A point mutation in the coding sequence of the beta-hexosaminidase alpha gene results in defective processing of the enzyme protein in an unusual GM2-gangliosidosis variant.

cDNA clones were isolated from cultured fibroblasts of a patient previously reported as having GM2-gangliosidosis due to defective processing of the precursor beta-hexosaminidase alpha chain. Sequence analysis of a clone containing the entire protein coding sequence showed a single nucleotide substitution, from G to A, at nucleotide residue no. 1444, which resulted in a change in amino acid residue no. 482, from the normal glutamic acid to lysine. This transversion was confirmed in two other cDNAs from the same unamplified library. The results collectively indicate that the change from the str…

Amino Acid Complexes of Zirconium in a Carbon Composite for the Efficient Removal of Fluoride Ions from Water.

Amino acid complexes of zirconia represent an entirely new class of materials that were synthesized and studied for the first time for the d … t appropriately by the two-site Langmuir model for both adsorbents. The adsorption capacities of Zr-glutamic acid-graphite oxide and Zr-aspartic acid-graphite oxide are 105.3 and 101.0 mg/g, respectively. We found that two distinct binding modes are combined in the Zr-amino acid complexes: at low solution concentrations, F- ions are preferentially adsorbed by c …

Cerebral excess release of neurotransmitter amino acids subsequent to reduced cerebral glucose metabolism in early-onset dementia of Alzheimer type.

A massive cerebral release of amino acids and ammonia was found in early-onset dementia of Alzheimer type. Aspartate and glycine were liberated in high concentrations, whereas glutamate remained rather unchanged. This excess cerebral protein catabolism is due to a 44% reduction in cerebral glucose metabolism. Whereas glutamate and other glucoplastic amino acids may substitute glucose, elevated aspartate may contribute to neuronal damage. The results are discussed with respect to a possible neuronal insulin/insulin receptor deficiency.

Hepatoprotective activity of carrot (Daucus carota L.) against carbon tetrachloride intoxication in mouse liver.

The effect of carrot extract on carbon tetrachloride (CCl4)-induced acute liver damage was evaluated. The increased serum enzyme levels (viz., glutamate oxaloacetate transaminase, glutamate pyruvate transaminase, lactate dehydrogenase, alkaline phosphatase, sorbitol and glutamate dehydrogenase) by CCl4-induction were significantly lowered due to pretreatment with the extract. The extract also decreased the elevated serum bilirubin and urea content due to CCl4 administration. Increased activities of hepatic 5'-nucleotidase, acid phosphatase, acid ribonuclease and decreased levels of succinic de

Glutamate accumulation in myelofibrosis microenvironment rewires mesenchymal stromal cells metabolic and epigenetic profiles.

BACKGROUND: Myelofibrosis (MF) is a clonal myeloproliferative neoplasm characterized by bone marrow (BM) fibrosis, osteosclerosis, and a proinflammatory tumor microenvironment (TME). The interplay between malignant hematopoietic stem cells and mesenchymal stromal cells (MSCs) drives disease progression. Although glutamate has been implicated in various cancers, its contribution to MF pathogenesis remains unclear. METHODS: High-performance liquid chromatography (HPLC) profiling was performed on sera from MF patients and healthy controls (HCs) to identify alterations in amino acid abundance. Th

Glutathione is a physiologic reservoir of neuronal glutamate.

Glutamate, the principal excitatory neurotransmitter of the brain, participates in a multitude of physiologic and pathologic processes, including learning and memory. Glutathione, a tripeptide composed of the amino acids glutamate, cysteine, and glycine, serves important cofactor roles in antioxidant defense and drug detoxification, but glutathione deficits occur in multiple neuropsychiatric disorders. Glutathione synthesis and metabolism are governed by a cycle of enzymes, the γ-glutamyl cycle, which can achieve intracellular glutathione concentrations of 1-10mM. Because of the considerable q

[Free and bound amino acids during germination of maize seeds with implemented foreign prolamines].

Wheat prolamines were preincorporated into the endosperm of maize seed, and concentrations of free and bound amino acids were measured in the endosperm and seedlings during seed germination. Incorporation of foreign prolamines lowered the decrease rate of free proline concentration in both the endosperm (sprouting and shoots) and the embryo (sprouting). The seedlings at the stage of shoots were characterized by higher concentrations of aspartic acid, glutamic acid, and tyrosine. At the stage of transition to autotrophic nutrition, the pool of amino acids that are physiologically important for …

Previously unrecognized missense mutation E126K of PSEN2 segregates with early onset Alzheimer's disease in a family.

Mutations in the gene PSEN2 are a rare cause of early onset Alzheimer's disease (EOAD). PSEN2 sequence variants are often only found in one patient and pathogenicity cannot be formally documented. Here we describe a previously unrecognized sequence change (c.376G>A) in PSEN2 in an EOAD patient and her likewise affected mother. This change results in the exchange of amino acid glutamic acid (E) by lysine (K) at position 126 of the protein (p.E126K). Pathogenicity of the mutation is shown by segregation with disease, evolutionary conservation of E126, and in silico analysis of the mutation.

Genetic differences of E gene of a Thai strain of Japanese encephalitis virus that determine small plaque size phenotype but not neurovirulence in suckling mice.

Two small plaque variants of Japanese encephalitis virus (JEV), S4P9 and S9P10, were recovered from the wild type of JEV strain KE-093 using plaque purification in combination with the temperature-shift induction technique. Growth patterns of the S4P9 and S9P10 in BHK-21 cells as well as neurovirulence in suckling mice were similar to that of KE-093. An amino acid substitution, lysine for glutamic acid, was present in envelope protein at residue E-83 in the small plaque variants. This study shows that small plaque phenotype is not always associated with attenuation in vivo.

N-acetylaspartylglutamate inhibits forskolin-stimulated cyclic AMP levels via a metabotropic glutamate receptor in cultured cerebellar granule cells.

The neuronal dipeptide N-acetylaspartylglutamate (NAAG) fulfills several of the criteria for classification as a neurotransmitter including localization in synaptic vesicles, calcium-dependent release after neuronal depolarization, and low potency activation of N-methyl-D-aspartate receptors. In the present study, the influence of NAAG on metabotropic receptor activation in cerebellar granule cells was examined in cell culture. Stimulation of granule cell adenylate cyclase with forskolin increased cyclic AMP (cAMP) several hundredfold above basal levels within 10 min in a concentration-depende

Production of R-(-)-mandelic acid from mandelonitrile by Alcaligenes faecalis ATCC 8750.

R-(-)-Mandelic acid was produced from racemic mandelonitrile by Alcaligenes faecalis ATCC 8750. Ammonium acetate or L-glutamic acid as the carbon source and n-butyronitrile as the inducer in the culture medium were effective for bacterial growth and the induction of R-(-)-mandelic acid-producing activity. The R-(-)-mandelic acid formed from mandelonitrile by resting cells was present in a 100% enantiomeric excess. A. faecalis ATCC 8750 has an R-enantioselective nitrilase for mandelonitrile and an amidase for mandelamide. As R-(-)-mandelic acid was produced from racemic mandelonitrile in a yiel…

Sex differences in response to kainic acid and estradiol in the hippocampus of newborn rats.

Premature and full-term human infants are at considerable risk of excitotoxic-mediated brain damage due to hypoxia-ischemia, infection or other trauma. Glutamate receptor activation is a major source of excitoxicity in the adult and developing brain, and the hippocampus is particularly vulnerable to damage. The seven-day-old rat is a widely used model of pediatric brain damage, in large part due to the relative insensitivity of the brain to exogenous glutamate treatment prior to this age. We have reexamined the possible role of glutamate in pediatric brain damage in the newborn rat using kaini

Inositolphospholipid-linked glutamate receptors mediate cerebellar parallel-fiber-Purkinje-cell synaptic transmission.

In slices of adult rat cerebellum inositolphospholipid turnover is stimulated markedly by glutamate and its rigid analogues quisqualate and ibotenate. The drug and amino acid specificity of the response reflects a quisqualate-preferring excitatory amino acid receptor. The absence of glutamate-enhanced inositolphospholipid turnover in mice with Purkinje-cell degeneration indicates that the inositolphospholipid-linked quisqualate receptor mediates parallel fiber-Purkinje cell synaptic transmission. The quantitative prominence of this synapse accounts for the massive enrichment of elements of the

[Effects of acoustic overstimulation and perilymph perfusion--with Joro spider toxin on the concentrations of amino acids in the perilymph].

Glutamate (Glu) is considered one of the most probable neurotransmitter candidates in primary afferent nerves of the cochlea. In the present study, the amino acid profile in the perilymph was determined in order to investigate the effects of acoustic overstimulation and perilymph perfusion with Joro Spider Toxin (JSTX), a specific antagonist of the quisqualate sensitive Glu receptor, on the stimulus-induced release of Glu. In the first series of experiments (control group; n = 10), samples of the scala tympani perilymph (PST) were collected from albino guinea pigs at 60 and 75 min after initia

Plasma amino acid profile in major depressive disorder: Analyses in two independent case-control sample sets.

Some amino acids act as neurotransmitters themselves, or are precursors of neurotransmitters. Previous studies reported inconsistent results regarding their changes in blood in major depressive disorder (MDD), which prompted us to examine plasma levels of amino acids and related molecules in two independent case-control sample sets. In total, 511 subjects were recruited. Sample set A consisted of 164 patients with MDD (147 currently depressed [dMDD]; 17 in remission, DSM-IV) and 217 healthy controls. Sample set B consisted of 65 patients (51 dMDD; 14 in remission) and 65 controls. Plasma amino

Loss of CD98HC phosphorylation by ATM impairs antiporter trafficking and drives glutamate toxicity in Ataxia telangiectasia.

Ataxia-telangiectasia is a rare genetic disorder characterized by neurological defects, immunodeficiency, cancer predisposition, radiosensitivity, decreased blood vessel integrity, and diabetes. ATM, the protein mutated in Ataxia-telangiectasia, responds to DNA damage and oxidative stress, but its functional relationship to the progressive clinical manifestation of this disorder is not understood. CD98HC chaperones cystine/glutamate and cationic/neutral amino acid antiporters to the cell membrane, and CD98HC phosphorylation by ATM accelerates membrane localization to acutely increase amino aci

Toxicity of ethacrynic acid in isolated rat hepatocytes.

Ethacrynic acid, a loop diuretic drug, caused lipid peroxidation in isolated rat hepatocytes. The thiobarbituric acid reactive substances (TBARS) formation showed a good correlation with the leakage of glutamic-oxaloacetic acid transaminase (GOT) from the hepatocytes. The addition of antioxidants such as N, N'-diphenyl-p-phenylenediamine (DPPD) and promethazine to the isolated rat hepatocyte suspension containing ethacrynic acid prevented the lipid peroxidation and decreased the GOT leakage to some extent. SKF-525A inhibited the oxidative metabolism of ethacrynic acid and decreased the TBARS f

Glutamate Potentiates Meropenem Killing of Carbapenem-Resistant Pseudomonas aeruginosa by Increasing Outer Membrane Permeability to Increase Uptake.

Carbapenem-resistant Pseudomonas aeruginosa (CR-PA) has been designated a "priority" pathogen by the World Health Organization, calling for the development of new control strategies. However, no such new drugs are currently available. In this study, we restored bacterial sensitivity to meropenem through a metabolic state-reprogramming approach. Glutamate was identified as the key reprogramming metabolite that converts an antibiotic-resistant metabolic state to an antibiotic-sensitive state in both lab-evolved and clinically isolated CR-PA. This reprogramming leads to increased meropenem uptake

Interactions of neurotoxins with non-NMDA glutamate receptors: an autoradiographic study.

Neurotoxic substances are discussed to cause neurodegeneration by acting as excitotoxins on glutamate receptors. We investigated the properties of L-beta-oxalyl-amino-alanine (L-BOAA) and 3,4, 6-trihydroxyphenlyalanine (6-OH-Dopa) at the alpha-amino-3-hydroxy-5-methyl-4-isoxazole-propionic acid (AMPA) glutamate receptor and that of L-BOAA and domoic acid at the kainate glutamate receptor in human hippocampus. (3H)AMPA binding in hippocampal subfields was inhibited by L-BOAA and 6-OH-Dopa with mean IC50-values in the low micromolar range. (3H)Kainate binding was inhibited by L-BOAA with similar
