Microplastics ingestion in the ephyra stage of Aurelia sp. triggers acute and behavioral responses.

For the first time, we report a correspondence between microplastics (MP) ingestion and ecotoxicological effects in gelatinous zooplankton (Cnidarian jellyfish). The ephyra stage of the jellyfish Aurelia sp. was exposed to both environmental and high concentrations of fluorescent 1-4 μm polyethylene MP (0.01-10 mg/L). After 24 and 48 h, MP accumulation, acute (Immobility) and behavioral (Frequency pulsation) endpoints were investigated. MP were detected by confocal and tomographic investigations on gelatinous body and mouth, either attached on the surface or ingested. This interaction was resp

Acute toxic effects of polyethylene microplastic on adult zebrafish.

To identify the physical effects, behavioral changes, and gene expression profiles of the phase 1 detoxification-related gene (cyp 1a) and oogenesis-related gene (vtg 1) induced by microplastics, high-density polyethylene microplastics of various sizes were used because of their dominance in coastal areas and effluent samples in Hong Kong. Adult zebrafish were used as the model organism to identify the upper and lower boundaries of microplastics ingestion and were exposed to individual polyethylene microplastics in five size ranges (10-22 μm, 45-53 μm, 90-106 μm, 212-250 μm, and 500-600 μm) at

Separation of oil sands process water organics and inorganics and examination of their acute toxicity using standard in-vitro bioassays.

Oil sands process water (OSPW) contains complex components of inorganics and organics. This is the first study that separated OSPW inorganic and organic fractions and examined their relative acute toxicity when compared with the original whole OSPW using an in-vitro cell-based bio-indicator system. The separation of OSPW inorganic and organic fractions would be conducive to the understanding of the toxic contribution of organic and inorganic fractions as well as the identification and treatment of organic fractions. In this research, we demonstrated that the highest organic fraction extraction

Impact of Wearing and Washing/Drying of Permethrin-Treated Clothing on Their Contact Irritancy and Toxicity for Nymphal Ixodes scapularis (Acari: Ixodidae) Ticks.

Permethrin-treated clothing is available as consumer products to prevent bites by tick and insect pests. We used bioassays to examine the impact of wearing and washing/drying of permethrin-treated shirts, pants, and socks, and wearing of treated shoes, on their contact irritancy and toxicity for nymphal Ixodes scapularis Say (Acari: Ixodidae) ticks, the primary vectors in the eastern United States of the causative agents of Lyme disease, human anaplasmosis, and human babesiosis. Pristine permethrin-treated clothing displayed strong contact irritancy and toxicity toward I. scapularis nymphs, wi

Sub-lethal effects induced by a mixture of different pharmaceutical drugs in predicted environmentally relevant concentrations on Lithobates catesbeianus (Shaw, 1802) (Anura, ranidae) tadpoles.

The increasing consumption of medications by humans has negative effects such as the increased disposal of these compounds in the environment. Little is known about how the disposal of a "drug mix" (DM) in aquatic ecosystems can affect their biota. Thus, we evaluated whether the exposure of Lithobates casteibeianus tadpoles to a DM composed of different medication classes (antibiotic, anti-inflammatory, antidepressant, anxiolytic, analgesic, and antacid drugs)-at environmentally relevant concentrations-may change their oral morphology, trigger behavioral disorders, and have mutagenic effects o

Cellular alterations in midgut cells of honey bee workers (Apis millefera L.) exposed to sublethal concentrations of CdO or PbO nanoparticles or their binary mixture.

Beside many beneficial applications in industry, agriculture and medicine, nanoparticles (NPs) released into the environment might cause adverse effects. In the present study, effects of exposure to sublethal concentrations of PbO and CdO NPs, either separately or in combination on honey bee (A. mellifera) workers were assessed. Honey bee workers were fed sugar syrup contained (20% of LC50) of CdO (0.01 mg ml-1) and PbO (0.65 mg ml-1) NPs either separately or combined for nine days under laboratory conditions. Control bees were fed 1.5 M sucrose syrup without NPs. Effects on histological and c

Toxicity of silver ions, metallic silver, and silver nanoparticle materials after in vivo dermal and mucosal surface exposure: A review.

Silver is used in different applications that result in contact with skin and mucosal surfaces (e.g., jewelry, wound dressings, or eye drops). Intact skin poses an effective barrier against the absorption of silver. Mucosal surfaces are observed to be less effective barriers and compromised skin is often a poor barrier. Silver can deposit as particles in the human body causing a blue-gray discoloration known as argyria. Urine and feces are reported pathways of excretion. Acute human mortality has been observed following an abortion procedure involving the intrauterine administration of 7 g sil

Satellite rats are redundant in embryo-fetal development studies.

Routinely in many laboratories, satellite rats are added to embryo-fetal development (EFD) studies for pharmaceuticals to assess toxicokinetic (TK) properties, because it is assumed that collection of multiple blood samples with relatively large volumes might affect the study outcome. With recent refinement of blood sampling techniques, this belief requires reevaluation. The current work showed successful implementation of jugular vein blood sampling in an EFD rat study without satellite animals, thereby reducing the number of rats in standard EFD studies for pharmaceuticals by 20%. Although n

Applicability of predictive toxicology methods for monoclonal antibody therapeutics: status Quo and scope.

Biopharmaceuticals, monoclonal antibody (mAb)-based therapeutics in particular, have positively impacted millions of lives. MAbs and related therapeutics are highly desirable from a biopharmaceutical perspective as they are highly target specific and well tolerated within the human system. Nevertheless, several mAbs have been discontinued or withdrawn based either on their inability to demonstrate efficacy and/or due to adverse effects. Approved monoclonal antibodies and derived therapeutics have been associated with adverse effects such as immunogenicity, cytokine release syndrome, progressiv

Induced tolerance from a sublethal insecticide leads to cross-tolerance to other insecticides.

As global pesticide use increases, the ability to rapidly respond to pesticides by increasing tolerance has important implications for the persistence of nontarget organisms. A recent study of larval amphibians discovered that increased tolerance can be induced by an early exposure to low concentrations of a pesticide. Since natural systems are often exposed to a variety of pesticides that vary in mode of action, we need to know whether the induction of increased tolerance to one pesticide confers increased tolerance to other pesticides. Using larval wood frogs (Lithobates sylvaticus), we inve

Toxic effects of copper ion in zebrafish in the joint presence of CdTe QDs.

Quantum dots (QDs) have strong adsorption capacity; therefore, their potential toxicity of the facilitated transport of other trace toxic pollutants when they co-exist to aquatic organisms has become a hot research topic. The lab study was performed to determine the developmental toxicities to the zebrafish after exposed to the combined pollution of Cadmium-telluride (CdTe) QDs and copper ion (Cu(2+)) compared to the single exposure. Our findings for the first time revealed that: 1) CdTe QDs facilitated the accumulation of Cu(2+) in zebrafish, 2) the higher mortality, lower hatch rate, and mor

Acute electrocardiographic ST segment elevation may predict hypotension in a swine model of severe cyanide toxicity.

Cyanide causes severe cardiac toxicity resulting in tachycardia, hypotension, and cardiac arrest; however, the clinical diagnosis can be difficult to make. A clinical finding that may precede or predict cyanide-induced hypotension may be a trigger to provide treatment earlier and improve outcomes in cyanide toxicity. Our primary objective was to determine if there is a clinically significant change in ST segment deviation measured on ECG during intravenous cyanide infusion that may predict cyanide-induced hypotension. As part of a larger study comparing antidotes for cyanide-induced shock, 30

Comparison of oral toxicological properties of botulinum neurotoxin serotypes A and B.

Botulinum neurotoxins (BoNTs) are among the most potent biological toxins for humans. Of the seven known serotypes (A-G) of BoNT, serotypes A, B and E cause most of the foodborne intoxications in humans. BoNTs in nature are associated with non-toxic accessory proteins known as neurotoxin-associated proteins (NAPs), forming large complexes that have been shown to play important roles in oral toxicity. Using mouse intraperitoneal and oral models of botulism, we determined the dose response to both BoNT/B holotoxin and complex toxins, and compared the toxicities of BoNT/B and BoNT/A complexes. Al

Influence of stability on the acute toxicity of CdSe/ZnS nanocrystals to Daphnia magna.

The acute toxicity of polymer-coated CdSe/ZnS quantum dots (QDs) to Daphnia magna was investigated using 48-h exposure studies. The principal objective was to relate the toxicity of QDs to specific physical and chemical aspects of the QD. As such, two different CdSe core diameters, 2 nm QDs (green-emitting) and 5 nm QDs (red-emitting), and two different surface coatings, polyethylene oxide (PEO) and 11-mercaptoundecanoic acid (MUA) were studied. The QDs were characterized before and after the 48-h exposure using fluorescence, ultrafiltrations (3 kDa), and inductively coupled plasma-atomic emis
