Keratose Hydrogel Drives Differentiation of Cardiac Vascular Smooth Muscle Progenitor Cells: Implications in Ischemic Treatment.

Peripheral artery disease (PAD) is a common vascular disorder in the extremity of limbs with limited clinical treatments. Stem cells hold gr … nitor cell (cVSMPC) differentiation and the therapeutic potential of the resultant VSMCs in a mouse hindlimb ischemic model of PAD. The results demonstrated that KOS but not collagen hydrogel was able to drive the majority of cVSMPCs into functional VSMCs in a defined Knockout serum replacement (SR) medium in the absence of differentiation inducers. This e …

Intravital Microscopy of Monocyte Homing and Tumor-Related Angiogenesis in a Murine Model of Peripheral Arterial Disease.

The therapeutic goal for peripheral arterial disease and ischemic heart disease is to increase blood flow to ischemic areas caused by hemody … asive options. Our group examines arteriogenesis after monocyte transplantation into mice using the hindlimb ischemia model. Previously, we have demonstrated improvement in hindlimb perfusion using tetanus-stimulated syngeneic monocyte transplantation. In addition to the effects on the collateral formation, tumor growth could be affected by this therapy as …

Arthroscopy of the normal cadaveric ovine femorotibial joint: a systematic approach to the cranial and caudal compartments.

OBJECTIVES: Preclinical studies using large animal models play an intergral part in translational research. For this study, our objectives were: to develop and validate arthroscopic approaches to four compartments of the stifle joint as determined via the gross and arthroscopic anatomy of the cranial and caudal aspects of the joint. METHODS: Cadaveric hindlimbs (n = 39) were harvested from mature ewes. The anatomy was examined by tissue dissection (n = 6), transverse sections (n = 4), and computed tomography (n = 4). The joint was arthroscopically explored in 25 hindlimbs. RESULTS: A cranio-…

Ontogenetic, gravity-dependent development of rat soleus muscle.

We tested the hypothesis that rat soleus muscle fiber growth and changes in myosin phenotype during the postnatal, preweaning period would be largely independent of weight bearing. The hindlimbs of one group of pups were unloaded intermittently from postnatal day 4 to day 21: the pups were isolated from the dam for 5 h during unloading and returned for nursing for 1 h. Control pups were either maintained with the dam as normal or put on an alternating feeding schedule as described above. The enlargement of mass (approximately 3 times), increase in myonuclear number (approximately 1.6 times) an…

[An experimental study on congenital foot deformity--with reference to a local disturbance in the limb bud using the exo utero survival method].

Experimental studies on abnormal developments in mammalian embryos have been limited by an inaccessibility to the embryo for surgical manipulations. Here we have developed the exo utero survival method proposed by Muneoka et al. in 1986 to produce models of various hindlimb anomalies by cauterizing a portion of the hindlimb bud of the mouse embryo. A total of 500 mice were used. The treatment was performed on time-pregnant JCL/ICR mice at 10.5 to 12.5 days of gestation (plug day = day 0). Polydactyly occurred in the group treated with pre-axial cauterization at 10.5 to 11.0 days of gestation, …

Trunk movements during locomotion in the marsupial Monodelphis domestica (didelphidae).

The small didelphid cmarsupial, Monodelphis domestica, uses a lateral sequence walk during slow treadmill locomotion and gradually shifts to … the pelvic girdle in the plane of yaw. Pelvic rotation is synchronized with the locomotor cycle of hindlimbs. Each side of the pelvis rotates forward during the recovery phase of the ipsilateral hindlimb and backward during the contact phase of this limb. Information on locomotor trunk movements in other limbed tetrapods is limited. The pattern of trunk b …

Aerobic Training Alleviates Muscle Atrophy by Promoting the Proliferation of Skeletal Muscle Satellite Cells in Myotonic Dystrophy Type 1 by Inhibiting Glycolysis via the Upregulation of MBNL1.

BACKGROUND: Skeletal muscle atrophy in myotonic dystrophy type 1 (DM1) is caused by abnormal skeletal muscle satellite cell (SSC) proliferat … cular mechanisms. HSALR mice were subjected to 4 weeks of aerobic training. After aerobic training, hindlimb grip, and myofiber mean cross-sectional area (CSA) detected by haematoxylin and eosin (HE) staining were performed. In DM1 primary SSCs, cell proliferation was assessed using Pax7 and MyoD immunofluorescence and CCK-8 assays, RNA foci were detected …

Lateral cervical nucleus in the cat: functional organization and characteristics.

1. The lateral cervical nucleus (LCN) was investigated with extracellular recordings in the anesthetized cat. A total of 556 LCN units were characterized; the locations of most of these were histologically verified. Half of these had receptive fields on the rostral third of the ipsilateral body surface including the face; 14% had fields on the thorax or abdomen, 33% had fields on the hindlimb or tail, and about 3% had receptive fields larger than one limb. 2. The LCN was observed to be somatotopically organized in experiments using angled microelectrode penetrations. Hindlimb units were dorsol…

Actinomycotic mycetoma in a cat.

Actinomycotic mycetoma, a chronic, progressive infection of the subcutaneous tissue characterized by tumefaction, draining sinuses, and grains, was diagnosed in the right hindlimb of a young adult, male cat. The organisms that cause actinomycetoma are soil or plant saprophytes that gain entrance to the skin through abrasion or traumatic implantation. Streptomyces griseus, an organism generally considered to be a saprophyte, was cultured bacteriologically. Despite extensive surgery and long-term antibiotic therapy, the infection persisted, and the cat was euthanatized.

Dynorphin induces partially reversible paraplegia in the rat.

Endogenous opioids have previously been implicated in the pathophysiology of spinal cord injury, but the specific opioid(s) involved has not been determined. Here we report that dynorphin administered intrathecally causes dose-related, partially reversible, hindlimb paralysis in the unanesthetized rat. Other opioids, including mu-selective and delta-selective synthetic enkephalins or beta-endorphin, failed to induce motor dysfunction. These findings suggest that dynorphin or dynorphin-related peptides may play a role in spinal cord injury.

Metastatic phycomycosis in a horse.

Extensive subcutaneous phycomycosis of the lower hindlimb developed over a period of 8 months in a Thoroughbred filly. There was no response to treatments such as trichlorphon, sodium iodide, or etisazole. Extension of the limb lesion over the following 3 months was associated with an enlarged inguinal lymph node, which was removed. Characteristic phycomycotic lesions were observed in the node, and a fungus having the morphologic features of Hyphomyces destruens was isolated from both the limb and nodal lesions.

Homeotic transformation of tails into limbs in anurans.

Anuran tadpoles can regenerate their tails after amputation. However, they occasionally form ectopic limbs instead of the lost tail part after vitamin A treatment. This is regarded as an example of a homeotic transformation. In this phenomenon, the developmental fate of the tail blastema is apparently altered from that of a tail to that of limbs, indicating a realignment of positional information in the blastema. Morphological observations and analyses of the development of skeletal elements during the process suggest that positional information in the blastema is rewritten from tail to trunk

Validation of a modified algometer to measure mechanical nociceptive thresholds in awake dogs.

This study was conducted to validate the use of a modified algometer device to measure mechanical nociceptive thresholds in six dogs. Dogs were administered morphine intravenously (IV) at 1 mg/kg or saline at equivolume in a crossover design with one-week washout period. Mechanical nociceptive thresholds were determined before, after the administration of treatments at 5 minutes, and hourly for 8 hours. Thresholds were recorded at the carpal pad, metacarpal foot pad, tibia, femur, and abdomen. Heart rates, body temperature, and respiration were recorded at similar time points. Thresholds incre

Skeletal development of Macrochelys temminckii (Reptilia: Testudines: Chelydridae).

Few descriptions of the development and sequence of chondrification and ossification of the entire skeleton of turtles exist, particularly compared to other groups of reptiles. In this study, the embryonic skeleton and its ontogenesis are described for the Alligator Snapping Turtle, Macrochelys temminckii (Chelydridae). Morphological descriptions utilize cleared and double-stained embryonic specimens and form the basis of comparison of the ontogenesis of the skeleton between this species and its extant sister taxon, Chelydra serpentina. The embryonic chondrocranium, as well as the sequences of
