Assessment of in vitro biofilm formation by Helicobacter pylori.

BACKGROUND AND AIMS: Biofilms are surface-bound communities of bacterial cells that are implicated in their survival. As with various bacteria studied to date, Helicobacter pylori can have an alternate lifestyle as a biofilm. We previously reported that strain TK1402 showed a strong biofilm-forming ability in vitro. However, the mechanisms of its biofilm development remain unclear. We analyzed the basic characteristics of the biofilm-forming ability in strain TK1402. METHODS: In order to characterize the biofilm-forming ability of the H. pylori strains, auto-aggregation, motility and hydropho…

Maintenance of Helicobacter pylori cultures in agar stabs.

BACKGROUND: Helicobacter pylori requires frequent passage at 37 °C with reduced oxygen tension to maintain viability, and recovery from frozen stocks can be unpredictable and slow. Agar stab cultures were assessed as a possible means of maintaining viability without the need to passage every 4-7 days. MATERIALS AND METHODS: Agar stabs prepared from either Brucella or Brain Heart Infusion media were inoculated deeply with H. pylori strains or H. felis and grown under varying conditions for up to 13 weeks. Subcultures were prepared from these stabs at various intervals to test for viability. R…

Helicobacter pylori's unconventional role in health and disease.

The discovery of a bacterium, Helicobacter pylori, that is resident in the human stomach and causes chronic disease (peptic ulcer and gastric cancer) was radical on many levels. Whereas the mouth and the colon were both known to host a large number of microorganisms, collectively referred to as the microbiome, the stomach was thought to be a virtual Sahara desert for microbes because of its high acidity. We now know that H. pylori is one of many species of bacteria that live in the stomach, although H. pylori seems to dominate this community. H. pylori does not behave as a classical bacterial …

[Periapoptotic markers in children with Helicobacter pylori infection].

UNLABELLED: Helicobacter pylori (Hp) infection influences cell metabolism and apoptosis in the epithelium and lymphocytes of gastric mucosa. It may cause difficulties in the elimination of bacteria and lead to chronic gastritis. THE AIM OF THE STUDY was to find if there is a relationship between periapoptotic markers such as Fas, FasL and Bcl-2 in gastric mucosa in children with chronic gastritis and with Hp infection. MATERIAL AND METHODS: Forty-nine children with chronic abdominal pain were included in the study. They, were divided into three groups: group I without Hp (22) group II with (2…

[Helicobacter pylori infection in children with celiac disease].

UNLABELLED: The purpose of this study is to evaluate symptomatology, endoscopic and histopathologic changes of Helicobacter pylori infection and gastritis lesions without Helicobacter pylori infection on children diagnosed with celiac disease. MATERIAL AND METHOD: 15 children under gluten-free diet were selected and, because of the recurrence of the dyspeptic syndrome, an upper digestive endoscopy associated with histopathologic exam was performed. Considering the histopathologic result we made two groups: first group (8 children with celiac disease and Helicobacter pylori infection) and seco…

Glaucoma and Helicobacter pylori infection: correlations and controversies.

A possible association between Helicobacter pylori infection and eye diseases, including Sjögren syndrome, blepharitis, central serous chorioretinopathy and uveitis, has been proposed. Glaucoma is the second leading cause of blindness in the world, after cataracts, and the leading cause of irreversible blindness, but many aspects of its pathogenesis remain unknown. H pylori infection may influence the pathophysiology of glaucoma by releasing various proinflammatory and vasoactive substances, as well as by influencing the apoptotic process, parameters that may also exert their own effects in th…

Role of endogenous cortisol on Helicobacter pylori colonization.

BACKGROUND AND AIMS: Helicobacter pylori infects the gastric mucosa and can lead to chronic gastritis, gastric ulcer and gastric cancer. Colonization of H. pylori in the gastric mucosa is influenced by a variety of host, bacterial and environmental factors. Host defense mechanisms have been affected by endogenous glucocorticoids. We aimed to investigate the relationship between H. pylori and endogenous glucocorticoid. MATERIALS AND METHODS: Forty cases with endoscopically and histologically proven H. pylori and 26 patients who did not have H. pylori on gastric biopsy samples were enrolled in …

Controversies in the Helicobacter pylori/duodenal ulcer story.

In patients with Helicobacter pylori-positive duodenal ulcer (DU), the organism must be eradicated to achieve rapid, stable healing. However, evidence is against much else that is commonly accepted. (1) Does H. pylori cause the ulcer? Evidence against includes archaeopathology, geographical prevalence, temporal relationships and H. pylori-negative DU patients. DU can recur after eradication of H. pylori infection, and DUs may remain healed after reduction of acid secretion despite persistent infection. The faster healing of ulcers when H. pylori has been eradicated is due to the organism's int…

Helicobacter pylori culture and antimicrobial resistance in Iran.

OBJECTIVE: Helicobacter pylori is considered as an important etiologic factor in pathogenesis of peptic ulcer disease, chronic gastritis and gastric cancer. To eradicate this micro-organism, numerous regimens containing various antimicrobial agents have been suggested. However, H pylori antimicrobial resistance is a leading factor to treatment failure and recurrence of the disease. The aim of the study was to evaluate the prevalence of H pylori resistance to metronidazole, clarithromycin, tetracycline, amoxicillin, erythromycin and furazolidone in authors pediatric patients. METHODS: Antral b…

Inverse associations of Helicobacter pylori with asthma and allergy.

BACKGROUND: Acquisition of Helicobacter pylori, which predominantly occurs before age 10 years, may reduce risks of asthma and allergy. METHODS: We evaluated the associations of H pylori status with history of asthma and allergy and with skin sensitization using data from 7663 adults in the Third National Health and Nutrition Examination Survey. Adjusted odds ratios (ORs) for currently and ever having asthma, allergic rhinitis, allergy symptoms in the previous year, and allergen-specific skin sensitization were computed comparing participants seropositive for cagA(-) or cagA(+) strains of H p…

A historical perspective of Helicobacter gastroduodenitis and its complications.

The discovery of Helicobacter pylori was one of the most notable in gastroenterology - and indeed medicine. The century before Marshall and Warren's discovery is peppered with isolated accounts of spiral-shaped bacteria in the stomach. The discovery of H. pylori, and the recognition of its importance, came about as a consequence of combined clinical, pathological and microbiological work. The discovery has transformed the treatment of peptic ulcers and other related diseases.

Recent Advances in the Treatment of Helicobacter pylori Infection.

The approach to the treatment of Helicobacter pylori infection has changed during the last years. In fact, during the last decade, the success rate of usual eradication regimens, based on a proton pump inhibitor plus clarithromycin associated with amoxicillin or metronidazole, declined from over 90% to about 80%, a critical threshold under which the eradication rate is considered unsatisfactory, according to current guidelines. This finding is mainly due to the raising prevalence of clarithromycin resistance, which is in turn linked to the widespread use of this antibiotic for respiratory trac…

Helicobacter pylori eradication for the prevention of gastric cancer.

Even though Helicobacter pylori infection is an obvious cause of chronic non-atrophic and atrophic gastritis, as well as gastric dysplasia and cancer, several recent controlled intervention trials for the prevention of gastric cancer by H. pylori eradication have yielded disappointing results. They showed that cancer eradication may still appear in relatively high frequency after successful treatment of H. pylori. One explanation for this observation is that H. pylori treatment has less influence in development of gastric cancer in atrophic gastritis than in development of cancers in non-atrop…

Expression of NF-kappaB in Helicobacter pylori infection.

Helicobacter pylori colonizes the gastric mucosa in humans and causes chronic gastritis. NF-kappaB has a key role as a mediator in mucosal inflammation. In this study, we examined the expression of NF-kappaB in the antral epithelial cells of H. pylori-infected and H. pylori-uninfected biopsies and examined these processes in relationship with grade and activity of gastritis, density of H. pylori, presence of the intestinal metaplasia, and atrophy. Fifty biopsies (35 H. pylori-positive patients and 15 H. pylori-negative controls) were studied. NF-kappaB immunohistochemical stain was performed. …

[Inflammation as a basis for Helicobacter pylori-associated diseases].

The molecular-cellular features of gastric mucosal inflammation caused by Helicobacter pylori are formulated. Chronic gastritis is considered as the morphological basis of gastric ulcer and non-cardial cancer of the stomach of intestinal type. The basic influences of the products of a pathogenicity island of bacterium (Cag A-protein) and VacA-cytotoxin on the gastric mucosa, as well as bacterial genetic polymorphism are discussed. The evolution of the phenotypes of chronic gastritis-- "ulcerative" "cancerous" is analyzed. Helicobacter pylori-associated inflammation may depend on the host's inf…
