Cephalotanes are a rare class of diterpenoids occurring exclusively in Cephalotaxus plants. The intriguing structures and promising biological activities for this unique compound class prompt us to investigate C. fortunei var. alpina and C. sinensis, leading to the isolation of six undescribed cephalotane-type diterpenoids and/or norditerpenoids, ceforloids A-F (1-6). Sulfolipid-1 (SL-1) is a lipid that is abundantly found in the cell wall of Mycobacterium tuberculosis (Mtb). MtbFadD23 is crucial in the SL-1 synthesis pathway. This article provides an introduction to the crystal phasing technique known as molecular replacement. The available software is reviewed, and the prospects for future developments are considered. Several examples are described in detail to illustrate potential problems. A brief account of past progress is included. The biogenic formation of hemozoin crystals, a crucial process in heme detoxification by the malaria parasite, is reviewed as an antimalarial drug target. We first focus on the in-vivo formation of hemozoin. A model is presented, based on native-contrast 3D imaging obtained by X-ray and electron microscopy, that hemozoin nucleates at the inner membrane leaflet of the parasitic digestive vacuole, and grows in the adjacent aqueous medium. This manuscript reports the improved synthesis of the α,α,α,α isomer of tetra-p-iodophenyl tetra-methyl calix[4]pyrrole and the X-ray characterization of two solvate polymorphs. Three novel tin(iv)corrole complexes have been prepared and characterized by various spectroscopic techniques including single crystal X-ray structural analysis. Packing diagrams of the tin(iv)corroles revealed that corrolato-tin(iv)-chloride molecules are interconnected by intermolecular C-HCl hydrogen bonding interactions.
