Microplastics (PET and PVC) disrupt palygorskite-mediated cadmium stabilization in paddy soil: Polymer-specific rhizosphere mechanisms and ecological consequences.

Microplastics (MPs) are emerging disruptors to soil heavy metal remediation, yet how different polymer types undermine mineral-mediated in-situ immobilization remains unclear. Using a multi-omics approach (16S rRNA gene sequencing, metagenomics, transcriptomics and metabolomics), we aimed to elucidate the polymer-specific mechanisms by which polyethylene terephthalate (PET) and polyvinyl chloride (PVC) residues destabilize palygorskite-immobilized cadmium (Cd) in paddy soil, at both the soil and microbial levels. PET and PVC differentially induced the remobilization of palygorskite-immobilized

The human reproductive tract microbiome: A novel source of live biotherapeutics.

Live biotherapeutics which are live microorganisms with clinically validated therapeutic benefits are rapidly emerging as innovative interventions for a broad spectrum of health disorders. The human reproductive tract, particularly the female vagina is home to specific microbial community that play a vital role in maintaining mucosal immunity, preventing pathogen colonization and supporting successful pregnancy outcomes. Disruption to this microbial balance have been strongly associated with conditions such as bacterial vaginosis (BV), sexually transmitted infections (STI), infertility and pre

Parkinson's disease and the gut microbiota connection: unveiling dysbiosis and exploring therapeutic horizons.

Parkinson's disease (PD) is a progressive neurodegenerative disorder marked by dopaminergic neuronal loss, α-synuclein aggregation, and sustained neuroinflammation. Emerging evidence supports the gut-brain-microbiota axis as a pivotal player in the disease's pathogenesis. Dysbiosis, disruptions in the gut microbial composition, has been consistently observed in individuals with PD, with notable reductions in beneficial, short-chain fatty acid-producing bacteria and elevations in pro-inflammatory microbial species. These alterations contribute to increased intestinal permeability, systemic infl

Gut Microbiota and Autism: Unlocking Connections.

Background: Autism Spectrum Disorder (ASD) is a multifactorial neurodevelopmental condition in which genetic predisposition interacts with environmental factors. Among these, the gut microbiota has emerged as a crucial modulator of the microbiota-gut-brain axis (MGBA), influencing neuroinflammation, neurotransmission, and behavior. This review aims to provide an updated and integrative overview of the relationship between gut microbiota, diet, and neurodevelopment in ASD. Methods: A comprehensive search was conducted in PubMed, Scopus, and Web of Science for articles published between 2010 and

The discovery of stromatolites developing at 3570 m above sea level in a high-altitude volcanic lake Socompa, Argentinean Andes.

We describe stromatolites forming at an altitude of 3570 m at the shore of a volcanic lake Socompa, Argentinean Andes. The water at the site of stromatolites formation is alkaline, hypersaline, rich in inorganic nutrients, very rich in arsenic, and warm (20-24°C) due to a hydrothermal input. The stromatolites do not lithify, but form broad, rounded and low-domed bioherms dominated by diatom frustules and aragonite micro-crystals agglutinated by extracellular substances. In comparison to other modern stromatolites, they harbour an atypical microbial community characterized by highly abundant re

Longitudinal Profiling of the Macaque Vaginal Microbiome Reveals Similarities to Diverse Human Vaginal Communities.

The vaginal microbiota plays an important role in women's reproductive and urogenital health. It is now well accepted that a "Работает" vaginal microbiome is dominated by Lactobacillus species. Disturbances in this microbial community can lead to several adverse outcomes, including pelvic inflammatory disease and bacterial vaginosis (BV), as well as increased susceptibility to sexually transmitted infections, miscarriage, and preterm births. However, vaginal communities, especially those of women in the developing world, can be comprised of a diverse set of microorganisms in the absence of over

TSA-ultrasound synergy enhances CRISPR-Cas9 gene editing efficiency in diploid yeast.

The CRISPR-Cas9 system is invaluable for microbial engineering. However, its efficiency remains limited in numerous microorganisms, especially in polyploid yeasts where the compact chromatin structures pose significant physical barriers, which are major constraints in microbial engineering. To address this limitation, we developed and validated a standardized synergistic protocol using the diploid industrial yeast Candida glycerinogenes as a model. This protocol combines TSA-induced chromatin decondensation with brief, low-intensity ultrasonication to enhance Cas9 accessibility. Key parameters

A synthetic microbiome drives a multi-omics response to remediate 1,4-dithiane-contaminated soil and simultaneously suppresses antibiotic resistance genes.

1,4-Dithiane, a degradation product of abandoned Japanese chemical weapons, is a persistent organic pollutant with ecological risks. A synthetic microbiome (SM) was constructed through pollution stress screening and ratio optimization, consisting of Shinella sp., Alcaligenes faecalis, Sphingomonas sp., and Stenotrophomonas sp. at an optimal ratio of 1: 1: 2: 2. The SM achieved a 1,4-dithiane degradation rate of 95.2% and reduced intermediate accumulation. Soil remediation experiments showed complete pollutant removal within 60 days, along with improved soil health: reduced bioavailability of h

A tungsten-specific maturation pathway governs cofactor assembly of a CO2-reducing formate dehydrogenase in Methylorubrum extorquens.

Tungsten-dependent formate dehydrogenases catalyze the reversible interconversion of carbon dioxide and formate and play key roles in microbial redox metabolism, yet the molecular basis for selective tungsten cofactor assembly in aerobic bacteria remains poorly understood. Here, we define a dedicated tungsten-specific maturation pathway that governs cofactor assembly and delivery to the carbon dioxide-reducing formate dehydrogenase MeFDH1 in Methylorubrum extorquens AM1. Targeted genetic deletions identified moeA1, mobB, and fdhD as essential for MeFDH1 activity and tungsten incorporation. Pro

Disentangling the Causal Role of Gut Microbiota in Bacterial Liver Abscess: A Mendelian Randomization Study with Clinical Validation.

Bacterial liver abscess (BLA), accounting for approximately 80% of all liver abscesses, is a severe suppurative infection of the liver. Although gut microbiota dysbiosis has been implicated in BLA pathogenesis, causal evidence remains limited. Here, we integrate Mendelian randomization (MR) and clinical cohort studies to systematically evaluate the causal role of gut microbiota in BLA. Using summary-level genetic data from MiBioGen, GWAS Catalog, and the Pan-UK Biobank, we identified several causal microbial taxa: Coprococcus, Veillonellaceae (including Dialister), and Klebsiella were positive

Single and mixture toxicity effects of legacy and emerging per- and polyfluoroalkyl substances on submerged plants and epiphytic biofilms.

Although per- and polyfluoroalkyl substances (PFASs), often referred to as "forever chemicals," pose persistent threats to aquatic ecosystems, the effects of multiple PFAS types on submerged macrophyte-biofilm symbiotic systems remain poorly understood. In this study, we systematically investigated the impacts of single and combined exposures to both legacy and emerging PFAS on submerged macrophytes, biofilms, and associated microbial risks. Our results show that the growth of Vallisneria natans was inhibited under both single and mixed PFAS stress. Photosynthetic performance and nutrient upta

Gut Microbiota in Rheumatoid Arthritis: Unraveling Pathogenic Mechanisms and Therapeutic Opportunities.

Rheumatoid arthritis (RA), a chronic autoimmune disorder driven by genetic-environmental interplay, manifests as progressive synovitis and irreversible joint damage. Despite mechanistic advances in disease-modifying antirheumatic drugs (DMARDs) and biologics, upstream mucosal triggers of autoimmunity remain elusive. Mounting evidence implicates gut microbiota dysbiosis as a pivotal environmental factor in RA pathogenesis through multifaceted mechanisms: (1) compromising intestinal barrier integrity, (2) facilitating molecular mimicry via cross-reactive microbial antigens, (3) skewing mucosal i

Environmental and physiological factors influencing the survival of resistant Salmonella under infection-related gut conditions.

SUMMARYThe human gastrointestinal tract is a dynamic and complex ecosystem that plays a pivotal role in maintaining host health and regulating microbial homeostasis. This environment significantly impacts the survival and evolution of antimicrobial-resistant organisms. Various innate factors, including pH, temperature, osmotic stress, oxygen levels, host secretions, immune response, bile salts, and nutrient gradients, influence microbial behavior, interspecies interactions, and the spread of resistance genes to ensure successful colonization. The potential of the bacteria to avoid or overcome

Inflammation and Gastric Cancer.

Gastric cancer remains a major killer globally, although its incidence has declined over the past century. It is the fifth most common cancer and the third most common reason for cancer-related deaths worldwide. Gastric cancer is the outcome of a complex interaction between environmental, host genetic, and microbial factors. There is significant evidence supporting the association between chronic inflammation and the onset of cancer. This association is particularly robust for gastrointestinal cancers in which microbial pathogens are responsible for the chronic inflammation that can be a trigg

Amoxicillin and thiamphenicol treatments may influence the co-selection of resistance genes in the chicken gut microbiota.

The aim of this study was to assess the dynamics of microbial communities and antimicrobial resistance genes (ARGs) in the chicken gut following amoxicillin and thiamphenicol treatments and potential co-selection of ARGs. To this purpose, the microbial community composition, using 16S rRNA NGS, and the abundance of ARGs conferring resistance to β-lactams and phenicols, using qPCRs, were determined. Results revealed that the administered antimicrobials did not significantly reduce the gut microbiota diversity, but changed its composition, with taxa (e.g. Gallibacterium and Megamonas) being enri

Biodegradation of tylosin in swine wastewater by Providencia stuartii TYL-Y13: Performance, pathway, genetic background, and risk assessment.

Microbial bioremediation offers a solution to the problem of residual antibiotics in wastewater associated with animal farms. Efficient degradation of antibiotic residues depends upon the genetic make-up of microbial degraders, which requires a comprehensive understanding of the degradation mechanisms. In this study, a novel, efficient tylosin (TYL)-degrading bacterium, Providencia stuartii TYL-Y13 (Y13) was isolated, which could completely degrade 100 mg/L TYL within 15 h under optimal operating conditions at 40 ℃, pH 7.0 %, and 1 % (v/v) bacterial inoculation rate. Culture-independent metagenomics supports discovery of uncultivable bacteria within the genus Chlamydia.

Advances in culture-independent methods have meant that we can more readily detect and diagnose emerging infectious disease threats in humans and animals. Metagenomics is fast becoming a popular tool for detection and characterisation of novel bacterial pathogens in their environment, and is particularly useful for obligate intracellular bacteria such as Chlamydiae that require labour-intensive culturing. We have used this tool to investigate the microbial metagenomes of Chlamydia-positive cloaca and choana samples from snakes. The microbial complexity within these anatomical sites meant that

The role and clinical implications of G6PI in experimental models of rheumatoid arthritis.

The antigens that trigger the pathogenic immune response in rheumatoid arthritis (RA) remain unknown. Until recently it was assumed that either viral or microbial antigens, or joint-specific antigens were the target of arthritogenic T and B lymphocytes in RA. Consequently, murine models of arthritis are induced by immunization with either joint-specific antigens such as type II collagen or microbial products such as streptococcal cell wall. In the K/BxN T-cell receptor transgenic mouse model arthritis is caused by a systemic autoimmune response to the ubiquitously expressed glycolytic enzyme g
