Next-Generation In Vitro Pulmonary Platforms for Respiratory Disease Modelling and Therapeutic Development: Current Advances and Future Prospects.

Pulmonary diseases such as Chronic obstructive pulmonary disease (COPD), asthma, pulmonary fibrosis, and acute respiratory infections remain a major global health challenge due to their complex pathophysiology and limited therapeutic options. Conventional 2D cultures and animal models have provided foundational insights; however, they often fail to accurately replicate the human lung's intricate architecture, immune interactions, and patient-specific variability. Recent advances in vitro technologies have transformed pulmonary research, enabling the generation of physiologically relevant and t

Screening of apical membrane antigen-1 (AMA1), dense granule protein-7 (GRA7) and rhoptry protein-16 (ROP16) antigens for a potential vaccine candidate against Toxoplasma gondii for chickens.

Toxoplasmosis is a zoonotic disease caused by the protozoan parasite, Toxoplasma gondii known to infect almost all animals, including birds and humans globally. This disease has impacted the livestock industry and public health, where infection of domestic animals increases the zoonotic risk of transmission of infection to humans, threatening public health. Hence the need to discover novel and safe vaccines to fight against toxoplasmosis. In the current study, a novel multiepitope vaccine was designed using immunoinformatics techniques targeting T. gondii AMA1, GRA7 and ROP16 antigens, consist

Testing the direct, indirect, and moderated effects of childhood animal cruelty on future aggressive and non-aggressive offending.

The relationship between childhood cruelty toward animals and subsequent aggressive offending was explored in 1,336 (1,154 male, 182 female) participants from the 11-wave Pathways to Desistance study (Mulvey, 2013). Aggressive and income offending at Waves 1 through 10 were regressed onto a dichotomous measure of prior involvement in animal cruelty and four control variables (age, race, sex, early onset behavior problems) assessed at Wave 0 (baseline). Results indicated that childhood animal cruelty was equally predictive of aggressive and non-aggressive (income) offending, a finding inconsist

Q Fever: current state of knowledge and perspectives of research of a neglected zoonosis.

Q fever is an ubiquitous zoonosis caused by an resistant intracellular bacterium, Coxiella burnetii. In certain areas, Q fever can be a severe public health problem, and awareness of the disease must be promoted worldwide. Nevertheless, knowledge of Coxiella burnetii remains limited to this day. Its resistant (intracellular and environmental) and infectious properties have been poorly investigated. Further understanding of the interactions between the infected host and the bacteria is necessary. Domestic ruminants are considered as the main reservoir of bacteria. Infected animals shed highly i

Leptospirosis in Southeast Asia: Investigating Seroprevalence, Transmission Patterns, and Diagnostic Challenges.

Leptospirosis remains a significant public health and economic burden in Southeast Asia, particularly in low- and middle-income countries where environmental, occupational, and socioeconomic factors contribute to its endemicity. Transmission is driven by close interactions between humans and infected animal reservoirs, alongside climatic conditions such as heavy rainfall and flooding. The region's high but variable seroprevalence reflects inconsistencies in diagnostic methodologies and surveillance systems, complicating disease burden estimation. Major gaps persist in diagnostic capabilities,

Presumptive risk factors for monkeypox in rural communities in the Democratic Republic of the Congo.

Monkeypox virus (MPXV), a close relative of Variola virus, is a zoonotic virus with an unknown reservoir. Interaction with infected wildlife, bites from peri-domestic animals, and bushmeat hunting are hypothesized routes of infection from wildlife to humans. Using a Risk Questionnaire, performed in monkeypox-affected areas of rural Democratic Republic of the Congo, we describe the lifestyles and demographics associated with presumptive risk factors for MPXV infection. We generated two indices to assess risk: Household Materials Index (HMI), a proxy for socioeconomic status of households and Ri

Geographical and Ecological Drivers of Zoonotic Viral Spillover: A Review of Emerging and Re-emerging Outbreaks.

Over the past two decades, outbreaks of zoonotic viruses have become increasingly frequent and severe, posing substantial threats to public health systems and the global economy. The viruses responsible for these outbreaks, such as SARS-CoV, MERS-CoV, Zika, Ebola, Nipah, avian influenza, and, most recently, SARS-CoV-2, typically originate in wildlife, highlighting the complex relationship between ecological systems and human activities. Human-wildlife interactions have markedly increased due to disruptions in environmental and geographic boundaries, primarily driven by urbanization, deforestat

Wild Felid Attacks on Humans in South and Central America: A 2014-2025 Case Series in French Guiana and Literature Review.

Wild felids are apex predators that play a crucial role in tropical ecosystems. In South America, interactions between humans and large felids-particularly jaguars (Panthera onca) and pumas (Puma concolor)-are increasingly reported, raising public concern. However, attacks on humans remain poorly documented, and their frequency is likely overestimated. This study assessed the scale of wild felid attacks in French Guiana and South America through a case series and literature review. We conducted a retrospective observational study of all wild felid attacks managed in healthcare facilities in Fr

Vaccination elicits HIV broadly neutralizing antibodies in primates.

The high antigenic diversity of HIV has been a major obstacle to development of a broadly protective vaccine. Nevertheless, protective HIV broadly neutralizing antibodies (bnAbs) exist and have been proposed as templates for vaccine development1-6. Germline targeting is a conceptually radical vaccine design approach to elicit bnAbs, aiming to prime rare bnAb-precursor B cells possessing predetermined human genetic and structural features shared with template bnAbs, and then guide B cell affinity maturation to potent bnAb evolution with heterologous boosters7-11. Although the approach has shown

Reproductive toxicity of micro- and nanoplastics: Insights from experimental and human studies.

The exponential rise in plastic production has driven widespread contamination by micro- and nanoplastics (MNPs) in the environment. These plastic particles and their chemical additives have been detected in water sources, human bodily fluids, and reproductive tissues. With global fertility rates declining, their role as potential contributors is under investigation. This scoping review compares findings from in vitro experiments, in vivo studies across animal models, and epidemiological data to assess potential reproductive hazards associated with MNP exposure. Forty original studies publishe

AFA and F17 adhesins produced by pathogenic Escherichia coli strains in domestic animals.

AFA and F17 are afimbrial and fimbrial adhesins, respectively, produced by pathogenic Escherichia coli strains in domestic animals. F17-related fimbriae are mainly detected on bovine and ovine E. coli associated with diarrhoea or septicaemia. The F17-G adhesin subunits recognize N-acetyl-D-glucosamine (GlcNAc) receptors present on bovine intestinal cells. Some F17 subtypes also bind to GlcNAc receptors present on human uroepithelial and intestinal Caco-2 cells or to the laminin contained in the basement of mammalian membranes. F17 is often associated with other virulence factors (aerobactin, s

Platelet-Activating Factor Promotes Neutrophil Activation and Platelet-Neutrophil Complex Formation.

Controlling excessive inflammation remains an unmet clinical need, for example, during sepsis or after severe injuries. Platelet-activating factor (PAF) is central in thromboinflammatory processes. However, its role in the interaction of platelets and neutrophils requires further insights. Therefore, we elucidated PAF-related neutrophil activation, including platelet-neutrophil complex (PNC) formation and investigated potential strategies to modulate PAF-related inflammation. For the translation of the PAF-mediated inflammation, we applied an animal-free human ex vivo whole blood model. The ne

Danshensu sodium salt alleviates muscle atrophy via CaMKII-PGC1α-FoxO3a signaling pathway in D-galactose-induced models.

Sarcopenia is an age-related muscle atrophy syndrome characterized by the loss of muscle strength and mass. Although many agents have been used to treat sarcopenia, there are no successful treatments to date. In this study, we identified Danshensu sodium salt (DSS) as a substantial suppressive agent of muscle atrophy. We used a D-galactose (DG)-induced aging-acceleration model, both in vivo and in vitro, to confirm the effect of DSS on sarcopenia. DSS inhibits the expression of muscle atrophy-related factors (MuRF1, MAFbx, myostatin, and FoxO3a) in DG-induced mouse C2C12 and human skeletal mus

The Search for Cryptic L-Rhamnosyltransferases on the Sporothrix schenckii Genome.

The fungal cell wall is an attractive structure to look for new antifungal drug targets and for understanding the host-fungus interaction. Sporothrix schenckii is one of the main causative agents of both human and animal sporotrichosis and currently is the species most studied of the Sporothrix genus. The cell wall of this organism has been previously analyzed, and rhamnoconjugates are signature molecules found on the surface of both mycelia and yeast-like cells. Similar to other reactions where sugars are covalently linked to other sugars, lipids, or proteins, the rhamnosylation process in th

Impact drugs targeting cardiometabolic risk on the gut microbiota.

PURPOSE OF REVIEW: Alterations in the gut microbiome composition or function are associated with risk factors for cardiometabolic diseases, including hypertension, hyperlipidemia and hyperglycemia. Based on recent evidence that also oral medications used to treat these conditions could alter the gut microbiome composition and function and, vice versa, that the gut microbiome could affect the efficacy of these treatments, we reviewed the literature on these observed interactions. RECENT FINDINGS: While the interaction of metformin with the gut microbiome has been studied most, other drugs that

The role of agrin as a downstream mediator of Slit3 in osteogenic-angiogenic interaction under osteoporosis.

Osteoporosis, characterized by an imbalance between bone formation and bone resorption, is currently treated mainly by inhibiting resorption or promoting formation. However, these therapeutic strategies are often limited in efficacy because they fail to simultaneously target the coupled osteogenic and angiogenic processes in bone metabolism. In recent years, Slit3 has emerged as a key regulator of osteo-angiogenic interaction and is now considered a promising therapeutic target for metabolic bone disease. In the present study, we performed in vitro and in vivo experiments, including employing

Investigation of varenicline and tropisetron in latent inhibition and novel object recognition in mice.

There is currently no treatment for cognitive-impairment associated with schizophrenia (CIAS). Animal and human studies suggest that nicotinic cholinergic agonists and 5-HT3 antagonists have potential therapeutic benefits. We investigated whether co-administration of varenicline, an alpha4beta2 nAChR partial agonist and alpha7 nicotinic receptor agonist, and tropisetron, a 5-HT3 antagonist and alpha7 nicotinic receptor agonist is pro-cognitive in male c57/Bl6 mice. We first assessed Latent inhibition (LI) an associative learning process of learning to ignore irrelevant stimuli. We report no ef

Engineering a NanoBiT biosensor for detecting angiotensin-converting enzyme-2 (hACE2) interaction with SARS-CoV-2 spike protein and screening the inhibitors to block hACE2 and spike interaction.

Infection with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) is facilitated by its trimeric surface spike protein, which binds to the human angiotensin-converting enzyme 2 (hACE2) receptor. This critical interaction facilitates viral entry and is a primary target for therapeutic intervention against COVID-19. However, it is difficult to fully optimize viral infection using existing protein-protein interaction methods. Herein, we introduce a nano-luciferase binary technology (NanoBiT)-based pseudoviral sensor designed to stimulate the dynamics of viral infection in both living ce
