Background Klebsiella pneumoniae (hereafter, Kp) is a major public health threat responsible for high levels of multidrug resistant (MDR) human infections. Besides, Kp also causes severe infections in the community, especially in Asia and Africa. Although most Kp infections are caused by endogenous intestinal carriage, little is known about the prevalence and microbiological characteristics of Kp in asymptomatic human carriage, and attached risk factors including environmental sources exposure. Methods Here, 911 pregnant women from communities in Madagascar, Cambodia, and Senegal were screened

Multidrug resistance (MDR) has emerged in hospitals due to the use of several agents administered in combination or sequentially to the same individual. We reported earlier MDR in Candida lusitaniae during therapy with amphotericin B (AmB), azoles, and candins. Here, we used comparative genomic approaches between the initial susceptible isolate and 4 other isolates with different MDR profiles. From a total of 18 nonsynonymous single nucleotide polymorphisms (NSS) in genome comparisons with the initial isolate, six could be associated with MDR. One of the single nucleotide polymorphisms (SNPs)

Antibiotic resistance has grown steadily in Vibrio cholerae over the last few decades to become a major threat in countries affected by cholera. Multi-drug resistance (MDR) spreads among clinical and environmental V. cholerae strains by lateral gene transfer often mediated by integrative and conjugative elements (ICEs) of the SXT/R391 family. However, in a few reported but seemingly isolated cases, MDR in V. cholerae was shown to be associated with other self-transmissible genetic elements such as conjugative plasmids. IncA/C conjugative plasmids are often found associated with MDR in isolates

Staphylococcus aureus is an important food-related pathogen associated with bacterial poisoning that is difficult to treat due to its multidrug resistance. The cfr and lsa(E) genes both cause multiple drug resistance and have been identified in numerous Staphylococcus species, respectively. In this study, we found that a methicillin-resistant S. aureus (MRSA) strain, 2868B2, which was isolated from a sample of frozen dumplings in Hangzhou in 2015, co-carried these two different multidrug resistance genes. Further analysis showed that this strain was resistant to more than 18 antibiotics and ex

Resistance of Plasmodium falciparum to chloroquine (CQ) is determined by the mutation at K76T of the P. falciparum chloroquine resistance transporter (pfcrt) gene and modified by other mutations in this gene and in the P. falciparum multidrug resistance 1 (pfmdr1) gene. To determine the extent of polymorphisms in these genes in field P. falciparum isolates from Yunnan province of China, we genotyped the pfcrt codon 76, pfmdr1 codons 86 and 1246. Our results showed that although CQ has been withdrawn from treating falciparum malaria for over two decades, 90.3% of the parasites still carried the

BACKGROUND: As an alternative method to the conventional therapies for Hunter's syndrome, which is a lethal lysosomal storage disorder, we have developed gene delivery vehicles using a series of retroviral vectors. The objective of this study was to develop a safe and efficient retroviral vector and to optimize conditions for efficient transduction of human bone marrow CD34+ stem cells using our vector.
METHODS: We constructed three types of MLV-based retroviral vectors expressing iduronate-2-sulfatase (IDS) which is deficient in patients suffering from Hunter's syndrome: MIN-IDS and MIM-IDS,

CD20 determinant expressed on B precursors is associated with regulation of proliferation, apoptosis and maturation of these cells. The acute lymphoblastic leukemia "common" type (cALL) based on expression of CD20 is subdivided in type I and II. However, the clinical significance of CD20 expression on cALL and significance of cALL type I and II discernment are not fully elucidated. The association of CD20 expression with the expression of multidrug resistance molecule (MDR), CD34, atypical immunophenotypes of leukemia cells and response to induction therapy were determined in the group of 147

Multidrug resistance (MDR) remains a major challenge in cancer treatment. MDR has been explained by single-gene mechanisms, such as overexpression of ATP-binding cassette (ABC) transporters, drug-target mutations, activation of bypass signaling pathways, and increased DNA repair. However, these mechanisms alone are insufficient to explain the heterogeneity, reversibility, temporal evolution, and environmental dependence of drug resistance observed in clinical settings. Increasing evidence suggests that MDR is better viewed as a complex and evolving biological phenomenon rather than the result

Klebsiella pneumoniae is widely recognized as an opportunistic hospital and community pathogen. It is one of the priority microorganisms included in the ESKAPE group, and its antibiotic resistance related to extended-spectrum β-lactamases (ESBL) is a global public health concern. The multi-drug resistance (MDR) phenotype, in combination with pathogenicity factors, could enhance the ability of this pathogen to cause clinical infections. The aim of this study was to characterize pathogenicity factors and biofilm formation in ESBL-producing K. pneumoniae from pediatric clinical infections. Capsul

PURPOSE: Antimicrobial resistance (AMR) is important in the pathogenesis and spread of Clostridioides difficile infection (CDI). However, very little is known about the association of AMR and C. difficile in South Africa. The present study aimed to investigate the phenotypic and genotypic antimicrobial profiles of clinical C. difficile isolates.
METHODS: Phenotypic antimicrobial susceptibility testing (AST) and whole genome sequencing (WGS) were used to characterize the isolates.
RESULTS: The AST showed that 43 isolates were susceptible to metronidazole and vancomycin. The WGS revealed a Pni

Antimicrobial resistance is a critical issue that is no longer restricted to hospital settings but also represents a growing problem involving intensive animal production systems. In this study, we performed a microbiological and molecular investigation of priority pathogens carrying transferable resistance genes to critical antimicrobials in 1-day-old chickens imported from Brazil to Uruguay. Bacterial identification was performed by matrix-assisted laser desorption ionization-time of flight (MALDI-TOF) mass spectrometry, and antibiotic susceptibility was determined by Sensititre. BACKGROUND: Klebsiella pneumoniae plays a critical role in hospital-acquired infections, which pose a serious threat globally. Older antibiotics such as fosfomycin are being reconsidered to combat MDR K. pneumoniae. In healthcare settings, bacteria are exposed frequently to antibiotics at sub-inhibitory concentrations (sub-MICs), which may alter their virulent characteristics. The aim of this research was to investigate the impact of fosfomycin at sub-MICs on the virulence determinants of MDR K. pneumoniae.
METHODS: The effect of sub-MIC fosfomycin treatment on biofilm formation was evaluated

INTRODUCTION: Acinetobacter baumannii is a highly problematic nosocomial pathogen, designated a "red alert" organism by the Infectious Diseases Society of America. Multidrug-resistant (MDR) strains are associated with mortality rates of 25-68%, particularly in critically ill patients. Efflux-mediated resistance, along with serum-induced upregulation of efflux pump genes, highlights the role of efflux systems in driving MDR phenotypes. Targeting these systems represents a promising therapeutic strategy.
METHODS: The efflux inhibitory potential of 13 plant-derived polyphenols was evaluated to r

Multidrug-resistant avian pathogenic Escherichia coli (MDR-APEC) causes critically high first-week mortality in chickens, resulting in substantial economic losses in the global poultry industry. This study aimed to isolate and characterize a novel bacteriophage as an effective safe alternative for the control of colibacillosis. A lytic bacteriophage CABI-SEA 2302 was obtained from E. coli lesion samples collected from commercial poultry farms. The selected phage was classified as a new species in the genus Tunavirus, subfamily Tunavirinae. Genomic analysis revealed that the phage contains 49,5

Acute infections caused by multidrug-resistant (MDR) Klebsiella pneumoniae immediately necessitate the development of novel therapeutic strategies. Phage therapy offers a viable alternative for combating MDR Klebsiella infections. Here, we report the isolation and comprehensive characterization of a novel lytic bacteriophage, BUCT791, recovered from hospital sewage. Phage BUCT791 exhibited a short latent period (20 min), a large burst size (∼275 PFU/cell), rapid adsorption efficiency (>90% within 15 min), and remarkable stability across a wide range of temperatures (4-50 °C) and pH values (3-1

The rapid rise of multidrug-resistant Acinetobacter baumannii (MDR A. baumannii) poses a serious threat in healthcare settings, as these bacteria can cause infections that are untreatable with conventional antibiotics. This crisis has driven the search for alternative therapies, one of which is bacteriophages. In this study, we isolated a novel bacteriophage, vB_AbaM_EA1 (E7 for short), from wastewater and evaluated its potential activity against clinical MDR A. baumannii strains. Phage E7 successfully infects (27.5%) of the 40 clinical MDR A. baumannii isolates we tested. Laboratory character
