Control of murine brown adipocyte development by GATA6.

Brown adipose tissue (BAT) is a thermogenic organ that protects animals against hypothermia and obesity. BAT derives from the multipotent paraxial mesoderm; however, the identity of embryonic brown fat progenitor cells and regulators of adipogenic commitment are unclear. Here, we performed single-cell gene expression analyses of mesenchymal cells during mouse embryogenesis with a focus on BAT development. We identified cell populations associated with the development of BAT, including Dpp4+ cells that emerge at the onset of adipogenic commitment. Immunostaining and lineage-tracing studies show

Multilineage differentiation potential in the infant adipose- and umbilical cord-derived mesenchymal stem cells.

BACKGROUND: This study aims to compare the biological properties of infant adipose-derived mesenchymal stem cells (infant ADSCs) from excised polydactyly fat tissue and umbilical cord-derived mesenchymal stem cells (UCSCs) in terms of proliferation and differentiation capabilities. The proliferation of infant ADSCs and UCSCs was analyzed by determining the fold changes of cell numbers and doubling time periods. METHODS: The state of senescence and replicative stress was compared by analyzing the expression of age-related genes, senescence-associated β-galactosidase (SA-β-gal) staining, and ph

The role of zinc finger proteins in the fate determination of mesenchymal stem cells during osteogenic and adipogenic differentiation.

Zinc finger proteins (ZFPs) constitute a crucial group of transcription factors widely present in various organisms. They act as transcription factors, nucleases, and RNA-binding proteins, playing significant roles in cell differentiation, growth, and development. With extensive research on ZFPs, their roles in the determination of mesenchymal stem cells (MSCs) fate during osteogenic and adipogenic differentiation processes have become increasingly clear. ZFP521, for instance, is identified as an inhibitor of the Wnt signaling pathway and RUNX2's transcriptional activity, effectively suppressi

Senescent immune cells release grancalcin to promote skeletal aging.

Skeletal aging is characterized by low bone turnover and marrow fat accumulation. However, the underlying mechanism for this imbalance is un … partially inactivates its downstream signaling pathways, thus repressing osteogenesis and promoting adipogenesis of bone marrow mesenchymal stromal cells. Heterozygous genetic deletion of Plexnb2 in skeletal stem cells abrogated the improved bone phenotype of Gca-knockout mice. Finally, we developed a grancalcin-neutralizing antibody and showed that its treatm …

Hedgehog Signaling Controls Bone Homeostasis by Regulating Osteogenic/Adipogenic Fate of Skeletal Stem/Progenitor Cells in Mice.

Skeletal stem/progenitor cells (SSPCs) can differentiate into osteogenic or adipogenic lineage. The mechanism governing lineage allocation of SSPCs is still not completely understood. Hedgehog (Hh) signaling plays an essential role in specifying osteogenic fate of mesenchymal progenitors during embryogenesis. However, it is still unclear whether Hh signaling is required for lineage allocation of SSPCs in postnatal skeleton, and whether its dysregulation is related to age-related osteoporosis. Here, we demonstrated that Hh signaling was activated in metaphyseal SSPCs during osteogenic different

Inflammation in adipose tissue and fatty acid anabolism: when enough is enough!

Recent findings in adipose tissue (AT) have uncovered negative interactions among obesity, lipogenesis, and fatty acid (FA) storage, perhaps … hich excessive fat storage, cell density, and diminished oxygen availability promote decreased lipo/adipogenesis and increased lipolysis and FA release. This response may be induced by an important inflammatory component that promotes angiogenesis and insulin resistance, but also by leptin and the increase of T3 in hyperplastic AT.

Dysfunctional adipose tissue-lymphatic crosstalk in obesity.

The lymphatic vasculature is recognized for its roles in immunity, lipid transport and fluid homeostasis, but has been ignored, until recently, for its involvement in adipose tissue metabolism, obesity and associated comorbidities. States of overnutrition, such as in obesity, are associated with impairment of lymphatic function, leading to localized lymphoedema, adipogenesis and insulin resistance. Conversely, compromised lymphatic vasculature integrity can induce adiposity and accelerate insulin resistance. Through the use of advanced 'omics' and bioinformatic technologies coupled with functi…

GPR146 in adipose tissue drives adipose-liver crosstalk and promotes hepatic steatosis in mice.

The limited therapeutic options for metabolic dysfunction-associated steatotic liver disease (MASLD) underscore the need for deeper mechanis … patic lipid accumulation by limiting free fatty acid (FFA) influx. Mechanistically, GPR146 promotes adipogenesis in preadipocytes via Gαq-PKC-AKT signaling, increasing lipid storage capacity, and enhances lipolysis in mature adipocytes through ERK activation, elevating circulating FFA. Together, these coordinated actions increase FFA delivery to the liver, pro …

MYCT1-IFITM2/3 interaction links endothelial endolysosomal trafficking to white adipose tissue expansion.

Vertebrates rely on a network of blood vessels to meet organ demands for oxygen and nutrients. While endothelial cells are known to transport excess nutrients to white adipose tissue (WAT) for energy storage, how their metabolic state impacts this process remains unclear. Here, we identify MYCT1 as a conserved, pan-endothelial protein essential for WAT expansion. Endothelial-specific MYCT1 deletion limited WAT expansion independently of angiogenesis, adipogenesis, or systemic metabolic parameters. Mechanistically, MYCT1 interacted with the transmembrane endolysosomal proteins IFITM2/3 to restr…

Cancer cachexia is a systemic metabolic disorder, with body weight loss and adipose tissue wasting as key features, and adipose tissue remod … y suppression of NF-κB signaling and activation of PPARγ, leading to reduced lipolysis and enhanced adipogenesis; these effects were partially attenuated by the PPARγ antagonist. Moreover, overexpression of OTOP1 in adipose tissue of LLC tumor-bearing mice alleviated adipose tissue wasting and improved lipid metabolism. These findings suggest a role for OTOP1 …

RNA-binding protein OTUD1 maintains skeletal homeostasis by determining mesenchymal stem cell lineage commitment.

Mesenchymal stem cells (MSCs) maintain bone homeostasis through osteogenic differentiation. During aging, MSCs undergo a fate shift toward adipogenesis rather than osteogenesis, but the post-transcriptional mechanisms remain unclear. Here, we identify ovarian tumor domain-containing protein 1 (OTUD1) as an RNA-binding protein that controls MSC fate by stabilizing osteogenic transcripts. OTUD1 directly binds and stabilizes BMP2 mRNA, thereby supporting osteogenic differentiation. In vivo, OTUD1 deficiency does not affect early skeletal development but progressively impairs bone homeostasis duri…

Bioactive structural features of natural polysaccharides: Inspiration for novel polymer strategies against metabolic disorders.

Obesity, diabetes, and hypertension are closely interconnected metabolic disorders that significantly increase the risk of cardiovascular complications and early mortality. Increasing attention has been directed toward natural polysaccharides as potential therapeutic agents due to their multifunctional biological activities and low toxicity. They regulate metabolic homeostasis through multiple pathways, including inhibition of adipogenesis and pancreatic lipase, modulation of carbohydrate-hydrolyzing enzymes, activation of insulin signalling pathways, regulation of the renin-angiotensin system…

Targeted screening of pancreatic lipase inhibitory peptides from quinoa protein hydrolysates by ligand fishing.

A ligand fishing strategy was developed to screen for pancreatic lipase (PL) inhibitory peptides from quinoa protein hydrolysate (QPH) using PL-immobilized Fe₃O₄@SiO₂ magnetic particles. An initial activity of 230 U/g, a PL loading capacity of 180 mg/g, an immobilization yield of 25 %, and retained 39.23 % of its activity after 10 reuse cycles. Three novel peptides (LNEILAHE, VETELEKR, TVSAPSPK) were identified by LC-MS/MS and exhibited moderate PL inhibitory activities with IC₅₀ values ranging from 0.48 to 0.92 mg/mL. They showed dose-dependent anti-adipogenesis effects in 3T3-L1 cells, with …

Histone deacetylase inhibition with givinostat: a multi-targeted mode of action with the potential to halt the pathological cascade of Duchenne muscular dystrophy.

Muscle repair and regeneration are complex processes. In Duchenne muscular dystrophy (DMD), these processes are disrupted by the loss of fun … igenetic changes and inhibition of muscle regeneration factors, chronic inflammation, fibrosis, and adipogenesis. HDAC inhibition has consequently been investigated as a therapeutic approach for muscular dystrophies that, significantly, works independently from specific genetic mutations, making it potentially suitable for all patients with DMD. miR-127-5p-enriched extracellular vesicles modulate the Hippo pathway to counteract steroid-induced osteonecrosis pathology of the femoral head.

Steroid-induced osteonecrosis of the femoral head (SONFH) is a debilitating condition characterized by bone tissue death and joint dysfuncti … teogenesis. EVs derived from SONFH-hBMSCs further inhibited osteogenic differentiation and promoted adipogenesis. However, miR-127-5p-enriched EVs reversed these effects, restoring osteogenic differentiation, improving bone microarchitecture, and enhancing gait performance in SONFH models. These findings suggest that miR-127-5p downregulation plays a critical …
