Chromosomal abnormalities and their clinical significance in acute lymphoblastic leukemia. Third International Workshop on Chromosomes in Leukemia.

Three hundred thirty newly diagnosed patients were studied to determine the frequency and type of chromosomal abnormalities in acute lymphoblastic leukemia (ALL) and their clinical significance. Analyses of banded chromosomes revealed clonal chromosomal abnormalities in 218 patients (66%), including all cases of B-ALL; and 70% of non-T, non-B ALL; but only 39% of T-ALL (p less than 0.001). Patients were classified into 10 groups according to karyotype: no abnormalities (34%), one of the following recurring structural abnormalities [the Philadelphia chromosome (12%), t(4;11) (5%), t(8;14) (5%),

Premature centromere division of a translocation-carrier autosome.

Premature centromere division of an aberrant chromosome 3 was found in a newborn girl with fetal hydrops and in her healthy father, both bearing a familial "balanced" 3p;19q translocation. The out-of-phase separation of a structurally abnormal chromosome may be of pathogenetic significance, the nature of which is not yet understood.

[Results of chromosomal studies in 270 couples with reproductive problems].

Cytogenetic study in couples with recurrent reproductive failure allows the detection of structural chromosome anomalies which don't have a phenotypic effect in carriers, but permit the explanation of the production of genetically unbalanced gametes. We present the investigation performed in 270 couples which demonstrated a chromosomal alteration in 9.2% of them. This allows genetic counselling in respect of the importance of the finding, recurrence risk and reproductive alternatives. In 8.6% we found some cellular lines with aneuploidies in sexual chromosomes, which don't represent a such con

Phenotype of two males with abnormal Y chromosomes.

Two infertile males with sex chromosomal abnormalities and mosaic karyotype, 45,X/46,X,dic(Yq) and 45,X/46,X,ring(Y), had considerably changed physical findings, including tooth sizes and craniofacial dimensions. Spermatogenesis was preserved with abnormal meiotic chromosomal behaviour. Mosaic karyotype and structurally changed Y chromosome in both cases had an influence on physical parameters. Tests were normally developed and spermatogenesis was preserved but depressed in later stages.

[Chromosome abnormalities in bone marrow cells and peripheral lymphocytes in a patient with Fanconi's anemia].

In direct preparations of bone marrow cells from an 18 years old patient with Fanconi's anaemia (FA) a low frequency of structural chromosome aberrations was observed (4% excluding gaps), while in cultured bone marrow cells (48 hours) and in cultured peripheral lymphocytes such aberrations were found to be about 3 times as many. The increase of chromosome abnormalities "in vitro" is discussed and the most recent observations on possible factors determining susceptibility to chromosome breakage in cases with FA are taken into account.

Epithelial neoplasm of the colorectum: correlation of histopathological features with cytogenetic studies.

Seven cases of primary rectosigmoid epithelial neoplasm, including a tubular adenoma, a villous adenoma with malignant change and five adenocarcinomas, were cytogenetically studied with short-term cultivation and GTG-banding. The tubular adenoma showed karyotypic normality. The other six malignant tumors were hyperdiploid in chromosomal modal number and showed multiple aberrations. The complexity of chromosomal aberrations increased proportionally to the pathologic stage. No common chromosome change to all of the tumors was found. The most common recurrent chromosome changes were extra gains o

The Biology and Evolution of Mammalian Y Chromosomes.

Mammals have the oldest sex chromosome system known: the mammalian X and Y chromosomes evolved from ordinary autosomes beginning at least 180 million years ago. Despite their shared ancestry, mammalian Y chromosomes display enormous variation among species in size, gene content, and structural complexity. Several unique features of the Y chromosome--its lack of a homologous partner for crossing over, its functional specialization for spermatogenesis, and its high degree of sequence amplification--contribute to this extreme variation. However, amid this evolutionary turmoil many commonalities h

Y chromosome haplogroups in autistic subjects.

The male to female ratio in autism is 4:1 in the global autistic population, but increases to 23:1 in autistic subjects without physical or brain abnormalities.(1) Despite this well-recognised gender difference, male predisposition to autistic disorder remains unexplained and the role of sex chromosomes is still debated. Numerical and structural abnormalities of the sex chromosomes are among the most frequently reported chromosomal disorders associated with autism. However, genome scans have failed to detect linkage on the X chromosome(2,3,4) and this approach cannot study the non-recombining

Complex genomic alterations and intellectual disability: an interpretative challenge.

BACKGROUND: Complex chromosomal rearrangements (CCRs) are structural rearrangements involving more than three chromosomes or having more than two breaks; approximately 70% are not associated with any clinical phenotype. Here, we describe a CCR segregating in a two-generation family. METHOD: A 4-year-old male was evaluated for developmental delay, mild intellectual disability and epicanthus. Karyotype, fluorescence in situ hybridisation (FISH) analysis and array comparative genomic hybridisation (aCGH) analysis were performed on the patient and of all family members. RESULT: Array CGH analysi

Chromosome abnormalities in diffuse large B-cell lymphomas: analysis of 231 Chinese patients.

Genome instability is a hallmark of cancer. Diffuse large B-cell lymphoma (DLBCL) is the most common form of non-Hodgkin lymphoma with high levels of chromosomal aberrations. The purpose of this study was to characterize chromosomal aberrations in Chinese DLBCL patients and to compare chromosomal abnormalities between germinal centre B-cell-like (GCB) and non-GCB subgroups. Fluorescence in situ hybridization, G-band cytogenetics and immunohistochemistry were performed in 231 cases of de novo DLBCL. We demonstrated that the rate of abnormal and complex karyotypes was 89.1% (139/156) and 92.8% (

Chromosome abnormalities in adult T-cell leukemia/lymphoma: a karyotype review committee report.

Karyotypes of 107 cases with adult T-cell leukemia/lymphoma (58 male, 49 female; 81 acute or lymphoma type, 26 chronic or smoldering type) were reviewed by a panel of cytogeneticists and were correlated with the subtypes of the disease. Clonal chromosome abnormalities were found in 103 (96%) cases, of which four had hypotetraploidy. Of 184 numerical abnormalities in the remaining 99 cases with near- or pseudodiploidy, trisomies for chromosomes 3 (21% of cases), 7 (10%), and 21 (9%), monosomy for X chromosome (38%) in the female, and loss of a Y chromosome (17%) in the male were more frequent t

Lymphocyte chromosome studies in humans exposed to chemical mutagens. The validity of the method in 67 patients under cytostatic therapy.

Eighty-three lymphocyte cultures from 67 patients exposed to high therapeutic doses of chemical mutagens and from 10 healthy controls were examined for structural aberrations of the chromatid type (gaps, breaks and exchanges) and chromosome type (rings, dicentrics, acentric fragments and abnormal chromosomes) in order to evaluate the reliability of this testing system. Most of the patients had received several drugs; a few had radiotherapy as well. Owing to insufficient yields of mitoses at shorter incubation periods a culture time of 72 h had to be chosen. Whenever possible, 100 mitoses were

Partial Trisomy and Partial Monosomy 21 Undetected by NIPT and FISH: A Case Report and Literature Review.

Non-invasive prenatal testing (NIPT) is a screening method that analyzes cell-free DNA (cfDNA) in maternal plasma to detect major aneuploidies. However, NIPT can fail to identify subtle chromosomal aberrations. We report a case in which chromosomal microarray analysis (CMA) revealed partial trisomy and partial monosomy of chromosome 21, despite negative results with both NIPT and fluorescence in situ hybridization (FISH). The neonate presented with characteristic phenotypes consistent with Down syndrome (DS). The same chromosomal imbalance was confirmed by postnatal peripheral blood CMA. This

[Genetic analysis of a family with super-male syndrome].

OBJECTIVE: To identify the genetic cause for a family featuring language retardation using combined cytogenetic and molecular genetic methods. METHODS: Following conventional G-banded karyotype analysis, the additional Y chromosome was identified by fluorescence in situ hybridization (FISH) and multiplex ligation dependent probe amplification (MLPA). Whole genome array comparative genomic hybridization (aCGH) was also carried out to detect minor structural chromosomal abnormalities. RESULTS: The proband's karyotype was determined as 47,XY,+?, and the unknown aberrant chromosome was identifie

Molecular cytogenetic analysis of a duplication Xp in a female with an abnormal phenotype and random X inactivation.

We describe a female infant with severe abnormal phenotype with a de novo partial duplication of the short arm of the X chromosome. Chromosome painting confirmed the origin of this X duplication. Molecular cytogenetic analysis with fluorescence in situ hybridization (FISH) was performed with YAC probes, further delineating the breakpoints. The karyotype was 46, X dup(X)(p11-p21.2). Cytogenetic replication studies showed that the normal and duplicated X chromosomes were randomly inactivated in lymphocytes. In most females with structurally abnormal X chromosomes, the abnormal chromosome is inac

[Pericentric inversion of the Y chromosome as an exclusion constellation].

A 37 year old second gravida received amniocentesis, because of advanced maternal age. Cytogenetic investigations of the fetal cells revealed a male karyotype with a pericentric inversion of the Y chromosome. Chromosome analysis of the father was also undertaken in order to decide whether this inversion was inherited or of de novo origin. The results of cytogenetic examinations of the mother as well as of two alleged fathers enabled us to detect an exclusion of paternity for one man and a positive hint of paternity for the other. The cytogenetic markers used were the inversion itself and a hig

Genomic organization and chromosomal localization of mouse proteinase 3 (Myeloblastin).

Proteinase 3 (PR3), is a matrix-degrading serine proteinase expressed in different hematopoietic cell lineages. The PR3 protein appears to regulate the myeloid differentiation and was found to be the autoantigen associated with Wegener granulomatosis. We have isolated and characterized the gene for mouse PR3 (mPR3) and determined its chromosomal location. The gene has been localized to Chromosome (Chr) 10. Comparison of mouse PR3 genomic structure with that of its human counterpart indicates that: 1) the mPR3 gene spans 7 kb organized in 5 exons and 4 introns, 2) the codons of His-Asp-Ser of t

Chromosomal alterations in lymphocytes of patients with Balkan endemic nephropathy and of healthy individuals after incubation in vitro with ochratoxin A.

The possible involvement of mycotoxins in chromosomal alterations in patients with Balkan endemic nephropathy (BEN) was investigated cytogenetically. Lymphocyte cultures from patients with BEN and from individuals from a nonendemic region were examined and compared with cultures from healthy people which had been incubated in vitro with noncytotoxic doses of ochratoxin A. Significantly increased numbers of various numerical and structural anomalies were found in patients with BEN. Chromosome X in female patients occurred in both monosomic and polysomic forms. A 'prosomization' effect was seen

Phenotypic expansion and structural analysis of the IQSEC2 p.Asp894Asn variant in a consanguineous Pashtun family.

X-linked Intellectual Disability (XLID) is one of the heterogenous neurodevelopmental disorders caused by a gene defect on the X chromosome. Clinical symptoms of ID are comprised of the disability of adapting to social environments and cognitive dysfunction which is often defined by having an IQ of less than 70. Whole exome sequencing revealed a hemizygous variant c.2680G > A (p.Asp894Asn) in IQSEC2 in the proband, further validated by Sanger sequencing. Subsequently, the three-dimensional structures of the wild and mutated type (Asp894Asn) IQSEC2 were deduced by structural bioinformatics appr
