CRISPR-Cas9 Gene Editing Protects from the A53T-SNCA Overexpression-Induced Pathology of Parkinson's Disease In Vivo.

Mutations in specific genes, including synuclein alpha (SNCA) that encodes the α-synuclein protein, are known to be risk factors for sporadic Parkinson's disease (PD), as well as critical factors for familial PD. In particular, A53T-mutated SNCA (A53T-SNCA) is a well-studied familial pathologic mutation in PD. However, techniques for deletion of the mutated SNCA gene in vivo have not been developed. Here, we used the CRISPR-Cas9 system to delete A53T-SNCA in vitro as well as in vivo. Adeno-associated virus carrying SaCas9-KKH with a single-guide RNA targeting A53T-SNCA significantly reduced A5

Neuropathological evidence of body-first vs. brain-first Lewy body disease.

Aggregation of alpha-synuclein into inclusion bodies, termed Lewy pathology, is a defining feature of Parkinson's disease (PD) and Dementia with Lewy bodies (DLB). In the majority of post mortem cases, the distribution of Lewy pathology seems to follow two overarching patterns: a caudo-rostral pattern with relatively more pathology in the brainstem than in the telencephalon, and an amygdala-centered pattern with the most abundant pathology in the "center of the brain", including the amygdala, entorhinal cortex, and substantia nigra, and relatively less pathology in the lower brainstem and spin…

Gut bacterial composition in a mouse model of Parkinson's disease.

The mechanism of neurodegeneration in Parkinson's disease (PD) remains unknown but it has been hypothesised that the intestinal tract could be an initiating and contributing factor to the neurodegenerative processes. In PD patients as well as in animal models for PD, alpha-synuclein-positive enteric neurons in the colon and evidence of colonic inflammation have been demonstrated. Moreover, several studies reported pro-inflammatory bacterial dysbiosis in PD patients. Here, we report for the first time significant changes in the composition of caecum mucosal associated and luminal microbiota and…

α-Synuclein Biomarkers for Parkinson's Disease.

α-Synuclein (α-syn) biomarkers show great promise as diagnostic tools for Parkinson's disease (PD). In recent years, a large body of evidence has validated their efficacy as diagnostic tools for PD and other synucleinopathies and has shown potential for use in patients with isolated prodromal symptoms of PD, such as rapid eye movement (REM) sleep behavior disorder and hyposmia, and further illuminates the pathophysiology of both idiopathic and genetic causes. Various detection methods have been deployed, predominantly immunohistochemistry and α-syn seed amplification assays. α-Syn has been sho

Towards biomarker-based diagnosis of Parkinson disease.

The current clinical diagnostic criteria for Parkinson disease (PD) have limitations and are inherently insensitive to the earliest stages of disease, when classical motor signs can be absent. Imaging and genetic tests are currently used to support or establish a diagnosis of PD, but no validated biomarker-based diagnostic framework currently exists. Substantial progress has been made in the field of molecular disease markers, most notably with the development and validation of seed amplification assays (SAAs), which enable detection of very low levels of pathological α-synuclein in the cerebr

Diagnostics of Synucleinopathies by Protein Amplification Methods: Methodological and Bibliometric Analysis.

Early preclinical diagnostics of neurodegenerative diseases (synucleinopathies, Alzheimer's disease, etc.) can be achieved through the detection of pathological amyloid aggregates in biological fluids. Protein amplification methods (PMCA, RT-QuIC) have become promising diagnostic tools by offering high sensitivity and specificity for detecting α-synuclein oligomers at the early disease stages and studying the fibril formation kinetics and mechanisms of amyloid aggregation. However, these methods have several significant limitations, as they are technically complex and time-consuming and lack s

Parkinson's disease as a multi-axis systems disorder: integrating molecular pathology and circuit-level therapeutics.

Parkinson's disease (PD) has generated extensive mechanistic insight, yet nearly all candidate disease-modifying therapies have failed in clinical trials. Antibodies targeting α-synuclein, mitochondrial antioxidants, lysosomal interventions, and anti-inflammatory agents frequently achieve target engagement without slowing clinical progression, creating a central paradox in PD therapeutics. Here we propose that this discrepancy arises because PD is not primarily a single-pathway neurodegenerative disorder but a multi-scale systems disease. Molecular pathology, organellar dysfunction, immune acti

The amplification of α-synuclein amyloid fibrils.

Amyloid fibrils formed by α-synuclein are a hallmark of a range of neurodegenerative diseases, notably Parkinson's disease, multiple system atrophy (MSA), and dementia with Lewy bodies, collectively known as synucleinopathies. Recent years have seen an increasing understanding of the structural architecture and diversity of α-synuclein amyloid fibrils. Furthermore, our mechanistic understanding of the formation of these structures has also experienced significant progress. Here, I provide a concise overview of the current state of knowledge of how α-synuclein amyloid fibrils can be amplified,

Fragmentation-Induced Disassembly and Reaggregation of α-Synuclein Amyloid Fibrils.

Aggregation of the protein α-synuclein (α-syn) is a defining pathological characteristic of Parkinson's disease (PD). Kinetic studies have provided increasingly detailed insights into the mechanisms of α-syn aggregation, highlighting the contributions of secondary nucleation and elongation to fibril growth. However, the understanding of the role of fibril breakage (fragmentation) remains sparse. We therefore established a modified thioflavin-T (ThT) kinetic assay in which ultrasonication steps were introduced when the conversion of α-syn monomers into amyloid fibrils had reached the plateau. Delving into the possible role of Ochratoxin A in Parkinson's disease: a computational study spotlighting the potential mechanisms of action.

Ochratoxin A (OTA) is a widespread food contaminant relevant to food safety that has various toxic effects including nephrotoxicity and neurotoxicity. Evidence suggests OTA may contribute to Parkinson's disease (PD) pathogenesis acting at multiple levels along the adverse outcome pathway (AOP), though molecular targets and underpinning mechanisms remain unclear. Using a computational pipeline based on molecular modelling techniques, which are well-established methods to elucidate protein-ligand interactions, this study investigated key events in the PD AOP where the current state-of-the-art su

Are we ready for a biological diagnosis of Parkinson's Disease?

The diagnosis of Parkinson's disease (PD) is currently based on clinical criteria, centered on the characteristic motor syndrome. However, motor manifestations become evident only after a significant proportion of nigro-striatal dopaminergic neurons have already undergone neurodegeneration. The recent "NSD-ISS" and "SynNeurGe" research frameworks have proposed new biological diagnostic criteria focusing on α-synucleinopathy, neurodegeneration, and genetic biomarkers, independent of clinical manifestations. These proposals intend to detect the disease at a "biologically early phase" to foster a

Seeding amplification assay: Limitations and insights for enhanced clinical and research applications.

The accurate diagnosis of synucleinopathies-neurodegenerative diseases marked by misfolded α-synuclein protein aggregates, such as Parkinson's disease, dementia with Lewy bodies, and multiple system atrophy-remains a critical challenge. Conventional clinical criteria, frequently verified only through postmortem examination, results in diagnostic delays that impede timely intervention. Seeding amplification assay (SAA) has emerged as a promising diagnostic tool, offering high sensitivity for detecting α-synuclein aggregates even in early disease stages. While SAA enables early diagnosis by ampl

A Novel Tool to Investigate the Early and Late Stages of α-Synuclein Aggregation.

The accumulation of an inherently disordered protein α-synuclein (α-syn) aggregates in brain tissue play a pivotal role in the pathology and etiology of Parkinson's disease. Aggregation of α-syn has been found to be complex and heterogeneous, occurring through multitudes of early- and late-stage intermediates. Because of the inherent complexity and large dynamic range (between a few microseconds to several days under in vitro measurement conditions), it is difficult for the conventional biophysical and biochemical techniques to sample the entire time window of α-syn aggregation. Here, for the

α-Synuclein in Parkinson disease and other neurodegenerative disorders.

With the aging population in the Western hemisphere, neurodegenerative parkinsonism and dementia will become two of the great public health challenges of this century. A major pillar in the effort to treat these conditions will be the shift from symptomatic treatment to disease modifying therapy. This step will absolutely require cheap and reliable biomarkers; patients will need to be diagnosed before irreversible change has occurred. α-Synuclein (αS) is a recent candidate biomarker for Lewy body neurodegeneration. It is a 140 amino acid protein that forms the pathological substrate in idiopat

Brain region-resolved pharmacodynamics of an antisense oligonucleotide targeting human SNCA 3'UTR in BAC-hSNCA rats.

BACKGROUND: Alpha-synuclein accumulation contributes to Parkinson's disease (PD), and lowering α-synuclein expression is a candidate disease-modifying approach. The brain-wide pharmacodynamic profile of human-targeting antisense oligonucleotides (ASOs) remains incompletely defined in humanized models that preserve the regulatory architecture of the human SNCA locus. METHODS: BAC-hSNCA rats were treated at an early stage, when human α-synuclein is elevated, but striatal dopamine content remains largely preserved. A single intracerebroventricular dose of an ASO targeting human SNCA 3'UTR (SNCA …

The Progress of Active Immunotherapy for Parkinson's Disease.

Parkinson's disease (PD) is a multifactorial neurodegenerative disorder defined by nigrostriatal dopaminergic neuron loss and the pathological aggregation of alpha synuclein, yet current clinical interventions remain largely symptomatic and fail to alter long-term disease progression. Emerging evidence demonstrates that immune dysregulation and chronic neuroinflammation contribute significantly to PD pathology, supporting the rationale for active immunotherapy as a disease modifying strategy. This review examines contemporary active immunotherapy platforms including peptide vaccines, genetic v

Parkinson's disease: genetics and neuroinflammatory insights.

Parkinson's disease is a fast-growing neurodegenerative disorder that causes dopaminergic neuron loss and build-up of alpha-synuclein. In terms of number of people affected, it has a large effect on society with an approximately 6.1 million people diagnosed till 2023. Two important factors in PD are neuroinflammation and genetic mutations. The build-up of alpha-synuclein activates neuroimmune cells which leads to the release of inflammatory mediators. Problems with immune system can worsen this inflammation, creating a cycle of ongoing nerve damage. This review discusses the complex interlink …

Thermodynamic stability modulates chaperone-mediated disaggregation of α-synuclein fibrils.

Aggregation of the intrinsically disordered protein alpha-synuclein into amyloid fibrils and their subsequent intracellular accumulation are hallmark features of several neurodegenerative disorders, including Parkinson's disease, for which no curative treatments currently exist. In this study, we investigate the relationship between fibril morphology, thermodynamic stability, and susceptibility to disaggregation by the human chaperone system comprising HSP70, DNAJB1, and Apg2. By varying assembly conditions and incubation times, we generated alpha-synuclein fibrils with diverse morphological a…
