phlebotomus sand flies and Leishmania transmission, thereby increasing leishmaniasis risk. To assess these dynamics, a geospatial classification and risk-indexing framework was developed that integrates remote sensing, supervised machine learning, and ecological reasoning on vector habitats. The analysis covered three Central Tunisian sites and their narrower environments - Gafsa, Kairouan, and Sidi Bouzid - from 1984 to 2020. Habitat clustering revealed marked spatial and tempo…

A sand fly salivary protein vaccine shows efficacy against vector-transmitted cutaneous leishmaniasis in nonhuman primates.

Currently, there are no commercially available human vaccines against leishmaniasis. In rodents, cellular immunity to salivary proteins of sand fly vectors is associated to protection against leishmaniasis, making them worthy targets for further exploration as vaccines. We demonstrate that nonhuman primates (NHP) exposed to Phlebotomus duboscqi uninfected sand fly bites or immunized with salivary protein PdSP15 are protected against cutaneous leishmaniasis initiated by infected bites. Uninfected sand fly-exposed and 7 of 10 PdSP15-immunized rhesus macaques displayed a significant reduction in …

Diagnosis & management of leishmania/HIV co-infection.

Leishmaniasis, a globally prevalent parasitic disease occurs in three forms viz., visceral, cutaneous and mucocutaneous, transmitted by the bite of infected female Phlebotomus sandflies. Visceral leishmaniasis (VL) has 100 per cent fatality rate, if left untreated. India has the largest burden of this disease. HIV infection is also increasing worldwide and several reports indicate rising trend of VL/ HIV co-infection, modifying the traditional anthroponotic pattern of VL transmission. Both VL and HIV tend to lower the cell mediated immunity (CMI) resulting in poor drug response and opportunist…

Zoonotic cutaneous leishmaniasis caused by Leishmania major: Do humans play a role in amplifying transmission?

Zoonotic cutaneous leishmaniasis (ZCL) caused by Leishmania (L.) major is the most widespread form of CL worldwide, with hundreds of thousands of cases annually. It is associated with significant morbidity and economic burden, particularly in North Africa and the Middle East. For over a century, the zoonotic transmission of ZCL, involving wild gerbils (Muridae: Gerbillinae) as the reservoir and Phlebotomus (Ph.) papatasi as the vector, has been considered an established fact, with no contradictory debate. However, the pronounced endophilic and anthropophilic behaviors of Ph. papatasi, the expo…

Sex-structured disease transmission model and control mechanisms for visceral leishmaniasis (VL).

BACKGROUND: Leishmaniasis are a group of diseases caused by more than 20 species of the protozoan that are transmitted through the bite of female sand fly. The disease is endemic to 98 countries of the world. It affects most commonly the poorest of the poor and mainly males. Several research has been conducted to propose disease control strategies. Effective medical care, vector control, environmental hygiene, and personal protection are the mainstays of the current preventative and control methods. The mathematical models for the transmission dynamics of the disease studied so far did not con
