Incidence and severity of G6PI-induced arthritis are not increased in genetically distinct mouse strains upon aging.

BACKGROUND: The incidence of rheumatoid arthritis is correlated with age. In this study, we analyzed the association of the incidence and severity of glucose-6-phosphate isomerase (G6PI)-induced arthritis with age in two different mouse strains. METHODS: Young and very old mice from two different arthritis-susceptible wild-type mouse strains were analyzed after a single subcutaneous injection of G6PI s.c. The metabolism and the function of synoviocytes were analyzed in vitro, the production of bioactive lipid mediators by myeloid cells and synoviocytes was assessed in vitro and ex vivo by UPL

The Action of the Hexokinase Inhibitor 2-deoxy-d-glucose on Cryptosporidium parvum and the Discovery of Activities against the Parasite Hexokinase from Marketed Drugs.

Cryptosporidium parvum is one of the major species causing mild to severe cryptosporidiosis in humans and animals. We have previously observed that 2-deoxy-d-glucose (2DG) could inhibit both the enzyme activity of C. parvum hexokinase (CpHK) and the parasite growth in vitro. However, the action and fate of 2DG in C. parvum was not fully investigated. In the present study, we showed that, although 2DG could be phosphorylated by CpHK to form 2DG-6-phosphate (2DG6P), the anti-cryptosporidial activity of 2DG was mainly attributed to the action of 2DG on CpHK, rather than the action of 2DG or 2DG6P

A new family of enzymes catalyzing the first committed step of the methylerythritol 4-phosphate (MEP) pathway for isoprenoid biosynthesis in bacteria.

Isoprenoids are a large family of compounds with essential functions in all domains of life. Most eubacteria synthesize their isoprenoids using the methylerythritol 4-phosphate (MEP) pathway, whereas a minority uses the unrelated mevalonate pathway and only a few have both. Interestingly, Brucella abortus and some other bacteria that only use the MEP pathway lack deoxyxylulose 5-phosphate (DXP) reductoisomerase (DXR), the enzyme catalyzing the NADPH-dependent production of MEP from DXP in the first committed step of the pathway. Fosmidomycin, a specific competitive inhibitor of DXR, inhibited

Identification, expression, function and localization of a DUF985 domain-containing hypothetical gene from amphioxus Branchiostoma belcheri.

The progress in genome sequencing has led to an increasing submission of uncharacterized hypothetical genes with the domain of unknown function, DUF985, in GenBank, and none of these genes is related to a known protein. We therefore underwent an experimental study to identify the function of a DUF985 domain-containing hypothetical gene BbDUF985 (GenBank Accession No. AY273818) isolated from amphioxus Branchiostoma belcheri (B. belcheri). BbDUF985 was successfully expressed in both prokaryotic and eukaryotic systems, and its recombinant proteins expressed in both systems definitely exhibited an

Novel EBP gene mutations in Conradi-Hünermann-Happle syndrome.

BACKGROUND: Conradi-Hünermann-Happle syndrome [X-linked dominant chondrodysplasia punctata type 2 (CDPX2); MIM no. 302960] is an X-linked dominant disorder of cholesterol metabolism that causes a wide spectrum of skeletal abnormalities and linear ichthyosiform skin lesions. Mosaicism is probably responsible for the variability of the phenotype. OBJECTIVES: To describe new mutations in patients with variable manifestations of the disease. METHODS: We studied three patients with CDPX2. We performed mutation analysis of the EBP (formerly known as CDPX2) gene and gas chromatography-mass spectros

Komrower Lecture. Congenital disorders of glycosylation (CDG): it's all in it!

Congenital disorders of glycosylation (CDGs) are due to defects in the synthesis of the glycan moiety of glycoproteins or other glycoconjugates. This review is devoted mainly to the clinical aspects of protein glycosylation defects. There are two main types of protein glycosylation: N-glycosylation and O-glycosylation. N-glycosylation generally consists of an assembly pathway (in cytosol and endoplasmic reticulum) and a processing pathway (in endoplasmic reticulum and Golgi). O-glycosylation lacks a processing pathway but is otherwise more complex. Sixteen disease-causing defects are known in

Inhibition of isoprene biosynthesis pathway enzymes by phosphonates, bisphosphonates, and diphosphates.

We have investigated the docking of a variety of inhibitors and substrates to the isoprene biosynthesis pathway enzymes farnesyl diphosphate synthase (FPPS), isopentenyl diphosphate/dimethylallyl diphosphate isomerase (IPPI) and deoxyxylulose-5-phosphate reductoisomerase (DXR) using the Lamarckian genetic alogorithm program, AutoDock. The docked ligand structures are predicted with a approximately 0.8 A rms deviation from the structures determined crystallographically. The errors found are a function of the number of atoms in the ligand (R = 0.91, p < 0.0001) and, to a lesser extent, on the re

Discovery of Two 17β-Estradiol Metabolic Microbes and Their Metabolic Enzyme from Cecal Contents of Hy-Line Brown Laying Hens.

17β-Estradiol (E2), a reproductive hormone closely associated with the decline in egg production of laying hens, has attracted considerable attention. Although microbial feed additives have significantly alleviated this decline, the direct interactions between the host microbe and E2 remain poorly understood. In the present study, two E2-metabolic strains (Brevibacillus borstelensis NRRL NRS-818 and Acinetobacter radioresistans) were isolated through gradient pressure acclimatization with E2. Further RNA-Seq analysis of these two strains grown in medium with or without E2 identified 14 upregul

TAL1 Suppresses High-Grade Serous Ovarian Cancer Progression by Transcriptionally Inhibiting TPI1-Mediated Glycolysis.

High-grade serous ovarian cancer (HGSOC) is often accompanied by a poor prognosis, which urges the development of new therapeutic approaches. This study attempts to explore the role of T-cell acute lymphocytic leukemia Protein 1 (TAL1)-triosephosphate isomerase 1 (TPI1) mechanism in glycolysis-driven progression of HGSOC. Bioinformatics analysis identified TPI1 as upregulated in HGSOC tissues. This was confirmed by RT-qPCR and Western blotting (WB) in HGSOC cells. Functional assays, including EdU, colony formation, and Transwell, showed that TPI1 knockdown or knockout suppressed HGSOC cell pro

PPIL2 suppression induces cellular senescence and inhibits proliferation in hepatocellular carcinoma via c-myc/p21 axis.

Hepatocellular carcinoma (HCC) is the most prevalent primary malignancy of the liver characterized by high mortality rates. While peptidylprolyl isomerase-like two (PPIL2) has been implicated in various malignancies, its functional contribution to HCC progression and the underlying molecular mechanisms remain poorly defined. In this study, we observed significant upregulation of PPIL2 in HCC tissues, which correlated with unfavorable patient prognosis. Functional assays showed that PPIL2 knockdown markedly inhibited HCC cell proliferation by inducing cell cycle arrest and cellular senescence.

Herbicides as Isoform-Selective Inhibitors of Human 3β-HSD1 and Rat 3β-HSD4: Structure-Activity Relationships and Implications for Progesterone Biosynthesis and Recurrent Pregnancy Loss.

Environmental exposure to herbicides has raised significant concerns. However, whether herbicides affect placental 3β-hydroxysteroid dehydrogenase/Δ5-Δ4 isomerase (3β-HSD) remains unclear. This study investigated the inhibitory effects of 14 herbicides, especially dinitroanilines, on human (h3β-HSD1) and rat (r3β-HSD4), key enzymes in progesterone biosynthesis. In vitro assays showed isopropalin and oryzalin potently inhibited h3β-HSD1 (IC50 39.56 and 52.99 μM) via mixed-type inhibition. Inplacental JAr cells, oryzalin and profluraline suppressed progesterone synthesis at 100 μM, while isoprop

Impaired antioxidant defense associated with flavonoid biosynthesis reprogramming in the mangrove Acanthus ilicifolius under high salinity.

INTRODUCTION: This study investigates how varying salinity levels influence the physiological and metabolic responses of the mangrove plant Acanthus ilicifolius, aiming to clarify its salt adaptation mechanisms. METHODS: Field experiments were conducted at three sites with stable salinity conditions (approximately 0.73, 2.26, and 3.09 g/L), combined with physiological assays, metabolomics, and transcriptomics. RESULTS: Increasing salinity elevated intracellular reactive oxygen species, reduced antioxidant enzyme activity, and aggravated membrane lipid peroxidation. Metabolomics revealed that

GPI inactivation mediates pentose phosphate pathway flux switch-on inducing temozolomide resistance in glioma stem cell.

Temozolomide (TMZ) resistance in glioblastoma (GBM) remains a substantial clinical challenge. Targeting glioma stem cells (GSCs) represents a promising strategy to overcome chemoresistance and tumor recurrence. In this study, we found that GSCs maintain chemoresistance by increasing pentose phosphate pathway (PPP) flux compared with differentiated tumor cells. Following TMZ treatment, the activity of glucose-6-phosphate isomerase (GPI), a key glycolytic enzyme that catalyzes the conversion of glucose-6-phosphate to fructose-6-phosphate, was significantly suppressed in GSCs. Mechanistically, At

Myeloid UCHL1 deficiency ameliorates sepsis-induced liver injury by suppressing M1 macrophage polarization via targeting Gpi ubiquitination.

Ubiquitin C-terminal hydrolase 1 (UCHL1), a critical deubiquitinating enzyme (DUB), cleaves ubiquitin/polyubiquitin chains from substrate proteins andhas been shown to promoteoxidative stress and inflammatory response. However, UCHL1's precise role in sepsis-induced liver injury (SILI) pathogenesis remains unknown. Here, we show elevated UCHL1 in cecal ligation puncture (CLP)-induced livers and lipopolysaccharide (LPS)-treated bone marrow-derived macrophages (BMDMs). Deficiency or pharmacological inhibition of UCHL1 with LDN-57444 (LDN) conferred protection against SILI by inhibiting oxidative

Multiomics analysis of GSTP1 knockdown pancreatic cancer cells reveals key regulators of redox and metabolic homeostasis.

Glutathione S transferase pi-1 (GSTP1) is a detoxification enzyme essential for oxidative homeostasis. In cancer, GSTP1 has been implicated in tumorigenicity, cell cycle progression, and chemoresistance. While GSTP1 depletion has been associated with decreased cancer growth in various models, the mechanism remains poorly understood. This study investigates GSTP1 as a therapeutic target for pancreatic ductal adenocarcinoma (PDAC) using inducible knockdown models. We demonstrate that GSTP1 loss disrupts redox balance, impairs cell survival, and induces metabolic adaptations. Multiomics analysis
