Rhythmic melatonin secretion in different teleost species: an in vitro study.

The rhythmic production of melatonin is governed by intrapineal oscillators in all fish species so far investigated except the rainbow trout. To determine whether the latter represents an exception among fish, we measured in vitro melatonin secretion in pineal organs of nine wild freshwater and six marine teleost species cultured at constant temperature and under different photic conditions. The results demonstrate that pineal organs of all species maintain a rhythmic secretion of melatonin under light:dark cycles and complete darkness, and strongly suggest that most fish possess endogenous in…

Synergistic effects of gentamicin, levofloxacin, and cefotaxime against Aeromonas salmonicida: In vitro and In vivo evaluation.

The increasing prevalence of multidrug-resistant (MDR) Aeromonas salmonicida (AS) threatens both animal and human health by limiting the effectiveness of conventional antibiotics. To identify effective therapeutic strategies, we evaluated the activity of gentamicin (GENTA), levofloxacin (LEV), and cefotaxime sodium (CFT) as individual treatments and in combination using zebrafish and rainbow trout infection models. In vitro checkerboard assays showed consistent synergistic effects in the GENTA-CFT combination, as confirmed by Combenefit analysis. In vivo experiments revealed that AS infection …

Protective responses of intestinal mucous cells in a range of fish-helminth systems.

Histopathological, immunofluorescence and ultrastructural studies were conducted on the intestines of four fish species infected with different taxa of enteric helminths. Brown trout (Salmo trutta trutta), eel (Anguilla anguilla) and tench (Tinca tinca) obtained from Lake Piediluco (central Italy) were examined. Brown trout and eel were infected with two species of acanthocephalans, and tench was parasitized with a tapeworm species. In addition to the above site, specimens of chub (Squalius cephalus) and brown trout infected with an acanthocephalan were examined from the River Brenta (north It…

Fish rhabdovirus cell entry is mediated by fibronectin.

Three monoclonal antibodies (MAbs) generated against rainbow trout gonad cells (RTG-2) have been selected for their ability to protect cells from the viral hemorrhagic septicemia virus (VHSV) infection, a salmonid rhabdovirus. Protection from infection was restricted to the salmonid-derived cell lines indicating species specificity of the blocking MAbs. Surprisingly, the blocking activity of these MAbs was also effective against other nonantigenically related fish rhabdoviruses. Indirect immunofluorescence and immunoelectron microscopy observations demonstrated that the three MAbs were all dir…

Combined Transcriptomic and Histological Profiling Uncover Hepatic Regulatory Hierarchy of Triploid Oncorhynchus mykiss Under Interactive Salinity, Temperature and Body Weight Regimes.

Salinity, temperature and body weight dominate seawater acclimation in rainbow trout (Oncorhynchus mykiss), yet few studies simultaneously explore their main effects and potential correlative interactive patterns in hepatic responses. A 60-day L9 (33) orthogonal trial was performed on triploid rainbow trout with three gradients of body weight, temperature and salinity. Hepatic transcriptomics revealed that salinity drove global transcriptional remodeling and high salinity induced far fewer DEGs than medium salinity. WGCNA screened a salinity-positive blue module (r = 0.408, p = 0.0346), while …

Regulation of pancreatic somatostatin gene expression by insulin and glucagon.

Rainbow trout were used as a model system to study the effects of insulin and glucagon on the expression of preprosomatostatins (PPSS). We previously showed that the endocrine pancreas of trout contains three mRNAs that encode for distinct somatostatin-containing peptides: PPSS I, which contains somatostain-14 (SS-14) at its C-terminus, and two separate PPSS IIs, PPSS II' and PPSS II'', each containing [Tyr7, Gly10]-SS-14 at their C-terminus. Rainbow trout injected (100 ng/g body weight) with insulin displayed elevated expression of PPSS II' and PPSS II'' mRNAs. Glucagon-injected (100 ng/g bod…

Lack of circadian regulation of in vitro melatonin release from the pineal organ of salmonid teleosts.

In many teleost species, the photoreceptive pineal organ harbors the circadian clock that regulates melatonin release in the pineal organ itself. However, the pineal organ of three salmonids (rainbow trout Oncorhynchus mykiss, masu salmon Oncorhynchus masou, and sockeye salmon Oncorhynchus nerka) did not exhibit circadian rhythms in melatonin release when maintained under constant darkness (DD) in vitro, suggesting that the pineal organs of all salmonids lack the circadian regulation of melatonin production. To test this hypothesis, the pineal organ of seven salmonids (common whitefish Coregon…

Vibrio damsela associated with diseased fish in Denmark.

A total of 26 Vibrio damsela strains were isolated in Denmark. Fifteen strains were isolated from the head kidney of rainbow trout (Oncorhynchus mykiss) in aquaculture, eight were from organs of two stingrays (Dasyatis pastinaca) held in captivity, two were from organs and pathological material of a nurse shark (Orectolobus ornatus) held in captivity, and one was from the water in the aquarium where the rays and shark were kept. This is the first report on the isolation of V. damsela in Denmark and the first report on this organism associated with diseased farmed rainbow trout. Ecological and …

Detection of triploidy in fish using the cytokinesis-blocked method for erythrocyte and hepatic cells.

The use of two cytochalasin-blocked methods with erythrocyte and hepatic cells was developed to identify spontaneous and/or induced triploid fish by observing the presence of trinucleated cells. Spontaneous triploids were found in two specimens out of fifteen commercial rainbow trout from a hatchery at 2 years of age. Some rare spontaneous triploid (3n) trinucleated cells were also found in the other thirteen diploid (2n) trout specimens under investigation. The use of this novel technique in a study of the triploidy in vitro, allows evaluation of the polyploid specimens under field and labora…

Conspecific migration and environmental setting determine parasite infracommunities of non-migratory individual fish.

Parasite infracommunities tend to be stochastic in nature, although environmental characteristics such as the type of water source in streams and host traits can have an effect on the biotic assemblages and by extension the parasite fauna. We examined the effect of water source and the rate of adult fish migration on the metazoan parasite infracommunities of conspecific juvenile brown trout, Salmo trutta L. among streams flowing into Lake Lucerne (Switzerland). Juvenile (1 to 2-year old) fish harboured higher parasite species richness in groundwater-fed than in surface water-fed streams, where…

Lessons from an outbreak of listeriosis related to vacuum-packed gravad and cold-smoked fish.

The first lesson learned from this outbreak was that vacuum-packed rainbow trout is not only an excellent medium for the growth of Listeria monocytogenes, but may also cause human listeriosis. Another lesson is that one single fish processing plant may spread multiple clonal types of L. monocytogenes by selling contaminated products to consumers. Thus, when investigating fish-borne outbreaks of listeriosis one should identify and type several isolates of L. monocytogenes from each food and environmental sample, since multiple clonal types might be present. The outbreak described in this paper …

Saturable triiodothyronine-binding sites in the pituitary nuclei of salmonid teleost fish.

High-affinity, limited-capacity, 3,5,3'-triiodo-L-thyronine (T3)-binding sites were established by in vitro saturation analysis in cell nuclei of the pituitary gland of arctic charr. The sites were extracted from the purified nuclei using 0.4 M NaCl and incubated with [125I]T3 in the presence of 0.2 M NaCl. T3 saturable binding attained equilibrium after 18-24 hr of incubation at 4 degrees. The association constant ranged from 6.7 to 20.1 liters.mol-1 x 10(9), indicating a T3 affinity greater than that for T3-binding sites in rainbow trout liver. The maximal binding capacity ranged from 0.93 t…

Thermal tolerance of cultured and wild Icelandic arctic charr (Salvelinus alpinus) at self-selected flow rates.

Climate change is predicted to change not only the temperature of many freshwater systems but also flow dynamics. Understanding how fishes will fare in the future requires knowing how they will respond to both extended variations of temperature and flow. Arctic charr have had their thermal tolerance measured, but never with respect to flow. Additionally, this circumpolar species has multiple populations exhibiting dramatic phenotypic plasticity which may mean that regional differences in thermal tolerance are unaccounted for. In Iceland, Arctic charr populations have experienced highly variabl

The first detection of infectious hematopoietic necrosis virus (IHNV) genogroup U in western Europe in the first IHNV outbreak in Finland.

Infectious hematopoietic necrosis virus (IHNV) was detected for the first time in Finland in November 2017. The virus was isolated from rainbow trout (Oncorhynchus mykiss) from a sea-cage farm in the Gulf of Bothnia. Sampling was carried out as part of a routine risk-based surveillance program. During inspections based on epidemiological investigations, five other IHNV-positive rainbow trout establishments were detected in 2017-2018. No symptoms indicative of IHN were reported, and only a few macroscopic and histopathological findings were recorded. To assess the pathogenicity of the Finnish i…

A role for tricellulin in the regulation of gill epithelium permeability.

The apical-most region of cell-to-cell contact in a vertebrate epithelium is the tight junction (TJ) complex. It is composed of bicellular TJs (bTJs) that bridge two adjacent epithelial cells and tricellular TJs (tTJs) that are points of contact between three adjoining epithelial cells. Tricellulin (TRIC) is a transmembrane TJ protein of vertebrates that is found in the tTJ complex. Full-length cDNA encoding rainbow trout TRIC was cloned and sequenced. In silico analysis of rainbow trout TRIC revealed a tetraspannin protein with several putative posttranslational modification sites. TRIC mRNA …
