Association of NAT2 genetic polymorphism with the efficacy of Neurotropin® for the enhancement of aggrecan gene expression in nucleus pulposus cells: a pilot study.

BACKGROUND: Intervertebral disc degeneration, one of the major causes of low-back pain, results from altered biosynthesis/turnover of extracellular matrix in the disc. Previously, we reported that the analgesic drug Neurotropin® (NTP) had an anabolic effect on glycosaminoglycan synthesis in cultured nucleus pulposus (NP) cells via the stimulation of chondroitin sulfate N-acetylgalactosaminyltransferase 1. However, its effect on the aggrecan core protein was not significantly detected, because of the data variance. A microarray analysis suggested that the effect of NTP on aggrecan was correlate

Papillary Renal Neoplasm with Reversed Polarity in a 64-Year-Old Male: A Rare Case Report and Review of the Literature.

BACKGROUND: Papillary renal neoplasm with reversed polarity is a rare entity, with its latest definition established only in 2019. Given its rarity, with less than 100 reported cases, further characterization is needed to enhance diagnostic accuracy and inform management strategies. CASE PRESENTATION: We report a case in a 64-year-old Chinese male with an incidentally discovered a size of 5.2cm×4.0cm left renal mass on imaging. The patient has a medical history of coronary heart disease and left lower limb numbness. Further evaluation via contrast-enhanced CT confirmed that a nodular slightly

Fine structural studies on a type of somatostatin-immunoreactive neuron and its synaptic connections in the rat neostriatum: a correlated light and electron microscopic study.

Somatostatin-immunoreactive neurons in the rat neostriatum were studied by correlated light and electron microscopy using the peroxidase-antiperoxidase immunocytochemical technique. Immunoreactivity was localized in neuronal perikarya and processes. The perikarya were of spindle or fusiform shape (average length 16.9 microns) and were found in all parts of the neostriatum. From each neuron there arose two to four straight immunoreactive dendritelike processes, which could frequently be traced as far as about 130 microns from their perikaryon. Immunoreactive varicose axonlike processes were occ

Fluorescent Probes for Sensing and Imaging within Specific Cellular Organelles.

Fluorescent probes have become powerful tools in biosensing and bioimaging because of their high sensitivity, specificity, fast response, and technical simplicity. In the last decades, researchers have made remarkable progress in developing fluorescent probes that respond to changes in microenvironments (e.g., pH, viscosity, and polarity) or quantities of biomolecules of interest (e.g., ions, reactive oxygen species, and enzymes). All of these analytes are specialized to carry out vital functions and are linked to serious disorders in distinct subcellular organelles. Each of these organelles p

Antiviral activity of human immunodeficiency virus type 1 protease inhibitors in a single cycle of infection: evidence for a role of protease in the early phase.

The antiviral activities of two substrate-based inhibitors of human immunodeficiency virus type 1 (HIV-1) protease, UK-88,947 and Ro 31-8959, were studied in acute infections. H9 and HeLaCD4-LTR/beta-gal cells were infected either with HIV-1IIIB or a replication-defective virus, HIV-gpt(HXB-2). Both inhibitors were capable of blocking early steps of HIV-1 replication if added to cells prior to infection. Partial inhibition was also obtained by addition of inhibitor at the time of or as late as 15 min after infection. The inhibitors were ineffective if added 30 min postinfection. The inhibitory

Exosomal miR-223-3p from mesenchymal stem cells targets FBXW7 to inhibit intervertebral disc degeneration: mechanism insights.

BACKGROUND: Intervertebral disc degeneration (IDD) represents a widespread musculoskeletal condition. Programmed cell death in nucleus pulposus cells (NPCs) significantly contributes to IDD pathogenesis. MicroRNAs (miRNAs) play critical roles in IDD development. Bone marrow mesenchymal stem cell (MSC)-derived exosomes can inhibit NPCs apoptosis and promote disc regeneration/repair by delivering miRNAs. METHODS: Rat bone marrow-derived mesenchymal stem cells (MSCs) were expanded in vitro, followed by exosome isolation via differential ultracentrifugation. Exosome characterization included asse

In vivo characterization of a retinal cellular biomarker of inflammation in multiple sclerosis.

Neuroinflammation plays a central role in the progression of multiple sclerosis (MS). While neuroinflammation contributes to neuronal damage, it also has regenerative roles, highlighting the complex relationship between immune responses and neurodegeneration. However, capturing the cellular processes involved in inflammation and regeneration in vivo remains a challenge. The retina, a part of the CNS tissue, provides a unique, non-invasive window into these processes. High-resolution retinal imaging techniques, such as adaptive optics scanning laser ophthalmoscopy (AOSLO), enable the observatio

Episodic exposure to ethanol during development differentially affects brainstem nuclei in the macaque.

Neuronal vulnerability to ethanol may be non-specific, i.e., vulnerability may be conferred by the developmental state of the population or by the site of derivation. To address these issues, the effect of developmental exposure to ethanol on three brainstem nuclei; the trigeminal motor (MoV), facial motor (MoVII) and medial superior olivary (MSO) nuclei was determined. MoVII and MSO are generated at the same time and from the same rhombomere, r4. MoV is generated earlier from r2. Macaca nemestrina were exposed to ethanol or a control solution one day per week for six or 24 weeks of gestation.

Two Pathways of p27Kip1 Degradation Are Required for Murine Lymphoma Driven by Myc and EBV Latent Membrane Protein 2A.

Epstein-Barr virus (EBV) latent membrane protein 2A (LMP2A), expressed in EBV latency, contributes to Burkitt lymphoma (BL) development in a murine model by acting as a constitutively active B cell receptor (BCR) mimic. Mice expressing both LMP2A and MYC transgenes (LMP2A/λ-MYC) develop tumors significantly faster than mice only expressing MYC (λ-MYC). Previously, we demonstrated the cell cycle inhibitor p27Kip1 is present at significantly lower levels in LMP2A/λ-MYC mice due to increased posttranslational degradation. P27Kip1 degradation can occur in the cytoplasm following phosphorylation on

NADPH-diaphorase activity in the olfactory system of the hamster and rat.

A comparative analysis of nicotinamide adenine dinucleotide phosphate (NADPH)-diaphorase activity in the olfactory bulb was conducted in the hamster and rat. The distribution and morphological features of NADPH-stained neurons were compared to those of glutamic acid decarboxylase-like (GAD-LI) and tyrosine hydroxylase-like (TH-LI) immunoreactive somata in order to relate NADPH-staining to neuronal classes with specific biochemical properties. Intense NADPH-staining was located in primary nerve fibers of the accessory and main olfactory systems, producing dense staining of individual glomeruli.

Evaluation of Histidine-Octamer-Modified Hyaluronic Acid as a Cytosolic Drug Delivery Material via CD44-Mediated Cellular Uptake and Endosomal Escape.

Background: Although intracellular delivery via endocytosis is a promising strategy for drugs acting in the cytosol or nucleus, many macromolecular therapeutics remain trapped within endosomal/lysosomal compartments, limiting efficient cytosolic delivery. In this study, we designed a novel functional hyaluronic acid (HA)-based polymer, HA-octahistidine (His8), by conjugating His8 to CD44-targeting HA, and evaluated its physicochemical properties, cellular uptake, and endosomal escape capability. Methods: HA-His8 was synthesized via 1-ethyl-3-(3-dimethylaminopropyl)carbodiimide/N-hydroxysuccini

Prognostic significance of tumor budding in pancreatic carcinoma: Digitalized image approach evaluation using artificial intelligence.

INTRODUCTION: Pancreatic carcinoma (PC) is a highly malignant and lethal tumor characterized by a dismal prognosis which raised the need to identify other prognostic factors for better patient risk stratification. Tumor budding (TB), defined as isolated single cancer cells or small clusters of up to four cells at the invasive front, is an emerging histoprognostic factor associated with aggressiveness in various malignancies. This study investigated the prognostic significance of tumor budding (TB) in pancreatic carcinoma using artificial intelligence. METHODS: In this retrospective multicente

Changes in Properties of Auditory Nerve Synapses following Conductive Hearing Loss.

UNLABELLED: Auditory activity plays an important role in the development of the auditory system. Decreased activity can result from conductive hearing loss (CHL) associated with otitis media, which may lead to long-term perceptual deficits. The effects of CHL have been mainly studied at later stages of the auditory pathway, but early stages remain less examined. However, changes in early stages could be important because they would affect how information about sounds is conveyed to higher-order areas for further processing and localization. We examined the effects of CHL at auditory nerve syna

How does the brain rapidly learn and reorganize view-invariant and position-invariant object representations in the inferotemporal cortex?

All primates depend for their survival on being able to rapidly learn about and recognize objects. Objects may be visually detected at multiple positions, sizes, and viewpoints. How does the brain rapidly learn and recognize objects while scanning a scene with eye movements, without causing a combinatorial explosion in the number of cells that are needed? How does the brain avoid the problem of erroneously classifying parts of different objects together at the same or different positions in a visual scene? In monkeys and humans, a key area for such invariant object category learning and recogn

Intracellular Nanopharmacology of Trastuzumab Deruxtecan Reveals Lysosome-Centered Organelle Vulnerabilities in HER2-Positive Breast Cancer Cells.

Trastuzumab-deruxtecan (T-DXd) is a clinically effective antibody-drug conjugate (ADC) with activity across HER2-amplified, HER2-low, and ultralow breast cancers. Despite its clinical success, how its nanoscale intracellular trafficking and processing shape therapeutic outcomes remains incompletely defined. Given the central role of HER2 internalization and lysosomal processing in ADC pharmacology, we investigated the spatiotemporal intracellular response to T-DXd in human HER2-positive breast cancer cell models. By integrating biochemical analyses with light and electron microscopy-based nano

Proteomic identification of non-erythrocytic alpha-spectrin-1 down-regulation in the pre-optic area of neonatally estradiol-17β treated female adult rats.

It is well established that sexually dimorphic brain regions, which are critical for reproductive physiology and behavior, are organized by steroid hormones during the first 2 weeks after birth in the rodents. In our recent observation, neonatal exposure to estradiol-17β (E2) in the female rat revealed increase in cyclooxygenase 2 (COX-2) level, sexually dimorphic nucleus (SDN)-pre-optic area (POA) size and down-regulation of synaptogenesis related genes in POA in the adult stage. In the present study, using the same animal model, the protein profile of control and neonatally E2-treated POA wa

Zinc enhances autophagic flux and lysosomal function through transcription factor EB activation and V-ATPase assembly.

The stimulation of autophagy or lysosomes has been considered therapeutic for neurodegenerative disorders because the accumulation of misfolded proteins is commonly observed in the brains of individuals with these diseases. Although zinc is known to play critical roles in the functions of lysosomes and autophagy, the mechanism behind this regulatory relationship remains unclear. Therefore, in this study, we examined which mechanism is involved in zinc-mediated activation of autophagy and lysosome. Exposure to zinc at a sub-lethal concentration activated autophagy in a concentration-dependent m

Modulation of proopiomelanocortin gene expression by ethanol in mouse anterior pituitary corticotrope tumor cell AtT20.

In humans, alcoholism is associated with a decrease in basal ACTH and cortisol levels, and blunted pituitary ACTH responses to administered corticotropin-releasing hormone (CRH) during active drinking and after long-term abstinence. Preclinical studies indicate that a persistent decrease in pituitary activation after chronic exposure to ethanol is due to a direct effect of ethanol on the corticotrope of the anterior pituitary. The present studies were undertaken to determine if ethanol has effects on proopiomelanocortin (POMC) gene transcription activity in mouse anterior pituitary corticotrop

The Yield-Associated TaCOL-B5 Locus Activates Its Tightly Linked Target TaDRS1 to Restrict Root Apical Meristem Proliferation and Deep Rooting in Wheat.

Rooting depth underpins subsoil water capture and drought resilience in wheat, yet the regulators coupling root apical meristem (RAM) activity to vertical root growth remain largely unknown. We characterise Deep Root Suppressor 1 (TaDRS1; TraesCS7B01G400700), a transcriptional target of the yield-associated CONSTANS-like locus TaCOL-B5 lying only 32 kb away on chromosome 7B. In the common recessive (Tacol-B5) background of most modern wheat, chromatin immunoprecipitation-qPCR and dual-luciferase assays show that the Tacol-B5 protein occupies a CCAAT-box region of the TaDRS1 promoter and activa
