Herbicidal Active Compound Ferulic Acid Ethyl Ester Affects Fatty Acid Synthesis by Targeting the 3-Ketoacyl-Acyl Carrier Protein Synthase I (KAS I).

Exploring new herbicide targets based on natural product derivatives is an important research aspect for the generation of innovative pesticides. Ferulic acid ethyl ester (FAEE), a natural product derivative from ferulic acid, has significant herbicidal activity mainly by inhibiting the normal growth of weed seedling roots. However, the FAEE target protein underlying its herbicidal activity has not been identified. In this study, we synthesized an FAEE probe to locate its site of action. We discovered that FAEE entry point was via the root tips. Fourteen major binding proteins were identified

A Mixture of Phenolic Metabolites of Quercetin Can Decrease Elevated Blood Pressure of Spontaneously Hypertensive Rats Even in Low Doses.

Quercetin is proven to decrease arterial blood pressure when given orally. Its bioavailability is, however, low and, therefore, its metabolites could rather be responsible for this effect. In particular, the colonic metabolites of quercetin, 3,4-dihydroxyphenylacetic acid (DHPA), 4-methylcatechol (4MC), and 3-(3-hydroxyphenyl)propionic acid (3HPPA), have been previously shown to decrease the blood pressure in spontaneously hypertensive rats (SHR). Interestingly, the mechanisms of action of these three metabolites are different. The aim of this study is hence to investigate if these metabolites

Deep Learning and Domain Expertise for Fabricating High-Performance Biosensors.

Transcription factor-based biosensors are essential for the scalable bioproduction of plant natural products. However, a major bottleneck in developing high-performance biosensors is the engineering of responsive promoters due to the vast sequence space and poorly understood sequence-function relationships. Here, we extend our previously published DeepSEED framework into an artificial intelligence-driven pipeline for efficient and extensible biosensor-responsive promoter optimization. This strategy integrates deep learning with domain-specific biological expertise. By fixing core promoter elem

The therapeutic role of Coccinia grandis in flavor-enhancing high-lipid diet-induced chronic kidney disease: a dual role of NF-kB and TGF-β/smad in renal fibrosis.

BACKGROUND: Coccinia grandis (CG), a plant used as traditional medicine, possesses antioxidant, anti-inflammatory, and anti-fibrotic activities. Chronic kidney disease (CKD) and renal fibrosis are public health issues, further promoted by monosodium glutamate in high-lipid diets (MH) via oxidative stress and inflammation. CG could potentially alleviate MH-induced CKD by modulating cell signaling. The purpose of this study is to hypothesize that CG, with known anti-inflammatory and antioxidant properties, might counteract MH-induced kidney dysfunction through its putative modulation of the sign

Propolis extract promotes translocation of glucose transporter 4 and glucose uptake through both PI3K- and AMPK-dependent pathways in skeletal muscle.

It is well known that propolis has the ability to prevent hyperglycemia. However, the underlying mechanism is not yet fully understood. We therefore investigated whether a Brazilian propolis ethanol extract affects glucose uptake and translocation of insulin-sensitive glucose transporter (GLUT) 4 in skeletal muscle cells. In L6 myotubes, the extract at 1 μg/mL significantly promoted GLUT4 translocation and glucose uptake activity. Regarding the mechanism of GLUT4 translocation, propolis extract induced both PI3K and AMPK phosphorylation in a dose-dependent manner in L6 myotubes. However, we co

Colon-targeted sinapic acid-loaded cationic guar gum - gellan gum hydrogel beads: A novel approach for treating inflammatory bowel disease.

Sinapic acid (SA), a naturally occurring phenolic compound found in various plants and grains, possesses significant anti-inflammatory properties, making it a potential therapeutic candidate for inflammatory bowel disease (IBD). However, its clinical application is hindered by extensive first-pass metabolism, necessitating an effective colon-targeted delivery system. The present study focused on developing and evaluating a microbially triggered, colon-targeted delivery system for SA. Hydrogel beads were fabricated using cationic guar gum (CGG) and anionic gellan gum (GG) through polyelectrolyt

Beta-Sitosterol-Conjugated Sinapic Acid-Engineered Nanoliposome: Biomucoadhesive and Enzyme-Responsive Targeted Oral Therapy in Ulcerative Colitis.

Developing oral drug delivery systems is promising for ulcerative colitis (UC). However, the key challenges, including formulation degradation under harsh gastric conditions, poor targeting efficiency, and limited colonic residence, lead to poor therapeutic efficacy that still needs to be tackled. Effective treatment requires a safe, efficacious, enzyme- and pH-responsive, biomucoadhesive oral drug delivery system to overcome these challenges. Therefore, we have developed chitosan-armored 1,2-dimyristoyl-sn-glycero-3-phosphoglycerol (DMPG) nanoliposomes amalgamated with synthesized beta-sitost

Ferulic Acid, Pterostilbene, and Tyrosol Protect the Heart from ER-Stress-Induced Injury by Activating SIRT1-Dependent Deacetylation of eIF2α.

Disturbances in Endoplasmic Reticulum (ER) homeostasis induce ER stress, which has been involved in the development and progression of various heart diseases, including arrhythmias, cardiac hypertrophy, ischemic heart diseases, dilated cardiomyopathy, and heart failure. A mild-to-moderate ER stress is considered beneficial and adaptative for heart functioning by engaging the pro-survival unfolded protein response (UPR) to restore normal ER function. By contrast, a severe or prolonged ER stress is detrimental by promoting cardiomyocyte apoptosis through hyperactivation of the UPR pathways. Prev

Primary Fatty Alcohols Are Major Components of Suberized Root Tissues of Arabidopsis in the Form of Alkyl Hydroxycinnamates.

Suberin is a complex hydrophobic polymer that acts as a barrier controlling water and solute fluxes and restricting pathogen infections. Suberin is deposited immediately outside of the plasmalemma in the cell wall of certain tissues such as endodermis of roots, aerial and underground periderms, and seed coats. Suberin consists of a variety of fatty acid derivatives polymerized with glycerol and phenolics. In this study, we show using liquid chromatography-tandem mass spectrometry and gas chromatography-mass spectrometry techniques that most of the fatty alcohols not covalently linked to the su

Hydrophobic nano-calcium superphosphate enhances leaf disease resistance via coordinated physical, biochemical, and phyllosphere responses.

Hydrophobic nano-calcium superphosphate (NPS1) is a novel nano-fertilizer that has shown great potential for enhancing plant growth and suppressing foliar diseases. However, the mechanisms underlying these effects remain unclear. We investigated the responses of grape leaves and phyllosphere microbiota to NPS1 under both open-field and rain-shelter cultivation systems. NPS1 application significantly reduced leaf wetness duration and the disease severity of downy and powdery mildew, increased leaf-associated phosphorus retention, which remained elevated at later sampling stages, while enhancing
