Sunlight-induced immunosuppression in humans is initially because of UVB, then UVA, followed by interactive effects.

Solar-simulated ultraviolet radiation (ssUV) suppresses immunity in humans. The ultraviolet B (UVB) waveband is recognized as immunosuppressive; however the relative significance of UVA to ssUV immunosuppression is unknown. We created dose and time-response curves for UVB-, UVA-, and ssUV-induced suppression of memory immunity to nickel in humans. UVB caused immunosuppression within 24 h. UVA immunosuppression required 48 h and was normalized by 72 h. UVB alone accounts for ssUV immunosuppression at 24 h, but both UVB and UVA contributed at 48 h. By 72 h neither waveband accounted for ssUV imm

Effect of DMSO on radiation-induced chromosome aberrations analysed by FISH.

The purpose of the present work was to determine if the described reduction in the frequency of radiation-induced chromosome aberrations by DMSO is homogeneous within different human chromosomes. Blood samples were irradiated with 4 Gy of X-rays in absence and presence of 0.5 M DMSO. FISH painting was carried out independently for human chromosomes 1, 2, 3, 4, 7, 11 and 12. The observed frequencies of apparently simple translocations and dicentrics for all these chromosomes, showed a homogeneous reduction when the irradiation was done in the presence of DMSO. Moreover, a better fit between the

[The relation of the characteristics of DNA replication to cell sensitivity to ionizing radiation in xeroderma pigmentosum in form II (XP2CP)].

Using DNA fiber autoradiography, DNA replication in cultured fibroblasts, derived from normal donors, and from XPII patient with increased sensitivity to ionizing radiation was estimated. Here evidence is provided on the fact that the fork movement, significantly decreased in XPII cells before irradiation, remains the same after exposure to X-rays. The density of replicon clusters simultaneously operating in tandem groups, which is initially much less in XPII cells compared to normal cells, also remains unchanged after exposure to X-rays (5 Gy), since the inhibition of DNA replication occurs t

The study of post-irradiation recovery of yeast: the 'premolecular period'.

Recovery of yeast cells after exposure to ionizing radiation was discovered in 1957. During the following decade, i.e., in the 'premolecular period' of studying this phenomenon, its basic features including dependence on cell ploidy, cellular energy exchange and radiation linear energy transfer (LET) were determined and a mathematical model of recovery was proposed. In addition, DNA double-strand breaks were shown to be the damage causing death of irradiated haploid and diploid cells. The results of these investigations led to the formulation of the concept that the ability of a cell to repair

Speculations on the ataxia-telangiectasia defect.

Ataxia-telangiectasia (A-T) is inherited as an monogenetic autosomal recessive disease. Ataxia appears around 1 year of age and progresses until the patient becomes wheelchair-bound, usually by age 10. This progress correlates with deterioration of Purkinje cells in the cerebellum. Sinopulmonary infections are common in patients from some countries but not others. One-third of the patients develop a neoplasm, usually lymphoid, sometime during their shortened lives. Conventional doses of radiation therapy for such cancers are contraindicated since A-T patients are hypersensitive to ionizing rad

Comparative human cellular radiosensitivity: II. The survival following gamma-irradiation of unstimulated (G0) T-lymphocytes, T-lymphocyte lines, lymphoblastoid cell lines and fibroblasts from normal donors, from ataxia-telangiectasia patients and from ataxia-telangiectasia heterozygotes.

We have measured clonal survival following gamma-irradiation of unstimulated (G0) T-lymphocytes from 35 donors, of 11 T-lymphocyte cell lines, of six lymphoblastoid cell lines, and of nine primary fibroblast strains for which we have G0 T-lymphocyte material from the same donor. Amongst the G0 lymphocytes we have results from nine normal donors, from eight cord bloods, from seven ataxia-telangiectasia (A-T) patients and from nine A-T heterozygotes. Although there is some variation between samples, G0 T-lymphocytes from normal donors appear to be slightly more radioresistant than T-lymphocyte l

A study of heat and radiation response of a malignant, melanin-producing cell line derived from C3H 10T1/2 cells transformed in culture by radiation.

The mouse C3H 10T1/2 cell line was transformed to the malignant state using ionizing radiation. One of the transformed lines (R25) that was isolated, displayed some properties similar to malignant melanoma cells. The cells became dark and pigmented after prolonged time in culture and this cell line produced tumors in C3H mice. The radiation survival curve of R25 had a large shoulder which was also observed for human melanoma cell lines. R25 was more resistant to heating at 45.0 degrees C than the normal cell line. Heating at 45.0 degrees C before irradiation resulted in a reduction of the surv

Effect of hypothermia on cell kinetics and response to hyperthermia and X rays.

Hyperthermia is a potent radio enhancer. Studies using hypothermia in combination with irradiation have given confusing results due to lack of uniformity in experimental design. This report shows that hypothermia might have potential significance in the treatment of malignant cells with both thermo- and radiotherapy. Reuber H35 hepatoma cells, clone KRC-7 were used to study the effect of hypothermia on cell kinetics and subsequent response to hyperthermia and/or X rays. Cells were incubated at 8.5 degrees C or between 25 and 37 degrees C for 24 hr prior to hyperthermia or irradiation. Hypother

An appraisal of the role of radiation therapy in the management of nonseminomatous germ-cell tumors of the testis in the era of effective chemotherapy.

Patients with nonseminomatous germ-cell tumors of the testis can be divided into two broad groups. The first includes patients with negative lymphograms or small-volume metastases confined to the abdominal nodes. The overall cure rate with orchiectomy and nodal irradiation is 80%. A policy of early detection of relapse and treatment with chemotherapy is advocated. Adopting this approach, no deaths have occurred in this group of patients treated in 1976 and 1977, and only one (associated with acute myeloblastic leukemia) occurred in 1975. The second group consists of patients with other stage c

Isolation of UV-sensitive variants of human FL cells by a viral suicide method.

A new method (viral suicide method) for the isolation of UV-sensitive mutants is described. Colonies of mutagenized human FL cells were infected with UV-irradiated Herpes simplex viruses and surviving ones which seemed to be deficient in host cell reactivation (HCR) were examined for their UV sensitivity. Nineteen of 238 clones examined were sensitive to UV irradiation at the time of the isolation. After recloning, four of these clones have been studied and two (UVS-1 and UVS-2) of them are stable in their UV sensitivity for 4 months in culture. UV sensitivity of UVS-1, UVS-2, and the parental

[Antioxidative activity of breast tissue in fibroadenoma, fibrocystic mastopathy and cancer].

The data obtained indicate that AOA of a tumor and adjacent normal tissues is reasonably changed as the tumor grows. The character of these changes is analogous to the AOA changes, observed while inducing experimental tumors in animals, which evidence some common physico-chemical mechanisms of tumors occurrence and growth in animals and man. This concept may be supported by a resemblance of the response (by AOA changes) to the irradiation effect on tumor and tumor-bearer tissues, obtained both on experimental and clinical materials. The fact, that in each separate case a correlation between tu

The short- and long-term effect of single high-dose intra-operative electron beam irradiation of retroperitoneal structures--an experimental study in dogs.

Intra-operative electron beam radiotherapy is a new treatment modality. The abdomen is the area of greatest potential applicability of IORT and therefore its effect on retroperitoneal structures was investigated in a canine model. The retroperitoneal structures tolerate 30 to 40 Gy well, with the exception of the radiation-sensitive ureter which showed fibrosis. Twenty to 25 Gy is within the safe dose limits for the clinical application of IORT.

Radiation effects on human heredity.

In experimental organisms such as fruit flies and mice, increased frequencies in germ cell mutations have been detected following exposure to ionizing radiation. In contrast, there has been no clear evidence for radiation-induced germ cell mutations in humans that lead to birth defects, chromosome aberrations, Mendelian disorders, etc. This situation exists partly because no sensitive and practical genetic marker is available for human studies and also because the number of people exposed to large doses of radiation and subsequently having offspring was small until childhood cancer survivors b

Moving beyond mean heart dose: The importance of cardiac substructures in radiation therapy toxicity.

Normal tissue tolerance dose limits to the heart have been established to reduce the risk of radiation-induced cardiac disease (RICD). Dose constraints have been developed based on either the mean dose delivered to the whole heart (MHD) or the dose delivered to a specific volume, for example, volume of heart receiving equal to or greater than 30 Gy (V30). There is increasing evidence that the impact of thoracic radiation on cardiac morbidity and mortality has been underestimated. Consequently, there is a need to reduce the dose delivered to the heart in radical radiotherapy treatment planning.

An Agent-Based Model of Radiation-Induced Lung Fibrosis.

Early- and late-phase radiation-induced lung injuries, namely pneumonitis and lung fibrosis (RILF), severely constrain the maximum dose and irradiated volume in thoracic radiotherapy. As the most radiosensitive targets, epithelial cells respond to radiation either by undergoing apoptosis or switching to a senescent phenotype that triggers the immune system and damages surrounding healthy cells. Unresolved inflammation stimulates mesenchymal cells' proliferation and extracellular matrix (ECM) secretion, which irreversibly stiffens the alveolar walls and leads to respiratory failure. Although a

Stem cell therapies for the treatment of radiation-induced normal tissue side effects.

SIGNIFICANCE: Targeted irradiation is an effective cancer therapy but damage inflicted to normal tissues surrounding the tumor may cause severe complications. While certain pharmacologic strategies can temper the adverse effects of irradiation, stem cell therapies provide unique opportunities for restoring functionality to the irradiated tissue bed. RECENT ADVANCES: Preclinical studies presented in this review provide encouraging proof of concept regarding the therapeutic potential of stem cells for treating the adverse side effects associated with radiotherapy in different organs. CT scanning for radiation therapy treatment planning of hepatoma.

One hundred forty-five patients with hepatoma had CT scanning for radiation therapy treatment planning. In order to demonstrate the anatomical distortions that occur with hepatoma and its effect on treatment planning, a control group of 50 colorectal cancer patients with normal livers was analyzed for comparison. The objectives of planning were to deliver as homogeneous a dose to the whole liver as possible and not to treat more than one of two functional kidneys or more than one-half of both functional kidneys. Conventional AP/PA portals were defined by physical examination, intravenous pyelo
