Canine Staphylococcus pseudintermedius sinonasal infection in human hosts.

BACKGROUND: Staphylococcus pseudintermedius is a gram-positive bacterium commonly found as part of the normal skin and nasal flora of healthy dogs. It may act as an opportunistic pathogen in dogs, but has also been shown to colonize the nasal mucosa of humans. We report 4 cases of chronic rhinosinusitis (CRS) refractory to aggressive medical management with cultures that grew S. pseudintermedius, with clinical improvement only after initiating culture-directed therapy. METHODS: Retrospective review of 4 patients with CRS treated at a tertiary academic medical center with sinonasal cultures gr

Remodeling memory T cells with chemotherapy and immune checkpoint inhibitors as host pre-conditioning to empower in vivo CAR-T therapy.

Chemotherapy and immune checkpoint inhibitors (ICIs) are widely utilized in cancer treatment, exerting not only direct cytotoxic effects on tumor cells but also significantly reshaping the systemic immune status of patients, particularly the composition and function of the T cell repertoire. This review begins with the analysis of the widespread depletion and dysfunction of early T cell subsets in cancer patients, particularly naïve T cells (Tn) and stem-like memory T cells (Tscm). Afterward, it details how chemotherapy and ICIs differentially regulate T cell homeostasis, including their abili

Combatting multidrug resistance in Klebsiella pneumoniae: mechanisms, global trends, and innovative therapeutic strategies.

Emergence of drug-resistant bacterial infections, particularly those caused by Klebsiella pneumoniae, represents a growing public health issue. K. pneumoniae is the leading cause of nosocomial infections, associated with diverse diseases and high mortality rates. Its ability to develop multiple resistance mechanisms, including biofilm formation, efflux pump activity, β-lactamase production, enzymatic modification, and porin loss, contributes to its resistance to conventional antibiotics. These challenges urge the ned for novel therapeutics and alternative therapies. While resistance rates rema

Does ABPA Contribute to Bronchiectasis? A Structured Evaluation of Competing Hypotheses.

Whether allergic bronchopulmonary aspergillosis (ABPA) causes bronchiectasis or merely represents Aspergillus fumigatus colonisation of damaged airways remains debated. Establishing causality is challenging because airway damage is predominantly driven by host immune responses rather than by direct fungal invasion. We systematically evaluate four competing hypotheses: (i) ABPA causes bronchiectasis, (ii) reverse causation, with bronchiectasis predisposing to ABPA, (iii) a shared underlying factor independently producing both conditions and (iv) a spurious association. We evaluate radiological,

Current and emerging therapeutic strategies targeting adherent-invasive Escherichia coli in Crohn's disease: a review.

Considerable evidence has accumulated over the past two decades demonstrating that Escherichia coli, specifically adherent-invasive E. coli, contributes to the pathogenesis of Crohn's disease. Adherent-invasive E. coli can adhere to and invade intestinal epithelial cells, survive, and replicate within macrophages, thereby enabling a key mechanism that induces chronic inflammation. Despite extensive knowledge of the molecular interactions between adherent-invasive E. coli and the host from these studies, translating this knowledge into targeted therapy remains limited. In this review, we summari

Oridonin mitigates bacterial pneumonia by regulating mitochondrial integrity and ferroptosis via targeting KEAP1/NRF2 signaling.

Methicillin-resistant Staphylococcus aureus (MRSA) is a highly virulent and drug-resistant pathogen frequently causing bacterial pneumonia. Currently, there are limited effective treatments available due to the rapidly evolving resistance of bacteria. Therefore, there is an urgent need to develop novel therapies that focus on host-pathogen interactions. Oridonin is a naturally occurring diterpenoid with multiple pharmacological effects, but its therapeutic potential in bacterial pneumonia, as well as its action mode, remains largely unknown. Here, we demonstrated that oridonin conferred protec

A Conceptual Review of Gut, Skin, and Oral Microbiota in Autoimmune Bullous Diseases: From Dysbiosis to Therapeutic Potential.

Autoimmune bullous diseases (AIBDs), including pemphigus and bullous pemphigoid, are chronic inflammatory skin disorders characterized by dysregulated immune responses mediated by autoantibodies that target adhesion molecules in the skin and mucous membranes. Emerging evidence highlights the pivotal role of host microbiota dysbiosis in AIBDs pathogenesis, offering novel insights into disease mechanisms and therapeutic strategies. This review systematically synthesizes the current findings on gut, skin, and oral microbiota alterations in AIBDs, emphasizing their contributions via the gut-skin a

Adjunctive Immunotherapeutic Efficacy of N-Formylated Internal Peptide of Mycobacterial Glutamine Synthetase in Mouse Model of Tuberculosis.

BACKGROUND: Host-directed therapies are a comparatively new and promising method for the treatment of tuberculosis. A variety of host pathways, vaccines and drugs have the potential to provide novel adjunctive therapies for the treatment of tuberculosis. In this connection, we have earlier reported the immunotherapeutic potential of N-formylated N-terminal peptide of glutamine synthetase of Mycobacterim tuberculosis H37Rv (Mir SA and Sharma S, 2014). Now in the present study, we investigated the immunotherapeutic effect of N-terminally formylated internal-peptide 'f- MLLLPD' of mycobacterial g

Echinocandins for the Treatment of Invasive Aspergillosis: from Laboratory to Bedside.

Echinocandins (caspofungin, micafungin, anidulafungin), targeting β-1,3-glucan synthesis of the cell wall, represent one of the three currently available antifungal drug classes for the treatment of invasive fungal infections. Despite their limited antifungal activity against Aspergillus spp., echinocandins are considered an alternative option for the treatment of invasive aspergillosis (IA). This drug class exhibits several advantages, such as excellent tolerability and its potential for synergistic interactions with some other antifungals. The objective of this review is to discuss the in vi

Preclinical evaluation of oncolytic vaccinia virus for therapy of canine soft tissue sarcoma.

Virotherapy using oncolytic vaccinia virus (VACV) strains is one promising new strategy for canine cancer therapy. In this study we describe the establishment of an in vivo model of canine soft tissue sarcoma (CSTS) using the new isolated cell line STSA-1 and the analysis of the virus-mediated oncolytic and immunological effects of two different Lister VACV LIVP1.1.1 and GLV-1h68 strains against CSTS. Cell culture data demonstrated that both tested VACV strains efficiently infected and destroyed cells of the canine soft tissue sarcoma line STSA-1. In addition, in our new canine sarcoma tumor x

Targeting the JAK/STAT pathway in chronic viral infections: opportunities and challenges for immunomodulation.

The discovery of the Janus kinase/signal transducer and activator of transcription (JAK/STAT) pathway as a key regulator of immune function and inflammation led to the development of JAK inhibitors (JAK-i), several of which are now approved for autoimmune and chronic inflammatory conditions. Recently, JAK-i have been repurposed for infectious diseases, most notably with baricitinib, a United States Food and Drug Administration (FDA)-approved JAK1/2 inhibitor for individuals hospitalized for COVID-19, demonstrating a survival benefit by dampening cytokine-mediated hyperinflammation. This has ge

Gp350-targeted CAR-T therapy in EBV-positive Burkitt lymphoma: pre-clinical development of gp350 CAR-T.

BACKGROUND: Epstein-Barr virus (EBV) is an oncovirus belonging to the herpesvirus family, associated with the pathogenesis of multiple malignancies, particularly Burkitt lymphoma (BL). The virus remains latent in host cells and plays a critical role in tumor progression through various mechanisms. A key glycoprotein, gp350, expressed during the lytic phase of EBV, is instrumental in viral entry into B cells and presents a unique antigenic target, making it a promising candidate for immunotherapeutic approaches, such as chimeric antigen receptor T-cell (CAR-T) therapy. METHODS: In this study,

Maintenance Strategies After Hematopoietic Cell Transplantation.

Hematopoietic cell transplantation (HCT) is an essential component of potentially curative therapy for patients with hematologic malignancies. High-dose chemotherapy with autologous (auto) stem cell rescue is used to overcome chemoresistance in multiple myeloma, non-Hodgkin lymphoma, and Hodgkin lymphoma. Alternatively, poor-risk acute leukemias rely on the graft versus leukemia effect of allogeneic (allo) products. Long-term remissions are feasible with both auto- and allo-HCT; however, disease relapse is the leading cause of death after HCT for many patients. In recognition of this, novel th

Oral tolerance: a new tool for the treatment of gastrointestinal inflammatory disorders and liver-directed gene therapy.

Oral tolerance is a method of downregulating an immune response by feeding antigens. The use of oral tolerance toward adenoviruses and colitis-extracted proteins for long term gene therapy and alleviation of experimental colitis, and the mechanisms of tolerance induction are presented. Adenoviruses are efficient vectors in liver-directed gene therapy; however, the antiviral immune response precludes the ability to achieve long term gene expression and prohibits the ability to reinject the recombinant virus. Oral tolerance induction via feeding of viral-extracted proteins prevented the antiaden

Technology-enabled activation of skin cancer screening for hematopoietic cell transplantation survivors and their primary care providers (TEACH).

BACKGROUND: Hematopoietic cell transplantation (HCT) is a curative option for a growing number of patients with hematologic diseases and malignancies. However, HCT-related factors, such as total body irradiation used for conditioning, graft-versus-host disease, and prolonged exposure to immunosuppressive therapy, result in very high risk for melanoma and non-melanoma skin cancer (NMSC). In fact, skin cancer is the most common subsequent neoplasm in HCT survivors, tending to develop at a time when survivors' follow-up care has largely transitioned to the primary care setting. The goal of this s

Host miRNA orchestration of mitophagy pathways during bacterial infection: current advances and future directions.

Pathogenic bacterial infections continue to impose a substantial global health burden. MicroRNAs (miRNAs), a class of small non-coding RNAs, have emerged as critical post-transcriptional regulators of host innate immunity and inflammatory responses during bacterial pathogenesis. Accumulating evidence indicates that miRNAs dictate the outcomes of infection by orchestrating fundamental cellular processes, notably mitophagy-a selective autophagic pathway essential for mitochondrial quality control and cytosolic surveillance. While recent studies have begun to elucidate the molecular interplay bet

Viral receptors as host-pathogen control nodes linking entry, signalling, and system-level pathogenesis.

Viral receptors have traditionally been viewed as docking sites that mediate viral attachment and entry. However, this perspective no longer captures the full complexity of receptor function during infection. Receptor engagement links viral binding to intracellular trafficking, signal transduction, barrier integrity, and ultimately disease outcome. Rather than acting solely as entry determinants, receptors influence endocytic routing, cellular permissiveness, tissue tropism, immune responses, and systemic dissemination. A functional distinction between attachment factors, entry receptors, co-r

PRRSV interaction with the adaptive immune system of host: an update review.

Porcine reproductive and respiratory syndrome virus (PRRSV), a highly variable RNA virus, causes a significant threat to the global swine industry. PRRSV suppresses both innate and adaptive immune responses in the host. While only the regulation of innate immunity by PRRSV has been summarized. This article systematically reviews the regulatory roles of PRRSV on the host's adaptive immune system, and summarizes its impact on the behavior of T cells, B cells, macrophages, monocytes, dendritic cells, and natural killer cells (NK cells). PRRSV evades antiviral immunity through multiple mechanisms,
