Discovery of Novel Nonpeptidic Proteasome Inhibitors Using Covalent Virtual Screening and Biological Evaluation.

Many reported proteasome inhibitors, including the three clinically approved inhibitors, bortezomib, carfilzomib, and ixazomib, have peptidic structures. In this study, using a hybrid and versatile strategy for covalent virtual screening by combining warhead screening and preprocessing with GOLD and CovDock software that were applied to the ZINC virtual library, we identified multiple proteasome inhibitors with new nonpeptidic structural scaffolds. Proteasome inhibition assays confirmed the inhibitory activities of these new compounds. These results demonstrate the effectiveness of our computa

Organometallic compounds in oncology: implications of novel organotins as antitumor agents.

Since the introduction of cisplatin in cancer therapy, metal complexes and organometallic compounds have been gaining growing importance in oncology. The impressive clinical effectiveness of cisplatin is limited by significant side effects and the emergence of drug resistance. Thus, novel classic and unconventional Pt(II) and Pt(IV) complexes have been introduced in therapy or are presently in advanced clinical trials. Moreover, innovative non-platinum metal-based antitumor agents, whose activity does not rely on direct DNA damage and may involve proteins and enzymes, have been developed. Gold

Waaler Rose titres may decrease but increase again during chrysotherapy.

In 62 patients with seropositive rheumatoid arthritis, treated with gold compounds, the relationship between serological and clinical response was analysed, based on data on the rheumatoid factor (RF) titre, ESR, number of swollen joints and joint tenderness, through 3 years. Three groups were distinguished. The RF titre became normal in 19, remained elevated in 22, became normal and subsequently again elevated in 21. Responders and non-responders to chrysotherapy occurred in all 3 groups. The findings suggest involvement of pathogenetic mechanisms related to IgM-RF, and of pathogenetic mechan…

Pharmacological therapy of atypical facial pain: actuality and perspective.

Atypical facial pain is a condition, which creates a difficult challenge for every physician, either for diagnosis or for choice of treatment. There are very few controlled studies and evidence-based data about what constitutes the gold standard treatment. Most physicians treat this condition with antidepressant agents, thinking that the psychiatric aspects, often present in these patients, play a fundamental role in the pathogenesis of the condition. However, antidepressant drugs are effective also in non-psychiatric patients, and new compounds, with no effect on psychological aspects, have r

Chemistry and Structure of Silver Molecular Nanoparticles.

Silver and gold molecular nanoparticles (mNPs) are a relatively new class of molecular materials of fundamental interest. They are high-nuclearity metal-organic compounds, with ligated metal cores, where the different character of bonding in the ligand shell and metal core gives rise to many of the unique properties of these materials. Research has primarily focused on gold mNPs, due to their good stability and the ease with which they may be synthesized and processed. To understand these materials as a general class, however, it will be necessary to broaden research efforts to other metals. G

Managing urinary incontinence in women - a review of new and emerging pharmacotherapy.

INTRODUCTION: The pharmacological treatment of urinary incontinence and overactive bladder (OAB) has been, for a longer time, based on antimuscarinic agents. In recent years, two other pharmacological principles have been introduced for the treatment of OAB and urgency urinary incontinence: the β3-adrenergic agent mirabegron and botulinum neurotoxin. Meanwhile, there is lack of effective drugs for the treatment of stress incontinence. AREAS COVERED: This literature review presents synthetic compounds aimed to treat female urinary incontinence that are in phase II-III clinical development. EX

A new look at auranofin, dextromethorphan and rosiglitazone for reduction of glia-mediated inflammation in neurodegenerative diseases.

Neurodegenerative disorders including Alzheimer's disease are characterized by chronic inflammation in the central nervous system. The two main glial types involved in inflammatory reactions are microglia and astrocytes. While these cells normally protect neurons by providing nutrients and growth factors, disease specific stimuli can induce glial secretion of neurotoxins. It has been hypothesized that reducing glia-mediated inflammation could diminish neuronal loss. This hypothesis is supported by observations that chronic use of non-steroidal anti-inflammatory drugs (NSAIDs) is linked with lo

Subcorneal pustular dermatosis-type IgA pemphigus induced by thiol drugs.

We report a case of subcorneal pustular dermatosis (SPD)-type IgA pemphigus arising in a 49 year-old woman with rheumatoid arthritis who had been treated with chrysotherapy. Scaly erythemic plaques containing vesicles and pustules occurred on her chest and abdomen during the course of anti-rheumatic treatments using prednisolone at 11 mg/day and thiol compounds (bucillamine and gold sodium thiomalate). Histological investigations revealed subcorneal pustules containing many neutrophils and a few acantholytic cells, and intercellular IgA deposits at the upper epidermis of the eruptions without

Experimental models of Parkinson's disease: insights from many models.

Toxin-induced and genetic experimental models have been invaluable in investigating idiopathic Parkinson's disease (PD). The neurotoxins--reserpine, 6-hydroxydopamine (6-OHDA), 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP), and methamphetamine--have been used to develop parkinsonian models in a wide variety of species. Both 6-OHDA and MPTP can replicate the neurochemical, morphologic, and behavioral changes seen in human disease. The unilateral 6-OHDA rat model is an excellent model for testing and determining modes of action of new pharmacologic compounds. The nonhuman primate MPTP-indu

Sulfhydryl drug-induced eruption: a clinical and histological study.

Sulfhydryl drug-induced skin eruptions were studied clinically and histologically in 23 patients. In this study, tiopronin, D-penicillamine, captopril and gold sodium thiomalate were considered to be sulfhydryl drugs, because they have a thiol group or release sulfhydryl compounds. The clinical features included skin eruptions that were maculopapular, erythema multiforme-like, eczematous, psoriasis-like, seborrheic dermatitis-like, Gibert-like, lichen planus-like, and pemphigus-like. These clinical findings were reminiscent of the wide variety of eruptions seen in cutaneous graft-versus-host r

Off-Stoichiometry Thiol-Ene Surface Functionalization: Example with Gold Nanoparticles.

Surface modification is essential in microfluidic applications due to the inherent hydrophobicity of polymers, which can lead to biofouling and reagent denaturation. Despite the development, challenges such as hydrophobic molecule absorption and limitations in scaling are still present. Off-stoichiometry thiol-ene (OSTE) materials have emerged as a promising alternative, offering advantages like rapid prototyping, minimal hydrophobic absorption, and customizable surface chemistries. While the thiol-ene polymerization mechanism is well understood, the fundamental understanding of thiol group bi

Identification of novel triazolylquinoxaline-based metal complexes supported by experimental and virtual screenings.

The formation of novel complexes from so far non-investigated ligands and different metal centers is important for the development of new functional materials such as (photo)catalysts or biologically active compounds. Still, promising strategies to quickly and systematically investigate the complexation behavior of selected ligands are rare. We developed an NMR-based screening approach to monitor changes within reaction mixtures containing metals and ligands on a small scale via a simple but reliable protocol. Based on the obtained data, we could draw conclusions on the formation of 2-(1',2',3

Indole-3-Carbinol Mechanisms Combating Chemicals and Drug Toxicities.

The toxicity of chemicals and drugs is a common crisis worldwide. Therefore, the search for protective compounds is growing. Natural compounds such as indole-3-carbinol (I3C) derived from cruciferous vegetables are preferred since they are safe for humans and the environment. This review focuses on I3C potential role in preventing and repairing damage caused by chemicals and drugs. Interestingly, I3C ameliorates hepatotoxicity induced by carbon tetrachloride (CCl4), diethylnitrosamine (DENA), alcohol, gold nanoparticles, and microbial toxins. Additionally, it inhibits carcinogenesis induced by

Virtual screening, identification and in vitro validation of small molecule GDP-mannose dehydrogenase inhibitors.

Upon undergoing mucoid conversion within the lungs of cystic fibrosis patients, the pathogenic bacterium Pseudomonas aeruginosa synthesises copious quantities of the virulence factor and exopolysaccharide alginate. The enzyme guanosine diphosphate mannose dehydrogenase (GMD) catalyses the rate-limiting step and irreversible formation of the alginate sugar nucleotide building block, guanosine diphosphate mannuronic acid. Since there is no corresponding enzyme in humans, strategies that could prevent its mechanism of action could open a pathway for new and selective inhibitors to disrupt bacteri

High-content imaging assay to evaluate Toxoplasma gondii infection and proliferation: A multiparametric assay to screen new compounds.

Toxoplasma gondii is an intracellular protozoan parasite widely distributed in animals and humans. Infection of host cells and parasite proliferation are essential steps in Toxoplasma pathology. The objective of this study was to develop and validate a novel automatic High Content Imaging (HCI) assay to study T. gondii infection and proliferation. We tested various fluorescent markers and strategies of image analysis to obtain an automated method providing results comparable to those from gold standard infection and proliferation assays. No significant difference was observed between the resul
