[Investigation of the bile acid metabolism of children with subtotal atrophy of intestinal mucosa (author's transl)].

A secretin-pancreozymin test was conducted with 43 gastroenterologically healthy children and 12 patients with subtotal atrophy of intestinal mucosa. While the pancreas enzymes were normal, the patients with coeliac disease reacted to the injection of the peptide hormones by producing a larger volume of secretion than did the control group. Despite the increased secretion there was at the same time a significantly higher concentration of bile acids in the duodenal juice. Analysis of the bile acid distribution indicated no deviation from the norm. There was, however, a striking discrepancy betw

Control of phosphate balance by the kidney and intestine.

The prevention and correction of hyperphosphatemia are major goals of the treatment of chronic kidney disease (CKD)-bone mineral disorders, and thus, Pi balance requires special attention. Pi balance is maintained by intestinal absorption, renal excretion, and bone accretion. The kidney is mainly responsible for the plasma Pi concentration. In CKD, reduced glomerular filtration rate leads to various Pi metabolism abnormalities, and Pi absorption in the small intestine also has an important role in Pi metabolism. Disturbances in Pi metabolism are mediated by a series of complex changes in regul

[Histological and histochemical investigations of large and small intestine in guinea pigs infected by entamoeba histolytica].

In order to gain better knowledge about pathomechanisms of amebiasis, histological and histochemical studies were performed on large and small intestine of guinea pigs infected by PS-2 Entamoeba histolytica strain. Results of these studies showed that the presence of E. histolytica in large intestine caused also pathomorphological changes of small intestine: partial atrophy of villi, epithelium destruction, epithelial metaplasia, lacteral dilation, infiltration of mucous membrane by lymphocytes, plasmocytes and macrophages. Influence of this parasite on lowering of investigated enzyme reaction

Identification and pharmacological characterization of a natural folipastatin as a CFTR inhibitor and potential anti-secretory agent in a human colonoid model.

BACKGROUND: Overstimulation of cAMP-induced intestinal chloride secretion is a major cause of secretory diarrheas affecting both children and adults. This study aimed to investigate an anti-secretory effect of folipastatin, a natural depsidone derived from Aspergillus unguis, on cAMP-induced transepithelial chloride secretion using both human intestinal epithelial cell lines (T84 cells) and human colonoids. METHODS: Short-circuit and apical chloride current measurements were performed to investigate the inhibitory effect of folipastatin on cAMP-induced chloride secretion. Mechanisms of action

Vagal control of the release of somatostatin, vasoactive intestinal polypeptide, gastrin-releasing peptide, and HCl from porcine non-antral stomach.

We studied the secretion of somatostatin and HCl and the release of vasoactive intestinal polypeptide (VIP) and gastrin-releasing peptide (GRP) from isolated, vascularly perfused, porcine non-antral stomach. Electric vagus stimulation increased acid secretion and the release of VIP and GRP and inhibited somatostatin secretion as determined in the venous effluent. Atropine abolished the HCl response and reversed the somatostatin inhibition to a three-fold increase, whereas GRP and VIP responses were unchanged. Both intra-arterial carbachol (10(-6) M) and GRP (10(-8) M) increased acid secretion

Lactobacillus Protects Against S. Typhimurium-Induced Intestinal Inflammation by Determining the Fate of Epithelial Proliferation and Differentiation.

SCOPE: The influence of the intestinal microbiota, such as Lactobacillus, on the intestinal mucosa, particularly intestinal stem cells, remains incompletely understood. In this study, mice and intestinal organoids are used to explore the regulatory effect of Lactobacillus on the proliferation and differentiation of intestinal epithelial cells. METHODS AND RESULTS: This study demonstrates that S. typhimurium causes intestinal epithelial damage and affected growth of intestinal organoids. S. typhimurium also colonizes the intestine and then causes pathological changes to the intestinal epitheli

Intestinal microflora and metabolites affect the progression of acute pancreatitis (AP).

Specific intestinal metabolites are closely associated with the classification, severity, and necrosis of acute pancreatitis (AP) and provide novel insights for in-depth clinical investigations. In this study, the gut microbiota and metabolites of 49 AP patients at different treatment stages and severities were analysed via 16S rDNA sequencing and untargeted metabolomics to investigate the trends in gut microbiota composition and metabolome profiles observed in patients with severe AP. These findings revealed an imbalance in intestinal flora homeostasis among AP patients characterized by a dec

Intestinal plasticity and metabolism as regulators of organismal energy homeostasis.

The small intestine displays marked anatomical and functional plasticity that includes adaptive alterations in adult gut morphology, enteroendocrine cell profile and their hormone secretion, as well as nutrient utilization and storage. In this Perspective, we examine how shifts in dietary and environmental conditions bring about changes in gut size, and describe how the intestine adapts to changes in internal state, bowel resection and gastric bypass surgery. We highlight the critical importance of these intestinal remodelling processes in maintaining energy balance of the organism, and in pro

Diagnosis and treatment of cystic fibrosis. An update.

Cystic fibrosis is the most common fatal inherited disease of Caucasians. At present, cystic fibrosis accounts for most cases of chronic progressive pulmonary disease and for many other clinical features in the first three decades of life. Thus, it is a challenge to both pediatricians and internists, particularly chest physicians. The diagnosis is based on the triad of chronic obstructive pulmonary disease, pancreatic insufficiency, and increased levels of electrolytes in the sweat. The cardinal test for confirmation of the diagnosis is the "sweat test," which is an excellent discriminant for

Mucin histochemistry of the columnar epithelium of the oesophagus: a retrospective study.

Reflux oesophagitis may be accompanied by replacement of squamous epithelium by columnar epithelium (Barrett's oesophagus). The columnar epithelium may resemble either gastric or intestinal mucosa, though ultrastructural and histochemical studies indicate that the intestinal type does not necessarily resemble normal small intestine. Twenty-two malignant and 10 benign oesophageal specimens were examined histologically and by mucin histochemistry in an attempt to classify the various types of columnar epithelium. An incompletely differentiated variant of intestinal metaplasia secreting sulphomuc

[Research progress on mechanism of Atractylodis Macrocephalae Rhizoma and its combinations in treating constipation].

Atractylodis Macrocephalae Rhizoma(Baizhu), the dried rhizome of Atractylodes macrocephala in the Compositae family, was first recorded in the Shen Nong's Materia Medica(Shen Nong Ben Cao Jing). It is warm in nature, sweet and bitter in taste, and belongs to the spleen and stomach meridians. Baizhu exerts effects of strengthening the spleen and benefiting Qi, drying dampness and promoting diuresis. As a core drug in the traditional Chinese medicine(TCM) theory of "strengthening the spleen and regulating the bowels", its mechanism in treating constipation goes beyond the traditional s

Cellular Plasticity, Reprogramming, and Regeneration: Metaplasia in the Stomach and Beyond.

The mucosa of the body of the stomach (ie, the gastric corpus) uses 2 overlapping, depth-dependent mechanisms to respond to injury. Superficial injury heals via surface cells with histopathologic changes like foveolar hyperplasia. Deeper, usually chronic, injury/inflammation, most frequently induced by the carcinogenic bacteria Helicobacter pylori, elicits glandular histopathologic alterations, initially manifesting as pyloric (also known as pseudopyloric) metaplasia. In this pyloric metaplasia, corpus glands become antrum (pylorus)-like with loss of acid-secreting parietal cells (atrophic gas

Salmonella Infection Causes Hyperglycemia for Decreased GLP-1 Content by Enteroendocrine L Cells Pyroptosis in Pigs.

Inflammatory responses have been shown to induce hyperglycemia, yet the underlying mechanism is still largely unclear. GLP-1 is an important intestinal hormone for regulating glucose homeostasis; however, few studies have investigated the influence of digestive tract Salmonella infection on enteroendocrine L cell secretions. In this study, we established a model of Salmonella-infected piglets by oral gavage in order to analyze the effects of Salmonella infection on enteroendocrine L cell function. Furthermore, in vitro lipopolysaccharide (LPS) was administered to STC-1 cells to clarify its dir

Intestinal epithelial barrier: The target for pathogenic Escherichia coli.

Diarrheagenic Escherichia coli strains are included in 9 pathotypes (pathovars) that present different virulence factors responsible for the patomechanism of infections they cause. As all other intestinal pathogens, E. coli exerts a significant effect on intestinal epithelium. To initiate the infection, these microorganisms have evolved countless strategies to subvert the epithelial barrier and efficiently colonize the intestinal epithelium. The barrier function of the intestinal epithelium is achieved by the presence of a tight junction protein network surrounding individual cells around thei

Tissue-specific tolerance mechanisms and lymph node co-drainage converge to shape T cell immunity in the upper digestive system and regulate pancreatic cancer progression.

The liver, pancreas, and duodenum share lymph nodes (LNs), providing a unique system to examine how tissue origin of self-antigens shapes T cell fate. Comparing mice expressing ovalbumin (OVA) from distinct subcellular compartments, we found that cytosolic OVA from liver or pancreas, but not gut, was immunologically ignored. High-dose hepatic secreted OVA triggered antigen-specific T cell deletion, whereas secreted pancreatic and intestinal OVA induced regulatory T (Treg) cells, revealing immunological ignorance, clonal deletion and Treg cell generation as tissue-specific tolerance mechanisms.

IL-4/IL-13-producing ILC2s are required for timely control of intestinal helminth infection in mice.

Infection of mice with Nippostrongylus brasiliensis (Nb) serves as a model for human hookworm infection affecting about 600 million people world-wide. Expulsion of Nb from the intestine requires IL-13-mediated mucus secretion from goblet cells and activation of smooth muscles cells. Type 2 innate lymphoid cells (ILC2s) are a major cellular source of IL-13 but it remains unclear whether IL-13 secretion from ILC2s is required for Nb expulsion. Here, we compared the immune response to Nb infection in mixed bone marrow chimeras with wild-type or IL-4/IL-13-deficient ILC2s. ILC2-derived IL-4/IL-13

Secretion of c-di-AMP by Listeria monocytogenes Leads to a STING-Dependent Antibacterial Response during Enterocolitis.

Stimulator of interferon genes (STING) acts as a cytoplasmic signaling hub of innate immunity that is activated by host-derived or bacterially derived cyclic dinucleotides. Listeria monocytogenes is a foodborne, facultative intracellular pathogen that secretes c-di-AMP and activates STING, yet the in vivo role of the STING pathway during bacterial pathogenesis remains unclear. In this study, we found that STING-deficient mice had increased weight loss and roughly 10-fold-increased systemic bacterial burden during L. monocytogenes-induced enterocolitis. Infection with a L. monocytogenes mutant

Protein digestion and absorption in young chicks and turkeys.

The site of digestion and absorption of protein was examined in chicks fed soybean meal and casein and in turkeys fed soybean meal, using 91YCl3 as a non-absorbed reference substance. In soy-fed chicks, peptides with molecular weight (MW) between 8,000 to 15,000 accumulated in the proximal small intestine, and low MW peptides accumulated distally. A similar pattern was observed in soy-fed turkeys, although with events occurring proximally to those in the chick. In casein-fed chicks, a gradual reduction in the peptide MW was observed with increasing distance from the pylorus. The rates of forma

Maternal gut microbiota and placenta-derived tissues microbes are important for initial gut microbial colonization in infants.

BACKGROUND: Early infant gut microbiota colonization, influenced by various factors, significantly affects future growth and development. However, results related to how the initial microbial ecology is established in the infant gut remain inconsistent. RESULTS: In this study, we collected maternal and infant feces, vaginal secretions, placental tissues, breast milk, amniotic membrane tissues, umbilical cord blood, and breast skin for homology comparisons and for exploring the main sources of infant intestinal microbiota. Our results revealed that early infant gut microbiota originated mainly

Influence of retention time on degradation of pancreatic enzymes by human colonic bacteria grown in a 3-stage continuous culture system.

Hydrolytic enzymes were measured in gut contents from four sudden death victims. Pancreatic amylase and total protease activities decreased distally from the small bowel to the sigmoid/rectum region of the large intestine, showing that considerable breakdown or inactivation of the enzymes occurred during gut transit. To determine whether pancreatic enzymes were substrates for the gut microflora, mixed populations of bacteria were grown in a 3-stage continuous culture system on a medium that contained pancreatic extract as the sole nitrogen source. The multichamber system (MCS) was designed to
