Rapidly progressive scalp and lung metastases with fatal pneumothorax in glioblastoma, IDH-wildtype, with MET and CDK6 amplifications: a case report of clinical course and postmortem autopsy, including genetic analysis.

We report a rare case of extracranial metastases of a glioblastoma, IDH-wildtype, in a 77-year-old man who initially presented with a right frontal tumor, and gross total resection and adjuvant chemoradiotherapy were performed. The tumor was histologically comprised of two cellular components: astrocytic and poorly differentiated astrocytic tumor cells, with each strongly and infrequently positive for glial markers. Importantly, both components were positive for Nestin and CD44, indicating stemness and migratory characteristics. Three-and-a-half months after surgery, the patient presented with

[Mutations in epigenetic and metabolic regulators in peripheral T-cell lymphoma].

Recent advances in sequencing technologies led us to the discovery of mutations in epigenetic and metabolic regulators in peripheral T-cell lymphoma (PTCL). Frequencies of these mutations were found to be especially high in PTCL with features of follicular helper T cells (TFH). These mutations are also identifiable in myeloid malignancies as well as PTCL, suggesting that abnormalities in epigenetic and metabolic pathways are likely to be the fundamental mechanisms underlying various types of hematologic malignancies. The downstream pathways of these mutations have been extensively analyzed in

[Histochemical study of the chemoreceptor function of the carotid body in comparison with the superior cervical ganglion].

The distribution and relative activities of some dehydrogenases and diaphorases in the rat's carotid body and ganglion cervicale superior were studied histochemically and cytophotometrically. The treatment of rats by the medium with decreased oxygen and increased carbon dioxide contents, a reliable rise of NAD--diaphorase activities was found in the interlobe tissue and (less expressed and late appearing) in the principal cells. In contrast, such changes were not observed in the ganglion cervicale superioir, that have not a special chemoreceptory function. It is supposed that these changes in

Myco-degradation of anthraquinone dyes by immobilized white-rot fungal consortium and toxicity assessment.

Anthraquinone (AQ) dyes widely used in textile industries cause serious environmental and health risk due to their persistence and toxicity. In this context, myco-remediation using white-rot fungal consortium (WRFC) immobilized in chitosan beads demonstrated mixture of AQ dyes (MAQD) biodegradation with enhanced dye decolorization efficiency (97.35%) within 8 h. Further, FTIR analysis confirmed structural disruption of MAQD, including aromatic ring cleavage and functional group transformation. Moreover, LCMS analysis revealed formation of several low molecular weight intermediate organic acids

Molecularly Guided Therapy and the Tumor Microenvironment in Advanced Cholangiocarcinoma: Current Evidence and Emerging Strategies Targeting FGFR2, HER2, and IDH1 with Pemigatinib, Zanidatamab, and Ivosidenib.

Cholangiocarcinoma (CCA) remains an aggressive malignancy characterized by late-stage diagnosis, substantial molecular heterogeneity, and limited therapeutic options. Advances in molecular characterization have identified actionable alterations and expanded systemic treatment beyond conventional cytotoxic chemotherapy. This review examines three clinically relevant and mechanistically distinct targeted strategies in advanced CCA. Pemigatinib, a selective fibroblast growth factor receptor 1-3 (FGFR1-3) inhibitor, targets oncogenic signaling driven by FGFR2 fusions or rearrangements, which occur

Recurrent Dedifferentiated Clival Chondrosarcoma Presenting 32 Years after Initial Treatment: A Case Report.

Background Skull base chondrosarcomas are rare and locally aggressive tumors with heterogeneous biology accounting for 0.1-0.2% of all intracranial tumors. Dedifferentiated chondrosarcoma is an exceptionally uncommon and aggressive variant, seldom seen at the skull base. We report a recurrent clival chondrosarcoma with a small, dedifferentiated component, and an unusually long latency after initial treatment. Case Report A 66-year-old woman was referred with a recurrent progressive clival lesion, initially suspected to be a chordoma. She had undergone subtotal resection followed by single-frac

Augmenting Radiation Sensitivity by Targeting PAR-Dependent Replication Fork Vulnerability in IDH- Mutant Glioma.

UNLABELLED: Mutations in isocitrate dehydrogenase 1 ( IDH1 ) drive the early stages of gliomagenesis while simultaneously imposing replication stress that creates targetable vulnerabilities. Using both in vitro and in vivo models, we show that inhibition of poly(ADP-ribose) glycohydrolase (PARG) induces a poly(ADP-ribose) (PAR)-dependent augmentation of radiosensitivity in IDH1 -mutant glioma cells. Metabolic repletion of NAD + fails to rescue this effect, indicating that the vulnerability cannot be explained solely by NAD + depletion. Instead, PARG inhibition profoundly alters replication for

Genome-wide CRISPR screen identifies PKMYT1 inhibition as a synthetic lethal vulnerability in IDH1-mutant glioma.

Mutations in isocitrate dehydrogenase (IDH) are frequently observed in several aggressive malignancies, including gliomas, chondrosarcomas, and acute myeloid leukemia. Mutant IDH enzymes exhibit neomorphic activity that produces the oncometabolite D-2-hydroxyglutarate (D-2-HG), leading to widespread DNA and histone hypermethylation. This aberrant epigenetic state remodels chromatin accessibility, thereby compromising both DNA repair and replication. However, the mechanisms underlying IDH mutation-induced DNA replication stress and the therapeutic potential of this vulnerability remain elusive.

Glioblastoma survival in rural America: a 10-year experience from a quaternary care center.

BACKGROUND: Glioblastoma (GBM) is the most aggressive primary brain tumor with dismal prognosis. Patients residing in rural and Appalachian regions encounter multiple barriers to timely diagnosis and access to multidisciplinary neuro-oncology care, but whether these barriers translate into worse survival remains uncertain. West Virginia (WV), a predominantly rural Appalachian state, is served by a primary academic medical center (WV University Ruby Memorial Hospital) with a dedicated neuro-oncology program. We performed a retrospective analysis of the electronic health record to characterize o

Acquired genetic and cell-state changes in IDH-mutant glioma progression.

Gliomas with mutant isocitrate dehydrogenase (IDH) are malignant brain tumours that typically arise in early to mid-adulthood and nearly always recur following treatment1,2. However, the genetic and cellular-state changes that drive IDH-mutant glioma progression under treatment remain incompletely understood. Here we integrated single-nucleus transcriptomic profiles, chromatin accessibility profiles and bulk DNA and RNA sequencing from 75 temporally separated gliomas across 35 patients comprising both the oligodendroglioma and astrocytoma IDH-mutant glioma tumour types. We show that malignant

[Immunotherapy and Targeted Therapy for Advanced Biliary Tract Cancer].

Biliary tract cancer (BTC), encompassing intrahepatic and extrahepatic cholangiocarcinoma as well as gallbladder cancer, represents a heterogeneous group of malignancies characterized by an aggressive clinical course and poor prognosis. Systemic chemotherapy with gemcitabine plus cisplatin has remained the standard first-line treatment for more than a decade because most patients are diagnosed at an advanced or unresectable stage, but the associated survival benefit is limited. Recent therapeutic advances have been driven by the integration of immunotherapy and molecularly targeted approaches.

Alpha-ketoglutarate accelerates granulocyte-monocyte progenitor differentiation and atherosclerotic plaque inflammation via oxoglutarate receptor 1.

Accumulating evidence shows that excess cholesterol and glucose uptake stimulates the expansion of hematopoietic stem/progenitor cells and myeloid progenitors, resulting in increased production of inflammatory cells and atherosclerotic progression. However, the role of other metabolites in plaque progression remains unclear. Hereby, we observed elevated α-ketoglutarate levels in granulocyte-monocyte progenitors (GMPs) of Ldlr-/- mice on a high-fat diet (HFD), determined by targeted metabolomics. On top of HFD, α-ketoglutarate administration further increased GMP proportion, myeloid cell produc

Substrate Specificity and Kinetic Mechanism of the NAD+-Dependent Deacylase from Mycobacterium tuberculosis, Rv1151c (Mt-Sirt).

The pathogen Mycobacterium tuberculosis (Mtb) relies on extensive metabolic remodeling and post-translational regulation of its proteins to retain persistence in its host, yet these biochemical functions remain largely uncharacterized, including the actions of lysine deacylases. Here, we characterized Rv1151c, the sole annotated NAD+-dependent deacylase in Mtb, which we designate Mt-Sirt. Using synthetic substrates, peptide libraries, and mutational analysis, we showed that Mt-Sirt exhibits strong selectivity for peptide substrates containing Nε-succinyl-lysine and long-chain fatty-acyl-lysine

Identification of key molecules and pathways in CaOx crystal-induced renal injury based on metabolomics and proteomics.

PURPOSE: Calcium oxalate (CaOx) crystal deposition is the major pathological process of kidney stone formation and causes renal tubular injury. However, the molecular mechanisms and metabolic alterations involved remain unclear. This study aimed to identify key molecules and pathways associated with CaOx crystal-induced renal injury. METHODS: A mouse model of renal CaOx crystal deposition was established by glyoxylate (Gly) administration. Kidney tissues from CaOx crystal-bearing and control mice were analyzed using integrated proteomic and metabolomic approaches. Differentially expressed pro

Murine modeling of IDH-mutant 1p/19q-codeleted oligodendroglioma reveals genotype specific phenotypes.

BACKGROUND: Oligodendroglioma is a primary central nervous system tumor classified by the presence of isocitrate dehydrogenase (IDH) mutations and codeletion of 1p/19q. A lack of faithful preclinical model systems for this disease has limited progress in understanding the biology of this genetically unique subtype of adult diffuse-type glioma and in developing new effective therapies. To address this unmet need, we sought to establish a mouse model that faithfully recapitulates the genetic and phenotypic features of human IDH-mutant 1p/19q-codeleted oligodendroglioma. METHODS: We performed in

Dynamic alterations of the intestinal microbiome and metabolome during transmissible gastroenteritis virus infection in weaned pigs.

Transmissible gastroenteritis virus (TGEV) infection induces diarrhea in piglets by targeting the small intestine, especially the jejunum and ileum. However, dynamic changes in the gut microbiota and metabolome during TGEV infection remain unclear. This study investigated these alterations and their association with intestinal damage in weaned pigs during early TGEV infection. Thirty 4-week-old pigs were allocated randomly into TGEV-inoculated and mock groups. On days 3, 5, and 7 postinoculation, intestinal tissue and fecal samples were collected. Full-length 16 S rRNA sequencing and ultrahigh
