Invasive candidiasis frequently involves medical device placement. On the surfaces of these devices, Candida can form biofilms and proliferate in adherent layers of fungal cells surrounded by a protective extracellular matrix. Due in part to this extracellular matrix, biofilms resist host defenses and antifungal drugs. Previous work (using neutrophils from healthy donors) found that one mechanism employed to resist host defenses involves the inhibition of neutrophil extracellular traps (NET) formation. NETs contain nuclear DNA, as well as antimicrobial proteins that can ensnare pathogens too l

Large, architecturally complex lichen symbioses arose only a few times in evolution, increasing thallus size by orders of magnitude over those from which they evolved. The innovations that enabled symbiotic assemblages to acquire and maintain large sizes are unknown. We mapped morphometric data against an eight-locus fungal phylogeny across one of the best-sampled thallus size transition events, the origins of the Placopsis lichen symbiosis, and used a phylogenetic comparative framework to explore the role of nitrogen-fixing cyanobacteria in size differences. Thallus thickness increased by >15

BACKGROUND: Gaseous fumigants are commonly employed to control fungal decay of cold-stored grapes. So far it is not clear if these fumigants, besides the direct interaction against fungal structures, induce transcriptional responses of defensive markers. In order to contribute to understanding the mechanisms by which these fumigants exert their effect, we studied the influence of ozone (O3) and sulfur dioxide (SO2) on the decay caused by Botrytis cinerea, and the quality and expression of the defense-related genes chitinase, β-1,3-glucanase and phenylalanine ammonia-lyase (PAL) in the table gr…

Ganoderma, a famous fungus, has traditionally been used in China for thousands of years. Fourteen previously undescribed triterpenes were isolated from the antler-shaped fruiting bodies of Ganoderma lucidum. Seven of these triterpenes showed inhibitory activity against influenza A virus (IAV) neuraminidase, with IC50 from 5.64 to 18.36 μM. One triterpene exhibited anti-IAV activity in Madin-Darby canine kidney (MDCK) cells. Four triterpenes significantly reduced IAV-induced production of inflammatory cytokines IL-1β or IL-6 in A549 cells. Our findings suggest that Ganoderma triterpenes provide

Ten previously undescribed grifolin derivatives (1, 2, and 4-11), including three pairs of enantiomers, and five known analogs (3 and 12-15), were isolated from the ethanol extract of Albatrellus dispansus. Their chemical structures were determined based on spectroscopic analysis, electronic circular dichroism calculations, and acid hydrolysis. In addition, all 15 isolated compounds were evaluated for cytotoxicity and antibacterial activities. Compound 15 showed mild antibacterial activity against MRSA with MIC values of 6.2 μM. Compound 13 showed moderate cytotoxicity against the human cancer

Hyporheic zones in erosion-prone loess watersheds are key interfaces for solute and contaminant transport and biogeochemical transformation. Heavy metals can alter sediment microbial communities, while microorganisms mediate nutrient and organic-matter cycling. However, the relative roles of metal gradients and sediment environmental conditions in structuring bacterial and fungal communities in these systems remain poorly resolved. This study investigated 16 sites across the Jinghe River Basin on the Loess Plateau during the hydrologically stable dry season, combining sediment metal and enviro

Fungal infections constitute a major global health concern, with annual mortality rates exceeding those of tuberculosis and malaria. This troubling situation has been exacerbated by the emergence of multidrug-resistant fungal strains, which have significantly undermined the effectiveness of current treatments. A number of antifungal medicines are now being utilised in clinical practice. Some of these treatments include azoles, polyenes, pyrimidines, and echinocandins. However, the therapeutic efficacy of these drugs is becoming increasingly limited due to the presence of unwanted side effects,

Polygalae Radix, a traditional Chinese medicine for insomnia and memory disorders, is highly susceptible to fungal contamination and mycotoxin production (especially by Aspergillus flavus) during storage, compromising its safety and efficacy. Therefore, in this study, high-throughput sequencing was employed to evaluate the dynamic changes in fungal communities during the storage of Polygalae Radix and to analyze common mycotoxin-producing genera. Furthermore, the inhibitory effects of peppermint essential oil (PEO) on A. flavus were assessed through fumigation treatments, combined with colony

Antimicrobial resistance, particularly in fungal infections, is a major global health problem, aggravated by the misuse of antifungal agents and the emergence of multidrug-resistant strains. This study focused on the synthesis and evaluation of novel 1,2,3-triazole derivatives as potential antifungal agents, capitalizing on the chemical stability and diverse biological activity of the triazole scaffold. Target compounds have been evaluated for their antibacterial and antifungal activity against a panel of Gram positive (e.g., S. aureus, B. subtilis) and Gram negative (e.g., E. coli) bacteria,

Sulfur metabolism is an essential aspect of fungal physiology and pathogenicity. Fungal sulfur metabolism comprises anabolic and catabolic routes that are not well conserved in mammals, therefore is considered a promising source of prospective novel antifungal targets. To gain insight into Aspergillus fumigatus sulfur-related metabolism during infection, we used a NanoString custom nCounter-TagSet and compared the expression of 68 key metabolic genes in different murine models of invasive pulmonary aspergillosis, at 3 time-points, and under a variety of in vitro conditions. We identified a set

BACKGROUND: Aspergillus fumigatus is a well-known opportunistic pathogen that causes a range of diseases including the often-fatal disease, invasive pulmonary aspergillosis (IPA), in immunocompromised populations. The severity of IPA is dependent on both host- and pathogen-derived signaling molecules that mediate host immunity and fungal growth. Oxylipins are bioactive oxygenated fatty acids known to influence host immune response and Aspergillus developmental programs. Aspergillus synthesizes 8-HODE and 5,8-diHODE that have structural similarities to 9-HODE and 13-HODE, which are known ligand

Common reed (Phragmites australis) is a native dominant plant in many coastal wetlands. To determine how rhizosphere effects and soil depth shape microbial communities, we sampled the rhizosphere and three bulk-soil layers (0-15, 15-30, and 30-45 cm) in a monodominant common-reed stand in a coastal salt marsh. Soil physicochemical properties and bacterial and fungal α-diversity, community composition, and assembly processes were evaluated using one-way ANOVA, principal coordinates analysis (PCoA), permutational multivariate analysis of variance (PERMANOVA), neutral community models, and phylog

PURPOSE: Androgenetic alopecia (AGA) is characterized by perifollicular micro-inflammation, although its precise trigger remains elusive. Given that the hair follicle harbors a distinct microbiota which may modulate local immune responses, this study aimed to comprehensively profile the bacterial and fungal microbiome within the deep hair follicles of AGA patients versus healthy controls, and to evaluate the influence of disease severity, age, sex, and geographical environment.
PATIENTS AND METHODS: We recruited 96 subjects (72 AGA patients and 24 healthy controls), collecting a total of 192

Fungal keratitis, a disease caused by fungal infection of the cornea, is an eye disease with a high rate of blindness. Despite voriconazole (VCZ) has a good therapeutic effect in the treatment of fungal infection, the use of VCZ in the eye is limited due to the poor solubility of VCZ and the special physiological structure of the eye. In this study, a soluble microneedle (MN) containing VCZ micelles was designed for the eye to effectively deliver VCZ for the treatment of fungal keratitis. The water-insoluble VCZ was first encapsulated into the micelle prepared by polyethylene glycol-15-hydroxy
