The ectoparasitic dagger nematode (Xiphinema index), vector of Grapevine fanleaf virus (GFLV), provokes gall formation and can cause severe damage to the root system of grapevines. Mycorrhiza formation by Glomus (syn. Rhizophagus) intraradices BEG141 reduced both gall formation on roots of the grapevine rootstock SO4 (Vitis berlandieri×V. riparia) and nematode number in the surrounding soil. Suppressive effects increased with time and were greater when the nematode was post-inoculated rather than co-inoculated with the arbuscular mycorrhizal (AM) fungus. Using a split-root system, decreased X.

Iron (Fe) deficiency directly affects crop growth and development, ultimately resulting in reduced crop yield and quality. Recently, long non-coding RNAs (lncRNAs) have been demonstrated to play critical regulatory roles in a multitude of pathways across numerous species. However, systematic screening of lncRNAs responding to Fe deficiency and their regulatory mechanism in plants has not been reported. In this work, 171 differently expressed lncRNAs (DE-lncRNAs) were identified based on analysis of strand-specific RNA-seq data from rice shoots and roots under Fe-deficient conditions. We also f

Ethylene biosynthetic enzymes, 1-aminocyclopropane-1-carboxylate (ACC) synthase (ACS) and ACC oxidase (ACO), participate in the floral sex differentiation of bitter gourd (Momordica charantia). However, the relationship between their structural features and developmental expression patterns remains to be further clarified. In this study, eight McACS and five McACO genes were identified using the Dali-11 reference genome. AlphaFold 3-based modeling showed structural differences between the two families, particularly regarding the diverse C-terminal flexibilities of McACS proteins. Targeted qRT-

Bamboo compost is a renewable organic material that can replace peat and enhance plant resistance to insect pests. However, its combined effects on tomato (Solanum lycopersicumL.) growth, defense responses, and underlying molecular mechanisms remain unclear. In this study, bamboo compost was incorporated into growing substrates at varying proportions (5-30%) to assess its impact on tomato seedling growth, physiological and biochemical traits, and the survival of the South American tomato leafminer, Tuta absoluta. Metabolomic and transcriptomic analyses were further integrated to characterize m

Plant photomorphogenesis is a light-induced developmental switch that combines massive reprogramming of gene expression and a general enhancement in RNA Polymerase II activity. Yet, transcriptome analyses have failed to demonstrate any tendency toward gene upregulation. To solve this conundrum, we use a spike-in RNA-seq experimental and bioinformatic pipeline, enabling to reconcile transcriptome dynamics with epigenomic and cytogenetic observations of Arabidopsis thaliana cotyledon photomorphogenesis. During the transition, a quasi-unilateral impact of light, with 94% of the differentially exp

BACKGROUND: Camellia sinensis 'Huangjinju' is an albino tea variety developed recently in China. Young leaves of 'Huangjinju' demonstrate bright yellow when cultivated under natural sunlight, but regreens under reduced light intensity. To elucidate the physiological and molecular mechanisms of this light-sensitive albinism, we compared leaf pigmentation, metabolites, cellular ultrastructure and transcriptome between plants cultured under natural sunlight and shade.
RESULTS: Shading treatment doubled the chlorophyll concentration and regreened albino leaves; carotenoid also increased by 30%. E

The salicylic acid (SA) plays a critical role during the establishment of systemic acquired resistance (SAR) in uninfected plant tissues after localised exposure to a pathogen. Here, we studied SA in Populus tomentosa infected by the plant pathogen Botryosphaeria dothidea. The accumulation of SA and methyl salicylate (MeSA) occurred in chronological order in P. tomentosa. The SA and MeSA contents were greater at infected than uninfected sites. Additionally, a gene expression analysis indicated that SA might be accumulated by phenylalanine ammonialyase (PAL) and converted to MeSA by SA carboxyl

Plant elicitor peptides (Peps) are conserved regulators of defense responses and models for the study of damage-associated molecular pattern-induced immunity. Although present as multigene families in most species, the functional relevance of these multigene families remains largely undefined. While Arabidopsis Peps appear largely redundant in function, previous work examining Pep-induced responses in maize (Zm) implied specificity of function. To better define the function of individual ZmPeps and their cognate receptors (ZmPEPRs), activities were examined by assessing changes in defense-asso

In plants, reactive oxygen species and, more particularly, hydrogen peroxide (H(2)O(2)) play a dual role as toxic by-products of normal cell metabolism and as regulatory molecules in stress perception and signal transduction. Peroxisomal catalases are an important sink for photorespiratory H(2)O(2). Using ATH1 Affymetrix microarrays, expression profiles were compared between control and catalase-deficient Arabidopsis (Arabidopsis thaliana) plants. Reduced catalase levels already provoked differences in nuclear gene expression under ambient growth conditions, and these effects were amplified by

Shoot branching, a major determinant of plant architecture and productivity, has long been attributed to the coordinated action of auxin, cytokinins (CKs), and strigolactones (SLs). Recent work shows that sugars act not only as metabolic resources but also as early developmental signals that regulate axillary bud outgrowth, the initial step in shoot branching. Sugar availability can rapidly trigger bud outgrowth, often preceding major hormonal changes. Mechanistically, sugars antagonize auxin and SL signaling while acting synergistically with CKs. These effects involve sugar signaling pathways

This study uncovers a novel concentration-dependent signaling network through which abscisic acid (ABA) modulates acid metabolism in grape berries. Treatment with 800 μM ABA significantly decreased malate and citrate synthesis and transport while enhancing glucose and fructose accumulation, optimizing the sugar-acid ratio. Multi-omics analysis revealed differential changes in 96 organic acids and identified a key malate transporter, VvALMT9. Through yeast two-hybrid screening, the VvPP2C-VvALMT9-VvSnRK2.3-VvCDPK interaction network was characterized in a dual regulatory mode. Under low-ABA con

Ammonium (NH4+) is an essential nitrogen source for plants, yet its excessive accumulation triggers toxicity, a process typically coupled to futile NH4+ cycling in roots cells. While the cytokinin-ARR10/12 signaling pathway is known to regulate vacuolar NH4+ sequestration, it is not known whether it also coordinates NH4+ efflux across the root-cell plasma membrane to maintain intracellular homeostasis. Here, we identify the auxin-amido synthetase GH3.5 as a direct transcriptional target of ARR10, using a combination of yeast one-hybrid (Y1H) assays, dual-luciferase reporter assays, and CUT&RUN

The antagonism between potassium (K+) and magnesium (Mg2+) is the primary cause of Mg2+ deficiency worldwide. Recent studies have demonstrated that the suppressive effect of K+ on Mg2+ uptake is significantly reduced as the K+/Mg2+ ratio increases, as both cations share non-specific cation channels. Concomitantly, the relative root/shoot translocation of Mg2+ increased. In contrast, there are indications that elevated tissue [K+] impedes the primary physiological functions of Mg2+. In this study on Zea mays L., the involvement of the only known specific Mg2+ uptake transporter ZmMGT10 and that

Combination of biotic and abiotic stress is a major challenge for crop and fruit production. Thus, identification of genes involved in cross-response to abiotic and biotic stress is of great importance for breeding superior genotypes. Lectins are glycan-binding proteins with a functions in the developmental processes as well as in the response to biotic and abiotic stress. In this work, a lectin like gene, namely ClLectin1, was characterized in Volkamer lemon and its expression was studied in plants exposed to either water stress, hormonal elicitors (JA, SA, ABA) or wounding to understand whet

Hydrogen sulfide (H2 S) is an important gaseous signal molecule that regulates plant growth and stress resistance. However, research on the H2 S synthase (HSase) genes is still limited in the model legume plant Medicago truncatula Gaertn. In the present study, a total of 40 HSase family members were first identified and analyzed in the M. truncatula genome, and these genes distributed across eight chromosomes and were clustered into five groups (I-V) based on their conserved gene structures and protein motifs. Expression analysis revealed that the MtHSase genes were expressed in all the tested

Plants have evolved complex mechanisms to defend themselves against pathogens. It has been shown that several defense responses are influenced by light, and the red/far-red light photoreceptor phytochromes (PHY) modulate plant defense responses in Arabidopsis. Blue light receptor cryptochromes (CRY) work together with PHY to regulate many light-controlled responses, including photomorphogenesis, floral induction, and entrainment of the circadian clock. We report here that the Arabidopsis blue light photoreceptor CRY1 positively regulates inducible resistance to Pseudomonas syringae under conti

Black yeast-like fungi of the genus Aureobasidium exhibit morphological plasticity, but whether distinct cellular states within the same genetic background are associated with different plant growth-promoting functions remains unclear. Here, yeast-like cells (YL), swollen cells (SC), and chlamydospores (CH) of Aureobasidium melanogenum ZN were characterized. YL was associated mainly with siderophore production and laccase activity, SC with extracellular polysaccharide accumulation, and CH with phosphate mobilization and higher ammonia and IAA production. Whole-genome and comparative genomic an

Bamboo, as a timber plant, holds significant environmental and economic value. Dendrocalamus farinosus is particularly valuable as it serves both as a source of bamboo shoots and timber, offering high yield, strong disease resistance, and superior fiber quality. Our previous study demonstrated that bio-organic fertilizers promoted the growth of D. farinosus and significantly altered the cellulose and lignin content, key components of the secondary cell wall in culms. However, the underlying regulatory mechanisms remain unclear. In this study, we used metabolomic and transcriptomic analyses to
