Physiology of invasive Plasmodium merozoites.

Malaria, a life-threatening disease caused by Plasmodium parasites, relies on a specialized stage, the merozoite to invade red blood cells and multiply in the bloodstream. While much research has focused on the molecular interactions between merozoite and host cell surface, invasion is a complex process that also requires the parasite to move actively, sense its environment, and respond to signals such as changes in ions or contact with the red blood cell. These cues trigger precise biological responses, including the secretion of specialized organelles and activation of gliding motility, enab

Equine cyathostomins-Hidden histories.

Cyathostomins (also known as small strongyles or small redworms) are ubiquitous parasites of equids. They are found in a wide range of climatic regions, and inhabit the large intestine of their hosts. While infections are often sub-clinical, clinical consequences ranging from chronic to acute, and most notably, acute larval cyathostominosis, can occur in specific circumstances. Control of cyathostomin infection is complicated by widespread drug resistance. This Chapter provides a broad review of cyathostomin biology and host-parasite relationships, and also highlights significant knowledge gap

Morphometric variability of Fasciola hepatica eggs among definitive host species: A multivariate approach.

Egg morphometry provides valuable information for investigating phenotypic variation and host-associated patterns in Fasciola hepatica. However, comparative studies evaluating egg morphometric variation across multiple definitive host species remain limited. Therefore, the aim of this study was to evaluate morphometric variation in F. hepatica eggs recovered from different definitive host species using univariate, multivariate, and host-level classification approaches. A total of 690 eggs obtained from 60 independent host individuals representing cattle, sheep, goats, horses, mules, donkeys, a

The long and winding road: On the endoplasmic reticulum of Plasmodium and implications for pathogenesis.

As a eukaryote, Plasmodium, the causative agent of malaria, is reliant on its endoplasmic reticulum (ER) for survival. However, as a protozoan pathogen, Plasmodium has also adapted its ER to support its parasitic lifestyle. This Pearl focuses on the interplay between conserved ER functions in Plasmodium such as cell signaling, protein synthesis/secretion, lipid production, and cellular stress, and their contribution to parasite pathogenesis. We further summarize the current knowledge about the role of the ER in pathogenic processes such as motility, egress/invasion, host-cell remodeling, and t

Recent advances in Cryptosporidium secreted proteins.

As an apicomplexan parasite, Cryptosporidium possesses the conserved secretory organelles, micronemes, rhoptry and dense granules in addition to the unique small granules. These specialized organelles house various effectors that are secreted upon invasion, helping to establish cryptosporidiosis, a leading cause of severe diarrhea among children. Only recently has its proteome been defined, laying the foundation to unravel host-parasite interactions that are poorly understood in Cryptosporidium. Recent technical advances in genetic manipulation have helped elucidate the role of a few secreted …

Success of parasitism of Nezara viridula and Halyomorpha halys (Hemiptera: Pentatomidae) by Trichopoda pennipes (Diptera: Tachinidae) in the southeastern United States.

Nezara viridula (L.) and Halyomorpha halys (Stål) are stink bug pests that damage fruit in a variety of crops throughout the southeastern United States. Trichopoda pennipes (F.) parasitizes N. viridula late instars and adults, however, little is known regarding parasitism of H. halys by this tachinid in systems where both stink bug species coexist. Thus, our main objective was to analyze patterns of T. pennipes parasitism for adult N. viridula and H. halys in corn, cotton, and soybean. We also assessed location of T. pennipes eggs on stink bug bodies as well as successful emergence from parasi

Host sphingolipids support Plasmodium berghei liver stage development.

During the asymptomatic liver stage, Plasmodium resides within a parasitophorous vacuole (PV) that protects the parasite from immune clearance while also restricting nutrient exchange with its host cell. Although it is known that Plasmodium must scavenge resources from its environment, the specific nutrients sequestered and the mechanisms for transporting them to the PV are poorly understood, particularly during the liver stage. In this study, we investigated the role of host lipids and discovered that sphingolipids are critical for both Plasmodium berghei liver stage development and invasion.

Deciphering the molecular targets of Plasmodium and Anopheline interactions for malaria control.

Malaria is a severe disease that is transmitted by female Anopheles mosquitoes and caused by the Plasmodium parasite. Despite a decrease in mortality rate, it continues to pose significant challenges such as resistance to antimalarial drugs and insecticides, which necessitates the need for novel malaria control and elimination strategies. To identify new molecular targets for malaria control, there is a need to understand the molecular interaction between mosquitoes and parasites. Plasmodium ookinetes interact with the mosquito midgut proteins during midgut invasion and sporozoites interact wi

The first phylogenetic reconstruction of Nippostrongylinae (Nematoda: Heligmonellidae) reveals 3 new genera, the polyphyletic nature of Carolinensis and Vexillata, and identifies 5 clades with varying associations with mammals.

The Nippostrongylinae is a group of strongylid nematodes that includes species typically associated with coprophagous mammals; in the New World, it is represented by 82 species within 11 genera. Two main morphological features, the synlophe and the caudal bursa, are used to evaluate the characteristics that allow identification and classification of the organisms in the taxon. However, the analysis of these characters often requires a partial or total destruction of specimens and therefore morphological variation is studied in only a very small subset of organisms per species. To evaluate the

Searching for a common host: parasitoids of Lema daturaphila on Datura stramonium in Central Mexico.

BACKGROUND: Natural enemies of herbivore insects can change the arms race between plants and insects. Their presence and abundance even can affect the co-evolution of interacting species. The annual herb Datura stramonium varies geographically in the extent of its direct defenses against herbivores. Its main specialist herbivore, Lema daturaphila, is adapted to cope with these defenses, but little is known about its natural enemies. Here, we determined the presence and incidence of L. daturaphila parasitoids as an initial step to explore other ecological and evolutionary relationships in a tri

Development of the Roe Deer-Fascioloides magna Association over Time.

The trematode Fascioloides magna is originally a parasite of North American deer species. Upon its arrival to Europe, F. magna met new intermediate and final hosts. Depending on the type of host, the clinical picture, pathological findings, epidemiology and outcome can vary significantly. As an aberrant host, it was long believed that the roe deer (Capreolus capreolus) fails to develop pseudocysts, and therefore the infected animal dies before the parasite can mature and start to produce eggs. In this study, 676 roe deer livers were collected in Croatia during the hunting years of four consecu

Observations on morphological, physiological, and behavioural changes induced by protozoan infections in certain storage insect pests.

Entomopathogenic protozoans are, to a large extent, host specific, and through acute or chronic infections they negatively alter host reproductive fitness, metabolism, immune response, juvenile hormonal balance, and host development. Hence, these entomopathogens are well-suited to reduce populations of stored product insect pests. The first step towards achieving successful suppression, natural or applied, of storage insect pest populations is the detection for these entomopathogenic protozoans in their habitats. Therefore, a preliminary survey of naturally occurring protozoan infections in st

Perforation of the host cell plasma membrane during Toxoplasma invasion requires rhoptry exocytosis.

Toxoplasma gondii is an obligate intracellular parasite. Proteins released during host cell invasion from apical secretory organelles known as rhoptries are delivered into the host cell cytosol to perform functions critical for parasite survival and virulence. How these effector proteins move across the host cell plasma membrane is unknown but may involve a previously noted temporary loss of host cell plasma membrane barrier integrity. Here, we use high-speed, multi-wavelength fluorescence imaging to spatially monitor the barrier integrity of the host cell plasma membrane, in real time, during

Reviving knowledge: A 70-year gap in the host-parasite relationship between Trichodectes galictidis (Ischnocera: Trichodectidae) and Galictis cuja (Molina, 1782) (Carnivora: Mustelidae).

Galictis cuja is one of the least known mustelids in South America, with its frequency and distribution poorly documented across the Neotropical region from Mexico to Brazil. In February 2024, a female was rescued by fishermen on Porto Beach in Imbituba, Santa Catarina. A clinical examination revealed ectoparasites on the animal, identified as Trichodectes galictidis, a louse species not detected in these animals for over 70 years. This study explores the revival of knowledge regarding the host-parasite relationship between Trichodectes galictidis and Galictis cuja, highlighting a significant

Host Proteins in Echinococcus multilocularis Metacestodes.

Metacestodes of Echinococcus multilocularis are the causative agents of alveolar echinococcosis, a neglected, life-threatening, zoonotic disease. To study these metacestodes in vitro, a model system using a culture medium conditioned by rat hepatoma cells is available. A key question is how the parasite interacts with the host and, in particular, which host-derived compounds are taken up. In this study, we focus on the uptake of host-derived proteins. Studies with artificially labeled proteins suggest that this uptake may occur independently of protein size or charge. Closer investigation usin

mGem: Extracellular vesicles in Leishmania-secret messengers driving infection and disease.

Leishmaniasis is a disease caused by Leishmania parasites, transmitted by insects, that occurs worldwide. The parasite and parasite-infected cells release extracellular vesicles (EVs), which are involved in numerous biological processes. EVs secreted by Leishmania modulate the host cell and, in turn, the immune response. In this review, we focused on two particular EV-related topics: (i) EVs as carriers of Leishmania virulence factors and implications in parasite biology, and (ii) the effects of Leishmania-derived EVs on the host's immune response.

Sea lice parasites and salmonids - a hormesis perspective.

Low-dose exposure to pesticides can have stimulatory effects on the biological traits of arthropods, a hallmark of hormesis, which induces adaptive responses. In this forum article we postulate that such responses could be induced in sea lice ectoparasitic copepods, offering a perspective for scientific advance and improved practical solutions.

Quantitative ethology of schistosome miracidia characterizes a conserved snail peptide that inhibits host recognition.

Over 700 million people are at risk of contracting schistosomiasis due to regular exposure to freshwater sources where infected snails, the obligate intermediate hosts of schistosomes, are endemic. Although mass drug administration of praziquantel effectively controls the disease in most regions, achieving elimination will require reducing populations of infected snails that shed the human-infective larval stage. Considerable research effort has focused on parasite development and immunological responses after snail penetration, but comparatively little is known about the molecular and behavio

Genetic diversity and population structure of parasite infrapopulations within and across hosts for two trophically transmitted trematode parasites.

Complex parasite life cycles frequently require trophic transfer of parasites from an intermediate host prey to a definitive host predator. This results in aggregated distributions of parasites in predator host populations, which are subsequently expected to host more genetically diverse parasite infrapopulations than lower trophic level hosts. Host dispersal and seasonal population dynamics, particularly in the case of first-intermediate hosts, are also expected to drive population genetic patterns within and across populations. To examine how parasite life history and host ecology influence

Submicrometre spatiotemporal characterization of the Toxoplasma adhesion strategy for gliding motility.

Toxoplasma gondii is a protozoan apicomplexan parasite that uses an adhesion-dependent mode of motility termed gliding to access host cells and disseminate into tissues. Previous studies on Apicomplexa motile morphotypes, including the T. gondii tachyzoite, have identified a cortical actin-myosin motor system that drives the rearward translocation of transmembrane adhesins, thus powering forward movement. However, this model is currently questioned. Here, combining micropatterning and tunable surface chemistry (to edit parasite surface ligands) with flow force and live or super-resolution imag
