Glutamate Excitotoxicity and Oxidative Stress in Epilepsy: Modulatory Role of Melatonin.

Epilepsy is thought to be associated with oxidative stress, glutamate excitotoxicity, and mitochondrial dysfunction. The enhanced synthesis … s have a significant role in suppressing seizures. Melatonin scavenges oxygen free radicals such as hydroxyl radical, peroxy radical, peroxynitrite anion, and superoxide radical and stimulates synthesis of superoxide dismutase and glutathione peroxidase, which are potent antioxidant enzymes. Melatonin administration has been shown to be effective in both experimen …

Cr (VI) increases tyrosine phosphorylation through reactive oxygen species-mediated reactions.

While Cr (VI)-containing compounds are well established carcinogens, the mechanisms of their action remain to be investigated. In this study we show that Cr (VI) causes increased tyrosine phosphorylation in human lung epithelial A549 cells in a time-dependent manner. N-acetyl-cysteine (NAC), a general antioxidant, inhibited Cr (VI)-induced tyrosine phosphorylation. Catalase, a scavenger of H2O2, sodium formate and aspirin, scavengers of hydroxyl radical (*OH), also inhibited the increased tyrosine phosphorylation induced by Cr (VI). SOD, an inhibitor of superoxide radical (O2*-), caused less i…

Peculiarities of the free radical processes in rat liver mitochondria under toxic hepatitis on the background of alimentary protein deficiency.

The rate of superoxide anion radical, hydroxyl radical and hydrogen peroxide generation, the level of oxidative modification of mitochondrial proteins in the liver of rats with toxic hepatitis was investigated on the background of alimentary protein deficiency. We did not find significant increases of the intensity of free radical processes in liver mitochondria of rats maintained on the protein-deficient ration. The most significant intensification of free radical processes in liver mitochondria is observed under the conditions of toxic hepatitis, induced on the background of alimentary prote…

Tunable delocalization of unpaired electrons of nitroxide radicals for sickle-cell disease drug improvements.

Hydroxyurea is a drug recently approved to treat sickle cell diseases. Hydroxyurea benefits the patients by increasing the level of fetal hemoglobin via a nitroxide radical pathway. Here, we report an unpaired-electron-delocalization approach to tune the stability of nitroxide radicals. In this approach, the substitution by an unsaturated alkyl group containing conjugated C=C double bonds for the hydrogen on the nitrogen atom attached to the hydroxyl of hydroxyurea can significantly increase its ability to generate nitroxide radical. Furthermore, the increase can be remarkably enhanced by incr

Quantitative 31P NMR detection of oxygen-centered and carbon-centered radical species.

Quantitative 31P NMR spin trapping techniques can be used as effective tools for the detection and quantification of many free radical species. Free radicals react with a nitroxide phosphorus compound, 5-diisopropoxy-phosphoryl-5-methyl-1-pyrroline-N-oxide (DIPPMPO), to form stable radical adducts, which are suitably detected and accurately quantified using (31)P NMR in the presence of phosphorus containing internal standards. Initially, the 31P NMR signals for the radical adducts of oxygen-centered (*OH, O2*-) and carbon-centered (*CH3, *CH2OH, CH2*CH2OH) radicals were assigned. Subsequently,

Hydroxyl radical was detected by use of DMPO only in the presence of oxygen. The DNA breaking activity of AAPH was found to be due primarily to the aliphatic carbon-centered radical. While the reactivity of carbon-centered radicals have received little attention, th …

Relationship between bilirubin free radical and formation of pigment gallstone.

In this paper, we summarize the main progresses made in our group in the field of the mechanism of pigment gallstone formation. It was found that after treatment with free radicals, bilirubin (BR) was changed into free radical itself, and a semiquinone free radical and a superoxide free radical bound with metal were recognized, which was detected by ESR (electron spin resonance). By the means of NMR (nuclear magnetic resonance) and IR (Infra-red spectra), it was postulated that bilirubin polymerized through the reaction between the vinyl group and the hydroxyl group under the attack of free ra

Cross-Radical Knowledge Sharing via Graph Neural Networks for Unified Prediction of Aqueous Organic Contaminant Second-Order Radical Reaction Rate Constants.

Selecting among hydroxyl (HO•), sulfate (SO4•-), and carbonate (CO3•-) radical advanced oxidation processes requires reliable intrinsic second-order rate constants, yet the corresponding curated data sets contain 1250, 493, and 248 records, respectively. Reframing this imbalance as a cross-radical few-shot learning problem, we introduce maco, a radical-conditioned graph neural network that uses shared molecular representation learning, oxidant/pH cross-attention, residual adapters, and a two-stage curriculum to transfer structural knowledge from the data-rich HO• task to the lower-data SO4•- a

Protection against chemically-induced oxidative gastrointestinal tissue injury in rats by bismuth salts.

Oxygen free radicals (OFR) are implicated in the pathogenesis of stress, chemically induced gastric lesions, and gastrointestinal injury. The concentration-dependent scavenging abilities of bismuth subsalicylate (SBS), colloidal bismuth subcitrate (CBS), and selected OFR scavengers, including superoxide dismutase (SOD), catalase, mannitol, and allopurinol were examined against biochemically or chemically generated superoxide anion, hydroxyl radical, and hypochlorite radical plus hypochlorous acid based on a chemiluminescence assay. Furthermore, both gastric (GM) and intestinal mucosa (IM) were…

Effects of UVC/H2O2 on dissolved organic matter derived from a cyanobacterial bloom.

Cyanobacterial blooms release complex metabolite mixtures, yet their degradation during water treatment remains poorly understood. This study investigates the degradation of cyanometabolite-rich natural bloom extracts using UVC/H2O2 to generate hydroxyl radicals (HO•). By applying LC-HR-MS/MS combined with computational metabolomics, we identified nine microcystins (MCs), four anabaenopeptins (APs), and sixteen microginins (MGs), proposing structures for five new congeners and two novel transformation products (TPs). UVC/H2O2 accelerated cyanopeptide degradation compared to individual control …

Cytotoxic and radical activities of metal-organic framework modified with iron oxide: Biological and physico-chemical analyses.

Metal-organic framework (MOF) modified with iron oxide, Fe3O4-MOF, is a perspective drug delivery agent, enabling magnetic control and production of active hydroxyl radicals, •OH, via the Fenton reaction. This paper studies cytotoxic and radical activities of Fe-containing nanoparticles (NPs): Fe3O4-MOF and its components - bare Fe3O4 and MOF (MIL-88B). Luminous marine bacteria Photobacteriumphosphoreum were used as a model cellular system to monitor bioeffects of the NPs. Neither the NPs of Fe3O4-MOF nor MOF showed cytotoxic effects in a wide range of concentrations (<10 mg/L); while Fe3O4 wa…

Degradable Tumor-Responsive Iron-Doped Phosphate-Based Glass Nanozyme for H2O2 Self-Supplying Cancer Therapy.

Tumor microenvironment (TME)-responsive chemodynamic therapy (CDT) mediated by nanozymes has been extensively studied both experimentally and theoretically, but the low catalytic efficiency due to insufficient H2O2 in the TME and the poor biodegradability of the nanozymes are still main challenges for clinical translation of nanozymes. Herein, we designed a H2O2 self-supplying nanozyme bearing glucose oxidase (GOX) and polyethyleneimine based on a degradable iron-doped phosphate-based glass (FePBG) nanomimic (FePBG@GOX), which can convert endogenous glucose into toxic hydroxyl radicals. The GO…

Nano- and micro-scale zerovalent iron-activated peroxydisulfate for methyl phenyl sulfoxide probe transformation in aerobic water: Quantifying the relative roles of SO4·-, Fe(IV), and ·OH.

Multiple reactive intermediates have been proposed to be involved in peroxydisulfate (PDS) activation by zerovalent iron (ZVI), including sulfate radical (SO4·-) produced via iron-oxide shell mediated electron transfer, ferryl ion species (Fe(IV)) formed from Fe(II)-PDS interaction, and hydroxyl radical (·OH) generated by ZVI aerobic oxygenation. In this study, evolution of the relative role of these intermediates in microscale and nanoscale ZVI (mZVI vs. nZVI) activated PDS processes is comparatively investigated by using a methyl phenyl sulfoxide (PMSO) probe that selectively reacts with Fe(…

Degradation of bisphenol A by electro-enhanced heterogeneous activation of peroxydisulfate using Mn-Zn ferrite from spent alkaline Zn-Mn batteries.

Mn-Zn ferrite (Mn0.6Zn0.4Fe2O4) was prepared by a gel method using spent alkaline Zn-Mn batteries as raw materials and employed as catalyst to degrade bisphenol A (BPA) by electro-enhanced heterogeneous activation of peroxydisulfate (PDS). The effects of initial pH, current density, PDS concentration, and Mn-Zn ferrite dosage on BPA removal were investigated. The formation of reactive radicals was verified by electron paramagnetic resonance (EPR) spectroscopy. The results of radical quenching experiments indicate that surface-bound sulfate and hydroxyl radicals played an important role in BPA …
