Pairing mild stress with increased serotonin-1B receptor expression in the nucleus accumbens increases susceptibility to amphetamine.

Both serotonin-1B (5-HT(1B)) receptors and stress modulate the behavioral and neurobiological effects of psychostimulant drugs. In order to … issipated, the resulting effects of this 5-HT(1B)/stress pairing on the acute locomotor response to amphetamine and on the development of psychomotor sensitization were examined. We report that the increasing expression of 5-HT(1B) receptors on the terminals of NAc shell neurons that project to the ventral tegmental area and repeatedly exposing rats to mild s …

Discriminative stimulus and subject-rated effects of methamphetamine, d-amphetamine, methylphenidate, and triazolam in methamphetamine-trained humans.

Methamphetamine abuse is a significant public health concern. Although widely studied in laboratory animals, little is known about the abuse-related behavioral effects of methamphetamine relative to other abused stimulants in controlled laboratory settings in humans. The aim of this study was to examine the discriminative stimulus, subject-rated, performance, and cardiovascular effects of methamphetamine in humans. In the present study, subjects first learned to discriminate 10 mg of oral methamphetamine from placebo. After acquiring the discrimination (> or = 80% drug-appropriate responding o…

Differential behavioral responses to chronic amphetamine in adult male and female rats exposed to postnatal cocaine treatment.

The impact of cocaine exposure during development on behavioral sensitization as measured by locomotor activity and stereotypy following repeated intermittent administration of amphetamine is examined. Male and female Sprague-Dawley rats were exposed to cocaine at 50 mg/kg/day during postnatal days (PND) 11-20 and, as adults (PND193-212), were administered seven daily injections of 2.0 mg/kg amphetamine. Both locomotor activity and stereotypic behavior were assessed following the first and seventh injections. Control males and females showed sensitized behavior following repeated amphetamine i…

Sensitization of amphetamine-induced stereotyped behaviors during the acute response: role of D1 and D2 dopamine receptors.

During the response to an injection of amphetamine, rapid changes occur in the ability of the drug to induce stereotyped behaviors. This enhanced responsivity does not involve changes in the caudate-putamen or nucleus accumbens extracellular dopamine response, but appears to require activation of dopamine receptors. In the present studies we examined the role that D1 and D2 dopamine receptors might play in the development and expression of the enhanced stereotypy response. In one series of experiments we used the dopamine agonists, SKF 82958 and quinpirole as relatively selective probes at D1 …

Modulation of amphetamine-stimulated [3H]dopamine release from rat pheochromocytoma (PC12) cells by sigma type 2 receptors.

An important regulatory mechanism of synaptic dopamine (DA) levels is activation of the dopamine transporter (DAT), which is a target for many drugs of abuse, including amphetamine (AMPH). sigma receptors are located in dopaminergic brain areas critical to reinforcement. We found previously that agonists at sigma2 receptors enhanced the AMPH-stimulated release of [3H]DA from slices of rat caudate-putamen. In the present study, we modeled this response in undifferentiated pheochromocytoma-12 (PC12) cells, which contain both the DAT and sigma2 receptors but not neural networks that can complicat…

Detection of thyrotrophin releasing hormone in rat brain in vivo using novel antibody microprobes: effects of amphetamine.

Antibody microprobes of novel design were used to monitor thyrotrophin releasing hormone (TRH) in rat brain before and after-parenteral administration of amphetamine. Specific antibodies to TRH were bound to the outside of glass microprobes by adsorption to a surface of activated charcoal embedded in epoxylite resin. In male Wistar rats anaesthetised with chloral hydrate a series of antibody microprobes were implanted in forebrain. Amphetamine (10 mg/kg i.p.) caused a highly significant decrease in the binding of 125I-TRH to microprobes, indicating an increase in extracellular TRH localised in…

The selective 5-HT3 receptor antagonist, ondansetron, augments the anorectic effect of d-amphetamine in nondeprived rats.

Previous behavioural studies have shown that 5-hydroxytryptamine3 (5-HT3) receptor antagonists either block or have no effect on amphetamine-induced effects. The present experiments investigated whether or not the highly selective 5-HT3 receptor antagonist ondansetron would affect the anorectic effect of a small dose (1 mg/kg) of d-amphetamine. Nondeprived male rats were tested in two feeding paradigms: consumption of a palatable sweetened mash and ingestion of a 3% sucrose solution. Ondansetron (10-100 micrograms/kg) did not antagonize amphetamine-induced anorexia; instead, in both paradigms …

Serotonergic psychedelics and amphetamine shift male rat ultrasonic vocalizations toward call patterns associated with positively valenced states.

Psychedelics and psychostimulants produce subjective and often positive affective effects in humans. In rats, positive affective states are often associated with frequency-modulated ultrasonic vocalizations (USVs). We examined whether the hallucinogenic 2,5-dimethoxy-4-iodoamphetamine (DOI; 1.25-2.5 mg/kg) and LSD (0.04-0.08 mg/kg), the non-hallucinogenic lisuride (0.03 or 0.3 mg/kg) and the psychostimulant amphetamine (1 mg/kg) modulate USV emission, activity, and head-twitch response (HTR) in male Wistar rats. USVs were detected using DeepSqueak and clustered in a custom, unsupervised manner…

Fatal Coronary Vasculitis and Acute Myocardial Infarction Associated With Chronic Amphetamine Use: A Forensic Autopsy Case Report.

Sudden unexpected death in young adults is a significant challenge in forensic pathology. While atherosclerotic cardiovascular disease is the most common cause of sudden cardiac death, rare chronic inflammatory conditions such as coronary vasculitis are important and often overlooked. Amphetamine use is emerging as a factor linked to severe cardiovascular complications, including arrhythmia, myocardial infarction, and vasculitis. We report the sudden death of a 27-year-old male with no medical history of cardiac illness. Postmortem autopsy revealed acute myocardial infarction with no evidence …

Sex-dependent effects of vasopressin microinjections in CA3 on nucleus accumbens neurochemical modulation and amphetamine-induced behavior.

Addiction to psychostimulants, such as amphetamine (AMPH), disrupts mesolimbic dopaminergic circuits, and effective pharmacological treatments remain limited. AMPH enhances dopamine (DA) release in the nucleus accumbens (NAc), a process tightly linked to context-dependent reinforcement. Emerging evidence implicates the dorsal CA3 subregion of the hippocampus (CA3d) in modulating reward-related processes through its interactions with limbic and mesolimbic circuits. While vasopressin (AVP) signaling has been shown to regulate addictive-like behaviors via extra-hypothalamic regions, its functiona…

Amphetamine use and Parkinson's disease: integration of artificial intelligence prediction, clinical corroboration, and mechanism of action analyses.

Parkinson's disease (PD) is an increasingly prevalent neurologic condition for which symptomatic, but not preventative, treatment is available. Drug repurposing is an innovate drug discovery method that uncovers existing therapeutics to treat or prevent conditions for which they are not currently indicated, a method that could be applied to incurable diseases such as PD. A knowledge graph artificial intelligence prediction system was used to select potential drugs that could be used to treat or prevent PD, and amphetamine was identified as the strongest candidate. Retrospective cohort analysis…

Amphetamine-induced dopamine release in rat: Whole-brain spatiotemporal analysis with [11C]raclopride and positron emission tomography.

Whole-brain mapping of drug effects are needed to understand the neural underpinnings of drug-related behaviors. Amphetamine administration is associated with robust increases in striatal dopamine (DA) release. Dopaminergic terminals are, however, present across several associative brain regions, which may contribute to behavioral effects of amphetamine. Yet the assessment of DA release has been restricted to a few brain regions of interest. The present work employed positron emission tomography (PET) with [11C]raclopride to investigate regional and temporal characteristics of amphetamine-indu…

Pharmacological characterization of 3,4-methylenedioxyamphetamine (MDA) analogs and two amphetamine-based compounds: N,α-DEPEA and DPIA.

3,4-methylenedioxyamphetamine (MDA) is a psychoactive compound chemically related to the entactogen MDMA. MDA shares some of the entactogenic effects of MDMA but also exerts stimulant effects and psychedelic properties at higher doses. Here, we examined the pharmacological properties of MDA analogs and related amphetamine-based compounds detected in street drug samples or in sport supplements. We examined the key pharmacological mechanisms including monoamine uptake inhibition and release using human embryonic kidney 293 cells stably transfected with the respective human transporters. Addition…

Diazepam reverses increased anxiety-like behavior, social behavior deficit, and dopamine dysregulation following withdrawal from acute amphetamine.

Psychostimulants such as amphetamine (AMPH) increase dopamine (DA) release from ventral tegmental area (VTA) neurons, which is associated with their acute reinforcing actions. This positive state is followed by a negative affective state during the withdrawal period each time the drug is taken (i.e., opponent process theory). AMPH withdrawal is accompanied by symptoms of anxiety and depression, which are associated with DA system dysfunction in humans and animal models. Most studies have focused on the negative affective state after withdrawal from chronic drug administration; yet, this negati…

[Determination of six amphetamine-type stimulants in urine samples using electro-enhanced single drop microextraction-gas chromatography].

A method for the simultaneous determination of six amphetamine-type stimulants (ATSs) in urine by electro-enhanced single drop microextraction-gas chromatography (EE-SDME-GC) with a flame ionization detector (FID) was developed. The influences of the EE-SDME parameters on the extraction efficiencies were investigated. The 5 mL-samples at pH 7 were extracted with 2 μL dichloromethane for 3 min under an applied potential of -2.5 V. The analysis was performed on a HP-5 capillary column (30 m×0.32 mm×0.25 μm) using a flame ionization detector. The six ATSs were linear in the range of 20-1000 μg/L …

Polysubstance use in treatment seekers who inject amphetamine: Drug use profiles, injecting practices and quality of life.

BACKGROUND: The injection of amphetamine is becoming increasingly common. However, there has been a lack of research examining people who inject amphetamine as the primary drug of use, limiting the potential to ensure services address the unique needs of this group. The current study used latent class analysis to identify classes of polydrug use among people who report injecting amphetamine during the past 12months. It also examined differences between classes and drug use patterns, injecting practices, quality of life and psychological distress. METHODS: Participants who were attending non-g…
