Accelerated functional losses in ageing congenital Minamata disease patients.

Severe methylmercury poisoning occurred in Minamata and neighboring communities in Japan during the 1950s and 1960s. A considerable number of children were born with conditions resembling cerebral palsy, later known as congenital Minamata disease. Although surviving patients are now in their 50s or 60s, few assessments of functional capacities in daily living have been performed. We assessed the changes in Activities of Daily Living (ADL) status of 11 patients over a 10-year period. For assessment of ADL, we applied the Barthel Index (BI) and the Functional Independence Measure (FIM). We obtai

Digital maze test reveals cognitive performance patterns associated with amyloid-β and tau.

OBJECTIVE: Digital cognitive tools provide novel ways to examine subtle cognitive processes not captured with traditional measures. This is particularly useful in examining subtle cognitive changes in early Alzheimer's disease (AD) stages. Here, we evaluate a digital maze test and its association to AD biomarkers in a largely cognitively unimpaired sample. METHOD: A total of 221 participants in the Harvard Aging Brain Study and affiliated studies completed a digital maze test, and neuropsychological battery including the trail making test. Cortical Amyloid-β and tau burden were visualized wit

Cerebral Haemodynamic Changes in Type 2 Diabetes Mellitus Following a Three-Month Yoga Intervention: A Randomized Controlled Trial.

Background and purpose Cerebral haemodynamics and cognitive performance may be adversely affected in type 2 diabetes mellitus (T2DM). Previous studies reported reduced cerebral blood flow (CBF) and altered cerebrovascular reactivity (CVR) in T2DM. Yoga, an ancient holistic health approach, is known to be beneficial for T2DM. We hypothesized that yoga practice may alter CBF and the flow resistance in the middle cerebral artery (MCA) and improve cognition in T2DM. Our secondary objective was to explore the relationship between changes in cerebral haemodynamics and cognition in T2DM. Materials an

What's in a score: A longitudinal investigation of scores based on item response theory and classical test theory for the Amsterdam Instrumental Activities of Daily Living Questionnaire in cognitively normal and impaired older adults.

OBJECTIVE: We aimed to investigate whether item response theory (IRT)-based scoring allows for a more accurate, responsive, and less biased assessment of everyday functioning than traditional classical test theory (CTT)-based scoring, as measured with the Amsterdam Instrumental Activities of Daily Living Questionnaire. METHOD: In this longitudinal multicenter study including cognitively normal and impaired individuals, we examined IRT-based and CTT-based score distributions and differences between diagnostic groups using linear regressions, and investigated scale attenuation. We compared chan

Effects of traumatic brain injury and posttraumatic stress disorder on Alzheimer's disease in veterans, using the Alzheimer's Disease Neuroimaging Initiative.

Both traumatic brain injury (TBI) and posttraumatic stress disorder (PTSD) are common problems resulting from military service, and both have been associated with increased risk of cognitive decline and dementia resulting from Alzheimer's disease (AD) or other causes. This study aims to use imaging techniques and biomarker analysis to determine whether traumatic brain injury (TBI) and/or PTSD resulting from combat or other traumas increase the risk for AD and decrease cognitive reserve in Veteran subjects, after accounting for age. Using military and Department of Veterans Affairs records, 65 …

Altered temporal features of visual processing in subjective cognitive decline.

OBJECTIVE: Subjective cognitive decline (SCD) is possibly the earliest known stage of Alzheimer's disease. Individuals with SCD report perceiving a decline in their cognitive functioning and express concern about it. However, when assessed with standard neuropsychological tests, they perform within normal limits. METHOD: The present study focuses on the temporal features of visual processing in SCD using the random temporal sampling technique. This method reveals the perceptual oscillations which occur during a visual task by presenting stimuli with randomly varying signal-to-noise ratio thro

Decoupling of local activity and interhemispheric connectivity in late mild cognitive impairment: A resting-state functional magnetic resonance imaging study.

OBJECTIVE: Mild cognitive impairment (MCI), a high-risk precursor to Alzheimer's disease, progresses through early (EMCI) and late (LMCI) stages. While distinct functional brain alterations between EMCI and LMCI have been reported, differences in regional brain activity and interhemispheric functional connectivity during resting-state remain incompletely characterized. The aim of the study was to systematically assess the differences in resting-state brain functional alterations between the LMCI patients and EMCI patients. METHOD: We assessed resting-state functional alterations in EMCI and L

Depression and anxiety modify cognitive concerns associated with amyloid and early clinical impairment.

OBJECTIVE: Cognitive concerns (CCs) are common for patients with Alzheimer's disease (AD) dementia. We explore anxiety, depression, and clinical impairment as moderators of CCs in early AD. Understanding these associations may help inform efforts to advance early identification of disease progression in AD. METHOD: Data sources, selected based on requirements for availability of biomarker data and clinical assessment data, generated a pooled sample of adults ages 65+ with known amyloid status and cognitive function ranging from unimpaired (Clinical Dementia Rating global score [CDR-GS] = 0, n

Language switching and speaking a nondominant language challenge executive control: Preliminary data for novel behavioral markers of Alzheimer's risk in Spanish-English bilinguals.

OBJECTIVE: The present study explored psycholinguistic analysis of spoken responses produced in a structured interview and cued linguistic and nonlinguistic task switching as possible novel markers of Alzheimer's disease (AD) risk in Spanish-English bilinguals. METHOD: Nineteen Spanish-English bilinguals completed an Oral Proficiency Interview (OPI) in both languages, cued-switching tasks, and a battery of traditional neuropsychological tests (in a separate testing session). All were cognitively healthy at the time of testing, but eight decliners were later diagnosed with AD (on average 4.5 y

How does emotion regulation change during psychotherapy? A daily diary study of adults in a transdiagnostic partial hospitalization program.

OBJECTIVE: Improvement in emotion regulation is a proposed transdiagnostic mechanism of change. However, treatment research is limited by disorder-specific investigations that assess a narrow number of emotion regulation strategies. Moreover, most assess pre-to-post-treatment change without examining short-term changes throughout psychotherapy that might influence treatment response. METHOD: To address these gaps, this study uses daily diary methodology to examine trajectories of change in use of six emotion regulation strategies during partial hospitalization psychiatric treatment. Treatment

Sex-related differences in the relationship between β-amyloid and cognitive trajectories in older adults.

Objective: We aimed to test the hypothesis that elevated neocortical β-amyloid (Aβ), a hallmark feature of Alzheimer's disease (AD), predicts sex-specific cognitive trajectories in clinically normal older adults, with women showing greater risk of decline than men. Method: Florbetapir Aβ positron emission tomography (PET) was acquired in 149 clinically normal older adults (52% female, Mage = 74). Participants underwent cognitive testing at baseline and during annual follow-up visits over a timespan of up to 5.14 years. Indirect effect of family climate on adolescent depression through emotion regulatory processes.

Adolescent depression is a serious public health concern, warranting examination of its development. A negative family emotional climate (NFEC) is one risk factor for the development of depressive symptoms. The specific emotion regulatory processes linking NFEC and depression, however, remain unclear. Cognitive reappraisal, a strategy that entails shifting one's thoughts about an emotion eliciting situation before the emotion is generated, expressive suppression, an emotion regulation strategy where individuals push down their expressions of an emotion after it is generated, and emotional iner

Autistic traits and cognitive abilities associated with two molecular causes of Silver-Russell syndrome.

Silver-Russell syndrome is a rare genetic imprinting disorder. Two molecular causes of Silver-Russell syndrome have been identified: loss of methylation on chromosome 11p15 (11p15 LOM) and maternal uniparental disomy for chromosome 7 (matUPD7). Current understanding of the cognitive and behavioral phenotypes associated with these two molecular subtypes is limited. This study aimed to address this gap in the literature. The Social Responsiveness Scale (SRS-2) was used to assess autistic traits in individuals with 11p15 LOM (n = 47) and matUPD7 (n = 32). A subset of participants with 11p15 LOM (

The European Society of Paediatric Oncology Ependymoma-II program Core-Plus model: Development and initial implementation of a cognitive test protocol for an international brain tumour trial.

It is increasingly accepted that survival alone is an inadequate measure of the success of childhood brain tumour treatments. Consequently, there is growing emphasis on capturing quality of survival. Ependymomas are the third most frequently occurring brain tumours in childhood and present significant clinical challenges. European Society of Paediatric Oncology Ependymoma II is a comprehensive international program aiming to evaluate outcomes under different treatment regimens and improve diagnostic accuracy. Importantly, there has been agreement to lower the age at which children with posteri

Information processing deficit in older adults with HIV infection: A comparison with Parkinson's disease.

OBJECTIVE: Individuals with HIV treated with antiretroviral therapy can expect to reach average life span, making them susceptible to combined disease and aging effects on cognitive and motor functions. Slowed processing speed in HIV is a concern for cognitive and everyday functioning and is sensitive to declines in aging. We hypothesized that information processing (IP) deficits, over and above that expected with normal aging, would occur in older HIV patients similar to those observed in Parkinson's disease (PD) patients, with both conditions affecting frontostriatal pathways. METHOD: Group

Does the serial position effect identify those at risk for dementia? Findings from the Canadian Longitudinal Study on Aging.

OBJECTIVE: Subjective cognitive decline (SCD), or a self-perceived decline in cognition without objective cognitive impairment, is a potential predictor of Alzheimer's disease. However, summary-level scores on objective neuropsychological measures often fail to detect subtle cognitive changes in this population. The serial position effect (SPE), where individuals tend to recall items presented at the beginning (primacy) and end (recency) of a word list better than ones in the middle, may provide greater sensitivity. Although altered SPE patterns have been observed in Alzheimer's disease, SPEs

Association between preoperative frailty and postoperative delirium and cognitive dysfunction in elderly patients undergoing surgery under general anaesthesia: A prospective observational study.

BACKGROUND AND AIMS: Advancing age is associated with poor physiological reserve to combat stressors of surgery and anaesthesia, which is termed frailty, and it leads to postoperative complications. Frailty has been found to have a strong association with postoperative delirium (POD) and postoperative cognitive dysfunction (POCD) in cardiac surgeries. This study aims to determine the association of frailty with POD and POCD in patients undergoing non-cardiac surgeries. METHODS: A prospective observational, cohort study was done at a tertiary-level hospital on 130 patients aged 55 years and ab
