Reference change values of FIB-4.

When comparing two analytical results for the same analyte, the clinicians may benefit from knowing the reference change values (RCVs) of the analyte. For Fibrosis-4 Index (FIB-4), a noninvasive test used for assessing the risk of liver fibrosis, no RCVs have been published for non-cirrhotic individuals. Therefore, we estimated RCVs for adults, using retrospectively collected data from outpatients with AST, ALT, and thrombocytes within the respective reference intervals. FIB-4 was calculated as (age × AST)/(thrombocytes × ALT0.5). From two FIB-4 values in each patient we calculated the RCVs pa

Probabilistically Weighted Multilayer Networks disclose the link between default mode network instability and psychosis-like experiences in healthy adults.

The brain is a complex system in which the functional interactions among its subunits vary over time. The trajectories of this dynamic variation contribute to inter-individual behavioral differences and psychopathologic phenotypes. Despite many methodological advancements, the study of dynamic brain networks still relies on biased assumptions in the temporal domain. The current paper has two goals. First, we present a novel method to study multilayer networks: by modelling intra-nodal connections in a probabilistic, biologically driven way, we introduce a temporal resolution of the multilayer

Maternal biological age assessed in early pregnancy is associated with gestational age at birth.

Maternal age is an established predictor of preterm birth independent of other recognized risk factors. The use of chronological age makes the assumption that individuals age at a similar rate. Therefore, it does not capture interindividual differences that may exist due to genetic background and environmental exposures. As a result, there is a need to identify biomarkers that more closely index the rate of cellular aging. One potential candidate is biological age (BA) estimated by the DNA methylome. This study investigated whether maternal BA, estimated in either early and/or late pregnancy,

Association of salivary levels of the bone remodelling regulators sRANKL and OPG with periodontal clinical status.

AIM: To determine the variations in salivary concentrations of sRANKL, osteoprotegerin (OPG) and its ratio, regarding the periodontal status. MATERIALS AND METHODS: Ninety-seven chronic periodontitis (CP) subjects and 43 healthy controls were selected. Periodontal status was assessed based on full-mouth clinical periodontal measurements. sRANKL and OPG salivary levels were analysed by ELISA. The association between these analytes and its ratio with CP was analysed individually and adjusted for confounding using a binary logistic regression model. RESULTS: sRANKL and sRANKL/OPG ratio were inc

A field study of the exposure-annoyance relationship of military shooting noise.

This article reports a field study on noise annoyance from military shooting with small, midsize, and heavy weapons that was carried out among 1002 residents living near eight different training grounds of the Swiss army. The goal of the study was to derive the exposure-annoyance relationship for military shooting noise in communities in the vicinity of average military training grounds. Annoyance was determined in a telephone survey by means of the 5-point verbal and 11-point numerical annoyance scale recommended by the International Commission on Biological Effects of Noise. Exposure was cal

Spinal stenosis: surgical versus nonsurgical treatment.

UNLABELLED: Lumbar spinal stenosis is being diagnosed increasingly in older people as advanced imaging studies become more widely available and clinicians become more familiar with the presentation of this common condition. For symptomatic patients, the goal of treatment is pain relief and improved function, but there is little empiric evidence to support many of the common interventions used. The relative risks and benefits of various surgical and nonsurgical treatments are poorly understood, and the result has been wide variations in the evaluation and treatment of spinal stenosis across geo

The physiological stress response and the role of cognitive coping in migraine patients and non-headache controls.

Our study investigated physiological response specificity and cognitive coping in migrainous patients during an achievement task situation. Thirty-three migrainous subjects and thirty-two non-headache controls were subjected to 40 min of demanding cognitive tasks and 20 min of recovery during which cranial and peripheral vasomotor responses were registered as well as electrodermal and myographic activity. Subjects of both groups were randomly assigned to two experimental conditions, a so-called 'spontaneous processing condition' and the 'positive coping treatment'. In this treatment condition

[Blood amylase: a biological marker in irradiation accidents? Preliminary results obtained at the Gustave-Roussy Institut (GRI) and a literature review].

The retrospective evaluation of the dose after an irradiation accident is of paramount importance; it allows an adequate selection of patients and the most appropriate treatment can then be proposed. Classical physical dosimetry often lacks precision for dose assessment in such accidents. Cytogenetics, usually more reliable, is not 100% accurate and cannot be used in some particular instances. At the Institut Gustave-Roussy, we studied amylasemia in 15 patients who received a total body irradiation (TBI) for bone marrow grafting, at various dose levels (10, 2 and 1.35 Gy). Hyperamylasemia was

Microbiome Engineering in Dairy Cattle: A Critical Review of Strategies for Disease Resistance, Productivity, and Sustainable Farming.

Dairy production currently faces three converging challenges: the escalation of antimicrobial resistance (AMR), rising global food demand, and stricter regulatory requirements for reducing enteric methane emissions. This review evaluates probiotics, prebiotics, fecal microbiota transplantation (FMT), metagenomic tools, and Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR)-based synthetic biology for dairy cow microbiome engineering, applying a Technology Readiness Level (TRL) 1-9 framework to assess the translational maturity of each strategy. The quantitative ranges below are

Quantitative assessment of neonatal health using dried blood spot metabolite profiles and deep learning.

Neonatal prematurity leads to considerable morbidity and mortality, partly because of acquired conditions such as bronchopulmonary dysplasia (BPD), intraventricular hemorrhage (IVH), necrotizing enterocolitis (NEC), and retinopathy of prematurity (ROP). Standard gestational age and birthweight-based classifications of prematurity inadequately capture the variation in newborns' health outcomes, creating an urgent need to develop risk stratification tools for vulnerable newborn infants to initiate the most appropriate care pathways as early as possible. We hypothesized that the metabolic profile

Estimating Accelerated Retinal Decline in Mental Health Disorders Through Normative Modeling.

BACKGROUND AND HYPOTHESIS: Several studies have found that retinal tissue is thinner in people with mental health disorders than in healthy controls (HC). However, most research has examined group-level differences and has therefore inadequately accounted for normative biological heterogeneity. A focus on single disorders has also made transdiagnostic comparisons challenging. We used normative modeling to estimate age-related decline of the human retina (and its expected variation) and compared it to the decline seen in schizophrenia (SZ), bipolar disorder (BD), and major depressive disorder (

The influence of temperament and perinatal factors on language development: a longitudinal study.

Early language development is characterized by large individual variation. Several factors were proposed to contribute to individual pathways of language acquisition in infancy and childhood. One of the biologically based explaining factors is temperament, however, the exact contributions and the timing of the effects merits further research. Pre-term status, infant sex, and environmental factors such as maternal education and maternal language are also involved. Our study aimed to investigate the longitudinal relationship between infant temperament and early language development, also conside

Effect of Pathway-Specific Polygenic Risk Scores for Alzheimer's Disease (AD) on Rate of Change in Cognitive Function and AD-Related Biomarkers Among Asymptomatic Individuals.

BACKGROUND: Genetic scores for late-onset Alzheimer's disease (LOAD) have been associated with preclinical cognitive decline and biomarker variations. Compared with an overall polygenic risk score (PRS), a pathway-specific PRS (p-PRS) may be more appropriate in predicting a specific biomarker or cognitive component underlying LOAD pathology earlier in the lifespan. OBJECTIVE: In this study, we leveraged longitudinal data from the Wisconsin Registry for Alzheimer's Prevention and explored changing patterns in cognition and biomarkers at various age points along six biological pathways. Comparing the Gut Microbiome in Autism and Preclinical Models: A Systematic Review.

Many individuals diagnosed with autism spectrum disorder (ASD) experience gastrointestinal (GI) dysfunction and show microbial dysbiosis. Variation in gut microbial populations is associated with increased risk for GI symptoms such as chronic constipation and diarrhoea, which decrease quality of life. Several preclinical models of autism also demonstrate microbial dysbiosis. Given that much pre-clinical research is conducted in mouse models, it is important to understand the similarities and differences between the gut microbiome in humans and these models in the context of autism. We conducte

Identity-by-descent analysis of a large Tourette's syndrome pedigree from Costa Rica implicates genes involved in neuronal development and signal transduction.

Tourette Syndrome (TS) is a heritable, early-onset neuropsychiatric disorder that typically begins in early childhood. Identifying rare genetic variants that make a significant contribution to risk in affected families may provide important insights into the molecular aetiology of this complex and heterogeneous syndrome. Here we present a whole-genome sequencing (WGS) analysis from the 11-generation pedigree (>500 individuals) of a densely affected Costa Rican family which shares ancestry from six founder pairs. By conducting an identity-by-descent (IBD) analysis using WGS data from 19 individ

YAP1-FAM118B Fusion Defines a Rare Subset of Childhood and Young Adulthood Meningiomas.

Meningiomas are a central nervous system tumor primarily afflicting adults, with <1% of cases diagnosed during childhood or adolescence. Somatic variation in NF2 may be found in ∼50% of meningiomas, with other genetic drivers (eg, SMO, AKT1, TRAF7) contributing to NF2 wild-type tumors. NF2 is an upstream negative regulator of YAP signaling and loss of the NF2 protein product, Merlin, results in YAP overexpression and target gene transcription. This mechanism of dysregulation is described in NF2-driven meningiomas, but further work is necessary to understand the NF2-independent mechanism of tum

Probing the clinical and brain structural boundaries of bipolar and major depressive disorder.

Bipolar disorder (BD) and major depressive disorder (MDD) have both common and distinct clinical features, that pose both conceptual challenges in terms of their diagnostic boundaries and practical difficulties in optimizing treatment. Multivariate machine learning techniques offer new avenues for exploring these boundaries based on clinical neuroanatomical features. Brain structural data were obtained at 3 T from a sample of 90 patients with BD, 189 patients with MDD, and 162 healthy individuals. We applied sparse partial least squares discriminant analysis (s-PLS-DA) to identify clinical and

Exome Sequencing of a Primary Ovarian Insufficiency Cohort Reveals Common Molecular Etiologies for a Spectrum of Disease.

CONTEXT: Primary ovarian insufficiency (POI) encompasses a spectrum of premature menopause, including both primary and secondary amenorrhea. For 75% to 90% of individuals with hypergonadotropic hypogonadism presenting as POI, the molecular etiology is unknown. Common etiologies include chromosomal abnormalities, environmental factors, and congenital disorders affecting ovarian development and function, as well as syndromic and nonsyndromic single gene disorders suggesting POI represents a complex trait. OBJECTIVE: To characterize the contribution of known disease genes to POI and identify mol
