Recently it was reported that there is an increase in monoamine oxidase B (MAO-B) activity in post-mortem brains of patients with Alzheimer's disease. It was postulated that this increase in MAO-B activity was due to gliosis associated with neuronal degeneration. The aim of the present investigation was to evaluate the effect on MAO of neuronal degeneration primarily affecting the cholinergic system. The specific cholinergic toxin AF64A (3 and 4.5 nmol) was injected bilaterally into the cerebral ventricles of rats. We then estimated MAO-A, MAO-B, dopamine (DA) uptake rates and choline acetyltr

Metaphit, a derivative of phencyclidine (PCP) differing by the meta substitution of an isothiocyanate group on the phenyl ring, was given into the lateral cerebral ventricle of mice alone and as a pretreatment to the subsequent i.p. injection of PCP and pentobarbital. Drug effects on motor performance as an index of neurologic impairment were quantified using the mouse platform test. The ED50 of metaphit to disrupt mouse platform behavior 5 min after i.c.v. administration was 0.48 mumol. An isobolographic analysis indicated that metaphit interacted additively with PCP and pentobarbital. Animal

Thyrotropin-releasing hormone (TRH) has previously been found to have a potent respiratory stimulant effect following intracerebroventricular administration. One part of the respiratory response, the shortening of inspiratory time, seems to be elicited from the raphe obscurus in the medulla. The prominent tachypnoea however is not elicited after local injections in the medullary region. In the present paper a micro-injection technique was employed to study respiratory actions of TRH in lightly anaesthetized rats kept in a whole body plethysmograph. Thyrotropin-releasing hormone in a dose of 10

The transport and metabolism of diphenhydramine was studied in vitro in the isolated rabbit choroid plexus and in vivo in New Zealand white rabbits and Sprague-Dawley rats. In vitro, [14C] diphenhydramine was accumulated by a saturable, energy-requiring system in choroid plexus. In vivo, 20 min after intraventricular injection into rabbits, [14C]diphenhydramine was cleared from cerebrospinal fluid much more rapidly than [3H]sucrose, a molecule transported in the central nervous system by simple diffusion. In vivo, employing the in situ rat brain perfusion technique, [14C]diphenhydramine was cl

The lateral septal area of anaesthetized rats was perfused using a push-pull cannula and the effect of the intracerebroventricular injection of bradykinin on the release of 3H-noradrenaline from the lateral septal area studied. A significant increase in 3H-noradrenaline release concomitant to a cardiovascular response was observed after bradykinin administration, suggesting the involvement of septal noradrenergic mechanisms in the response to the peptide. Morphine, subcutaneously administered, blocked the increase in the effluent radioactivity and the cardiovascular response to bradykinin, sug

Effects of intracerebroventricular injection of bombesin on gastric mucosal glycoproteins and acid secretion were studied to clarify its action mechanism for experimental ulcers in rats. Bombesin (10 micrograms) prevented stress ulcers and indomethacin ulcers. Aspirin (300 mg/kg p.o.) reduced the high molecular weight glycoproteins extracted from the gastric mucosa, and increased the degraded glycoproteins extracted from gastric juice in pylorus-ligated rats. Bombesin inhibited the aspirin-induced changes of the glycoproteins. Bombesin also inhibited basal gastric acid secretion. These results

Rats were injected in one lateral cerebral ventricle (i.c.v.) with apamin (100 ng per animal). The resulting desynchronisation pattern in the electrocorticogram (ECoG) and the symptoms of poisoning were monitored before and after transsection at different levels, and following morphine. Apamin acts primarily on the brain stem and spinal cord, i.e. structures possessing a sensory input, and then indirectly on the higher integrating systems. There is no general parallelism between receptor density and locus of action.

The organization of the corticocuneate pathway was investigated in monkeys by using the anterograde and retrograde axonal transport of either horseradish peroxidase (HRP) or wheat germ agglutinin conjugated to horseradish peroxidase (WGA-HRP). Injection of either tracer into the precentral cortex (centered on area 4) results in heavy anterograde labeling in the tegmental region, which lies immediately ventrolateral to the cuneate nucleus, particularly at levels caudal to the obex. On the other hand, injections of the same tracers involving areas 3b, 1, and 2 cause anterograde labeling mainly w

Thyrotropin-releasing hormone (TRH), injected into the central nervous system (CNS) in rats, has been shown to elicit systemic hyperglycemia. In the present study, central TRH administration significantly decreased the plasma glucose in mice. The hypoglycemic response could be blocked by pretreatment with the muscarinic cholinergic antagonist, atropine methyl bromide, or the diabetogenic beta-cytotoxin, alloxan, implicating the involvement of the parasympathetic system and insulin-secreting cells in the endocrine pancreas. The role of TRH in the CNS in the autonomic regulation of glucose homeo

Ouabain was infused into the third cerebral ventricle of fully conscious, pentobarbital-anaesthetized or urethane-anaesthetized rats. Convulsive seizures occurred in the freely moving rats. The blood pressure of the pentobarbital-anaesthetized rats rose inconsistently. However, in those animals, anaesthetized with urethane, a consistent cardiovascular response was observed with an initial bradycardia, followed by a progressive increase in both the blood pressure and the heart rate. The hypertension was associated with a rise in the CSF potassium level. Tachyphylaxis to these responses was obse

The effects of dermorphin, a new potent opioid peptide, on structural and ultrastructural changes in the crop sac and in pituitary lactotrophs were assessed in pigeons (Columba livia). A single administration of dermorphin into the third cerebral ventricle (1 microgram) and the same dose repeated for 3 consecutive days both produced maximal crop sac stimulation, as documented by the presence of milk-like material as well as by scanning and transmission electron microscopy of crop sac mucosa. In addition, marked stimulatory effects were also observed in pituitary lactotrophs, as shown by stimul

The effect of centrally administered indomethacin on hemorrhage-induced vasopressin release was studied in the morphine-sedated, urethan/chloralose-anesthetized dog. Ventriculocisternal perfusion of indomethacin 1) significantly reduced the amount of prostaglandin E2 in the effluent from the cisterna magna, 2) significantly enhanced the vasopressin response to volume depletion, and led to a greater fall in mean arterial blood pressure during severe hemorrhage. The results suggest that central prostaglandins may have an inhibitory effect on vasopressin secretion during volume depletion.

The influence of centrally administered meperidine, normeperidine and pentazocine on the excitability of brain was studied by measuring the threshold for flurothyl-induced convulsions in rats. All three opioids are reported to lower seizure thresholds when given subcutaneously to rats in this test. Dose-and time-dependent changes in the seizure threshold occurred after intracerebroventricular injection of pentazocine (10-160 micrograms), meperidine (25-150 micrograms) and normeperidine (50-150 micrograms). Rapid increases in the seizure threshold were associated with pentazocine and meperidine

The effect of an intracerebroventricular injection of an antimuscarinic drug, ganglionic blocking agent, alpha and beta adrenergic blocking drug, antiserotonin agent or antihistamine upon salivation produced by nicotine similarly injected in the cat was investigated. Atropine and hexamethonium abolished the salivation evoked by nicotine. On the other hand, salivation induced by nicotine was not significantly altered by yohimbine, practolol, methysergide and antazoline. It is concluded that the salivation produced by nicotine is mediated via central receptors which have mixed nicotinic and musc

Cats were anaesthetized with a mixture of alpha-chloralose and urethane and artificially ventilated. The spinal cord was transected at the C-1 level and the fourth cerebral ventricle cannulated. Phentolamine (500 microgram) administered into the fourth cerebral ventricle depressed the reflex vagal bradycardic responses elicited by intravenous pressor doses of noradrenaline. Enhancement of reflex bradycardia occurred following intracerebroventricular administration of L-DOPA (3.0 mg) which was reversed by subsequent administration of phentolamine into the fourth cerebral ventricle of spinal cat

Influences on renal water, electrolyte, and arginine vasopressin (AVP) excretions of 1 h infusions (20 microliters/min) of a neutral (L-alanine) and two basic (L-lysine and L-arginine) amino acids into the lateral cerebral ventricle were studied in hydrated goats, and were compared to effects of control infusions of hypertonic (0.25 M) NaCl. L-alanine (0.11 M) dissolved in hypotonic NaCl caused more pronounced inhibition of the water diuresis and greater increase in AVP excretion than did the control infusions, but, in comparison to the latter, the responses developed very slowly. The effects

Since there is degeneration of substantia nigra concomitant with that of locus coeruleus (LC) in patients with Parkinson's disease, the study was performed to determine the role of central norepinephrine (NE) on harmaline induced tremor. The duration of harmaline (10 mg/kg IP) induced tremor was significantly reduced by intraventricular administration of L-thero-3,4-dihydroxyphenylserine (200 micrograms/rat) and 1-NE (50 micrograms/rat) was increased NE levels in the cerebral cortex, striatum, diencephalon, cerebellum and brain stem. Electrical stimulation of bilateral LC suppressed harmaline-

5-Hydroxytryptamine (10 microgram), gamma-amino-butyric acid (20 microgram) and carbachol (1 microgram) were injected through the soft skull into the lateral cerebral ventricle of guinea pigs aged 2 to 12 days, at the slightly subneutral ambient temperature of 30 degrees C. 5-Hydroxytryptamine (5-HT) produced no immediate change in oxygen consumption, whereas colonic temperature fell rapidly. One hour after injecting 5-HT, when colonic temperature had decreased by about 0.5 degrees C, oxygen consumption increased and after a lag of 30--40 min was followed by an increase in colonic temperature.

BACKGROUND: High-risk and recurrent medulloblastoma (MB) is associated with significant mortality. The murine monoclonal antibody 3F8 targets the cell-surface disialoganglioside GD2 on MB. We tested the efficacy, toxicity, and dosimetry of compartmental radioimmunotherapy (cRIT) with intraventricular 131 I-labeled 3F8 in patients with MB on a phase II clinical trial.
METHODS: Patients with histopathologically confirmed high-risk or recurrent MB were eligible for cRIT. After determining adequate cerebrospinal fluid (CSF) flow, patients received 2 mCi (where Ci is Curie) 124 I-3F8 or 131 I-3F8

BACKGROUND: Reactive gliosis has been implicated in the pathogenesis of communicating hydrocephalus. Because the activation of Wnt/β-catenin signaling pathway is considered as a significant factor to contribute to brain development, neurodegenerative process, and reactive gliosis, we target this pathway for intervention by using sFRP-l and investigated the expression of β-catenin, cyclin D-1, and glial fibrillary acidic protein (GFAP) in the brain of experimental hydrocephalic rats in terms of protein and gene expression.
METHODS: Therefore, 30 adult SD rats were randomly divided into the nor
