Ciprofloxacin-loaded polymeric nanoparticles incorporated electrospun fibers for drug delivery in tissue engineering applications.

Presented work focuses on the development of biodegradable polymer nanoparticles loaded with antibiotics as drug delivery systems deposited on electrospun scaffolds for tissue engineering. The innovative ciprofloxacin-loaded poly(DL-lactide-co-glycolide) NPs ensure a continuous slow release and high local concentration at the site of action for an optimal therapy. The local delivery of antibiotics as an integrated part of electrospun scaffolds offers an effective, safe, and smart enhancement supporting tissue regeneration. Presented data provides solid scientific evidence for fulfilling the re…

Evaluation of Sterilization/Disinfection Methods of Fibrous Polyurethane Scaffolds Designed for Tissue Engineering Applications.

Sterilization of a material carries the risk of unwanted changes in physical and chemical structure. The choice of method is a challenge-the process must be efficient, without significantly changing the properties of the material. In the presented studies, we analyzed the effect of selected sterilization/disinfection techniques on the properties of nanofibrous polyurethane biomaterial. Both radiation techniques (UV, gamma, e-beam) and 20 minutes' contact with 70% EtOH were shown not to achieve 100% sterilization efficiency. The agar diffusion test showed higher sterilization efficiency when us

Polycaprolactone fibrous electrospun scaffolds reinforced with copper doped wollastonite for bone tissue engineering applications.

The bone defects healing are always associated with post implantation infections; hence biomaterials rules significant role for orchestration of defective bone. In this study, we synthesized biocomposite scaffold by combining polycaprolactone (PCL), wollastonite (Ws) and metal ions (Cu) by electrospinning technique. The manufactured scaffolds (PCL/Ws andPCL/Cu-Ws) were subjected to physio-chemical characterization by scanning electron microscopy, energy dispersive X-ray spectroscopy, Fourier Transform Infra Red Spectroscopy (FTIR) and XRD. The surface topography of the scaffolds was found to b

Marine spongin incorporation into Biosilicate® for tissue engineering applications: An in vivo study.

Biomaterials and bone grafts, with the ability of stimulating tissue growth and bone consolidation, have been emerging as very promising strategies to treat bone fractures. Despite its well-known positive effects of biosilicate (BS) on osteogenesis, its use as bone grafts in critical situations such as bone defects of high dimensions or in non-consolidated fractures may not be sufficient to stimulate tissue repair. Consequently, several approaches have been explored to improve the bioactivity of BS. A promising strategy to reach this aim is the inclusion of an organic part, such as collagen, i

Human Stem Cell Based Tissue Engineering for In Vivo Cartilage Repair: A Systematic Review.

Pure chondral defects represent the most clinically significant articular cartilage injuries. To inform the development of clinically suitable tissue-engineering strategies for chondral repair using cells from a human patient, the combination of human stem cells (HSCs), biomaterial scaffolds, and growth factors has been widely harnessed in preclinical animal models. Due to the large heterogeneity in study designs and outcome reporting in such studies, we aimed to systematically review literature pertaining to HSC based tissue engineering strategies in animal models of chondral repair such that…

Noninvasive ultrasound imaging for assessment of intact microstructure of extracellular matrix in tissue engineering.

Development of artificial tissues or organs is one of the actual tasks in regenerative medicine that requires observation and evaluation of intact volume microstructure of tissue engineering products at all stages of their formation, from native donor tissues and decellularized scaffolds to recipient cell migration in the matrix. Unfortunately in practice, methods of vital noninvasive imaging of volume microstructure in matrixes are absent. In this work, we propose a new approach based on high-frequency acoustic microscopy for noninvasive evaluation and visualization of volume microstructure i…

Indirect 3D bioprinting and characterization of alginate scaffolds for potential nerve tissue engineering applications.

Low-concentration hydrogels have favorable properties for many cell functions in tissue engineering but are considerably limited from a scaffold fabrication point of view due to poor three-dimensional (3D) printability. Here, we developed an indirect-bioprinting process for alginate scaffolds and characterized the potential of these scaffolds for nerve tissue engineering applications. The indirect-bioprinting process involves (1) printing a sacrificial framework from gelatin, (2) impregnating the framework with low-concentration alginate, and (3) removing the gelatin framework by an incubation…

Cryogel composites based on hyaluronic acid and halloysite nanotubes as scaffold for tissue engineering.

We present here preparation of mechanically strong and biocompatible cryogel composites based on hyaluronic acid (HA) and halloysite nanotubes (HNTs) of various compositions, and their applications as scaffold for different cell growing media. Uniaxial compression tests reveal that the incorporation of HNTs into HA cryogels leads to a ~2.5-fold increase in their Young moduli, e.g., from 38 ± 1 to 99 ± 4 kPa at a HA:HNTs weight ratio of 1:2. Although HA:HNTs based cryogels were found to be blood compatible with 1.37 ± 0.11% hemolysis ratio at a HA:HNTs weight ratio of 1:2, they trigger thrombog

Multiple and Promising Applications of Strontium (Sr)-Containing Bioactive Glasses in Bone Tissue Engineering.

Improving and accelerating bone repair still are partially unmet needs in bone regenerative therapies. In this regard, strontium (Sr)-containing bioactive glasses (BGs) are highly-promising materials to tackle this challenge. The positive impacts of Sr on the osteogenesis makes it routinely used in the form of strontium ranelate (SR) in the clinical setting, especially for patients suffering from osteoporosis. Therefore, a large number of silicate-, borate-, and phosphate-based BGs doped with Sr and produced in different shapes have been developed and characterized, in order to be used in the

Halloysite-alkaline phosphatase system-A potential bioactive component of scaffold for bone tissue engineering.

Alkaline phosphatase (ALP) was sucessfully incorporated into the halloysite (HAL) nanotubes, as confirmed by FTIR-ATR and XRD analyses. The loading efficiency (LE) of ALP was found to be 13.5%, while its encapsulation efficiency (EE) was estimated to be around 27%, as determined using the Bradford test. The influence of the immobilization in HAL on the enzyme activity was measured using standard ALP activity assay. Immobilized ALP effectively induced the bomineralization process, as showed by SEM, EDS, and XRD studies. As a result, calcium phosphate was produced in the form of hydroxyapatite c

Electrospun collagen/poly(L-lactic acid-co-ε-caprolactone) scaffolds for conjunctival tissue engineering.

Conjunctival injuries are general but intractable ocular surface diseases, the sequelae of which are particularly challenging to treat. A promising therapy for conjunctival injuries is to employ biodegradable scaffolds to deliver conjunctival epithelial cells for repairing damaged or diseased conjunctiva. In the present study, an ultrathin porous nanofibrous scaffold was fabricated by using collagen and poly(L-lactic acid-co-ε-caprolactone) (PLCL) and displayed a thickness of 20 µm, with a high porosity and an average fiber diameter of 248.83±26.44 nm. Conjunctival epithelial cells seeded on t

Skeletal muscle regeneration on protein-grafted and microchannel-patterned scaffold for hypopharyngeal tissue engineering.

In the field of tissue engineering, polymeric materials with high biocompatibility like polylactic acid and polyglycolic acid have been widely used for fabricating living constructs. For hypopharynx tissue engineering, skeletal muscle is one important functional part of the whole organ, which assembles the unidirectionally aligned myotubes. In this study, a polyurethane (PU) scaffold with microchannel patterns was used to provide aligning guidance for the seeded human myoblasts. Due to the low hydrophilicity of PU, the scaffold was grafted with silk fibroin (PU-SF) or gelatin (PU-Gel) to impro…
