Hemidystonia secondary to pediatric thalamic glioblastoma: a case report.

INTRODUCTION: Thalamic tumors are rare and uncommonly manifest as movement disorders, including hemidystonia. Despite this association, little is known about the evolution of hemidystonia. CASE DESCRIPTION: We report on a 11-year-old boy who complained of hypaesthesia and fine motor problems in the left hand. A magnetic resonance imaging showed a large mass in the right thalamus. Stereotactic biopsy revealed a WHO grade 4 astrocytoma, and the patient underwent normofractioned radiochemotherapy with proton-beam radiation and temozolomide. Three months later, a spastic hemiparesis developed on …

Gliosarcoma occurrence after surgical clipping of aneurysm - coincidence or causal relationship?

Gliosarcoma is relatively rare brain tumor of glial and sarcomatous origin. The occurrence of this highly malignant brain tumor in the vicin … s pathologically confirmed after complete tumor extirpation. Despite postoperative radiotherapy and temozolomide chemotherapy, reoccurrence and subarachnoid metastasis unavoidably emerged. Brain injury, co-existence of two pathologies, metal ions released from aneurysm clips, and radiation might have led to tumorigenesis.

Effect of chemotherapeutic drugs and cytochalasin B on tunneling nanotubes in U87 MG cells.

Intercellular communication in the tumor microenvironment has a major impact on therapeutic outcomes. In particular, highly aggressive cance … eletal component, is required for developing TNT networks. In contrast, the chemotherapeutic agents temozolomide (TMZ) and cytarabine (AraC) do not affect TNTs and actin content. As TNT networks are closely linked to increased therapy resistance of certain tumors, we believe that the lack of suppression of TNTs is a major problem in current glioblastoma therap …

Potent Therapeutic Activity of NEO212 in Preclinical Models of Human and Canine Leukaemia and Lymphoma.

Haematological cancer types, such as leukaemia and lymphoma, represent diseases that are life-threatening to canine and human patients alike, and better treatments are needed. We are developing a novel anticancer agent, NEO212, a conjugate of two cancer drugs, the alkylating agent temozolomide (TMZ) and the monoterpene perillyl alcohol (POH). NEO212 has revealed robust therapeutic activity in preclinical tumour models harbouring different human cancer types. In the comparative preclinical study presented here, a two-species (canine and human) and two-cancer (leukaemia and lymphoma) analysis wa…

Cystathionine gamma-lyase (CTH) inhibition attenuates glioblastoma formation.

PURPOSE: Glioblastoma (GBM) is the most common type of adult brain tumor with extremely poor survival. Cystathionine-gamma lyase (CTH) is on … ion and associated with worse overall survival in all grades of gliomas. Patients not responding to temozolomide have also higher CTH expression. In mouse or human GBM cells, pharmacological inhibition (PAG) or CTH knockdown (siRNA) attenuates GBM cell proliferation, migration and stem cell formation frequency. MAJOR CONCLUSIONS: Inhibition of CTH could be a …

Selective Vulnerability of Senescent Glioblastoma Cells to BCL-XL Inhibition.

UNLABELLED: Glioblastoma (GBM) is a rapidly fatal malignancy typically treated with radiation and temozolomide (TMZ), an alkylating chemotherapeutic. These cytotoxic therapies cause oxidative stress and DNA damage, yielding a senescent-like state of replicative arrest in surviving tumor cells. Unfortunately, recurrence is inevitable and may be driven by surviving tumor cells eventually escaping senescence. A growing number of so-called "senolytic" drugs have been recently identified that are defined by their ability to selectively eliminate senescent cells. A growing inventory of senolytic dru…

Identification of TNFRSF12A as a key regulator of stemness and immunosuppression in a novel glioma subtype via multi-omics analysis and experimental validation.

Glioma stem cells (GSCs) drive tumor heterogeneity, therapy resistance, and immunosuppression. This study identified molecular subtypes of glioma based on stemness-related genes and validated a key hub gene through bioinformatics and experiments. A stemness gene set was compiled from single-cell data, GeneCards, and MSigDB. Consensus clustering (NMF) on 157 glioma samples from GSE4290 revealed two subtypes (C1 and C2), with C1 showing higher stemness scores. C1 exhibited enriched DNA repair, proliferation, and metabolic pathways, plus greater temozolomide (TMZ) resistance and M2 macrophage inf…

Recurrent non-functioning pituitary adenomas: a review on the new pathological classification, management guidelines and treatment options.

At least 50% of surgically resected non-functioning pituitary adenomas (NFPA) recur. Either early or late adjuvant radiotherapy is highly efficacious in controlling recurrent NFPA but associates potentially burdensome complications like hypopituitarism, vascular complications or secondary neoplasm. Reoperation is indicated in bulky tumor rests compressing the optic pathway. To date, no standardized medical therapy is available for recurrent NFPA although cabergoline and temozolomide show promising results. Guidelines on the management of recurrent NFPAs are now available. The new 2017 WHO pitu…

Discovery and Optimization of a Novel Brain-Penetrant Colchicine-Binding Site Ligand for Treatment of Glioma.

The limited number of approved drugs for treatment of brain cancers is very apparent. Temozolomide and Vorasidenib continue to be the only approved brain-permeable, orally available drugs. A novel class of highly brain-penetrant tubulin-binding ligands was discovered through rigorous structure-activity-relationship (SAR) studies. The lead compounds were validated through tubulin polymerization assays and co-crystallization, demonstrating destabilization of microtubules by binding to the colchicine site of tubulin. The ligands maintain core interactions with tubulin like those of colchicine; ho…

Patient-derived organoids predict personalized drug response and reveal alternative therapeutic options in glioblastoma.

Glioblastoma remains a highly aggressive brain tumor with limited treatment options and poor prognosis. Temozolomide (TMZ) is the only approved first-line therapy, but frequent resistance limits its efficacy, highlighting the urgent need for alternative treatments. We establish 18 glioblastoma organoid (GBO) lines that preserve the histopathological and genomic features of the parental tumors, enabling comparison with clinical outcomes. GBO-based drug sensitivity testing (GBO-DST) showed a strong correlation with progression-free survival and outperformed O6-methylguanine-DNA methyltransferase…
