On processes of peer influences in adolescent drug use: a developmental perspective.

Data from longitudinal studies of adolescents carried out over the last ten years are reviewed to provide an integrated and dynamic perspective on the nature of friendships and processes of peer influence in adolescent drug involvement, within a general developmental perspective. Four interrelated questions are examined: What individual attributes are especially important in the formation of friendships among adolescents? Which of two processes, selection or socialization, account for the similarity in values and behaviors observed in ongoing friendship dyads, and how important is similarity i

Maternal education and adolescent drug use: a longitudinal analysis of causation and selection over a generation.

Current evidence indicates that in the USA illegal drug use among adolescents between the 1980s and 1990s became significantly more prevalent in families with lower maternal education in comparison to families with higher maternal education. In this study, we examine whether this inter-generational change results from either (a) a changing influence of socioeconomic status on drug use, as predicted by the inter-generational social 'causation' hypothesis, or (b) a negative influence of drug use on socioeconomic status, as predicted by the inter-generational social 'selection/drift' hypothesis.

DISCOVERY AND VALIDATION OF GENES DRIVING DRUG-INTAKE AND RELATED BEHAVIORAL TRAITS IN MICE.

Substance use disorders (SUDs) are heritable disorders characterized by compulsive drug use, but the biological mechanisms driving addiction remain largely unknown. Genetic correlations reveal that predisposing drug-naïve phenotypes, including anxiety, depression, novelty preference, and sensation seeking, are predictive of drug-use phenotypes, implicating shared genetic mechanisms. Because of this relationship, high-throughput behavioral screening of predictive phenotypes in knockout (KO) mice allows efficient discovery of genes likely to be involved in drug use. We used this strategy in two

Clinical and biobehavioral perspectives: Is medication overuse headache a behavior of dependence?

Medication overuse headache (MOH), previously known as analgesic abuse headache or medication misuse headaches, is a common form of chronic headache disorder that has a detrimental impact on health and society. Although it has been widely accepted that overusing abortive medications is paradoxically the cause of MOH and drug discontinuation is the treatment of choice, ongoing debates exist as to whether drug consumption per se is the cause or consequence of headache chronification. Certain features in MOH such as their compulsive drug-seeking behavior, withdrawal headaches and high relapse rat…

Manipulation of interoceptive signaling biases decision making in rhesus macaques.

Several influential theories have proposed that interoceptive signals, sent from the body to the brain, contribute to neural processes that coordinate complex behaviors. We altered the physiological state of the body using compounds that have minimal effect on the brain and evaluated their effect on decision making in rhesus monkeys. We used glycopyrrolate, a nonspecific muscarinic (parasympathetic) antagonist, and isoproterenol, a beta-1/2 (sympathetic) agonist, to create a sympathetic-dominated state in the periphery, that was indexed by increased heart rate. Rhesus monkeys were trained on t

Pharmacotherapy, pharmacogenomics, and the future of alcohol dependence treatment, part 1.

PURPOSE: The neurobiological basis of alcohol dependence, established pharmacotherapies for alcohol dependence, pharmacotherapies under investigation, and obstacles to treatment are discussed. SUMMARY: Alcohol binds to hydrophobic pockets of proteins, changing their three-dimensional structure and their function. Proteins that are particularly sensitive to alcohol include ion channels, neurotransmitter receptors, and enzymes involved in signal transduction. Established pharmacologic treatments, notably disulfiram and naltrexone, combined with behavioral therapies, may reduce the amount of dri

Double successful buprenorphine/naloxone induction to facilitate cardiac transplantation in an iatrogenically opiate-dependent patient.

Buprenorphine/naloxone is used for the treatment of opioid dependence. In the following case, a potential use for the medication combination is explored in the arena of transplant surgery. Psychiatry was consulted for a 29-year-old woman with iatrogenic opioid dependence after bilateral ventricular assist device placement for congenital cardiomyopathy. Her ejection fraction was less than 15% and she was considered a poor candidate for transplant due to drug-seeking behaviors. We transitioned her onto buprenorphine/naloxone to prevent abuse and control symptoms, qualifying her for cardiac trans…

BDNF in ventrolateral orbitofrontal cortex to dorsolateral striatum circuit moderates alcohol consumption, seeking and relapse.

BDNF plays a crucial role in shaping the structure and function of neurons. In rodents, BDNF signaling in the dorsolateral striatum (DLS) is part of an endogenous pathway that protects against the development of phenotypes associated with alcohol use. Dysregulation of BDNF levels in the cortex or dysfunction of BDNF/TrkB signaling in the DLS of rodents results in the escalation of alcohol drinking and compulsive alcohol intake. The major source of BDNF in the striatum is the prefrontal cortex. We identified a small ensemble of BDNF-positive neurons in the mouse ventrolateral orbitofrontal cort

Negative allosteric modulation of CB1 cannabinoid receptor signalling decreases intravenous morphine self-administration and relapse in mice.

The endocannabinoid system interacts with the reward system to modulate responsiveness to natural reinforcers, as well as drugs of abuse. Previous preclinical studies suggested that direct blockade of CB1 cannabinoid receptors (CB1R) could be leveraged as a potential pharmacological approach to treat substance use disorder, but this strategy failed during clinical trials due to severe psychiatric side effects. Alternative strategies have emerged to circumvent the side effects of direct CB1 binding through the development of allosteric modulators. We hypothesized that negative allosteric modula

High frequency DBS-like optogenetic stimulation of nucleus accumbens dopamine D2 receptor-containing neurons attenuates cocaine reinstatement in male rats.

Previous work indicated that deep brain stimulation (DBS) of the nucleus accumbens shell in male rats attenuated reinstatement of cocaine seeking, an animal model of craving. However, the potential differential impact of DBS on specific populations of neurons to drive the suppression of cocaine seeking is unknown. Medium spiny neurons in the nucleus accumbens are differentiated by expression of dopamine D1 receptors (D1DRs) or D2DRs, activation of which promotes or inhibits cocaine-related behaviors, respectively. The advent of transgenic rat lines expressing Cre recombinase selectively in D1D

Alternation in dopamine D2-like and metabotropic glutamate type 5 receptor density caused by differing housing conditions during abstinence from cocaine self-administration in rats.

BACKGROUND: Environmental conditions have an important function in substance use disorder, increasing or decreasing the risks of relapse. Several studies strongly support the role of the dopamine D2-like and metabotropic glutamate type 5 receptors in maladaptive neurobiological responses to cocaine reward and relapse. AIMS: The present study employed cocaine self-administration with yoked-triad procedure in rats to explore whether drug abstinence in different housing conditions affects the drug-seeking behaviour and the dopamine D2-like and metabotropic glutamate type 5 receptor density and a

Minocycline increases firing rates of accumbal neurons and modifies the effects of morphine on neuronal activity.

Accumulating evidence indicated that minocycline, a glial cell modulator, is able to modify a variety of morphine effects. Here, we investigated minocycline effects on electrical activity of nucleus accumbens (NAc) neurons using single unit recording in urethane-anesthetized rats. In addition, we investigated whether minocycline can modify the effects of morphine on NAc neural activity during reinstatement of morphine-seeking behavior. Minocycline increased the NAc firing activity in intact animals. Electrophysiological recording in morphine-treated animals was performed, following the acquisi

Cocaine reinstates extinguished food responding in male cynomolgus monkeys with a history of self-administering cocaine under a concurrent drug versus food choice paradigm.

The rate-dependency principle postulates that "stimulants" tend to increase low baseline rates of response and decrease high baseline responses. One exception to this principle is that stimulants do not typically increase responses that has been extinguished. However, drug self-administration studies showed that stimulant pretreatments reinstated extinguished drug-maintained responses. The goal of this study was to determine the effects of noncontingent cocaine on extinguished food-maintained responding in 3 male cynomolgus monkeys with extensive cocaine self-administration experience. Monkeys

Factors associated with willingness to participate in a pharmacologic addiction treatment clinical trial among people who use drugs.

BACKGROUND AND OBJECTIVES: Although new medications are needed to address the harms of drug addiction, rates of willingness to participate in addiction treatment trials among people who use drugs (PWUD), have not been well characterized. METHODS: One thousand twenty PWUD enrolled in two community-recruited cohorts in Vancouver, Canada, were asked whether they would be willing to participate in a pharmacologic addiction treatment trial. Logistic regression was used to identify factors independently associated with a willingness to participate. RESULTS: Among the 1,020 PWUD surveyed between Ju

Long-term effects of adolescent THC exposure on anxiety-like behavior and cognitive function in male rats.

RATIONALE: As cannabis legalization and decriminalization have expanded, it is increasingly perceived as a low-risk substance. Concurrently, the potency of cannabis products and the number of young adults seeking treatment for cannabis use disorder have risen markedly. A growing body of research links early onset, frequent use, and high potency cannabis products to a range of adverse outcomes. OBJECTIVE: This study aimed to examine whether frequent cannabis use during late adolescence affects cognitive functioning and increases the risk of developing anxiety in adulthood. METHODS: Male Sprague

Sustained reduction of attentional bias to smoking cues by smartphone-delivered attentional bias modification training for smokers.

OBJECTIVE: Cigarette smoking is thought to be at least partially maintained by the attentional bias (AB) toward smoking cues that develops as a consequence of drug dependence. This study evaluated the impact of smartphone-delivered, in-home attentional bias modification (ABM) to reduce AB to smoking cues and to reduce smoking behavior and withdrawal-related symptoms when used as an adjunct to conventional smoking cessation treatment. METHOD: Participants (N = 246) were treatment-seeking smokers who completed up to 13 days of either ABM designed to train attention away from smoking cues, using

Modulation of relapse-like drinking in male Sprague-Dawley rats by ligands targeting the α5GABAA receptor.

Preclinical evidence suggests a key role for GABAA receptors containing the α5 subunit (i.e., α5GABAA receptors) in the abuse-related effects of alcohol, including the reinforcing and discriminative stimulus effects, as well as cue-induced alcohol-seeking behavior. However, the contribution of this GABAA receptor subtype to relapse-like drinking behavior remains unknown. The present study evaluated the capacity of ligands targeting α5GABAA receptors to modulate the alcohol deprivation effect (ADE), a model of relapse-like drinking. Groups of Sprague-Dawley rats underwent repeated cycles of lon
